Abstract
Description
本発明は、M.tuberculosisに対する高められたT細胞応答を可能とする免疫診断方法、特にin vitroで行われる免疫診断方法を提供することにより当該技術分野での要求を達成するのを助け、更に詳しくは、本発明は、送達システムとして遺伝子的に解毒されたBordetella sp.のCyaA(Bordetella sp. CyaA)を使用する診断試験及び免疫モニタリングのための新規なシステムを提供する。
実施例に示されたとおり、与えられた抗原により予め刺激されたT細胞は、CyaAに含まれた同じ抗原によりin vitroで再刺激されうる。この発見に基づいて、本発明は、CyaAを含むタンパク質として、M.tuberculosis免疫優勢タンパク質(immunodominant proteins)ESAT−6及びCFP−10並びに他のタンパク質を送達することができる送達システムを提供することによるTBに対する診断試験及び免疫モニタリングを含む。
ウシの研究のための物質及び方法
ウシツベルクリン(PPD−B)及びトリツベルクリン(PPD−A)は、Veterinary Laboratories Agency-WeybridgeにおけるTuberculin Production Unitから得られそして10μg/mlで培養において使用された。組換えESAT−6は、Dr.A.Whelen(VLAWeybridge)により供給され、組換えCFP−10は、Lionex Ltd., Braunschweig, Germanyから得られた。CyaA、CyaA−CFP−10及びCyaA−ESAT−6は、Dr.C.Leclerc, Institute Pasteur, Parisにより提供された。タンパク質の同一バッチを全体にわたり使用した。
末梢血単核細胞(PBMC)を、ヘパリンを加えた血液からHistopaque−1077(Sigma)勾配遠心により単離し、そして組織培養培地(5%CPSR−1(制御されたプロセス血清代替物−1型、Sigma Aldrich, Poole,UK)、非必須アミノ酸(Sigma Aldrich)、5×10-5M2−メルカプトエタノール、100U/mlペニシリン及び100μg/mlストレプトマイシンサルフェート)を補充されたRPMI 1640)(Life Technologies, Paisley, Scotland, U.K.))中で培養した。直接ELISPOTsを以前に記載されたとおりに数えた。簡単に言えば、ELISPOTプレート(Immunobilon−Pポリフッ化ビニリデン膜、Millipore, Molsheim, France)を、ウシIFN−γ特異的モノクローナル抗体2.2.1で4℃にて一晩コーティングした。結合しなかった抗体を洗浄により除去し、そしてウエルをRPMI1640培地中の10%FCSでブロックした。次いで、組織培養培地(5%CPSR−1を補充されたRPMI1640)中に懸濁させたPBMC(2〜5×105/ウエル))を加えそして加湿したインキュベーター中で37℃及び5%CO2で24時間培養した。IFN−γに対して特異的なウサギ血清でスポットを発生させ、次いでウサギIgGに対して特異的なアルカリホスファターゼ−コンジュゲーテッドモノクローナル抗体(Sigma Aldrich)とインキュベーションさせた。モノクローナル抗体2.2.1は、親切にも、Dr.D.Godson、(Veterinary infectious Disease Organization, Saskatoon, SK, Canada)により提供された。スポットをBCIP−NBT基質(Sigma Aldrich)により可視化した。
完全な(entire)マイコバクテリア抗原Cfp10又はEsat−6を有する組換えCyaAの構築及び精製
組換えDNA構築のため及びCyaAに挿入された抗原の発現のために、Escherichia coli XL1−Blue(Stratagene)を使用した。pT7CACT1(Osicka et al., 2000)に由来する適切なプラスミドで形質転換されたバクテリアを、アンピシリン150μg/mlを補充されたLuria−Bertani培地中で37℃にて成育させた。Mycobacterium tuberculosisH37Rv遺伝子esat−6及びcfp−10のオープンリーディングフレームを、下記のプライマー:
実験的に感染したウシのIFN−γ応答
