Abstract

Description
図1〜3および5では、すでに述べた通り、本発明の光学アッセイ装置11は使用者の利益となる任意の検体の検出に使用できる。例えば、検体52は、有機物でも無機物でも可能である。有機物の検体52は、たとえば、バクテリア、ウィルス、胞子などの生きている病原体または死んでいる病原体、あるいは毒素、小分子、またはタンパク質のような他の生化学化合物または有機化合物が挙げられる。無機物の検体52は、例えば、金属などの化学元素、あるいは注目すべき重要な無機化合物等がある。
図1〜3で示すように、カップ10は土台12と側壁14からなる。側壁14は、内外の光学表面32,34を持つ導波管光導波管の部分28、内外の光反射面31a、31bのある反射面部分30、内外面31e、31dのあるレンズサポート部分31cから成り、レンズサポート31cは、反射面30と近位端36の間に延びている。
図3がよく示しているように、液体密封シール17は、カップ16とカップ10の側壁14の遠位端22の間に設置され、例えば試液55などのような、カップ10に入れられたあらゆる液体の漏れを防ぐ。シール17は、ガスケットのようなあらゆる適切な構造が可能である。あるいは、分離したシール17を削除し、たとえば、カバー16と遠位端22の間の液体密封を備えたり、あるいは、カバー16と遠位端22の間に、レーザージョイニングや超音波溶接などで液体密封ジョイントを備えるなど、望ましいシールを適切な方法で備えることができる。
反射体30の長さは、側壁14の近位端36と導波管28の間に部分的にまたは完全に伸びる。反射体30は、内外表面31aと31bの間に横たわる対称の光学表面38を持っている。内外表面31a、31bは、近位端36から導波管28に移動する際、各々の長さの部分または全体に沿って、対称の光学表面38に向かって収束する。内外表面31a、31bは両方とも、収束するものとして図示されているが、そのうちの一つだけが収束することもあり、内外表面31a、31b両方が収束する場合は、個々の長さの、部分または全てに沿って同量で収束することはない。反射体30は、表面31aか表面31bのいずれか一方の光学的な反射表面を含む。
カップ10(および、関連する光源26と検出器60)は、あらゆる方法で、ここで説明されているあらゆるアッセイと共に、またカップ10のA軸のあらゆる適切な方向に使用される。
使用中、カップ10内の必要な液体を回転させることは望ましい。なぜなら、そうした液体の回転は、例えば、液体およびその構成要素に、カップ10内にすでにある液体またはその他の物質との反応、導波管28の内表面32との反応、あるいは導波管28の内表面32上の検出コーティング50(例:検出層51a、51b、51c)との反応する機会を提供するからである(図5参照)。例えば液体が試液55の場合、液体が循環していない場合と比較すると、液体の循環は、試液55内のあらゆる検体52が、検出コーティング50の捕獲レイヤー51bと反応するための機会を提供するのに役立っている。
本明細書で説明される多くのアッセイでの目標の一つは、アッセイ内で使用される試薬の中には高価なものもあることから、カップ10内で使用される液体の量を減らすことにある。カップ10内の薄い液体レイヤー51cは、アッセイを実行する際に求められる高価な試薬の量を効果的に減らすことから、この目標を達成する助けになるものとして有利に使用される。
図1ないし13に移り、光源26は、ここで開示されたカップ10、210、210a、310、410と共に使用され、例えばレーザー、発光ダイオード(LED)、蛍光、あるいは白熱光などの検査光24のための発光体27を含む。光源26から放たれた検査光24は、1つ以上の波長を含み、干渉性または非干渉性である。
カップ10の側壁14における反射体30の内外表面31a、31bのいずれか、または両方は、光源26から反射された検査光24のうち、少なくともいくつかの光線が、反射体30の対称の光学表面38に応じて予め厳密に決められた角度内で導波管28に入射するように選択された各々のプロファイルを持っている。個々の表面プロファイル31a、31bは、非球面であることも非球面でないこともあり、また同一であることもないこともある。
