Abstract
Description

(定義)

システムは、種々の細胞の応答をモニターするために、細胞ベースのバイオセンサーと連結して使用することができる。バイオセンサーは、マイクロ位置決め装置、ナノ位置決め装置又はマイクロマニピュレーター、又は光学ピンセットのような位置決め装置に連結されたピペット、毛細管又はカラムのようなセンサー(静止でも可動でもよい)を保持する機構を使用するセンサーチャンバー内に配置されている、あるいは流れ又は表面張力を制御し、細胞ベースのバイオセンサーをチャンバー内の溶液に曝露することによって、全細胞又はその一部(例えば、細胞膜パッチ)を含むことができる。バイオセンサーは、基板を動かす、すなわち、バイオセンサーに対するチャネルの位置を変化させることによって、又は細胞を動かす(例えば、マイクロ位置決め装置の走査又は流れ及び/又は表面張力を変化させる)ことによって、基板の種々のチャネルにわたって走査することができる。
他の態様では、イオン開口型チャネルは、電圧開口型チャネルである。電圧開口型チャネルは、膜貫通の電圧閾値に応答して開放する。電圧開口型ナトリウム、カリウム及びカルシウムチャネルは、全て、活動電位(又は、神経パルス)を軸索に下げて、他の神経細胞(又は、ニューロン)に伝導するために必須である。これらのイオンチャネルは、一般に、リジンを有する膜貫通配列及び/又は高アルギニンS4共有配列を含む。S4配列内の正のアミノ酸は、配列を含むイオンチャネルを異なる電圧条件のもとで開閉させる、細胞膜を横切る「感知」電圧であると考えられている。
一態様では、センサーは、基板上に固定化された、センサー素子、好ましくは、細胞標的(例えば、細胞内受容体、酵素、シグナリングタンパク質、細胞外タンパク質、膜タンパク質、核酸、脂質分子など)である分子を含む。基板は、センサーチャンバーそれ自身の底面であっても、あるいは、チャンバーの底面上に置かれた基板であっても、又はチャンバー内に静止して置かれた(例えば、マイクロ位置決め装置を介して)、可動性の又は静止した基板であってもよい。
好ましい態様では、本発明による基板は、サンプル及び/又は緩衝液のセンサーチャンバーへの微小流体移送に適応している。
サンプル−ウェルプレート(例えば、96ウェルプレートのような工業規格のマイクロプレート)内に含まれるサンプル(すなわち薬物など)を、好ましくは、当該技術分野で公知の自動ロボットアレー分注器(例えば、「Beckman′s Biomek 1000&2000 automated workstations, Beckman Coulter,Inc., Fullerton, CA」から入手可能)を使用して、操作し、転送する。
本発明の中核をなすものは、流体中に含まれる化合物又は試薬の複雑な操作のために、異なる溶液のセンサーチャンバー内のセンサーへの繰返し及び迅速な送達を可能にする方法で、ミクロ加工されたチャネルの二次元(2D)及び三次元(3D)ネットワークを使用することである。例えば、当該システムで使用される微小流体は、リガンドを、受容体を含む細胞ベースのバイオセンサーにプログラム可能に送達するシステムを可能にする。これは、サンプル(例えば化合物ライブラリー)のHTS選別用に使用されるシステムがバイオセンサーの応答における化合物の効果をモニターすることを可能にする。一態様では、細胞ベースのバイオセンサーの電気特性を、電圧クランプ又はパッチクランプ技術を使用してモニターする。
本発明による基板の複数のマイクロチャネルに存在する隣接する流体流は、低いレイノルズ数を有し、拡散による混合を最小限受ける。例えば、拡散係数が約5×10−6cm2/sの小分子は、10μm拡散するのに約0.1秒かかるが、拡散時間は距離の2乗で変化する(x2=2Dt(式中、Dは拡散係数である))ため、100μmの拡散に10秒かかる。同様に、D〜10−6cm2/sの代表的なタンパク質に関しては、10μmの拡散に0.5秒かかり、100μの拡散に50秒かかるだろう。
複数のマイクロチャネルを使用する場合、マイクロチャネルの出口とオープンボリューム貯蔵部との間の界面での流体の多数の流れの流動プロフィールの理解は必須である。図16A及び16Bは、単一チャネル(図16A)及び多数のチャネル(図16B)からの、500μMの蛍光染料(フルオロレスセイン)を含む流体の流動プロフィール顕微鏡写真を示す。蛍光トレーサーの励起は、落射照明構成で、488−nmラインのアルゴンイオンレーザーを使用して行い、トレーサーの蛍光を、CCDカメラを使用して集め、画像化した。図16Aに示すように、隣接するマイクロチャネル及び流体流がない場合、流体は、単一チャネルから流れ出し、半円形様式で、チャネル出口で分散する。図16Bは、複数のチャネル出口から流れ出す、相互連結した緩衝液及びフルオレセイン流体流の流動プロフィールを示す。マイクロチャネルの寸法は、100μm幅、50μm厚さであり、チャネル間の間隔は25μmであった。図16A及び16Bにおけるマイクロチャネル中の流速は4mm/秒であった。
