従って、本発明は、AdipoR1及びAdipoR2のin vivoにおける機能を解明するための手段及びその用途を提供することを目的とする。
また、本発明は、上記非ヒト動物由来の、アディポネクチン受容体1及び/又はアディポネクチン受容体2の機能が欠失した細胞を提供するものである。
さらに、本発明は、上記非ヒト動物よりなるメタボリックシンドローム、インスリン抵抗性、インスリン分泌不全、糖尿病、動脈硬化症、ミトコンドリア機能低下、酸化ストレス又は高血圧症のモデル動物を提供するものである。
さらに本発明は、上記非ヒト動物に被検物質を投与することを特徴とするアディポネクチン受容体1及び/又はアディポネクチン受容体2作動薬、AMPキナーゼ活性化薬、PPARα活性化薬、インスリン分泌促進薬、グルコキナーゼ作動性糖取り込み促進薬、インスリン分泌促進薬、抗炎症薬、抗酸化ストレス薬、ミトコンドリア機能改善薬、高血圧治療薬、又はメタボリックシンドローム治療薬のスクリーニング法を提供するものである。
さらに本発明は、上記非ヒト動物に被検食品を投与することを特徴とするメタボリックシンドローム、肥満、糖尿病、動脈硬化、ミトコンドリア機能低下、酸化ストレス、高血圧症に有効な機能性食品の評価法を提供するものである。
さらに、本発明は、被検物質の存在下に、前記細胞を培養することを特徴とする、アディポネクチン受容体1及び/又はアディポネクチン受容体2作動薬、AMPキナーゼ活性化薬、PPARα活性化薬、インスリン分泌促進薬、グルコキナーゼ作動性糖取り込み促進薬、抗炎症薬、抗酸化ストレス薬、メタボリックシンドローム治療薬、ミトコンドリア機能改善薬又は高血圧症治療薬のスクリーニング法を提供するものである。
ここで遺伝子機能の欠失時期特異的に制御する例としては、任意の各発生・成長段階や、検討したい化合物の動物投与の前後など、が挙げられ、いつでも自由自在である。このような制御は、例えば、Cre/LoxP法をもとにして、キメラCre組換え酵素(CreERT)を発現させ、その遺伝子組換え活性を薬剤投与により時間的にコントロールすることによってなされる。その結果LoxPを導入した遺伝子は時間特異的に組換えをおこすことによって行なわれる。
かかるターゲティングベクターの構築は、市販のプラスミドベクターを利用して行うことができる。
AdipoR1遺伝子欠損動物はAMPキナーゼの経路などが異常となり、インスリン抵抗性を来たし、さらにインスリン分泌にも異常をきたし、糖尿病やメタボリックシンドローム、癌のモデル動物となる。例えば、肝臓においては、AMPキナーゼによって通常抑制されている糖新生に関わる酵素(PEPCKやG6Paseなど)や脂肪合成に関わる分子(SREBP1cやSCD1など)の発現が上昇しているのが認められた。さらに脂肪酸燃焼の活性が低下しているのが認められた。そして空腹時血糖値・インスリン値の上昇と糖負荷後の血糖値の上昇、すなわちインスリン抵抗性と耐糖能障害、メタボリックシンドロームが認められた。インスリン抵抗性の責任臓器としては、高インスリン正常血糖クランプ法により、主には肝臓であることも分かっている。さらに、単離膵島を用いた実験により、インスリン分泌にも不全があることが明らかとなっている。さらに、AdipoR1の発現が低下すると、癌細胞の増殖が促進されることも明らかとなっている。
AdipoR2遺伝子欠損動物はPPARαの経路などが異常となり、さらに肝臓への糖の取り込みに重要なグルコキナーゼの発現の低下、脂肪組織や肝臓での酸化ストレスの増大と慢性炎症の惹起などによりインスリン抵抗性を来たし、糖尿病やメタボリックシンドローム、癌のモデル動物となる。例えば、通常は、PPARαによって発現が誘導されている脂肪燃焼に関わる酵素(ACOなど)やエネルギー消費に関わる分子(UCP2など)の発現が低下しており、実際に脂肪酸燃焼の活性も低下しているのが認められた。さらに、酸化ストレスの消去に関わる酵素(SOD1やcatalaseなど)の発現の低下と炎症性サイトカイン(TNFαなど)やケモカイン(MCP−1など)の発現の上昇が認められ、各々の臓器障害を惹起しているのが認められた。そしてインスリン値の上昇、すなわち全身性のインスリン抵抗性、メタボリックシンドロームが認められた。さらに、AdipoR2の発現が低下すると、動脈硬化が促進や癌細胞の増殖が促進されることも明らかとなっている。