実験的に感染させたウシからPBMCを調製し、そして抗原(組換えESAT−6、CFP−10、CyaA−ESAT−6、CyaA−CFP−10及びCyaAコントロール)の段階希釈物とインキュベーションさせた。抗原で誘発されたIFN−γ応答を、鋭敏なELISPOTアッセイを使用して24時間の培養の後に決定した。抗原を加えないで(培地コントロール)見出されたスポット形成性細胞(SFC)の数を、CyaA−ESAT−6及びCyaA−CFP−10刺激後に誘発されたSFCの数から減じた。データがその後いかに表されそして比較されたかを説明するために、1頭の仔ウシで試験したCFP−10についての代表的結果を図1に示す。この仔ウシでは、CyaA−CFP−10は、組換えCFP−10より高いピーク応答を誘発し(水平線aとbにより示された値の比較により示されたとおり)、そして、組換えタンパク質で誘発された「半最大」(half maximum)(ピーク応答の50%)応答(線c)に必要な濃度を示す垂線d及びeにより示されるとおりより有効に認識された。
CyaA−CFP10の認識はCD11bにより媒介される
CyaA−CFP10の認識がCD11b依存性機構(マウスで最近示されたとおりの)を介して媒介されるかどうかを決定するために、感染した仔ウシからのPBMCを、ウシCD11bに対して特異的な同じアイソタイプ(IgG1)の2つのmAb(親切にも、Dr C.Howard,IAH,Compton,UKにより提供された)の存在下にCyaA−CFP−10で刺激した。これらのmAbの1つ、(ILA15)は、SFCの数が有意に減少したので、CyaA−CFP−10のCD11bとの相互作用を妨害したが、これに対して、非ブロッキングアイソタイプコントロールmAb(CC94)は妨害しなかった(図3、一側ウイルコクスンマッチドペアー検定one-tailed Wilcoxon matched pairs test)により決定して、試験された各濃度についてp<0.02)。これらの結果は、マウス系における如く、CyaAはウシのAPC上のCD11bと相互作用するという証拠を与える。
全血IFN−γ試験におけるCyaA−ESAT−6及びCyaA−CFP−10の成績(BOVIGAMアッセイ)
全血アッセイ(BOVIGAM試験)のフォーマットにおいて現場での診断アッセイとしてIFN−γ試験を適用した。このフォーマットにおいて、農場で集められた血液にヘパリンを加え、それをツベルクリン又は特異的抗原とインキュベーションした。24時間のインキュベーション期間の後に、血漿上清中の抗原で誘発されたIFN−γの量をELISAにより決定した。ESAT−6及びCFP−10とのCyaA融合タンパク質の性能を決定するために、第2バッチの8頭の実験的にM.bovisに感染した仔ウシから血液を得た。これらの血液サンプルを4及び20nMのESAT−6、CFP−10、CyaA−ESAT−6及びCyaA−CFP−10で刺激した。24時間後に行ったELISAアッセイの結果を図4に示す。ELISPOTにより測定されたPBMC応答について上記に示されたとおり、組換えCFP−10タンパク質と比較して有意により強いIFN−γ応答が、両試験濃度でCyaA−CFP−10で観察された(両濃度でp=0.078)。この増加した応答は、血液を4nM濃度の抗原で刺激したとき、特に明白であった(メディアンOD450単位、CyaA−CFP−10:313、CFP−10:105)。CyaA−ESAT−6及びESAT−6の応答は、20nMでは有意に異ならなかったが、4nMでのCyaA−ESAT−6による刺激後に有意に高められた応答が観察された(p=0.015、メディアンOD450単位、CyaA−ESAT−6:486;ESAT−6:260)。
ヒトの研究のための材料及び方法
ヒトの研究は、Harrow Local Research Ethics Committee (Harrow LREC 1645 and 2414)からの倫理的承認を伴って行われた。結核を有する患者及びそれらの健常な接触者は、Northwick Park Hospital, Harrow (North West London Hospitals NHS Trust)から動員された。異なる臨床表現型を有する3グループの人々が選ばれた。第1グループは、明白な(即ち、培養又は生検ポジティブ)結核を有する成人であった(n=21、14M、7F、平均年齢35.1歳)。第2グループは、正常な胸部X線写真を有するがそれにもかかわらず強くポジティブなTST反応(Heaf Grade 3以上)を示し、かくしてLTB1を有していそうだと思われる無症候成人からなっていた(n=44、26M、18F、平均年齢34.7歳)。