Abstract
Description
(ペプチド)ドメイン:それを含む配列中に、欠失−変異分析によって確定され得る少なくとも5つのアミノ酸を含むペプチド配列。このようなドメインは、一般に、機能又は役割に関与しており、その変異が機能の喪失を引き起こす不可欠なアミノ酸の存在によって特徴付けられる。例としては、核の多くのタンパク質中に存在するNLS(核局在化シグナル)ドメイン、細胞透過ドメイン、NES(核外移行シグナル)ドメイン、タンパク質−タンパク質相互作用ドメイン及び触媒ドメインを挙げることができる。
プレイ:試験ペプチドをコードする配列と融合しているGAL4の活性化ドメインを含有するプラスミド、例えば、pGAD424由来のプラスミド。
実施例1は、この試験に対して想定され得る作業条件に関して、さらに詳しい情報を提供する。
ウイルス:HIV−1−LAIというリンパ球向性株(Barre−Sinoussi et al;1983);
プロトコール;5つの濃度の試験ペプチド中で、細胞を30分間前処理した後、HIV−1−LAI株に感染させる。培養を通じて、このペプチドを培地中に維持し、感染から7日後に細胞上清を集め、逆転写活性を測定する。本試験を実施するために使用できる作業条件、濃度及び緩衝液に関して、実験の部に、さらに詳しい情報が記載されている。
i.本発明の相互作用ポリペプチドの存在下に標的分子を置くことと;
ii.前記標的分子と前記相互作用ポリペプチド間の相互作用を検出することと;
iii.候補分子を添加することと;
iv.前記標的分子と前記相互作用ポリペプチド間の相互作用に対する修飾を検出することと;
を含む。
そのN末端が転写因子(本ケースではGAL4)の活性化ドメインと融合され、そのC末端が塩基性Tatドメインの9つのアミノ酸と融合されたランダムヘプタペプチドのライブラリーを、酵母発現ベクター中に構築した。酵母中で目的のタンパク質を発現させることができるベクター中で、目的のタンパク質(ウイルス又は細胞性)をコードする配列を、DNAの結合ドメインをコードする配列(本ケースではLexA)の下流にクローニングする。
酵母環境中で選択されたモチーフがヒト細胞の生化学的状況下でも機能することを確かめるために、上述した試験と類似の試験をHeLa細胞中で行う。ヒト細胞中で、Flagエピトープ、核局在化シグナル及びタンパク質Vp16の活性化因子ドメインをコードする配列の下流をヒト細胞中での発現を可能とするプラスミド中の酵母ベクターから、ヘプタペプチドモチーフを再クローニングする。
短い直鎖ペプチドは、細胞外及び細胞内培地中での安定性が乏しい場合がある。この問題を克服するために、本発明者らは、ヒト細胞中において安定で、豊富に存在する小タンパク質に、ヘプタペプチド/塩基性Tatアミノ酸配列を会合させることを決定した。それらの選択は、まず、ユビキチンファミリーのメンバーに的を絞った。この構築物は、ユビキチン(ub)そのもの及び相同タンパク質SUMO−1を用いて作製した。これらのタンパク質をコードする配列を、哺乳類細胞に対する発現ベクター中に導入した。喪失されたジグリシンモチーフの場所で、SUMO−1又はユビキチンの下流に、ヘプタペプチド/塩基性Tatアミノ酸配列を導入する。このジグリシンモチーフは、前記ポリペプチドによって修飾されたタンパク質のリジンの側鎖に通常結合される。SUMO−1特異的Tatヘプタペプチド融合タンパク質(SHPT)及びRev(SHPR)の適切な発現を確認することができた。Ub構築物の場合、ユビキチンに対して誘導された良い抗体が存在せず、実施されなかった。
これらの結果に従って、本発明者らは、細胞中に透過し、細胞外培地から始まる標的タンパク質の機能を阻害する能力を評価できるようにするため、相互作用ペプチドを大量に作製するためのシステムを開発した。細菌中での産生を可能とするベクター中に、抗Rev SUMO−1−ヘプタペプチド/塩基性Tatドメイン配列をクローニングした。ヘパリンカラム、次いでゲルろ過上で精製するためのプロトコールを確定した。最初の結果は、SHPRが大量に産生され、この方法によって精製できることを示した。