受容体モジュレーター(アゴニスト又はアンタゴニスト)及び緩衝液の相互嵌合の流れを横切ってパッチクランプの細胞を迅速に走査する能力は、要求される流れ条件下でのパッチ細胞の機械的安定性と、走査速度に依存する。ここで、流体条件の範囲でのパッチクランプの細胞の「ギガシール(giga seal)」及びイオンチャネル活性の安定性を記載する。

図5D〜Fに示すように、2つの平衡するチャネルによって作られるU字型流体流を、細胞とパッチクランプのマイクロピペットとの間の最適なシールを作るために使用することができる。
走査は、(センサーを静止させて基板を動かすことによって、基板を静止させてセンサーを動かすことによって、又は基板及びセンサーの両方を異なる速度及び/又は異なる方向に動かすことによって)機械的に、あるいは異なるマイクロチャネルの圧力低下によって、調整することができる。図8A〜Iに、これらの方法がそれぞれどのように行われるかを概略的に示す。センサーの機械的な動き及び走査は、センサーを配置するマイクロ位置決め装置を移動させることによって、簡単に達成することができる。例えば、細胞ベースのバイオセンサーを、吸引によって、物理的にマイクロピペットに接続することができ、これは、次にマイクロマニピュレーターに接続する。殆どのマイクロマニピュレーターを、手動で、あるいは自動電気的に制御することができ、そのように事前プログラムされた動きは、簡単に達成することができる。プログラムされうる数多くのパラメータとして、等速走査の線速度;可変型速度走査の加速率;二次元及び三次元の両方における走査の軌道;繰返し走査の回数が挙げられるが、これらに限定されるものではない。センサーチャンバー内の1つ又はそれ以上のセンサーからのリアルタイムシグナルフィードバックに基づく走査に関して、プログラム可能なパラメータには、シグナル検出と走査設定の変更との間の時間遅延が含まれるが、これに限定されるものではない。走査用アウトプットに対する正しいフィードバックパラメータを決定するために、種々のシグナル加工及びコンピュータ機能を操作することができる。
(1)リガンド曝露時間を、走査速度及びリガンド流れの幅よりもむしろ、内部灌流時間(例えば、緩衝液のパルス間の時間)によって決定する。
(2)緩衝液灌流及び再感作時間も、緩衝液流れにおける滞留時間よりもむしろ、灌流パルスの持続時間によって決定する。
(3)リガンド流れをより高い充填密度の数で提供することができるので、その結果、1実験で多くの数のリガンドを走査することができる。
本発明によるシステム他の特徴は、流体をチャネルを通ってセンサーチャンバーへ迅速に送達することができ、化合物をセンサーの微小環境に導入して、当該微小環境から迅速に排出できることである。
この設計は、マイクロチャネル中に含まれる溶液とセンサーチャンバー(及び/又は細胞処置チャンバー)とを、迅速かつ効率的に置き換え及び交換することができるという事実に基づく。迅速な溶液交換は、種々の異なるマイクロチャネルネットワーク幾何学構造を用いることにより、達成することができる。一態様では、複数のマイクロチャネルは、合流し、又はセンサーチャンバーへ送り込まれ、一方他の態様では、複数のマイクロチャネルは、それ自身センサーチャンバーへ合流する、単一チャネルへ合流する。複数のマイクロチャネルは、それぞれ、サンプル用の相互嵌合チャネルと送達緩衝液とを含みうる。好ましい態様では、当該設計は、パッチクランプシステムと一体化する。3つの例示的な構造を以下に記載する。
この構造では、数多くの(例えば、96〜1024個)のマイクロチャネルを、約10μm〜約10mmの範囲の寸法で、センサーを収容するチャンバーに合流する、放射状スポークのように配置する。使用されるマイクロチャネルの数は、工業規格のマイクロタイタープレート、例えば96〜1024個のウェル、中のサンプルウェルの数に適合するように選択される。ウェルプレートからのインプットの数に適合するマイクロチャネルの数に加えて、少なくとも2個の付加的なマイクロチャネル(1つは灌流/再感作用の緩衝液の送達のため、もう1つは廃液除去のため)があるのが好ましい。マイクロピペットを使用するパッチクランプに関し、この構造は、また、細胞にアクセスするためのオープンボリューム領域も含む。しかし、チップベースのパッチクランプ測定(例えば、国際公開公報第99/31503号及び第01/25769号の記載)に関しては、いかなるオープンボリューム領域も無い方が好ましいであろう。マイクロチャネルからの流体流れによって細胞が除去されるのを防ぐため、細胞(複数を含む)は凹んだ部分あるいは細胞(複数を含む)の寸法に適合するウェル中にに置かれるのが好ましい。細胞の寸法より非常に小さな寸法を有する膜パッチに関し、ストークス抵抗により加えられた力は対象物(すなわちパッチ)の寸法に反比例するので、流体流によるバッチの除去は問題ではない。