以上よりアディポネクチン/AdipoR経路が骨格筋においてMEF2、PGC−1αを介し、ミトコンドリア機能調節を行っている可能性が示唆された。また、酸化ストレスの消去に関わるSOD2の低下が認められたことより、アディポネクチンのAMPK、PGC−1αを介した、酸化ストレス消去作用が存在する可能性が示唆された。
アディポネクチンが関与する疾患の治療薬としては、AdipoR1−AdipoR2作動薬、AMPキナーゼ活性化薬、PPARα活性化薬、インスリン分泌促進薬、グルコキナーゼ作動性糖取り込み促進薬・インスリン分泌促進薬、抗炎症薬、抗酸化ストレス薬、メタボリックシンドローム治療薬(肥満治療薬、糖尿病治療薬、動脈硬化治療薬等)ミトコンドリア機能改善薬、高血圧治療薬が挙げられる。また機能性食品としては、メタボリックシンドローム、肥満、糖尿病、動脈硬化、ミトコンドリア機能改善、酸化ストレス、高血圧症に有効な機能性食品が挙げられる。
AdipoR1遺伝子(配列番号1)及びAdipoR2遺伝子(配列番号4)を含むクローンをクローニングし、AdipoR1をコードしているエクソン2、3、4をneo耐性遺伝子に置き換えたターゲティングベクター(図1)、及びAdipoR2をコードしているエクソン3をneo耐性遺伝子に置き換えたターゲティングベクター(図2)を構築し、それぞれJ1ES細胞(129/Sv)にエレクトロポレーション法により導入してトランスフェクトした。ターゲティングベクターには、ネガティブセレクションのマーカーとして、ジフテリア毒素Aフラグメント(DT−A)を用いた。J1ES細胞(129/Sv)は、ターゲティングベクター導入に先立って、マウス繊維芽細胞等をフィーダー細胞として、これにES細胞用の液体培地(GIBCO社)を加えて共培養した。培養、エレクトロポレーション、相同組み換え体の選択は、公知の方法(J.Biol.Chem. 277, 25863−25866(2002))に則って行った。トランスフェクトされた細胞を、マイクロインジェクション法(Hogan,B.et al.”Manipulating the Mouse Embryo”Cold Spring Habor Laboratory Press,1988)により黒色の毛色を有するC57BL/6マウスの初期胚に導入し、キメラ胚を作製し、得られた胚を、仮親(偽妊娠動物)の子宮に移植し発生させ、キメラマウスを作製し、C57B1/6雌マウスとの交配によりF1、さらにF2を作製し、AdipoR1遺伝子欠損マウス、AdipoR2遺伝子欠損マウスを得た。遺伝子型の確認は、サザンブロッティング及びPCR法により行った。サザンブロッティングの結果を図5に示す。PCR法によるAdipoR1遺伝子の確認で用いたプライマーは、forward primer 1(5'- GCA GGG TAA GCT GAT TAG CTA TG -3':配列番号7)、forward primer 2(5'- ATA GAT CTC TCG TGG GAT CAT TG -3':配列番号8)、reverse primer(5'- TTA CTG CAC TTC TTC TGC TGG A -3':配列番号9)であり、AdipoR2遺伝子の確認で用いたプライマーは、forward primer 1(5'- AGC CTA CTG CCT ACT GTA TTG T -3':配列番号10)、forward primer 2(5'- ATA GAT CTC TCG TGG GAT CAT TG -3':配列番号11)、reverse primer(5'- ACT CTT CTA ACC TTC ATC AGG AG -3':配列番号12)である。
なお、用いたC57B1/6雌マウスは、8〜10週齢であり、CLEA Japanから購入した。マウスの飼育は、12時間の明暗周期で行い、餌としては、CE−2(CLEA Japan;タンパク質25.6%、食物繊維3.8%、灰分50.5%、脂肪分4%、水分9.2%を含有する)を与えた。
前記のターゲティングベクター構築において、欠損させるAdipoR1及び/又はAdipoR2遺伝子領域の5’側にloxP遺伝子を3’側にloxP遺伝子で挟んだマーカー遺伝子(ネオマイシン耐性遺伝子等)を組み込んだターゲティングベクターを作製し、ES細胞に導入した。ES細胞はマーカーによる選択の後、サザンブロッティングあるいはPCR法によるジェノタイプ解析を行って相同組換えを確認した。