HIV−1は、細胞の重要な因子に対するその作用がウイルス粒子の迅速且つ効果的な産生を確保する幾つかの制御タンパク質を発現する。20年にわたる熱心な研究の後、これらの制御タンパク質の様々な作用を誘導する分子的機序を理解する上で、著しい進歩が遂げられた。特に、これらの制御タンパク質のうち2つ、すなわち、Tat及びRevタンパク質に興味が持たれた。
A.ランダムペプチドライブラリー:
1−pGAD−CR構築物:
このプラスミドは、pGAD424(clontech)の誘導物であった。このベクターをEcoRI及びBgllIで消化した。
ランダム配列断片は、以下の2つのオリゴヌクレオチドをハイブリダイズし、DNAポリメラーゼで修復し、酵素BamHI及びXmaIで消化することによって作製された。
GGCCCCAGCGTCACb−5’
(bはビオチンを表す。)
ビオチン化された末端を抽出するために、この消化産物を常磁性ストレプトアビジンビーズと混合した。
1.pLex9ベクター:
このプラスミドは、pGBT9(clontech)の誘導物であった。GAL4 DNAの結合ドメインをLexAに対する結合ドメインと交換した(Farjot et al., 1999)。
pSG−Tatベクター(Veschambre et al., 1995)から得られた、修復されたNcoI/BamHI断片を、pLex9のSamI及びBamHI部位の間にクローニングした。
これらの構築物は、LexAのものと融合された、完全なRev配列(plexRev)又はNESのもののみ(pLex−NES)を含有した(Farjot et al, 1999)。
A.pSG−FNV−P構築物:
これらの構築物は、プラスミドpSG−FNV(Desbois et al.,1996)の誘導物である。以下のオリゴヌクレオチドを用いて、pGAD−CR−P構築物から、ヘプタペプチドを含有する領域を増幅した。
5’−CCTGAGAAAGCAACCTGACC−3’(オリゴ3’)
SalI及びBglIIでこのPCR断片を消化し、pSG−FNVベクターのXhoI及びBglII部位の間に挿入し、プラスミドpSG−FNV−Pを得た。
1.ベクターpSG−LexA−Revの構築:
このベクターは、哺乳類細胞中で融合物LexA−Revを発現する(Farjot etal., 1999)pSG5(Green et al, 1988)の誘導物である。
Tatをコードする配列を、pSG−LexA中のLexAをコードする配列の下流に導入した。
このプラスミドは、LexAに対する5つの結合部位の制御下にあるSEAP配列を有していた(Farjot et al,1999)。
A.野生型pTL1−SUMO−1構築物:
SUMO−1配列は、以下のオリゴを用いて、cDNA(HGMP−UKから得られたIMAGEクローン)から増幅した。 5’−GGGTCGACGTCCATATGTCTGACCAGGAGG−3’
5’−AAAAGATCTCTAAACTGTTGAATGACC−3’
増幅された断片を、SalI及びBglIIで消化し、pSG5の誘導物である発現ベクターpTL1のXhoI及びBglII部位の間に挿入した。
この構築物は、SUMO−1と融合したヘプタペプチドの挿入を可能とし、ジグリシンモチーフを置換した。以下のオリゴヌクレオチドを用いてSUMO−1配列を増幅した。

増幅された断片をSalI及びBglIIで消化し、ベクターpTL1のXhoI及びBglII部位の間に挿入した。
ヘプタペプチド及び塩基性Tatドメインを含有する配列を、II.Aに記載されているように増幅し、SalI及びBglIIで消化し、ベクターpTL1−SUMO−CPのXhoI及びBglII部位の間に挿入した。
