Tm=81.5C+16.6(LogM)+0.41(%GC)−0.61(%form)−500/L
式中、Mは一価のカチオンのモル濃度であり、%GCはDNA中のGおよびCヌクレオチドのパーセンテージであり、%formはハイブリダイゼーション溶液中のホルムアミドの割合であり、Lは塩基対のハイブリッドの長さである。100%同一ハイブリッドに対して計算したものよりTmが減少された各1Cについて、許容されるミスマッチの量は、約1%まで増加する。したがって、特定の塩およびホルムアミド濃度での、任意の所定のハイブリダイゼーション実験に対して使用したTmが、Meinkothの等式にしたがって、100%ハイブリッドについて計算したTmよりも10C低い場合、約10%までのミスマッチが存在する場合でさえも、ハイブリッド形成が起こる。
ここで前記記述とともに、非限定様式にて本発明を例示する以下の実施例に対し、参考文献を作製する。
CXCR4を、HIV−由来レンチウイルス遺伝子伝達系を用いて、ヒトCBおよびMPB CD34+濃縮細胞上で過剰発現させた。形質導入細胞を、CXCR4表面発現に関して解析した。
ヒト細胞−臍帯血(CB)細胞および成人移行末梢血(MPB)細胞を、インフォームドコンセントの後に得た。CD34+細胞濃縮を、先に記述された[Kollet(2001)Blood 97:3283-3291]ように、磁気ビーズ分離を用いて実施した。CXCR4発現を、精製した抗ヒトCXCR4(クローン12G5、R&D、ミネアポリス、MN)およびヤギ抗マウスIgG FITCの二次F(ab’)2断片(ジャクソン(Jackson)、ウエストグローブ、PA)を用いた、フローサイトメトリーによって促進した。
幹細胞および前駆細胞は、適切なサイトカインカクテルを供した場合に、骨髄および赤血球系統にインビトロで多系統分化が可能である(cancer research1977 vol61 ページ1でのMetcalf dの最近の結果)。したがって、本発明者らは、分化した系統における、形質導入細胞の、インビトロでの分化する能力に対する、トランスジーン発現の効果を評価した。
CFUアッセイ−エクソビボ培養中の形質導入後の、ヒト前駆細胞のレベルならびにトランスジーン発現の維持を検出するために、半固体培養を、先に記述された52ように実施した。簡単に記すと、CB CD34+形質導入細胞(3×103細胞/ml)を、0.9% メチルセルロース(シグマ)、30%FCS、5×10-5M 2ME、50ng/ml SCF、5ng/ml IL−3、5ng/ml GM−CSF(R&D)、および2u/ml エリスロポエチン(オルソ バイオ テック(Orto Bio Tech)、Don Mills,Canada)中にプレートした。培養物を、5%CO2を含む加湿雰囲気中、37℃にてインキュベートし、14日後、GFP+に関しては、位相差顕微鏡を用いて、ならびに形態基準を用いて、骨髄または赤血球コロニーを記録した。
ケモカインは、細胞骨格配置、とりわけアクチン重合化を導く細胞内事象のカスケードを活性化することによって、細胞運動性を誘導する54。挿入したレセプターの機能性を決定するための方法として、本発明者らは、運動性機構のSDF−1誘導活性化に対するCXCR4過剰発現の効果を試験した。
アクチン重合化アッセイ−形質導入細胞を、示した時間、37℃にて、血清−不含RPMI中で、SDF−1α(300ng/ml、ペプロテック(Peprotech)、Rocky Hill、NJ)で刺激した。反応を、RTにて10分間、3容量の3.7%パラホルムアルデヒドを加えることによって停止させ、続いて、PBSで洗浄し、0.1% Triton−Hepes(20mM Hepes、300mMサッカロース、50mM NaCl、3mM MgCl2、0.1% Triton)で、2分間、氷上にて透過化処理をした。ついで細胞を、RTにて30分間、FITC−Phalloidin(2mg/ml、シグマ)で染色し、洗浄し、フローサイトメトリーによって解析した。