図2、3、5に移って、試液55中のあらゆる特定な種類の検体52用の適切な検出コーティング50は、導波管28の内表面32の1部分または全部の上に適切に提供される。あるいは、導波管28の内表面32の部分または全部の上には検出コーティング50はない。
図1ないし3および5を再び参照すると、本発明を使うためには、特定な種類の検体52を含む液体(例えば、試液55、試薬、またはその他の液体)が、カップ10に加えられる。カバー16は、カップ10上に適宜確保されるものと仮定される。
本明細書で説明されている検出器60(図1ないし3)、60a(図17)、60b(図18)、60c(図18a)、460(図6)は、互いに同一であるか、各土台、位置、量(数)、サイズ、形、デザイン、素材、組成、構造、製造、物理特性、次元、仕様、バリエーション、操作、方法、および用途に関して、この開示によって明らかにする相違点を除き少なくとも互いに類似していることが理解される。検出器60、60a、60b、60c、および460はそのいずれもが、本明細書中で開示されたカップ10、210、210a、310、410と共に使用できる。
本明細書でいう「指標剤」(例えば指標剤77)とは、エバネセント光または暗視野照明検査光24a、24bによって検査される場合に、検体52の、存在、量、数、または少なくとも一つの標的識別特性に応じて放たれる、または放たれない信号光58のあらゆる特性に基づき検出される適切な任意の物質を意味する。指標剤77には、例えば、あらゆる適切な要素、分子部分、ナノ粒子、ナノスフェア、ナノロッド、液晶分子、着色されたマイクロ球、または色が変化する微生物(ウイルスやバクテリア属等)などの要素が含まれ得る。
図1ないし3および5を参照してみると、光学アッセイカップ10は、検出コーティング50を含んでいる。検出コーティング50は、検体52(または、図5にあるように、検体52のための任意の指標剤77でも)は、例えば、導波管28の内表面32近く、またはその上に位置する状況では、除外されることがある。検体52(またはそれら分析物ための特定な指標剤77)が内表面32近くまたはその上に位置している場合、それらは、試液55、空気、水、または任意の適当な試薬などの、カップ10中の液体によって、全体的にあるいは部分的に取り囲まれているか、覆われている。簡潔にするために、検出コーティング50を含むカップ10について少し詳しく述べることにする。何故なら、本明細書で開示されるすべてを考慮すれば、本発明対象とする技術について通常の技量を持つ者は、必要に応じて同じまたは類似する所感を検出コーティング50を含まないカップ10に応用することができ、かつそのようなカップ10をカップ10の内部ボリューム68内にあるあらゆる適切な検体52を検出するのに簡単に応用できるからである。
図5を参照すると、エバネセント検査のためのエバネセント検査光24aは、検査光24のための反射角0iを、求めるエバネセント電界56を生成するために使われるセンサー導波管29の選択された内表面64、66、67または70の臨界角で、またはその近辺で、しかし超過しないよう適切な方法で選択することにより生成される。
本発明に応じた暗視野照明による検査がセンサー導波管29と共に利用される場合、エバネセント検査と関連した問題の多くは軽減または排除される。
また、再び図1ないし3およびと5を参照すると、一般に、試液55に存在する可能性のある特定な種類の検体52を検出するためにカップ10の使用者が採用する特定の方法は、検出コーティング50を含み得る各レイヤー51a、50b、51cの数、厚み、組成、および構成などの検出コーティング50の望ましい特性を決定する。