また、三次元放射状スポークホイール配列も、センサーチャンバーに入る流体を効率的に置き換えるため、使用することができる。この構造では、1つ又はそれ以上のセンサー(例えば、細胞)を、放射状チャネルを含む基板と廃液貯蔵部を含む基板との間に挟まれたフィルター膜上に置く。この形式では、流体は、放射状チャネルが残る上層から(例えば、放射状チャネルへ送り込むインプットチャネルを通って)センサー(複数を含む)を過ぎ、次いでフィルターを通って、廃液チャネルに流れるように強制される。したがって、フィルターは、センサー(複数を含む)を速い流体流れで灌流させることができ、一方、センサー(例えば、細胞のような)を、流れに持っていかれないように又は排除されないように保持する。更に、流体を、センサーのそばを流すようにして、センサーを取り巻く流体の全てを置き換えるようにする。
この設計では、1つは化合物の送達ため、もう1つは廃液のための、2個だけのチャネルを1つ又はそれ以上のセンサー(例えば、パッチクランプの細胞)に隣接して直接配置するのが好ましい。全てのインプットチャネルを分離し、各インプットチャネルの出口へ合流し、中央センサーチャンバーへ送り込むよりも、チャネルを分岐状幾何学的構造に配置する。96〜1024ウェルプレートと接するように、センサー(複数を含む)に隣接する単一送達チャネルを、複数のインプットマイクロチャネルに連結し、各インプットチャネルは、96〜1024ウェルプレートの異なるウェルからインプットを受け取る。この形式は、化合物を送達するチャネル及び廃液チャネルをセンサー(複数を含む)に非常に接近した近接度で配置することができ、それによって、システムからの迅速な応答を保障するという利点を有する。多数の化合物を迅速に連続してセンサー(複数を含む)へ送達することは、化合物を含む流体を、センサーチャンバーに直接送り込む単一チャネルへの制御され、多重化された送達によって達成される。
(スキーム1:個々のマイクロチャネルを処理するための隔壁の使用)
このスキームでは、各マイクロチャネルに連結する貯蔵部を、隔壁、例えば、シーリング用ポリジメチルシロキサン(PDMS)又は当該技術分野で公知のその他の適切な材料を用いて密閉する。隔壁は気密シールを形成するため、隔壁を通って挿入される針又はチューブを介する正圧の適用(例えば空気又は窒素による)により、流体は、センサーチャンバー内の1つ又はそれ以上のセンサー上のマイクロチャネルへ流れ出る(例えば、放射状マイクロチャネルが合流するスポークホイールの中央へ)。隔壁を通って挿入される、針又はチューブを通る小さい吸引による負圧の適用により、流体は、反対方向に回収される(例えば、スポークホイールの中央のチャンバーから、マイクロチャネルに送り込む貯蔵部へ)。
個々の隔膜を使用する先の設計は、大きな柔軟性及び制御をもたらすが、化合物送達と流体流の配列が予め決められたある用途に関しては、より単純な設計が、実施の簡素化及び簡便性を提供する。このスキームでは、例えば、単一隔壁を介して全貯蔵部に均質にかけられる加圧空気を使用することによって、あるいは入口と出口貯蔵部との間の高さの違いにより起こる重力流動の使用によって、同じ正圧を全ての貯蔵部に適用する。各マイクロチャネルから1つ又はそれ以上のセンサーへの化合物の迅速な逐次的送達は、各マイクロチャネルの流体抵抗を変更することによって達成され、これは、各マイクロチャネルの幅及び長さを変えることにより容易に達成される。
この構造では、アレーウェルプレートのそれぞれのウェルからの化合物を、対応するマイクロチャネルに連結する外部チューブ又は毛細管によって導入する。これらの外部チューブ又は毛細管に結合する外部バルブを、流体流を制御するために使用することができる。多数の適切な外部バルブが存在し、手動で、機械的に、電子的に、空気圧で、磁気的に、流体的に又は化学手段(例えば、ヒドロゲル)によって作動されるものが挙げられる。