第3のコントロールグループは、TBへの証明された暴露(documented exposure)を持たず、その皮膚試験反応はネガティブである健常な成人からなっていた(n=7、3M、4F、平均年齢37.6歳)。最初の2つのグループは、M.tuberculosis特異的抗原に対するT細胞反応性の機会を最大にし、かくして組換えトキソイド及びCyaAトキソイドに対する応答の比較を可能とするように選ばれた。すべての対象は、次いでBritish Thoratic Societyガイドラインに従ってアドバイスされそして、必要であれば、処置された(Thorax 55:887-901, 2000)。
M.tuberculosis特異的T細胞を再刺激するのに必要なESAT−6又はCFP−10の用量はCyaA送達により10〜20倍減少する
CyaAトキソイドとして組換え分子中に挿入された抗原の同等な用量(即ち、同じモル量のタンパク質)を使用して、in vitroでのESAT−6及びCFP−10及びそれぞれのCyaAトキソイドの最適刺激用量を決定した。次いでオーバナイトELISPOTアッセイにおいてIFN−γSFCの数を数えた。CyaAトキソイドによるこれらの実権は、抗原性刺激が挿入されていない同じ量のCyaAトキソイド(モックトキソイド)を含有するウエルにおけるIFN−γSFCの数を減じることによりコントロールされた。ESAT−6に対応する9人の健常なTST+veドナーにおいて、この分子の最適認識が500nMの用量で観察された。抗原がCyaA−ESAT−6として提示される場合は、10倍少ないESAT−6(50nM)が必要であった(図5A)。
低く応答する対象におけるIFN−γSFCの検出はCyaA送達により高められる
図5に示された結果に基づいて、500nMrESAT−6及びCyaA−ESAT−6では50nM、及び250nMCFP−10及び25nMCyaA−CFP−10を、同等な効力に基づいて、更なる実験のために選んだ。CyaAによる送達がIFN−γSFCの数を増強させるかどうかを決定するために、より大きいグループの患者及び健常な感作された対象を研究した。患者と健常な感作された対象との間にいかなる刺激に対する応答の頻度又は大きさにも差はなく、それゆえ結果を分析のために組み合わせた。68人のドナーのうち63人はrESAT−6に応答し(>10SFC/106PBMC)(平均IFN−γSFC/百万PBMCは107.7±16.2であった)、そして64人はCyaA−ESAT−6に応答し(107.5±12.9);52人はrCFP−10に応答し(104.4±14.3)そして62人はCyaA−CFP−10に応答した(94.9±11.3)。かくして、組換え体又はトキソイドによるPBMCの刺激の後のIFN−γSFCの頻度間の全体の差は明白ではなかった。しかしながら、CFP−10に応答する対象の割合は76.4から91.1%に増加した。
CD4+及びCD8+応答はCyaA送達により高めることができる
CyaAトキソイドを認識したT細胞サブセットを規定するために、PBMCからのCD4+又はCD8+T細胞を先に免疫磁性枯渇を行うことにより集団を濃縮した(enriched)。残りの細胞をELISPOTアッセイにおいてセットアップし、そしてESAT−6又はCFP−10及び同じ抗原を組み込む解毒したCyaAで一晩刺激した。8人のドナーをCyaA−ESAT−6について試験し、5人のドナーをCyaA−CFP−10について試験し、そして対応する組換え抗原について試験した。CD4+及びCD8+応答の両方共組換え抗原に対して見られ、CD4+が優勢であった(図7)。組換え抗原に対する応答と比較すると、CyaAトキソイドに対する応答は、3つの例でCD4に向けてシフトし、2つの例でCD8に向けてシフトしていた(図7)。残りの8つの場合には正味の変化はなかった。
IFN−γSFCの高められた検出は、MHCを経由する提示のためにプロセッシングされなければならないCyaAへの抗原の共有結合会合(covalent associrtion)を必要とする
M.tuberculosisタンパク質がCyaAに共有結合で連結されなければかどうかを決定するために、ESAT−6をモックCyaAトキソイドと混合することの効果を試験した。4人の対象からのPBMCをESAT−6(500nM)、CyaA−ESAT−6(50nM)又はrESAT−6(500nM)とCyaA(50nM)の混合物でセットアップした。これらの刺激剤についてのメディアンIFN−γSFC/百万は、それぞれ、113、147及び58であり、これは増強が起こるためには、抗原と担体との共有結合連結が必要である(データは示されていない)ということを示している。事実、rESAT−6のCyaAとの単純な混合物は、rESAT−6に対する応答を実際には減少させたかも知れないと思われる。
