Abstract
Description
[Ala1Ψ(CH2NH)Val2、Nle8,18、Aib12、2−Nal23、Tyr34]bPTH(1−34)NH2(配列番号4)、
[Ser1Ψ(CH2NH)Val2、Nle8,18、Aib12、2−Nal23、Tyr34]hPT(1−34)NH2(配列番号42)、
[Ser1Ψ(CH2NH)Val2、Aib12]hPTHrP(1−34)NH2(配列番号43)が挙げられる。
a)モデル樹立
Honoreら,Neuroscience.(2000)98(3):585−98に記載されているようにマウス肉腫細胞を使用して成体C3Hマウスで転移性骨疾患の動物モデルを樹立した(Clohisyら,J Orthop Res.(1996)14(3):396−402も参照)。要約すると、マウスCCL−11(NCTCクローン2472)溶骨性肉腫細胞を遠位大腿骨に移植した。移植部位を歯科用グレードアマルガムで補填した。傷口を外科用クリップで止めた。行動試験を妨げないように6日目に外科用クリップを外した。試験後から開始してビヒクル10μl/kg又は80μg/ml濃度のPTH(1−34)を含有するビヒクル10μl/kgを偽移植マウスと肉腫移植マウスに毎日皮下注射し、肉腫細胞移植後6日目に放射線写真を撮影した。移植後14日目にPTH(1−34)又はビヒクル投与を終了し、最終行動試験を実施し、動物を屠殺した。
骨破壊を測定するために、骨癌誘導前(基線ないしBL)と、骨癌誘導後6、10、及び14日目にFaxitron X線撮影装置(Model MX−20)を使用してX線写真を撮影した。X線照射中にマウスが動かないように軽く麻酔した。マウス大腿骨のX線写真を最大解像度(2400dpi)でスキャンし、ディジタルグレースケールフォーマット(TIFF)で保存した。グレースケールレベルはアマルガム領域を白(グレースケール値255)に設定することにより全X線写真で標準化し、グレースケールの下限はスキャン画像の最も暗い画素に設定した。グレースケール値はスキャン画像の明度に対応する。
Lugerら(2001)により記載されているように自発四肢使用を評価した。マウスを空のマウスパンに入れ、連続動作でパンを横切って歩行する様子を観察した。右(肉腫移植)後肢について、ひきずる動作及び/又はかばう動作を以下の尺度で評価した。0=完全な不使用、1=部分的不使用、2=ひきずる動作とかばう動作、3=ひきずる動作、4=正常歩行。
転移性骨腫瘍動物と偽処置動物の大腿骨のX線写真評価を図1に示す。正常動物にPTH(1−34)を投与すると、14日目に多少の骨密度増加を示した。肉腫をもつ動物にビヒクルを投与すると、14日目に骨密度は著しく低下した。他方、肉腫をもつ動物にPTHを投与すると、14日目にほぼ正常な骨密度を示し、17日目に正常未投与動物よりも密度が増加した。従って、これらのデータによると、PTH(1−34)は骨転移性癌の増殖による骨密度低下を低減する。
Claims (10)
- 有効量を個体に投与すると骨量減少を低減するか、骨折を低減するか、又は疼痛を軽減する薬剤の製造における骨転移癌又は骨原発癌の増殖に付随する症状を改善するためのPTH受容体アゴニストの使用。
- 前記有効量を投与すると疼痛を軽減する請求項1に記載の使用。
- 前記PTH受容体アゴニストがPTH又はそのアナログである請求項1又は2に記載の使用。
- 前記PTH又はそのアナログがPTH(1−34)、好ましくは酢酸テリパラチドである請求項3に記載の使用。
- 前記PTH受容体アゴニストが副甲状腺ホルモン関連蛋白質又はそのアナログである請求項1又は2に記載の使用。
- 前記個体が骨転移癌をもつ請求項1又は2に記載の使用。
- 前記骨転移癌が乳癌、前立腺癌、肺癌、腎臓癌、甲状腺癌又は骨髄腫である請求項6に記載の使用。
- 前記個体が骨原発癌をもつ請求項1又は2に記載の使用。
- 前記骨原発癌が肉腫である請求項1又は2に記載の使用。
- PTH受容体アゴニストの有効量が0.1〜1000μg、好ましくは20〜200μgである請求項1から9のいずれか一項に記載の使用。