標識位置78(例:液体レイヤー51c)を含むフィルムで拡散支配の拡大では、無次元アッセイ反応時間は、アッセイ反応時間とレイヤー51c内の標識剤77の拡散係数の積を、レイヤー51cの厚みの2乗で割ったものである。標識位置78が、無次元アッセイ反応時間の平方根として、拡大し、無次元アッセイ反応時間が約1.0に等しくなると、拡大に変化が起こることが図5からわかる。その時点で、標識剤77の拡散プロセスは三次元の球形の拡張から二次元での横に向かった拡張に変わる。これはサイズにおいて十分な培養時間が与えられていて、そうした秒または分単位の培養時間は、レイヤー51cの厚みやレイヤー51c内の指標剤77の拡散係数により変わるという場合に、指標剤77の点源さえ、濃縮した標識剤77の球形またはディスク型の、サイズにおいてレイヤー51cの厚みと比較できる、標識位置78を作成する。例として、レイヤー51cの厚さが12ミクロンで、指標剤77のレイヤー51cにおける拡散係数が1x10−7cm2/secの場合、1.0の無次元時間に到達するために必要な経過時間は、わずか14.4秒である。この拡散係数は水または水性のゲルのための典型的な小分子拡散係数である。
本明細書中で考察されてきた通り、高密度の検体52は、スピニングカップ10によって得られる遠心力を用いて捕獲レイヤー51bに向かって、最終的にはその上に、遠心濃縮することができる。さらにまた、同様に考察されてきた通り、検体52は、捕獲レイヤー51bによる検体52の親和性精製によって捕獲レイヤー51b近くに、あるいはその上に、またはその内に濃縮され得る。
本発明に関連して検出アッセイとして適切な任意の競合アッセイを利用できる。競合アッセイは、例えば、カップ10の試液55は、検出されるべき特定の種類でラベル付けされていない検体52である。蛍光化合物など適切な指標剤77を使って適切な方法でラベルを付されている(例えば、最初の試薬培養過程などで)溶液を含んでいるラベルの付された検体52を、カップ10の試液55に加える。2番目の試薬培養過程中に、指標剤77でラベル付けされた検体52は、これにより、以前に準備された獲得レイヤー51b上の抗体結合サイト用ラベルの付いていない検体52と比較できることになる。
本発明に関連した検出アッセイとして、適切な任意のサンドイッチ式免疫検定を使用することができる。サンドイッチ式の免疫検定とは、例えば、カップ10中の試液55の初期の培養によって、特定な種類の検体52が以前に準備された獲得レイヤー51b上に備えられたあらゆる表面捕獲サイトによって捕捉できる(例えば、結合する)ようなものである。洗浄過程の後、捕獲検体52は、妥当な標識剤77を使用して適切にラベル付けするなどの適切な方法で、光学的に検出できるものとして提供される。カップ10はそこでエバネセント光か暗視野照明を使用した検査を受け、信号光58は、獲得検体52の存在、量、数、または少なくとも1つの標的識別特性に応じて、導波管28の外表面34から発せされる。
本発明に関連した検出アッセイとしてあらゆる適切なELISAが利用される。ELISAは、例えば、試液55中の特定な種類の検体52が、以前に準備されている獲得レイヤー51bを使用しているカップ10内の表面の捕捉場所に結合している場合、初期の捕獲培養手順として関与する。捕獲検体52はその後、活性酵素を、最初の試薬培養手順で付着させるといった適切な任意の方法で、化学的に活性化させられる。2番目の試薬培養手順では、2番目の試薬に最初に存在していた特定な種類の分子上での検体52の活性酵素の活動によるなどの適切な方法で、蛍光化合物などの適切な任意の標識剤77が形成される。
本発明に関連する分析アッセイとして、カップ10の試液55に存在する検体52に対し、進行中のすべての代謝活性を検出する適切な任意の方法が利用できる。例えば、特定な種類の検体52は、捕獲培養手順中に、事前に準備された獲得レイヤー上の適切な表面捕獲に最初に結合する場合がある。捕獲検体52は、その後、捕獲検体52または任意の代謝産物を、染料など適当な代謝指標剤77で染色するなど、適切な方法や、適切な代謝指標剤77を使用して検出される。染料は、例えば、捕獲検体52の代謝生産物の1つ以上を蛍光性に、または着色し、および捕獲検体52が、細胞であるか否か、どのような種類の細胞か、生きているか否かを検出する。