A.pTL1−Ub−CP構築物:
ユビキチンに対する配列は、以下のオリゴヌクレオチドを用いて、cDNA(HGMP−UKから得られたIMAGEクローン)から増幅した。
5’−CACGAAGATCTACACTCGAGCTCTCAGACGCAGGACC−3’
増幅された断片をEcoRI及びBglIIで消化し、pTL1のEcoRI及びBglII部位の間に挿入した。
II.Aに記載されているように、ヘプタペプチドと塩基性Tatドメインを含有する配列を増幅し、SalI及びBaglIIで消化し、pTL1−Ub−CPのXhoI及びBglII部位の間に挿入した。
pTL1−SUMO−PプラスミドをHindIII及びBglIIによって消化した。ヘプタペプチド/塩基性Tatドメイン部分と融合したSUMO−1配列を含有する断片を、pFLAGMacベクター(IBI FLAG(R) Biosystems)のHindIII及びBglII部位の間に挿入した。
PTL1−Ub−PプラスミドをEcoRI及びBglIIによって消化した。ペプチド/塩基性Tatドメイン部分と融合したUb配列を含有する断片を、ベクターpFLAG−2(IBI FLAG(R) Biosystems)のEcoRI及びBglII部位の間に挿入した。
5% CO2を含有する湿った雰囲気中において、5%のウシ胎児血清を補充したDMEM培地中でHeLa及びCOS7細胞を培養した。形質移入は、カルシウム/リン酸共沈法を用いて行った。
2LのLB培地中で、0.9の光学密度まで培養されたBL21−CodonPlusTM−RP(Stratagene)Eコリ細胞を形質転換するために、セクションVIに記載したベクターを使用した。18℃で30分後、0.1mMのIPTGを培地に添加し、これを18℃で一晩継続した。Rocheの「完全」アンチプロテアーゼ(Complete antiprotease.Roche)、0.5mg/mLのリゾチームおよび20u/mLのベンゾナーゼ(Sigma)を補充した溶解緩衝液:200mMのNaCl、0.1Mのtris−HCl、pH7.4、10mMのMgCl2中で、この細菌に音波処理を施した。4000rpmで30分間の遠心後、5mLのヘパリンHyperDカラム(Biosepra)に上清をかけた。溶出は、DE 600緩衝液(600mMのKCl、20mLのHepes、pH7.9、10μMのZnCl2、1.5mMのMgCl2、1mMのEDTA及び1mMのDTT)を用いて行った。Hiload 16/60 Superdex 200ゲルろ過カラム(Amersham Pharmacia Biotech)によって、溶出産物を分画した。SHPを含有する画分を合わせて、DE50緩衝液(KCl濃度が20mMであることを除いて、DE600緩衝液と同じ組成)に対する透析後、5mLのmonoQ HyperDカラム(Biosepra)にかけた。PBS 1×緩衝液に対して、フロースルーを透析し、凍結乾燥によって10倍濃縮し、滅菌ろ過を行った。
使用した技術は、本分野において慣用されているものであった(ポリアクリルアミドゲル、二フッ化ポリビニリデン膜、PVDF、Sigma製のFLAGに対して誘導され、1:1000希釈され、Amersham Biosciences製ECLキットで可視化されるモノクローナル抗体)。
PBMC細胞には、HIV−1−LA1(Barre−Sinoussi et al, Science, 1983,220, 868−871)というリンパ指向性株を感染させ、MDM細胞には、マクロファージに対して指向性を有するHIV−1/Ba−L(Gartner et al, Science, 1086, 233,215−219)という株を感染させた。PHA−Pで活性化された臍帯血単核細胞(UBMC)とともに、これらのウイルスをインビトロで増幅した。可溶性因子を除去するために、無細胞上清を360000gで、10分間遠心し、残留物を細胞培養培地中に再懸濁した。PHA−Pで活性化されたPBMC細胞を用いて、ウイルス株を滴定し、Karberの式を用いて、50%組織培養感染量(TCID50)を計算した。