CyaA−CFP−10に対する応答は、簡単な全血IFN−γ産生アッセイにおいても高められる
全血培養物の上清中に分泌されたIFN−γの検出は、より少ない血液を必要とし、そしてex−vivoIFN−γELISPOTアッセイよりも現場条件に潜在的により多く適用可能である。しかしながら、このような全血アッセイは、特異性を保持しているが、ELISPOT検出よりも感度が低いと思われる。ゆえに、ESAT−6又はCFP−10を有するCyaAトキソイドに対する高められた応答がこの欠陥を補償することができるかどうかを検査した。33人の患者及び健常な感作された対象を、IFN−γ産生について2つのリードアウトアッセイ(read-out assay)を使用して並行して試験した。すべての33人のドナーはrESAT−6に応答し、そして31人のドナーは、rCFP−10に応答した。CyaA−ESAT−6及びCyaA−CFP−10に対するELISPOT及び全血IFN−γ応答は正の相関があった(r=それぞれ、0.58及び0.64、両場合にp<0.001、図9A)。ドナーは、全血アッセイにおいて、遊離抗原に対するそれらの応答に従って、低い(<250pg/mlIFN−γ)、中間(250〜1000pg/mlIFN−γ)又は高い応答者(>1000pg/mlIFN−γ)に層別化される。結果は、ELISPOTアッセイにより見出されたとおり抗原認識に対するCyaA送達の同様な効果を示した。かくして、低く応答する対象では、CyaA−CFP−10の存在下に産生されるIFN−γの量は、遊離rCFP−10の存在下におけるよりは平均して27.7±9.5倍高かった(p=0.021、図9B)。CyaA−ESAT−6に対する応答は同じ一般的傾向を示したが(平均5.6±3.2倍)、この効果は統計的有意には達しなかった。
マイコバクテリアでプライミングされたT細胞のr−CyaA−ESAT−6による特異的及び有効なin vitro刺激
M.tuberculosisのRD1染色体領域を安定に補充された、1×106又は1×107CFUのBCG株(BCG::RD1と呼ばれる)(Pym,2002;Pym,2003)で感染させた(皮下又は静脈内)C57BL/6マウスの脾細胞は、ESAT−6:1〜20ペプチド又はrCyaA−ESAT−6構築物によりin vitro刺激すると十分なレベルのIFN−γを産生した(図10)。産生されたこれらのIFN−γレベルはM.tuberculosisの精製されたタンパク質誘導体(PPD)による刺激後に産生されたIFN−γレベルに匹敵していたことは注目する価値がある。このT細胞応答の特異性は、(i)無関係のMal−E:40〜54ペプチド又はr−CyaA−OVA:257〜264ネガティブコントロールによるこれらの細胞の刺激はIFN−γの放出を誘発しなかった及び(ii)コントロールBCGで感染したマウスの脾臓細胞(BCG::pYUB412)はESAT−6:1〜20又はr−CyaA−ESAT−6によるin vitro刺激後に検出可能なIFN−γを産生しなかったという観察により確立された(図10)。
本発明の使用の範囲を証明するための物質及び方法
マウス。Iffa CREDO (L'Arbresle, France)からの雌のC57BL/6(H−2b)マウスを6〜10週齢で使用した。C57BL/6バックグラウンドへの雌のTAP1ノックアウトマウス(Van kaer, L., et al., 1992)はA.Bandeira(Institut Pasteur, paris, France)からの寄贈品でありそして我々の動物施設において繁殖させた。