外部バルブで流体流を制御するより、使用することができる多数のチップベースのバルブがある。これらのチップベースのバルブは、外部バルブ用に使用したものと同じ原理のいくつかに基礎をおくことができ、あるいは、全く異なる、例えば、ボールバルブ、泡弁、電気運動バルブ、ダイアフラム型バルブ、及びワンショットバルブであってもよい。チップベースのバルブを使用する利点は、これらが、元来、微小流体システムとの一体化に適していることである。特に関係の深いものとして、先に記載した設計のいくつか(例えば、分岐状チャネル形式)を実施するのに好ましい、一方向受動バルブがある。
システムは、細胞の電気特性における変化を測定することによって、細胞の応答をモニターするために使用することができる。一態様では、チップのセンサーチャンバーは、細胞ベースのバイオセンサーを含み、システムは、チャネルから溶液流へのバイオセンサーの応答をモニターするための検出器を含む。モニターすることのできる1つの応答は、イオンチャネルのゲート開閉に応答する、バイオセンサーの電気特性の変化である。例えば、バイオセンサーの膜をわたって流れる電流の変化を、電圧固定法を使用して測定することができる。電流は、数ピコアンペア(pA)(例えば、単一イオンチャネル開口部に関して)から数μA(ゼノパス卵母細胞のような大きな細胞の細胞膜に関して)の範囲でありうる。
本発明は、数多くの異なる生物活性分子及び化合物(例えば、候補薬物)の、標的分子を含むセンサーへの送達を制御する微小流体システムを使用するための電位差を利用する。評価することのできる適切な分子/化合物として、薬物、刺激物、毒素、タンパク質、ポリペプチド、ペプチド、アミノ酸、タンパク質の類縁体及び改変体、ポリペプチド、ペプチド及びアミノ酸、抗体及びその類縁体、免疫剤(例えば、抗原及びその類縁体、ハプテン、発熱物質など)、細胞(例えば、真核細胞、原核細胞、感染細胞、トランスフェクト細胞、組換え細胞、細菌、酵母、生殖細胞など)及びその部分(例えば、細胞核、オルガネラ、分泌促進物質、細胞膜の部分)、ウィルス、受容体、受容体のモジュレーター(例えば、アゴニスト、アンタゴニストなど)、酵素、酵素モジュレーター(例えば、阻害物質、補助因子など)、酵素基質、ホルモン、代謝物及びその類縁体、核酸(例えば、オリゴヌクレオチド、ポリヌクレオチド、フィブリノチド、規制的な配列及び/又はイントロン、及び/又はコード領域を含む遺伝子又は分画、アレル異性体、RNA、アンチセンス分子、リボザイム、ヌクレオチド、アプタマー)及びその類縁体及び改変体、化学的及び生物的兵器剤、金属クラスタ、及び無機イオンが挙げられるが、これらに限定されるものではない。
化合物(アゴニスト又はモジュレーター又は薬物のような)を広い範囲のイオンチャネルに結合することは、これらのチャネルにおける構造の変化を誘引し、細胞膜にわたるイオンのフラックスを起こさせるばかりでなく、イオンチャネルに脱感作を起こす、すなわち、持続性、リガンド結合、未切断、非導電性状態に置く原因にもなる(例えば、「Jones and Westbrook, 1996, GL Trends Neurosci. 19:96-101」を参照されたい)。普通、多くのタイプのイオンチャネルの脱感作が数ミリ秒の間に起こり、中枢神経系におけるシナプス情報を加工し修正する機構の1つと考えられる。脱感作は、神経伝達物質又は他の神経モジュレーターによるイオンチャネルの過剰な活性によって起こされた刺激毒性プロセスを予防する、負のフィードバック機構としても作用しているかもしれない(例えば、「Nahum-Levy, et al., 2000, Biophys J. 80:2152-2166」;「Swope et al., 1999, Adv. Second Messenger Phosphoprotein. Res. 33,:49-78」を参照されたい)。
用量応答曲線は、薬物の作用及び力価に関する種々の情報を提供する。マイクロピペットを含む、従来の方法を使用して用量応答曲線を得ることは、しばしば、時間がかかり面倒である可能性がある。細胞(複数を含む)周辺の微小環境の迅速で制御された操作に関し微小流体を使用する、本発明は、用量応答測定に特異的に適用される。用量応答関係は、ほとんどの場合、直線−対数プロットでS字型曲線を描き、ヒルロジスティック関数により記載することができる。
I=Imax/[1+(EC50/C)n]
式中、Iは全細胞電流であり、Cはリガンドの濃度であり、Imaxは最高電流(すなわち、全チャネルが開放状態にあるときのもの)であり、EC50値は最大値の半分(すなわち、受容体集団の半分が活性化された時のものであり、しばしばリガンドの解離常数、KD値に等しい)であり、そしてnは受容体に結合するリガンドの化学量を反映するヒル係数である。
(部分アゴニスト)