本発明に関連した検出アッセイとして、カップ10の試液55中の検体52を染色する適切な任意の方法が利用される。例えば、検体52は捕獲培養過程で、事前に準備された捕獲レイヤー51bの上の適切な任意の表面捕獲サイトと結合する。捕獲検体52はその後、蛍光性のまたは着色染料などの、適切な任意の染色指標剤77で、適切な方法で染色される。
本発明に関連した検出アッセイとして、カップ10の試液55中にある検体52のための適切なDNAまたはRNA分析が利用される。例えば、検体52は、検体のDNAまたはRNAの特定なセグメントを遊離するなど、核素材を遊離し、適切で任意な方法でまず断裂させる。そのようにして遊離されたDNAまたはRNAのセグメントは、次に適切な方法で検出アッセイを利用し検出される。例えば、カップ10中の捕獲レイヤー51bに結合した捕獲分子は、任意の競合アッセイまたはサンドイッチ式の免疫検定が行われ、そこでは遊離されたRNAまたはDNAセグメントを補完するRNAまたはDNAセグメントとなり得る。検体52を断裂させた後、遊離されたDNAまたはRNAセグメントは、蛍光化合物など適切な任意の核酸指標剤77を含む試薬で、適切な方法(例えば、試薬の培養手順中など)でラベル付けがなされる。
カップ10の試液55中の検体52を断裂させて産出された遊離RNAまたはDNAのセグメントの数が希望する特定の検出アッセイの性能にとって適切でない場合、それらの遊離RNAまたはDNAセグメントの数をin situ,で増幅させるために使われるいくつかのテクニックがある。
図7ないし9を見ると、少なくとも1種類の検体52について試験するために、または、少なくとも1種類の検体52の少なくとも1つの標的識別特性を試験するために円周方向に分割されたカップ210が使用されている。カップ210は、本明細書で説明されたすべてのアッセイで使用することができ、カバー16またはカバー316がある。
円周方向の対となる導波管228とそれぞれの試験セグメント82または容器84のあらゆる特定の組み合わせはすべて、適切なそれぞれの境界86によって互いに分けられている。特定の境界86はすべて、一つ以上のその他の境界86と異なっていることがあり、例えば、隣接する試験セグメント82の組み合わせ間の境界86は、隣接する容器84の組み合わせ間の境界86とは異なっている。単一の境界86は、円周方向の導波管228とそのそれぞれの試験セグメント82それぞれの容器84の組み合わせを分けることができる。
図10に移り、軸方向に分割されたカップ210aは、本明細書で説明された任意のアッセイで使用することができ、カバー16またはカバー316がある。
概念の出発点として、カップ210aは、理論的には少なくとも2つのカップ210のための円周方向の導波管228ができるやり方で、カップ10の側壁14の導波管28が、円周方向に、少なくとも2つの導波管228aになったと見ることができる。そして、円周方向の導波管228aの少なくとも1つの内表面232aは、内表面232aが少なくとも2つの軸方向の試験セグメント88を含むように、カップ210aのA軸に多少平行している縦の方向で軸方向に分割されている。

- この前記アッセイ装置は、さらに前記カップを回転させるよう作動する回転装置を含む請求項1ないし3のいずれか1のアッセイ装置。
- 前記回転装置は、前記検体を少なくとも前記導波管の前記内表面の少なくとも一部分に遠心濃縮するように作動する請求項4のアッセイ装置。
- 前記カップは、さらに、前記導波管の前記内表面の少なくとも一部分上の一番奥の非液体検出レイヤーを包含し、この一番奥の非液体検出レイヤーは、内表面を包含し、また、前記回転装置は、前記一番奥の非液体検出レイヤーの前記内表面上の前記検体を遠心濃縮するように作動する請求項4のアッセイ装置。
- 前記回転装置は、前記検査中の前記カップを回転させるように作動する請求項4ないし6のいずれか1のアッセイ装置。
- 前記アッセイ装置は、さらに、前記導波管検査光の入力を発生するように作動する光源と、この光源と前記カップとを互いに関連付けて設置できるように作動する取付け装置を含む請求項1ないし7のいずれか1のアッセイ装置。
