Abstract
Description
a.血漿のサンプルを処理すること、
b.HPLCによってアッセイすること
を行うことを特徴とする、血漿からのオリゴ糖の分析方法である。
・溶媒A:NaH2PO4(2.5mM)、これはH3PO4の添加によってpH2.9にされる
・溶媒B:NaClO4(1N)、NaH2PO4(2.5mM)、これはH3PO4の添加によってpH3.0にされる。
溶出グラジエントは下記の通りにすることができる。
T=0分:%B=3;T=40分:%B=60;T=60分:%B=80
直径が2mmであるカラムについては、選ばれる流速は、例えば、0.22ml/分であり、カラム温度が40℃である。
手順
血漿の処理
体積が100μlから1000μlの間である血漿サンプルを採血し、適する場合には内部標準物質を後者に加える。
ろ過された血漿をアニオン交換HPLCによって分析する。使用されたカラムは、長さが25cmで、内径が2mmのIonPAc As11カラム(Dionex)である。使用された検出器は、ダイオードアレイを備えるUV分光光度計である。
溶媒A:NaH2PO4(2.5mM)(これはH3PO4の添加によってpH2.9にされる)
溶媒B:NaClO4(1N)、NaH2PO4(2.5mM)(これはH3PO4の添加によってpH3.0にされる)

Claims (23)
- 下記の2つの段階:
a.サンプルを処理すること、次いで
b.高速液体クロマトグラフィー(HPLC)によってアッセイすること
を行うことを特徴とする、オリゴ糖を血漿から分析するための方法。 - 血漿サンプルがろ過によって処理されることを特徴とする、請求項1に記載の方法。
- 30kDaよりも大きい分子量を有する分子が濃縮物に保持されることを特徴とする、請求項2に記載の方法。
- コントロールのオリゴ糖がろ過前に血漿に加えられることを特徴とする、請求項3に記載の方法。
- 濃縮物が塩溶液で少なくとも1回洗浄されることを特徴とする、請求項4に記載の方法。
- 塩溶液が塩化カリウム溶液であることを特徴とする、請求項5に記載の方法。
- 濃縮物の添加の後での塩溶液の濃度が1M以上であることを特徴とする、請求項5または6のいずれか一項に記載の方法。
- 濃縮物の添加の後での塩溶液の濃度が2M以上であることを特徴とする、請求項5または6のいずれか一項に記載の方法。
- 濃縮物が塩溶液で少なくとも1回洗浄され、次いで、水により洗浄されることを特徴とする、請求項2に記載の方法。
- ろ液および適する場合には洗浄液の全体が注入され、HPLCによって分析されることを特徴とする、請求項1から9のいずれか一項に記載の方法。
- クロマトグラフィーがアニオン交換クロマトグラフィーであることを特徴とする、請求項10に記載の方法。
- クロマトグラフィーが、第四級アンモニウムが共有結合による結合によってグラフト化されているアニオン交換カラムでのアニオン交換クロマトグラフィーであることを特徴とする、請求項10に記載の方法。
- 200nmまで透明である移動相が使用されることを特徴とする、請求項11に記載の方法。
- 移動相が、過塩素酸ナトリウムの、メタンスルホン酸塩のまたはリン酸塩の溶液であることを特徴とする、請求項13に記載の方法。
- 移動相が過塩素酸ナトリウムの溶液であることを特徴とする、請求項13に記載の方法。
- アセチル化糖を選択的に検出することを可能にする検出方法を含むことを特徴とする、請求項11に記載の方法。
- アセチル化糖を選択的に検出するための方法が、非アセチル化糖類の吸収性が相殺されるような方式で選ばれた2つの波長における吸収の違いをシグナルとして処理することによって行われることを特徴とする、請求項16に記載の方法。
- アセチル化糖を選択的に検出するための方法が、202nmから230nmおよび234nmにおける吸収を測定することによって行われることを特徴とする、請求項17に記載の方法。
- アセチル化糖を選択的に検出するための方法が、202nmから230nm、234nmおよび280nmにおける吸収を測定することによって行われることを特徴とする、請求項18に記載の方法。
- β−不飽和オリゴ糖をアッセイすることにおける請求項1に記載の方法の使用。
- エノキサパリン、オクタパリン(octaparin)、ベミパリン(bemiparin)またはチンザパリン(tinzaparin)をアッセイすることにおける請求項1に記載の方法の使用。