AdipoR1及び/又はAdipoR2遺伝子両端にloxP遺伝子を組み込んだターゲッティングベクターを導入したloxP導入組換え動物と、Cre発現ベクターを導入したCre発現組換え動物を別個に作製し、両者を交配することによって、Cre−loxP組換え動物を作製した。
図3にマウスAdipoR1コンディショナル遺伝子欠損用のターゲティングベクターを示す。このコンストラクトは、Cre蛋白の発現に応じて、時期特異的、部位特異的にマウスAdipoR1遺伝子を欠損させるため、AdipoR1遺伝子のイントロン5に、前後をFLTサイト及びloxPサイトではさまれたneo耐性遺伝子に置き換えたターゲティングベクターを作成し、挿入した。さらに、AdipoR1遺伝子のイントロン2に、loxPサイトを挿入した。
図4にマウスAdipoR2コンディショナル遺伝子欠損用のターゲティングベクターを示す。このコンストラクトは、Cre蛋白の発現に応じて、時期特異的、部位特異的にマウスAdipoR2遺伝子を欠損させるため、AdipoR2遺伝子のイントロン7に、前後をFLTサイト及びloxPサイトではさまれたneo耐性遺伝子に置き換えたターゲティングベクターを作成し、挿入した。さらに、AdipoR2遺伝子のイントロン4に、loxPサイトを挿入した。
マウスAdipoR1−AdipoR2コンディショナルダブル遺伝子欠損マウスはAdipoR1コンディショナル遺伝子欠損マウスとAdipoR2コンディショナル遺伝子欠損マウスの交配によって得られた。
実施例1で得られたAdipoR1遺伝子欠損マウス(以下、AdipoR1-/-マウス又はR1KOと称する)は、AMPキナーゼの経路などが異常となり、インスリン抵抗性を来たし、さらにインスリン分泌にも異常をきたしていた。例えば、肝臓においては、AMPキナーゼによって通常抑制されている糖新生に関わる酵素(PEPCKやG6Paseなど)や脂肪合成に関わる分子(SREBP1cやSCD1など)の発現が上昇しているのが認められた。さらに脂肪酸燃焼の活性が低下しているのが認められた。そして空腹時血糖値・インスリン値の上昇と糖負荷後の血糖値の上昇、すなわちインスリン抵抗性と耐糖能障害、メタボリックシンドロームが認められた。
以下、及び試験例3及び4に、これらの結果を含め、得られた欠損動物の特性についての実験結果を詳細に示す。
次に、血漿中のグルコース及びインスリン量を調べた(図7)。グルコースの測定には、グルコースBテスト(和光純薬工業株式会社)を、インスリンの測定には、株式会社シバヤギの抗体を用いた。図7に示すように、AdipoR1-/-マウス(R1KO)では、血漿中のグルコース及びインスリンの量は、野生型(WT)と比較して顕著に多かった。
実施例1で得られたAdipoR2遺伝子欠損マウス(以下、AdipoR2-/-マウス又はR2KOと称する)は、PPARαの経路などが異常となり、さらに肝臓への糖の取り込みに重要なグルコキナーゼの発現の低下、脂肪組織や肝臓での酸化ストレスの増大と慢性炎症の惹起などによりインスリン抵抗性を来たしていた。例えば、通常は、PPARαによって発現が誘導されている脂肪燃焼に関わる酵素(ACOなど)やエネルギー消費に関わる分子(UCP2など)の発現が低下しており、実際に脂肪酸燃焼の活性も低下しているのが認められた。さらに、酸化ストレスの消去に関わる酵素(SOD1やcatalaseなど)の発現の低下と炎症性サイトカイン(TNFαなど)やケモカイン(MCP−1など)の発現の上昇が認められ、各々の臓器障害を惹起しているのが認められた。そしてインスリン値の上昇、すなわち全身性のインスリン抵抗性、メタボリックシンドロームが認められた。
以下、及び試験例3、4に、これらの結果を含め、得られた欠損動物の特性について実験結果を詳細に示す。
また、これらのマウスを観察したところ、体重は通常と相違なかった。
インスリン抵抗性は、AdipoR1-/-・AdipoR2-/-マウスについても同様の方法で調べた。結果をAdipoR1-/-マウス及びAdipoR2-/-マウスに並べて図14に示す。AdipoR1-/-・AdipoR2-/-マウスでは、これら単独の遺伝子欠損マウスよりもインスリン抵抗性が悪化していた。
以上よりアディポネクチン/AdipoR経路が骨格筋においてMEF2、PGC−1αを介し、ミトコンドリア機能調節を行っている可能性が示唆された。また、酸化ストレスの消去に関わるSOD2の低下が認められたことより、アディポネクチンのAMPK、PGC−1αを介した、酸化ストレス消去作用が存在する可能性が示唆された。