Tat及びRevに結合するペプチド配列の同定
まず、ツーハイブリッド酵母システムスクリーニングによって、制御タンパク質Tat及びRevに対する相互作用ペプチドの探索を行った。ベイトは、Tat、Rev又はRevのNESの全配列と融合したLexAに対する配列を含有していた(図7A)。プレイは、GAL4活性化ドメイン、7アミノ酸のランダム配列及びTat形質導入ドメイン(図7B)という3つの異なる部分を含んでいた。21の縮重位の系列を含む、合成オリゴヌクレオチドから調製されたDNA断片をクローニングすることによって、プレイに対する発現ベクターのライブラリーを構築した。
次いで、細胞外培地から同定されたSHPの活性を調べた。まず、細胞毒性を調べた。最大1μMの濃度まで、これらの分子は、インビトロで培養され、PHA及びIL−2で活性化された末梢血単核細胞(PBMC)の集団における細胞死のパーセントを変化させないように見えた。
最後の工程では、SHPがHIV−1の複製を実際に阻害できるかどうかを調べた。これは、PHA−Pによって活性化されたPBMC中及びMDM中で行った。PHP−Aによって活性化されたPBMCのモデルでは、HIV−1−LA1の複製は第7日目に最適であった。このため、様々なSHPの効果をこの時点で検査した。AZT及びインジナビルを対照として用いた。予想どおり、HIV−1−LA1の複製は、AZT及びインジナビルによって効果的に阻害された(図12A及び表2a)。SHPT−8、抗HIV活性を全く有しなかったが、SHPR−15は低い活性を有していた。これに対して、SHPR−142及びSHPR−190は、PHA−Pで活性化されたPBMC細胞中での複製を減少した。HIV−1の複製は、2μMのSHPR−142(73±6%、表2a参照)並びに1及び2μMのSHPR−190(それぞれ、80±7%及び100%、表2a参照)によって減少された。50%有効量の比較によって、SHPR−190がSHPR−142より活性が高いことが確認される(表2bを参照)。

Revに対して誘導されたSHPの作用機序並びにSHPR−142及び−190中の各位置に存在する残基の重要性を最大限決定するために、変異分析を行った。
Revタンパク質は、特定のRNA配列:RREに結合することによって、HIV−1 mRNAの細胞質への移行を引き起こす。本発明者らは、RevのSHPR阻害剤の結合が、このRev/RRE会合を妨害するかどうかという問いに対して答えを出した。この目的のために、RRE要素に対応するRNA分子をプローブとして用いたゲル遅延実験を行った。細菌中で産生されたRevタンパク質は、このRNAに結合し、非変性ポリアクリルアミドゲル移動の遅延を誘発する。SHPR−190及び−142タンパク質の添加は、この会合を阻害しなかったが、さらに大きな遅延を引き起こし、SHPR−Rev複合体がなおRREに結合できることを示している。

- 以下の要素:
(a)配列X1X2X3X4X5X6X7を有するヘプタペプチドモチーフと;
(b)形質導入ドメインと;
を含み、キメラポリペプチドであること、アミノ酸X7が前記ポリペプチドのC末端から5ないし35のアミノ酸であること、及びドメイン(b)がモチーフ(a)に関してC末端側に位置すること、を特徴とする相互作用ポリペプチド。 - 前記モチーフがランダムプロセスによって規定される配列を有することを特徴とする、請求項1に記載の相互作用ポリペプチド。
- (c)安定化ドメイン;をさらに含み、前記安定化ドメインが前記ポリペプチドのヘプタペプチドに関してN末端側に位置することを特徴とする、請求項1に記載の相互作用ポリペプチド。