薬物分子の受容体介在応答を活性化する能力は、無であるか又は全てであるかの特性と言うより段階的特性である。同じ受容体上で作用する、一連の化学的に関連するアゴニストを細胞上でテストする。最大応答(すなわち、高濃度においてアゴニストが生成することができる最も大きな応答)は、一般的に、アゴニストのそれぞれで相違する。ある化合物(「完全アゴニスト」として知られる)は、最大応答を作り出すことができるが、「部分アゴニスト」と言われる他のものは、準最大応答しか作り出すことはできない。従って、「部分アゴニスト」は、完全アゴニストが受容体に結合するのを阻害することによって、「弱いアンタゴニスト」として作用する。従って、最大応答を作り出す公知のアゴニストとともに規定されたイオンチャネルを使用することによって、アゴニストの活性のグレードをモニターすることができる(例えば、図24を参照)。
一態様では、システムを、イオンチャネル活性のアンタゴニストを選別するために使用する。評価を行うことができる適切なイオンチャネルとして、(i)脱感作しないイオンチャネル、(ii)脱感作するイオンチャネル、(iii)脱感作するが、活性化された時大きな電流変動を媒介するイオンチャネル、及び(iv)脱感作特性がアロステリックモジュレーター(例えば、レクチン)の不可逆的結合によってブロックされているイオンチャネルが挙げられる。アンタゴニストを検出するために、バイオセンサーによって発現されたイオンチャネル又は受容体は、アンタゴニストを適用している間、その前、又はその後に、アゴニストにより活性化される、あるいは「テストされる」ことが必要である。例えば、異なるアンタゴニストを、公知の薬理特性を有する明確なアゴニストと共に適用することができる。異なる濃度でのアンタゴニストを、一定濃度のアゴニストと共に、マイクロチャネルに負荷することができる。
このタイプの競合的作用は、受容体上の同じ結合サイトでのアゴニストとアンタゴニストとの間の競合を言う。可逆的競合的拮抗作用は、同じ最大応答を保ちながら、用量応答曲線の勾配において、より高い濃度へのシフトと、曲線の勾配を特徴とする。不可逆的競合的拮抗作用では、細胞をアゴニストに曝露した時、アンタゴニストに占有されている場合には変化は見られない。
非競合的拮抗作用は、アンタゴニストが、ある時点で、アゴニストによる応答の生成に導く出来事の連鎖をブロックする状態を表す。このタイプの拮抗作用では、アゴニスト及びアンタゴニストがそれぞれ、受容体/イオンチャネル上の異なるサイトに結合し、あるいはアンタゴニストが単純にイオンチャネル細孔をブロックする。純粋な効果は、アゴニストの用量応答曲線の勾配と最大値とを減少させることである。
このタイプの拮抗作用は、ある濃度を超えるアゴニストの自己抑制を言う。すなわち、アゴニストは、充分高い濃度で、それ自身の作用を拮抗し始める。ベル型の用量応答曲線は、しばしば、この種の拮抗作用の存在を示す。
他の態様では、システムを、前シナプス発現イオンチャネルのモジュレーターを検出するために使用する。前シナプスで局在化するイオンチャネルを検討する方法として、しばしば、タンパクリポソーム又は平面的リン脂質二重層に挿入されたシナプトソーム(すなわち、脳組織を均質化することにより生成されるピンチオフ神経末端)のパッチクランプの記録が挙げられる(例えば、「Farley and Rudy, 1988, Biophys. J. 55:919-934; Hirashima and Kirino, 1988, Biochim Biophys Acta 946:209-214」を参照されたい)。Hirashima及びKirino,1988(上記を参照)の方法は、本発明によるシステムにおけるパッチクランプ用のバイオセンサーとして使用することのできる、前シナプス発現イオンチャネルを含む、ジャイアントタンパクリポソームを産生する単純で迅速な方法であるため、特に好ましい。
従来の薬物発見のアプローチは、しばしば、リガンドの生物活性、これは続いてその組織薬理及び生理学的役割を特定化するために使用される、の発見によって始まる。代表的には、リガンドが特定化された後、対応する受容体を、HTS用途における薬物選別用の標的として同定する。オーファン受容体(すなわち、未規定の生物活性を有する受容体)を特定化するための比較的新しい方法は、「逆薬理学的」アプローチと言われることが多い。
10年以上前に開発された、膜電気容量を測定するパッチクランプ方法(例えば、「Neher and Marty, 1982, Proc.Natl.Acad.Sci. USA 79:6712-6716」を参照されたい)は、エキソサイトーシスの基本機構及び制御を検討するのに力強いツールを提供する。