AdipoR1-/-マウス(R1KO)、AdipoR2-/-マウス(R2KO)及びAdipoR1-/-・AdipoR2-/-マウス(DoubleKO)について、肝臓におけるアディポネクチン誘導のAMPキナーゼ活性を調べた(図25)。キナーゼ活性測定においては、全長のアディポネクチンを30μg/10gの濃度で肝臓に10分間処理した。野生型で見られたアディポネクチン投与によるAMPKリン酸化は、R1KO及びDoubleKOでは阻害されていた。
AdipoR1-/-マウス、AdipoR2-/-マウス及びAdipoR1-/-・AdipoR2-/-マウスについて、肝細胞に対するアディポネクチンの結合性を、以下のような放射性リガンド結合試験により行った。まず、Na[125I](2000Cimmol-1、アマシャムファーマシアバイオテック)存在下でIODOビーズによって組換え体アディポネクチンのTyrを125Iで標識した。後記実施例2で得た肝細胞を3.75×105cell/12wellの濃度で植えた。48時間後、125I標識したアディポネクチン(5000カウント/min・ng)と非標識の競合アディポネクチンを含む結合バッファー(氷冷リン酸緩衝生理食塩水(PBS)、0.1%ウシ血清アルブミン)で4℃一晩インキュベートした。その後細胞をウェルあたり1mlの氷冷PBSを用いて20回洗浄し、800μLのPBS中でトリプシン分解した。細胞懸濁液の125I標識を測定した。細胞に結合したものの放射活性は、γ−カウンターを用いて求めた。非特異的結合は、200倍過剰の非標識アディポネクチンを用いて求めた。特異的結合は、合計結合量から非特異的結合量を減じて求めた。3回の測定の平均値として、結果を図26に示す。野生型で見られたアディポネクチンの特異的結合活性は、これらの遺伝子欠損マウスでは低下していた。
試験例1及び2で得たAdipoR1遺伝子やAdipoR2遺伝子欠損の8週齢の雌マウスからコラゲナーゼかん流法により肝細胞を単離した。これを、コラーゲンI被覆12ウェル培養皿に3.75×105ずつ植えた。培地としては、10%ウシ胎仔血清、10nMデキサメタゾン、1nMインスリン、100Unit/mlペニシリン、100μg/mlストレプトマイシンを含むウィリアム培地Eを用いた。5%CO2存在下、37℃で48時間インキュベートしたのち、PBSによって細胞を2度洗浄し、ウシ胎仔血清を含まないウィリアム培地Eによって6時間培養した。

- アディポネクチン受容体1及び/又はアディポネクチン受容体2の機能が欠失した非ヒト動物。
- アディポネクチン受容体1及び/又はアディポネクチン受容体2の機能が、時期特異的及び/又は部位特異的に欠失した非ヒト動物。
- 非ヒト動物がマウスである請求項1又は2記載の動物。
- 請求項1〜3のいずれか1項記載の非ヒト動物由来の、アディポネクチン受容体1及び/又はアディポネクチン受容体2の機能が欠失した細胞。
- 請求項1〜3のいずれか1項記載の非ヒト動物よりなるメタボリックシンドローム、インスリン抵抗性、インスリン分泌不全、糖尿病、動脈硬化症、ミトコンドリア機能低下、酸化ストレス又は高血圧症のモデル動物。
- 請求項1〜3のいずれか1項記載の非ヒト動物に被検物質を投与することを特徴とするアディポネクチン受容体1及び/又はアディポネクチン受容体2作動薬、AMPキナーゼ活性化薬、PPARα活性化薬、インスリン分泌促進薬、グルコキナーゼ作動性糖取り込み促進薬、抗炎症薬、抗酸化ストレス薬、ミトコンドリア機能改善、高血圧症治療薬又はメタボリックシンドローム治療薬のスクリーニング法。
- 請求項1〜3のいずれか1項記載の非ヒト動物に被検食品を投与することを特徴とするメタボリックシンドローム、肥満、糖尿病、動脈硬化、ミトコンドリア機能低下、酸化ストレス又は高血圧症に有効な機能性食品の評価法。
- 被検物質の存在下に請求項4記載の細胞を培養することを特徴とする、アディポネクチン受容体1及び/又はアディポネクチン受容体2作動薬、AMPキナーゼ活性化薬、PPARα活性化薬、インスリン分泌促進薬、グルコキナーゼ作動性糖取り込み促進薬、抗炎症薬、抗酸化ストレス薬、メタボリックシンドローム治療薬、ミトコンドリア機能改善薬又は高血圧症治療薬のスクリーニング法。