MalE及びOVAエピトープを同時に有するCyaAタンパク質をコードするハイブリッドCyaA対立遺伝子を構築するために、pT7CACT1由来のプラスミドの組にコードされそしてそれぞれCD4+MalEエピトープ(Loucka,J., et al)又はCD8+OVAエピトープ(Osicka, R.,et al., 2000)をコードするオリゴヌクレオチドインサートを有するCyaA対立遺伝子の組換えのために、CyaA対立遺伝子に沿った適切な独特の制限部位を使用した。CyaA遺伝子における両オリゴヌクレオチドの挿入及び配向(orientation)は、プラスミドの制限分析により立証され、対応する発現されたCyaAタンパク質の長さは、7.5%SDS−PAGEにより立証された。この研究で使用された組換えCyaAは、アミノ酸108と109の間にNGKLIAYPIAVEALS配列を有し(CyaA−MalE)、アミノ酸336と337の間にSIINFEKL配列を有し(CyaA−OVA)を、又はそれらのそれぞれの挿入部位に両配列を有する(CyaA−MalE−OVA)。すべての構築物を残基188と189の間にジペプチド配列の挿入により遺伝子的に解毒された。
免疫感作されたマウスからの脾臓細胞を、CyaA注射の7日後に単離し、そして同系の照射されたナイーブ脾臓細胞の存在下に、OVA257-264ペプチド(1μg/ml)により5日間in vitro再刺激した。細胞傷害性活性を、先に記載されたように(Fayolle, C., et al., 1996)5時間のin vitro[51Cr]−放出アッセイにおいて決定した。簡単に言えば、OVA257-264ペプチド50μMをロードされたEL4(H−2b)腫瘍細胞をH−2bエフェクター細胞のための標的細胞として使用した。種々のエフェクター対標的比を使用しそしてすべてのアッセイを二重に行った。各アッセイにおいて、ペプチドの不存在下にインキュベーションされたEL−4細胞を、非特異的溶解のためのコントロールとして使用した。各ウエルにおける[51Cr]−放出を、マイクロベータートリラックス液体シンチレーションカウンター(MicroBeta Trilux liquid scintillation Counter)(Wallac, Turku, Finland)を使用して計数した。特異的溶解の百分率は、100×(実験での放出−自然に起こる放出)/(最大放出−自然に起こる放出)として計算された。最大放出は、標的細胞に10%TritonX−405を加えることにより得られ、そして自然に起こる放出はCM中でインキュベーションされた標的細胞により決定された。
CyaA−MalEは、MHCクラスII提示経路へのCD4+T細胞エピトープ送達においてMalEタンパク質より有効である
レポーターとしてMalECD4+T細胞エピトープを使用して、組換えCyaA−MalEは、脾臓細胞のMHCクラスII提示経路にNGKLIAYPIAVEALS MalE100-114ペプチドを送達することが以前に示された(Loucka, J., et al., 2000)。MalEタンパク質に比べてこの送達の有効性を次に評価した。CD11bポジティブである(データは示されていない)BMDCsを使用して、位置108にMalE NGKLIAYPIAVEALS CD4+T細胞エピトープを有するCyaAの提示をMalEタンパク質と比較した。APCsを各タンパク質の段階希釈物とインキュベーションし、そしてそれらの表面におけるI−Ab−NGKLIAYPIAVEALS複合体の出現を、CRMC3、このMHC−ペプチド複合体に対して特異的なCD4+T細胞ハイブリドーマ、によりモニタリングした(Lo-Man, R., et al., 2000)。予想されるとおり(図11A)、CyaA−MalEとインキュベーションされたBMDCsは、CRMC3によるIL−2分泌を有効に刺激した。更に、CyaA−MalEと同じレベルのT細胞ハイブリドーマ刺激に達するためには、100倍高い濃度のMalEタンパク質が必要であった。脾臓細胞で先に示されたとおり(Loucka, J., et al.,2002)、CyaA−MalEとインキュベーションされたBMDCsはまた、CRMC3を刺激することにおいて、遊離MalE100-114ペプチドをロードされたBMDCsよりも10倍有効でもあった。
CyaA−MalE−OVAは、MHCクラスI及びII提示のためのモデルCD4+及びCD8+T細胞エピトープを同時に送達する