選別手順において使用される細胞が、広範なイオンチャネルタイプを発現する場合、細胞の活性化されたイオンチャネルのイオン透過性特性の特定化を、細胞での薬物の相互反応を特定化するために使用することができる。イオンチャネルの透過性特性に関する情報を、電流対電圧関係を評価することによって決定することができ、異なる保持電位差でのアゴニスト誘発電流の測定によって作ることができる、逆転電位差をモニターすることによって決定することができる。逆転電位と細胞内及び細胞外濃度に関する知識を使用することによって、相対イオンチャネル透過性特性を異なるモデルから決定する。
アゴニストによって活性化されたイオンチャネルからの電流−トレースの分析を、アゴニスト−イオンチャネル相互反応の更なる特性確認のため、集合及び単一チャネルレベルの両方で行うことができる。全細胞パッチクランプで記録された総電流のために得た自己相関機能のフーリエ変換により、単一又は二重ローレンツ機能により適合されうる電力スペクトルを得る。これらの適合は、電流応答(すなわち、コーナー周波数)の周波数依存性の分析を通して、平均単一チャネル導電率及びイオンチャネル反応速度(例えば、平均単一チャネル開放時間)に関する情報を提供する。原則として、より困難で時間のかかる方法ではあるが、アウトサイドアウトパッチクランプ構造から得られる記録も、同じ受容体イオンチャネル複合体により媒介される単一チャネル開口間隔及び異なる導電率レベルを測定するため、分析することが可能である。

図19は、SF6におけるディープ反応性イオンエッチングにより、シリコン中に加工したマイクロチャネルの実施例を示す。フォトリソグラフィ用のマスクを、標準電子ビーム書込み法を使用し、JEOL JBX−5DII電子ビームリソグラフィシステムで製作した(溶媒;反射性4″クロムマスク及びシップリーUV5レジスト、50keVアーク電圧、用量;15μC/cm−2、曝露電流;5nA)。レジストを2000rpmで60秒スピンコートし、250nmのレジストを得、曝露前に、熱プレートで、130℃、10分間ソフトベークを施した。曝露後、パターンを、オーブン中で130℃20分焼成し、シップリーMF24−A中で60秒現像し、脱イオン水で濯いで、反応性イオンエッチング装置(Plasmatherm RIE m−95、30秒、50W、250mTorr、10ccm O2)でエッチングした。クロムをバルザークロムエッチ#4で1〜2分、エッチングし、マスクを、シップリー1165除去装置を使用して残存するレジストから引き剥がして、アセトン、イソプロパノール及び脱イオン水で濯いだ。700nmの熱成長二酸化ケイ素を有し、総厚さが380μmの3インチ[100]、両面が研磨された低Nドープされたシリコンウェハを、反応性イオンエッチング装置Plasmatherm RIE m−95中で清浄し(30秒、50W、250mTorr、10ccm O2)、シップリーS−1813フォトレジストを用い、4000rpmでスピンコートし、1.3μmのレジストを得、これを110mJ/cm−2の用量に400nmの波長で、Carl Suss MA6マスクアライナーを使用して曝露した。ウェハを、脱イオン水で濯いだシップリーMF319中で45秒間現像して、反応性イオンエッチング装置(Plasmatherm RIE m−95、30秒、50W、250mTorr、10ccm O2)中で灰化した。ウェハを、10分間130℃でハードベークを施し、二酸化ケイ素をSioTech緩衝性酸化物エッチング液でエッチングして、脱イオン水で濯いだ。ウェハをアセトンで、残存するレジストから引き剥がして、イソプロパノール及び脱イオン水で濯いだ。ウェハのもう一方側を、シップリーAZ4562フォトレジストを用い、3000rpmで30秒間スピンコートし、約8μmのレジストを得、ソフトベークを、熱プレート上で100℃3分間行い、480mJ/cm−2の用量に、400nmの波長で、Carl Suss MA6マスクアライナーを使用して曝露した。パターンを、シップリーMF312と脱イオン水との50:50混合物中で200秒間現像し、脱イオン水で濯ぎ、反応性イオンエッチング装置(Plasmatherm RIE m−95、30秒間、50W、250mTorr、10ccm O2)で灰化した。
マイクロチャネルを、重合体、ポリジメチルシロキサン(PDMS)でモールドし、これをガラスカバーガラス上に不可逆的にシールし、4つの壁を有する封鎖チャネルを形成した。
