Abstract
Description
- MMP-2 メタロプロテアーゼ;
- MHC Iにより提示されるTエピトープを含む前記メタロプロテアーゼの断片;
- 前記メタロプロテアーゼまたは前記断片をコードするポリヌクレオチド;
から選択される分子の、抗腫瘍免疫療法用、特にMMP-2を発現する黒色腫の治療用の医薬品を製造するための使用である。
このペプチドは、MMP-2を発現するHLA-A*0201黒色腫細胞に関する特異的CTL応答を誘導でき、よってHLA-A*0201患者の治療用の医薬品を得るために特に用いることができる。
例えば、ある特定のMHC I対立遺伝子と複合体を形成し得るペプチドは、共通して、「アンカー残基」とよばれる特異的アミノ酸残基をある位置に有することが知られている。「一次アンカー残基」とよばれるアミノ酸を含む特異的アンカーモチーフは、よって、種々のMHC I対立遺伝子について規定されている。一次アンカーモチーフの外側に位置する残基(二次アンカー残基)が、MHCに対するペプチドの親和性に好ましいかまたは好ましくない影響を及ぼし得ることも示されている。
これらは、特に、複数特異的(polyspecific)なCTL応答を生じ得るマルチエピトープ組成物であることができ、この目的のために、該組成物は1種以上の他の免疫原性エピトープをも含む。これらの他のエピトープは、MMP-2に由来するかまたは1以上の他の抗原に由来することができる。
これらは、さらに、MHC II分子により提示されかつT-ヘルパー応答を誘導し得る少なくとも1種のエピトープをも含み得る。
本発明による抗原提示細胞は、MHC Iを介して抗原を提示し得る全ての細胞から得ることができる。特に、該細胞は、専門の抗原提示細胞、例えば樹状細胞から得ることができる。
これらのペプチドは、これらのCTLの特異的弁別を行うために用いることもできる。このようにして単離されたCTLは、インビトロで増幅され、大量に(10億のオーダーで) 患者に再注入され得る。

CTLクローンを、黒色腫に罹患した患者からの腫瘍浸潤リンパ球(TIL) (HLA A*0201、B*0801、Cw*0701)を、自己由来腫瘍細胞で(Pandolfinoら, 1992, Eur. J. Immunol., 22(7), 1795〜802により記載されたプロトコルに従って)刺激することにより得た。これらのCTLクローンは、自己由来の腫瘍細胞を特異的に溶解し、これらの細胞の刺激に応答してTNFαおよびIFNγを分泌し得る。
CTLクローンM134.12を、M134自己由来系統ならびに種々のHLA A*0201患者からの黒色腫由来の種々の同種異系の系統(M17、M113、M147、M153、M204、FM25、FM29、IPC 277/5、M25、M44、M88、M102、M117、M171、M199、M200)の存在下で共培養した。
Y軸に沿って、pg/mlでのTNFαの濃度である。
X軸に沿って、試験した種々の系統である。
CTLクローンM134.12により認識される抗原を同定するために、cDNAライブラリをM134腫瘍細胞系統のmRNAから構築した。
ポリ(A)+ mRNAを、M134細胞からFast Track 2.0キット(Invitrogen Corp., Oxon, UK)を用いて抽出した。cDNAを、精製mRNAからキット(Stratagene Inc., La Jolla, CA)を用いて合成した。新しく合成されたcDNAをEco RIアダプタに連結し、次いでXho Iで消化し、最後に、真核発現ベクターpcDNA3.1 (Invitrogen Corp.)のEco RIおよびXho I部位に挿入した。得られた組換えプラスミドは、E. coli XL1株(Stratagene Inc., La Jolla, CA)にエレクトロポレーションした。エレクトロポレーションの後、60,000に近いアンピシリン耐性クローンを単離した。これらのクローンのスクリーニングを許容するために、574群(各群は100のアンピシリン耐性細菌クローン)をつくった。各群のプラスミドDNAを、QIAprep Spin Miniprepキット(Qiagen S.A., Courtaboeuf, France)を用いるアルカリ溶解により抽出した。