先のレポートにおいて、3つの異なるCD8+T細胞エピトープを有するCyaAは、これらのエピトープに対してin vivo保護CTL応答を同時に誘発することが示された(Fayolle, C., et al., 2001)。ゆえに、CyaAは、BMDCsにCD4+及びCD8+T細胞エピトープの両方を送達して、特異的T細胞ハイブリドーマにAg提示することができるかどうかを決定した。ゆえに、CyaA−MalE−OVA、即ち、MalE(クラスII拘束性)及びOVA(クラスI拘束性)エピトープの両方を有する組換えCyaAを、提示アッセイにおいてCyaA−MalE及びCyaA−OVAと比較した。この比較を可能とするために、MalECD4+T細胞エピトープを、CyaA−MalE及びCyaA−MalE−OVAのアミノ酸108と109の間に挿入した。OVACD8+T細胞エピトープを、CyaA−OVA及びCyaA−MalE−OVAのアミノ酸336と337の間に挿入した。BMDCs上のKb−SIINFEKL複合体の存在を検出するために、B3Z、OVA257-267ペプチドに対して特異的なCD8+T細胞ハイブリドーマ(Karttunen., J., et al., 2002)を使用した。図11A及び11Cに示されたとおり、CyaA−MalE−OVAとインキュベーションされたBMDCsは、CRMC3及びB3ZT細胞ハイブリドーマの両方を刺激した。更に、CyaA−MalE−OVAは、MHCクラスII提示経路へのMalE100-114ペプチド送達においてCyaA−MalEと同じく有効であった。CyaA−MalE−OVA又はCyaA−OVAのBMDCsとのインキュベーションの後の、B3Zへの同等なKb−SIINFEKL複合体提示も観察された。これらの結果は、CyaAは、そのACドメインに挿入されたエピトープをMHCI及びII分子に同時に送達すること、及び1つのエピトープの送達の有効性がCyaAのACドメインへの他のエピトープの挿入により影響されないことを確証する。特に、MHCクラスII提示の増強は、OVA及びMalEエピトープの両方を有するCyaAで依然として観察された。
CyaAとBMDCs上のCD11bとの相互作用は、レポーターCD4+T細胞エピトープの送達の増強のために必要である
CyaA送達におけるMHCクラスII拘束性提示の増強は、このタンパク質と、BMDCs上に発現されているそのCD11b受容体(Guermonprez, P., et al., 2001)との特異的相互作用により説明することができる。この仮説を検定するために、BMDCsを最初に10μg/ml抗CD11bmAbs又は同じ濃度のアイソタイプコントロールmAbsとインキュベーションした。図2Aに示されたとおり、APCsと抗CD11bmAbsとの予備インキュベーションは、CyaA−MalE−OVA送達の後のCRMC3へのMalE100-114ペプチドの提示を完全に且つ特異的に妨害した。予想されるとおり、BMDCsとmAbsとの予備インキュベーションは、ハイブリドーマへのMalEタンパク質の提示に影響を与えなかった。抗CD11bとのBMDCsインキュベーションは、B3Zへの遊離OVA257-264ペプチド提示に影響を与えることなく、CyaA−OVA−MalEからのKb−SIINFEKL複合体の発生を阻止することも確証された(図12B)。これらの結果は、CyaAがMHCクラスI及びクラスII経路の両方に送達される高い有効性がCyaA−CD11b特異的相互作用に依存していることを示す。
CyaA−MalE−OVAによるMHCクラスII提示経路へのMalEペプチド送達は、プロテアソーム活性もTAPトランスポーターも必要としない
先の研究は、CD11bとのCyaAの相互作用は、標的細胞の細胞質ゾルへの直接のACドメイントランスロケーションをもたらすことを証明した。このドメインをその後にプロセッシングしてMHCクラスI拘束性提示のためのペプチドを発生させることは、プロテアソームを必要としそしてTAPトランスポーターに依存する(Guermonprez, P., et al., 1999)。ある内因性Agsは、プロテアソーム及びカルパインを必要とする別の経路によるMHCクラスII提示のために細胞質ゾルにおいてプロセッシングされることが報告された(lich., J.D., et al., 2000)。次いで、細胞質ゾル中に放出されたペプチドは、あまり理解されていない機構に沿ってエンドサイトーシス区画に輸送される。ゆえに、CRMC3へのMHCクラスII拘束性MalE100-114ペプチド提示のためのCyaA−MalE−OVAのプロテアソーム要求を試験した。BMDCsを、ラクタシスチン、20Sプロテアソーム阻害剤(fenteany, G.,et al., 1995; Craiu, A., et al., 1997)と1時間インキュベーションし、次いでAgsを加えた。
MHCII提示のためのCyaA−MalE−OVAプロセッシングは、エンドソームプロテアーゼ及び液胞酸性化を必要とする