(1)PDMSでモールディングするために使用したシリコンは、フォトレジストに対する良好な接着を補償するために、最初にウェハを清浄し、次いで、ネガ型フォトレジスト(SU8−50)の層(〜50μm)をウェハ上にスピンコートして製作した。次いで、ネガ型フォトレジストのこの層を、ソフトベークして、フォトレジスト中に含まれていた溶剤を蒸発させた。マスクアライナーを有するフォトリソグラフィを、電子ビーム書込み法を使用して調製する、適切なパターンを有するフォトマスクを使用して行った。次いで、曝露されたウェハを焼成して、曝露されていないフォトレジストを適切な現像液(例えば、酢酸プロピレングリコールメチルエーテル)中で洗い流すことによって現像した。
(4)次いで、マイクロチャネル特徴部を含む硬化PDMSモールドを、不可逆的にシールして、酸素プラズマ中で約1分間酸化して、ガラス基板を得た。我々がこの実施例使用したチャネル寸法は、幅約100μm、深さ50μmであった。
本発明が使用するHTSを実施するための好ましい実施形態の1つが緩衝液と、各リガンド流は異なる薬物に対応するリガンドとの相互連結流にわたるパッチクランプの細胞を迅速に走査することである。これらの用途では、先に検討したように、マイクロチャネルから排出する流体の流速と、パッチクランプの細胞の走査速度の両方が重要である。図21A〜Dに7−チャネル構造の出口をわたって走査された後のパッチクランプの全細胞の応答を示す。各チャネルの幅は、100μmであり、厚さは50μmであり、そして内部チャネル間隔は25μmである。この7−チャネル構造は、図16Bで示した構造と一致している。マイクロチャネルの製作及びパッチクランプの手順は実施例2に記載した手順と同一である(前記を参照)。使用したパッチクランプの細胞は、PC−12細胞であり、マイクロチャネルの出口から10〜20マイクロメーター離して配置した。チャネル1、3、5及び7をPBS緩衝液で満たし、チャネル2、4及び6はアセチルコリンで満たした。流体流の流速は、6.8mm/秒であった。
実施例3で使用したチャネル構造及び実験設定を、各リガンド流れにおけるリガンドの濃度が予め決められた量によって異なっている条件で、用量応答測定を行うために使用することができる。図23は、そのような実験の1つであって、3つの異なる濃度(1μM、12μM及び200μM)のニコチンをパッチクランプの細胞に適用する実験の結果を示す。この7−チャネル構造では、チャネル1、3、5及び7をPBS緩衝液で満たし、チャネル2、4及び6を、それぞれ、1μM、12μM及び200μMのニコチンで満たした。使用した流速は3.24mm/秒であり、そして細胞走査速度は250μm/秒であった。パッチクランプの細胞を、マイクロチャネルの出口から10〜20μm離して置いた。
市販のパッチクランプ用の微小流体装置(Dynaflow 16(Cellectricon AB,Goteborg Sweden))を使用して、電気生理学的実験を行った。装置は50×57μm(w×h)の寸法の16チャネルであって、オープンボリュームへの出口の点で22μm厚の壁によって分離されている。16個のサンプル貯蔵部の容積は、80μlで、オープンボリュームの寸法は、30×35mmである。実験の前に、装置を、マイクロピペットを使用して異なる溶液で負荷した。2mm厚のポリカーボネート蓋を、両面接着テープ(3M,Stockholm,Sweden)を使用して、サンプル貯蔵部の上に接着し、密閉システムを作った。蓋を、PEチューブを有するシリンジに連結し、シリンジポンプ(CMA/100、マイクロインジェクションポンプ(Carnegie Medicine,Cambridge,UK))を使用して、蓋に囲まれる空気を圧縮して、チャネル内の圧力誘導流でウェルを起動した。
接着WSS−1細胞を、ペトリ皿で4〜8日間(DMEM/F12)培地(当該培地は抗生物質、アンチミコチン(antimycotin)(0.2%)、ウシ胎児血清(10%)及びL−グルタミンを補充)で培養した。実験の前に、細胞を洗浄し、HEPES−食塩緩衝液(HBS)(HEPES:10(単位:mM)、NaCl:140、KCl:5、CaCl2:1、MgCl2:1、及びD−グルコース:10を含み、pH7.4)中で解離した。細胞培養で使用した全ての化学薬品は、シグマ−アルドリッチ(Sigma-Aldrich Sweden AB, Stockholm, Sweden)から入手した。
全細胞構造(保持電位差:−60mV、サンプリング周波数:5kHz、フィルター周波数:1KHz)で、パッチクランプの実験を行った。細胞浴溶液(HBS)は、10mMのHEPES、140mMのNaCl、5mMのKCl、1mMのCaCl2、1mMのMgCl2、10mMのD−グルコースを含んでいた(pH7.4)。パッチクランプの電極溶液は、100mMのKCl、2mMのMgCl2、1mMのCaCl2、11mMのEGTA及び10mMのHEPESを含み、pHをKOHで7.2に調整した。実験は、室温(18〜22℃)で行った。