CD34+細胞上のCXCR4の過剰発現が、低SDF−1濃度に対して、それらをより応答しやすくしうることが仮定された。これを検証するために、形質導入細胞のインビトロ遊走を、異なるSDF−1濃度に対して実施した。さらに、サイトカインとの相乗効果での低濃度のケモカインSDF−1が、ヒトCD34+細胞の増殖、ならびにヒトおよびマウス両方の前駆細胞の生存を増強することが示された25-28。したがって、CXCR4過剰発現細胞の増殖における、低濃度でのSDF−1の効果をモニタした。
遊走アッセイ−先に記述したのと同様(12)。簡単に記すと、10ng/mlまたは125ng/mlのいずれかのSDF−1αを含む、10% FCS含有RPMI(600μl)を、Costar 24−ウェルトランスウェル(コーニング(ポアサイズ5μm)、NY)の下部チャンバーに加えた。100μlの培地中の1×105形質導入CD34+細胞を上部チャンバーにのせ、37℃にて4時間遊走をさせた。遊走している細胞を下部チャンバーより回収し、FACSCaliburを用いて、30秒間計数した。対照自発遊走は、SDF−1αなしで下部チャンバーで実施した。
SDF−1は高濃度(1μg/mlおよびそれ以上)で最終的に細胞表面へ再循環されうる細胞表面CXCR4分子のエンドサイトーシスを介した脱感作および内部化を誘導することが記述されてきた55。したがって、本発明者らは、CXCR4を過剰発現している細胞における高SDF−1濃度の効果を試験した。
CXCR4細胞表面発現−CXCR4−形質導入CB CD34+細胞を、1μg/ml SDF−1とともに一晩インキュベートした。細胞表面CXCR4発現を、細胞を抗ヒトCXCR4−PE(12G5、BD ファーミンゲン、サンディエゴ、CA)で標識することにより特定し、フローサイトメトリー(FACSCalibur、ベクトン ディッキンソン(BD)、サンジョゼ、CA)によって解析した。
ヒトCB CD34+細胞のSDF−1/CXCR4−依存生着におけるCXCR4過剰発現の効果を評価するために、形質導入前駆細胞をNOD/SCIDマウスに移植した。
マウス NOD/LtSz−Prkdcscid(NOD/SCID)マウスを、先に18に記述したように、交配し、維持した。すべての実験はワイズマン インスティチュート(Weizmann Institute)の動物ケア委員会によって許可された。8〜10週齢のマウスに亜致死的放射をし(375cGy、60Co供給源より)、放射24時間後に、示した[2×105細胞/マウス(生着)および5×105細胞/マウス(ホーミング)]ように、ヒト細胞を移植した。
最初の一組の実験で、本発明者らは、対照GFP形質導入臍帯血CD34+細胞、またはCXCR4過剰発現CD43+細胞のいずれかに対して、CXCR4の最初のN−末端細胞外ドメイン内に局在する(図7)、ヒトCXCR4−a位の残基22〜25に特異的なmAb 6H8の結合を解析した。本発明者らは、内因性CXCR4のみを発現しているGFP(対照)形質導入CB CD34+細胞が、ヒトCXCR4の第二細胞外ドメインに結合するmAb 12G5に結合する(データは示していない)にもかかわらず、mAb 6H8に結合せず、一方でCXCR4過剰発現CB CD34+細胞は、抗体12G5および6H8の両方で陽性に染色されたことを発見した。これらのデータは、CXCR4の過剰発現が、CXCR4走化性機能において部分的に役割を果たすことが示された6H8エピトープの変質または開裂を妨げることを示唆している(Brelot et al, J Biol.Chem.275:23736-23744)。

Claims (40)
- 多量の未熟な初期前駆細胞を有する幹細胞を含む細胞の集団を製造するための方法であって、幹細胞を回収し、その細胞内に、CXCR4の配列を含むDNA断片を導入することを含む方法。
- 細胞の集団が、低濃度のSDF−1に応答してCXCR4シグナル伝達の改善を示す請求項1記載の方法。
- 細胞の集団が、高濃度のSDF−1に応答してCXCR4シグナル伝達の改善を示す請求項1記載の方法。
- 幹細胞が、造血幹細胞である請求項1記載の方法。
- 造血幹細胞が、CD34+濃縮されている請求項4記載の方法。