- 前記ヘプタペプチドがツーハイブリッド酵母試験においてHIV−1のRevタンパク質と相互作用することができ、前記ポリペプチドがHIV−1ウイルスの複製をインビボで阻害することができる、請求項15に記載のポリペプチド。

- 他の動物種におけるHIV又は相同ウイルスによる感染を治療するための薬物を製造するための、請求項1から18の何れか一項に記載のポリペプチドの使用。

ii.前記細胞の表現型の修飾を検出することと;
iii.必要に応じて、前記ポリペプチドのヘプタペプチドモチーフの配列を決定することと;

- 前記レポーター遺伝子がツーハイブリッドシステムのレポーター遺伝子であることを特徴とする、請求項36に記載のスクリーニング方法。
- i.請求項1から14又は19から22の何れか一項に記載のポリペプチドを作製することと;
ii.作製されたポリペプチドの存在下に標的を置くことと;
iii.ポリペプチドの前記標的との相互作用を検出することと;
iv.必要に応じて、ヘプタペプチドモチーフの配列を決定することと;
を含む、所定の細胞内標的と相互作用する分子を同定するために分子をスクリーニングする方法。 - 前記相互作用が、例えば、ツーハイブリッドシステムを用いて、表現型の変化を観察することによって、又は存在物のアッセイによって検出される、請求項38に記載のスクリーニング方法。
- 標的分子を含有する細胞を、請求項1から14の何れか一項に記載の相互作用ポリペプチドとインビトロで接触させることを含み、前記ポリペプチドが前記標的分子と相互作用するヘプタペプチドモチーフを有する、細胞内標的分子の特性を調節する方法。
- 表現型スクリーニングにおけるプレイとしての、請求項23に記載の分子の使用。
- 前記表現型スクリーニングがツーハイブリッドスクリーニングであることを特徴とする、請求項41に記載の使用。
- 前記スクリーニングにおけるベイトがHIV−Revタンパク質又はそのドメインの一つをコードする配列を含むことを特徴とする、請求項42に記載の使用。
- 前記スクリーニングにおけるベイトが7から15個のアミノ酸のドメインをコードする配列を含むことを特徴とする、請求項42に記載の使用。
- 前記ドメインがNLS(核局在化シグナル)であることを特徴とする、請求項44に記載の使用。
- 前記ドメインがNES(核外移行シグナル)であることを特徴とする、請求項44に記載の使用。
- 前記NESがHIV−Revタンパク質のNESであることを特徴とする、請求項46に記載の使用。
- 前記NESが以下の配列:LQLPPLERLTL(配列番号8)を有することを特徴とする、請求項46に記載の使用。
- i.請求項1から14又は19から22の何れか一項に記載の相互作用ポリペプチドの存在下に標的分子を置くことと;
ii.前記標的分子と前記相互作用ポリペプチド間の相互作用を検出することと;
iii.候補分子を添加することと;
iv.前記標的分子と前記相互作用ポリペプチド間の相互作用に対する修飾を検出することと;
を含む、分子内標的分子と相互作用できる候補分子をスクリーニングする方法。 - 前記標的分子と前記相互作用ポリペプチド間の前記相互作用が、例えばツーハイブリッド試験において、レポーター遺伝子を使用することによって観察される、請求項49に記載の方法。
- 前記標的タンパク質がRevタンパク質であり、前記相互作用ポリペプチドが請求項15から18の何れか一項に記載のポリペプチドである、請求項49又は50に記載の方法。
- i.19から22の何れか一項に記載のポリペプチドの集団の異なるメンバーをコードする配列を含む核酸分子を作製することと;
ii.標的分子をベイトとして含む、酵母ツーハイブリッドシステムを用いて前記分子をスクリーニングすることと;
iii.相互作用を検出することと;
iv.必要に応じて、ヒト細胞における相互作用を確認することと;
を含む、細胞内標的分子の特性を調節することができる分子を同定する方法。