外因性可溶性Agのインターナリゼーション後に、MHCクラスII提示のためのペプチドリガンドは、逐次に活性化されるプロテアーゼの組によるタンパク質のタンパク質分解によりエンドソーム及びリソソームにおいて発生される(Villadangos, J. 2001)。CyaA−MalE−OVA送達後にMalE100-114ペプチド提示を発生させるのにカテプシン活性が必要であるので、他のエンドサイトーシスプロテアーゼがI−Ab−MalE100-114複合体形成のために必要であるかどうかを試験した。ロイペプチン(Umezawa, H. 1976)、セリン及びシステインプロテアーゼの阻害剤は、CyaA−MalE−OVAがAgとして使用されるとき、CRMC3へのMalE100-114ペプチド提示を完全にブロックしたが、遊離ペプチドの提示に影響を与えなかった(図14A)。セリンプロテアーゼはMalEエピトープのN−末端を発生させるのに実際に必要でありうることが言及されるべきである。ペプスタチン(Umezawa, H. 1976; Mizuochi, T., et al., 1994)、アスパラギン酸プロテアーゼの阻害剤、も、CyaA−MalE−OVA送達後のMalE100-114ペプチド提示を部分的に阻害した。遊離MalE100-114ペプチド提示は、薬物の存在下に影響を受けないままであった。これらの結果は、エンドサイトーシスプロテアーゼがMHCクラスIIペプチド発生のためのACドメイン分解に関与していることを示す。ゆえに、これらの結果は、CyaAACドメインが古典的エンドサイトーシス経路に達してプロテアーゼによりプロセッシングされることを示す。
CyaA−MalE−OVAによるMalEエピトープ送達は、ブレフェルジンAによるゴルジ障害及びシクロヘキシミドによるタンパク質合成阻害に対して感受性である
APC表面におけるMHCクラスIIペプチド複合体の提示は、外因性Agの分解を必要とし、しかも発生したペプチドのMHCクラスII分子との会合も必要とする(Gordon, S.V., et al., 1999)。古典的エンドサイトーシス経路では、新たに合成されたMHCクラスII分子が必要である。これらの分子は、ゴルジをとおってERを去りそしてトランスゴルジ網(TGN)に達し、それによりそれらはエンドサイトーシス経路に向けて送られる。
CyaA−MalE−OVAによるMalEエピトープ送達は、アクチンフィラメント重合に依存しないがクラスリン被覆小孔を必要とする
CyaAのインターナリゼーション及びそのACドメインに挿入されたOVAペプチドのその後のMHCクラスI拘束性提示は、ファゴサイトーシスに依存しないことが既に示されている(Guermonprez,P., et al., 2000a, b)。しかしながら、CyaAのいくらかの分子は、他の分子が古典的外因性Agとして捕捉されそしてプロセッシングされてMHCクラスII拘束性ペプチドを生じようと、そのACドメインをAPCs細胞質中にトランスロケーションさせることは排除できない。アクチン依存性捕捉が、有効なMHCクラスII拘束性提示のためのMalEエピトープ送達に関係しているかどうかを最初に試験した。この実験では、サイトカラシンB(CCB)、アクチンフィラメント重合を阻止しそしてマクロピノサイトーシス、ファゴサイトーシス及びカベオレ媒介エンドサイトーシスを害する薬物(Gottlieb, T.A.., et al., 1993)、を使用した。図16に示されたとおり、CCBは、CyaA−MalE−OVA送達後のMalE又はOVA257-264ペプチドのそれぞれの特異的T細胞ハイブリドーマへのMalEの提示もOVA257-264ペプチドの提示も阻害しなかった。予測されるとおり、MalEタンパク質の提示は阻害剤により完全に妨害されたが、これに対して遊離MalE100-114ペプチドは依然としてCRMC3に提示された。これらの結果は、MHCクラスI及びMHCクラスII分子へのACドメイン送達が、BMDCsによるCyaAファゴサイトーシス、マクロピノサイトーシス又はカベオレ媒介エンドサイトーシスを必要としないことを示す。
CyaA−MalE−OVAは、in vivoでのOVA特異的CD8+T細胞応答及びMalE特異的CD4+T細胞応答を誘発する
種々のCD8+T細胞エピトープに対するCTL応答(Fayolle, C., et al. 2001)及びMalECD4+T細胞エピトープに対する増殖応答(Loucka, J., et al., 2002)を誘発するためのCyaAの高い有効性は既に証明されている。CyaAがAg提示のためにBMDCsにCD4+及びCD8+T細胞エピトープの両方を送達するための効力のあるビークルであることを証明するin vitro研究の後、これらのエピトープの同時のin vivo送達におけるCyaA−MalE−OVAの有効性を試験した。マウスを、アジュバントなしで、CyaA−MalE、CyaA−OVA、CyaA−MalE−OVA、又はCyaAE5、50μgにより静脈内経路により免疫感作させた。T細胞応答を注射の7日後にモニタリングした。