- 未熟な初期前駆細胞が、CD34+/CD38-/low系統のものである請求項1記載の方法。
- 幹細胞を回収することが、幹細胞の移行手法を誘導した後に実行され、および/または手術手法を含む請求項1記載の方法。
- 所定の閾値以上のCXCR4レベルを有する幹細胞をFACSにより単離することをさらに含む請求項1記載の方法。
- 幹細胞が、脊髄および赤血球系統へ分化可能である請求項1記載の方法。
- CD34+/CD38-/low系統の未熟な初期前駆細胞の量が、集団の約1〜5%である請求項6記載の方法。
- CD34+/CD38-/low系統の未熟な初期前駆細胞が、集団の約3%以上の量である請求項6記載の方法。
- 低濃度のSDF−1が、約50ng/ml以下である請求項2記載の方法。
- 改善されたシグナル伝達が、低濃度のSDF−1によって仲介される細胞遊走の増強として現れる請求項2または12記載の方法。
- 改善されたシグナル伝達が、低濃度のSDF−1によって仲介される細胞増殖の増強として現れる請求項2または12記載の方法。
- 高濃度のSDF−1が、約1マイクログラム/ml以上である請求項3記載の方法。
- 改善されたシグナル伝達が、SDF−1による脱感作の減少によって現れる請求項3または15記載の方法。
- 幹細胞内にCXCR4の配列を含むDNA断片を導入することによって製造され、多量の未熟な初期前駆細胞を発現し、低濃度および/または高濃度のSDF−1に応答して改善されたCXCR4シグナル伝達能力を示す幹細胞を含む集団。
- 幹細胞が、造血幹細胞である請求項17記載の幹細胞の集団。
- 脊髄および赤血球系統へ分化可能である請求項17または18記載の細胞の集団。
- 造血幹細胞が、CD34+造血幹細胞である請求項19記載の細胞の集団。
- 未熟な初期前駆細胞が、CD34+/CD38-/low系統のものである請求項17記載の細胞の集団。
- 多量のCD34+/CD38-/lowが、集団の約1〜5%である請求項21記載の細胞の集団。
- 多量のCD34+/CD38-/lowが、集団の約3%以上である請求項21記載の細胞の集団。
- 低濃度のSDF−1が、約50ng/ml以下である請求項17記載の細胞の集団。
- 高濃度のSDF−1が、約1マイクログラム/ml以上である請求項17記載の細胞の集団。
- 必要とする対象の標的組織に対する幹細胞のホーミングを増加させるための医薬の製造における請求項17〜25のいずれか記載の細胞の集団の使用。
- 必要とする対象の標的組織の再増殖を増加させるための医薬の製造における請求項17〜25のいずれか記載の細胞の集団の使用。
- 前記標的組織が、骨髄、血管、心臓、肺、肝臓、膵臓、腎臓、神経系、皮膚、骨および骨格筋からなる群より選択される請求項26または27記載の細胞の集団の使用。
- 移植を促進するための請求項26または27記載の細胞の集団の使用。
- 移植が化学治療プロトコールにしたがう請求項29記載の細胞の集団の使用。
- 移植が自家移植である請求項29記載の細胞の集団の使用。
- 移植が、自系細胞の移行を伴う請求項31記載の細胞の集団の使用。
- 移植が異種移植である請求項29記載の細胞の集団の使用。
- 移植が移行幹細胞で実施される請求項29に記載の細胞の集団の使用。
- 細胞または組織置換が必要な対象における疾患の治療方法であって、それを必要とする対象に、治療的に有効量の請求項17〜25のいずれか記載の細胞の集団を提供することを含む方法。
- インタクトな6H8エピトープを含むCXCR4を発現している幹細胞を含む、細胞の集団を製造する方法であって、幹細胞を回収すること、および細胞にCXCR4の配列を含むDNA断片を導入することを含む方法。
- CXCR4の配列を含むDNA断片を導入することによって製造される、インタクトなCXCR4 6H8エピトープを含む幹細胞を含む細胞の集団。
- 必要とする対象における移植のための医薬の製造における請求項37記載の細胞の集団の使用。
- 細胞または組織置換を必要とする疾病を治療する方法であって、それを必要とする対象に、治療的に有効量の請求項37記載の細胞の集団を提供することを含む方法。