用量応答曲線を得た。基板の他の全てのマイクロチャネルを1、5、10、20、50、100及び500μMのGABAで負荷し、各アゴニスト曝露後、クリアランスのため、緩衝溶液と相互連結した。チャネル出口にわたって、パッチクランプの細胞を昇順(低い→高い、1μMのGABAへの曝露から始める)で、あるいは降順(高い→低い、500μMのGABAへの曝露から始める)で走査した。走査は、10の異なる等速走査を使用して行い、30ミリ秒から10秒(各texpに関しn=3〜6回の走査)の範囲の曝露時間texpを得た。曝露時間及び洗浄時間twashを、各走査の間同等に保った。twashの影響を分離して調べ、30秒までのtwash、>1秒のtexpのデータに最小限の影響があることが示された。上昇と下降走査の間の待機時間trestは、常に>3分で、その間細胞は緩衝溶液で灌流した。図30A〜Fは、6個の異なる曝露時間に関し、同じ細胞から得たペアワイズピーク電流用量応答曲線を示す。
濃度0.01、0.1、1、5、10、100μMで競合的アンタゴニストビククリンを使用した。ビククリンサンプルを100μMのGABAとともに負荷し、効果を検査した。アゴニストでの実験のように、毎秒、清浄のためチャネルを緩衝溶液で満たし、折り返し走査を行う前に、少なくとも3分間の緩衝液中での静止時間を採用した。100ミリ秒及び3秒のtexpに関し、用量応答曲線を得た。アンタゴニストを昇順で適用した時、それぞれ、5.17±0.4μM(texp=100ミリ秒)及び8.46±1.4μM(texp=3秒)のIC50値を得た。対照的に、降順適用では、3.56±0.9μM(texp=100ミリ秒)及び1.84±0.4μM(texp=3秒)のIC50値を得た。したがって、上昇及び下降走査を比べると、長い曝露時間でブロッキング効果において要素5相違に近いものがある。全体として、結果は、受容体の活性化記憶は、特定の薬物の力価に影響を与えることを示す。また、用量応答実験を、GABAで、前のとおり固定濃度1μMビククリンで行い、ほぼIC20値に対応した。図33A及びBは、この方法で得た用量応答曲線は、純粋なGABA応答に比べて、より高い濃度で、左に変位していることを示す。昇順走査に関し、用量応答曲線は、降順走査より大きく変位する。さらに、アンタゴニストの添加は、純粋なGABA応答に関して、動的領域及び同調性において観察されるいくらかの違いを一掃する。如何なる科学的な理論に縛られるものではないが、これは固定濃度のアンタゴニストは異なる初期濃度のアゴニストの効果を特により長い曝露時間で平均化するということを意味する可能性がある。また、ある程度、受容体は、濃度及び時間依存性シグナルを解読する能力を、アンタゴニストの添加によって失ってしまうということも示唆している。図34a及びbは、パッチクランプの実験の結果を示す。GABA(1、5、10、20 50 100及び500mM)と固定濃度1μMのビククリンでの用量応答実験のために、パッチクランプの実験を16個の負荷チャネル微小流体チップを用いて行い、これはほぼIC20値に対応した。これらのサンプルチャネルを、他の各チャネル内で、緩衝液チャネルと相互連結し、曝露時間を100ミリ秒又は3秒のいずれかに設定した。
Claims (58)
- a)アゴニストによって活性化される受容体又はアンタゴニストによって不活性化される受容体を含有する細胞ベースのバイオセンサーを含んでなるチャンバー、及び
サンプル、アゴニスト、アンタゴニストの1つ又はそれ以上を送達する、複数の送達チャネル(各々のチャネルは、実質的に分離した水性流をチャンバーへ送達するための出口を含む)、を包含する基板を提供すること;及び
b)バイオセンサーを、2つ又はそれ以上の出口から出る流体流に、順次曝露すること;
を含んでなる、受容体を調節する、制御する、調製する又は検討する方法。 - 前記チャンバーが、緩衝液、少なくとも1つのアゴニスト、少なくとも1つのアンタゴニスト、少なくとも1つのサンプル、又はこれらの組合せを包含する、請求項1に記載の方法。
- 前記曝露することが、選定された濃度のサンプルにバイオセンサーを選択的に曝露することである、請求項1に記載の方法。
- 前記曝露することが、選定された時間選択的に行われる、請求項1に記載の方法。
- 前記チャネルに、1つ又はそれ以上の緩衝液を提供することを更に含んでなる、請求項1に記載の方法。
- 前記バイオセンサーを、1つ又はそれ以上の緩衝液に曝露することを更に含んでなる、請求項1に記載の方法。
- 前記バイオセンサーの1つ又はそれ以上の緩衝液への曝露することが、1つ又はそれ以上のサンプルへの曝露の間に散在する、請求項1に記載の方法。