Claims (45)
- 哺乳動物の疾患をin vitro診断又は免疫モニタリングする方法又はいかなる他のT細胞応答も免疫モニタリングする方法であって、
(A)
(1)Bordetella CyaA又はその断片、及び
(2)前記哺乳動物のT細胞を以前に刺激した疑いのある抗原のペプチド、
を含む組換えタンパク質に前記動物のT細胞を曝露し、そして
(B)T細胞の活性化の変化を検出する、
ことを含む方法。 - 組換えタンパク質が1種以上のペプチドを含む、請求項1に記載の診断又は免疫モニタリング方法。
- 組換えタンパク質が1種以上の抗原を含む、請求項1に記載の診断又は免疫モニタリング方法。
- Bordetella CyaAが、Bordetella pertussis、Bordetella parapertussis又はBordetella bronchisepticaに由来する、請求項1〜3のいずれか1項に記載の診断又は免疫モニタリング方法。
- Bordetella CyaA又はその断片が解毒されている、請求項1〜4のいずれか1項に記載の診断又は免疫モニタリング方法。
- CyaA及び組換えタンパク質のペプチドが遺伝子的に融合されているか又は化学的に結合している、請求項1〜5のいずれか1項に記載の診断又は免疫モニタリング方法。
- 疾患が非ヒト動物の疾患である、請求項1〜6のいずれか1項に記載の診断又は免疫モニタリング方法。
- 非ヒト動物が雌ウシである、請求項7に記載の診断又は免疫モニタリング方法。
- 疾患がヒトの疾患である、請求項1〜6のいずれか1項に記載の診断又は免疫モニタリング方法。
- 疾患が結核である、請求項9に記載の診断又は免疫モニタリング方法。
- 組換えタンパク質が融合タンパク質である、請求項10に記載の診断又は免疫モニタリング方法。
- 融合タンパク質がCyaA−ESAT−6又はCyaA−CFP10である、請求項11に記載の診断又は免疫モニタリング方法。
- 疾患がメラノーマである、請求項1〜9のいずれか1項に記載の診断又は免疫モニタリング方法。
- 抗原が、感染性物質、アレルゲン又はがん細胞からの抗原である、請求項1〜9のいずれか1項に記載の診断又は免疫モニタリング方法。
- ワクチン接種された個体又は動物の免疫モニタリングのために使用する、請求項1〜14のいずれか1項に記載の方法。
- 組換えタンパク質がBordetella CyaAの断片を含む、請求項1〜15のいずれか1項に記載の診断又は免疫モニタリング方法。
- 組換えタンパク質が、Bordetella CyaAにおける任意の許容的部位に局在する、以前に前記細胞を刺激したことがありうる抗原のペプチドを含む、請求項1〜16のいずれか1項に記載の診断又は免疫モニタリング方法。
- T細胞の活性化の変化が、IL−2、IL−4、IL−5又はIFN−γ産生の変化である、請求項1〜17のいずれか1項に記載の診断又は免疫モニタリング方法。
- 試験サンプルが末梢血単核細胞(PBMC)、全血又は全血の画分である、請求項1〜18のいずれか1項に記載の診断又は免疫モニタリング方法。
- T細胞活性化の変化の検出が、ELISPOTアッセイ、ELISA又はT細胞活性化を検出するための他のアッセイにより達成される、請求項1〜19のいずれか1項に記載の診断又は免疫モニタリング方法。
- 哺乳動物における疾患の診断試験用又は、疾患により生じたT細胞応答を含む、哺乳動物におけるT細胞応答を免疫モニタリングするためのキットであって、
(A)
(1)Bordetella CyaA又はその断片、及び
(2)前記哺乳動物のT細胞を以前に刺激した疑いのある抗原のペプチド、
を含む組換えタンパク質、及び
(B)T細胞の活性化の変化を検出するための試薬、
を含むキット。 - 組換えタンパク質が1種以上のペプチドを含む、請求項21に記載のキット。
- 組換えタンパク質が1種以上の抗原を含む、請求項21に記載のキット。
- Bordetella CyaAが、Bordetella pertussis、Bordetella parapertussis又はBordetella bronchisepticaに由来する、請求項21〜23のいずれか1項に記載のキット。