グルコース1 g/lおよび10%の胎児ウシ血清、抗生物質とL-グルタミンを含有するDMEM (Sigma)培地で培養したCOS-7細胞を、ベクターpHLA-A*0201単独またはM134 cDNAライブラリの群の1つに由来するプラスミドDNAとの組み合わせでトランスフェクションした。トランスフェクションは、DEAE-デキストラン-クロロキンプロトコル(Brichard, Exp. Med. 1993, 1(78): 489〜495)に従って行なった。2×104 COS-7細胞を100 ngのプラスミドpHLA-A*0201でトランスフェクションし、コトランスフェクションのために100 ngのM134ライブラリのDNAでトランスフェクションした。トランスフェクションの48時間後、これらのCOS-7細胞を用いてCTLクローンM134.12 Tを刺激した。共培養の6時間後、培養上清を回収して、そのTNF濃度を、上記のように、WEHI 164クローン13についてその細胞傷害性を測定することにより決定した。
M134細胞系統の存在下で得られたものに匹敵するTNFα分泌を誘導したpNA134-Aとよばれるこれらのプラスミドの1つが選択された。
プラスミドpNA134-Aの挿入断片は、MMP-2のアミノ酸501〜661をコードする。
全MMP-2 cDNAを含むプラスミドも構築した。
これらの結果は、CTLクローンM134.12により認識されるTエピトープがMMP-2のアミノ酸556と593との間に位置することを示す。
Y軸に沿って、相対的溶解のパーセンテージである。
X軸に沿って、nMでのペプチドLGLPPDVQRV (●)、GLPPDVQRV (■)、LPPDVQRV (▲)またはGLPPDVQR (×)の濃度である。
MMP-2が、子宮内膜、肝臓または大動脈のような組織により(Khasigov, Biochemistry, 2001)、またマクロファージ、栄養膜(Yamamoto, Cancer Res., 1996)、IL-2-活性化Tリンパ球(Leppert, JI, 1995 Esparza J., Blood, 1999)、線維芽細胞、ケラチノサイト、軟骨細胞、内皮細胞、単球または骨芽細胞(Birkedal-Hansen, Crit. Rev. Oral. Biol. Med., 1993)のような多数のタイプの細胞により構成的に発現されることが知られている。
一方、他の癌細胞系統および非癌細胞系統はMMP-2およびHLA-A2を発現するが、これらはCTLクローンM134.12により認識されない。
Claims (13)
- - MMP-2 メタロプロテアーゼ;
- MHC Iにより提示されるTエピトープを含む前記メタロプロテアーゼの断片;
- 前記メタロプロテアーゼまたは前記断片をコードするポリヌクレオチド;
から選択される分子の、抗腫瘍免疫療法用の医薬品を製造するための使用。 - MMP-2メタロプロテアーゼの8〜11の連続するアミノ酸の断片からなることを特徴とする、MHC Iにより提示されるTエピトープを構成する免疫原性ペプチド。
- 配列GLPPDVQRVで規定されることを特徴とする請求項2に記載の免疫原性ペプチド。
- 請求項2および3のいずれか1つに記載のペプチドをコードするポリヌクレオチド。
- 請求項2および3のいずれか1つに記載の少なくとも1つのペプチドあるいは請求項4に記載のポリヌクレオチドを含む組成物。
- 請求項2および3のいずれか1つに記載のペプチドあるいは請求項4に記載のポリヌクレオチドの、医薬品を得るための使用。
- 前記医薬品が、黒色腫の治療用である請求項6に記載の使用。
- インビトロで請求項2および3のいずれか1つに記載のペプチドをロードされたことを特徴とする、MHC I分子を発現する単離された抗原提示細胞。
- 請求項2および3のいずれか1つに記載の免疫原性ペプチドをコードする配列を含むポリヌクレオチドでトランスフェクションされたことを特徴とする、MHC I分子を発現する抗原提示細胞。
- MMP-2メタロプロテアーゼまたはその断片の、黒色腫に罹患した個体から得られた生物学的試料中のMMP-2に指向された細胞傷害性Tリンパ球を検出するための使用。
- 黒色腫に罹患した患者から採取した細胞傷害性Tリンパ球から、MMP-2タンパク質またはその断片を認識する細胞傷害性Tリンパ球を選択し、そのようにして選択されたTリンパ球をインビトロで増幅することを含むことを特徴とする、MMP-2メタロプロテアーゼに指向された細胞傷害性Tリンパ球を得る方法。
- 請求項11に記載の方法により得ることができる、MMP-2メタロプロテアーゼに指向された細胞傷害性Tリンパ球の調製物。
- - 請求項2および3のいずれか1つに記載の免疫原性ペプチド;
- 請求項4に記載のポリヌクレオチド;
- 請求項8および9のいずれか1つに記載の抗原提示細胞;
- 請求項12に記載の細胞傷害性Tリンパ球の調製物
から選択される活性成分を含む医薬品。