- このアッセイ装置は、さらに、前記信号光の出力の少なくとも一部分を受け取り、この受け取った前記信号光に応じて電気出力信号を発生する前記光検知器を包含する請求項1ないし8のいずれか1のアッセイ装置。
- 前記側壁は、さらに、反射体を含み、この反射体は、焦線を持ち、また、この反射体は、反射体の前記焦線からの反射体検査光の入力を受けて受信し、それへの反応として反射体検査光の出力を発生するように作動し、さらに、前記導波管検査光の入力は、前記反射体検査光の出力の少なくとも一部分を含む請求項1ないし9のいずれか1のアッセイ装置。
- このアッセイ装置は、さらに、前記反射体のための光源を含み、この光源は、前記反射体の焦線と交差するよう向けられている光軸を持つ請求項10のアッセイ装置。
- 前記カップの前記内部ボリュームは、前記導波管の前記内表面の少なくとも一部分にある検体検出コーティングを包含する請求項1ないし11のいずれか1のアッセイ装置。
- 前記検体検出コーティングの少なくとも一部分は、非液体検出レイヤーを包含する請求項12のアッセイ装置。
- 前記検出コーティングの少なくとも一部分は、液体検出レイヤーを包含する請求項12のアッセイ装置。
- 前記液体検出レイヤーの少なくとも一部分は、前記試液を包含する請求項14のアッセイ装置。
- 前記検体検出コーティングの少なくとも一部分は、内表面を持つ非液体検出レイヤーを包含し、また、前記液体検出レイヤーは、前記非液体検出レイヤーの前記内表面の少なくとも一部分の上に位置する請求項14もしくは15のアッセイ装置。
- 前記カップは、さらに、前記検体検出コーティングの少なくとも一部分でセンサーを含むセンサー導波管を包含し、この前記センサー導波管は、前記導波管検査光の入力の少なくとも一部分を受け入れるように作動し、また、このセンサー導波管は、前記検体検出コーティングの少なくとも一部分を、前記導波管検査光の入力の少なくとも一部分を使って検査を受けさせるように作動し、そして、前記側壁の前記導波管の前記外表面の少なくとも一部分は、前記検体の機能の一部として前記信号光を出力するように作動する請求項12ないし16のいずれか1のアッセイ装置。
- 前記センサー導波管は、さらに、前記側壁の前記導波管の少なくとも一部分を包含する請求項17のアッセイ装置。
- 前記導波管は、少なくとも2つの円周方向の導波管を包含し、各円周方向の導波管は、円周方向の弧の幅、外表面、および円周方向の試験セグメントを含む内表面を包含し、前記円周方向の導波管の前記各外表面の少なくとも一部分は、前記検体の機能の一部として前記信号光を出力するるように作動する請求項1ないし18のいずれか1のアッセイ装置。
- 前記検体検出コーティングの少なくとも一部分は、前記円周方向の試験セグメントの少なくとも一部分上にある、請求項12ないし16のいずれか1に従属する請求項19のアッセイ装置。
- 前記カップは、さらに、少なくとも2つのセンサー導波管を包含し、各センサー導波管は、前記円周方向の試験セグメントの各々の上の前記検体検出コーティングレイヤーの少なくとも一部分を包含し、各センサー導波管は、前記導波管検査光の入力の少なくとも一部を受けるように作動し、また、前記各センサー導波管は、前記円周方向の試験セグメントの各々の上の前記検体検出コーティングレイヤーの少なくとも一部分に、前記導波管検査光の入力の少なくとも一部分を使用して、検査を受けさせるように作動する請求項20のアッセイ装置。
- 前記各センサー導波管は、さらに、前記円周方向の導波管の各々の少なくとも一部分を包含する請求項21のアッセイ装置。
- 前記試液内の前記検体は、少なくとも2種類あり、前記円周方向の各導波管の前記外表面の少なくとも一部分は、前記検体の前記種類に応じて前記信号光をそれぞれ出力するように作動する請求項19ないし22のいずれか1のアッセイ装置。
- 前記検体は、少なくとも2つの標的識別特性を包含し、前記各円周方向の導波管の前記外表面の少なくとも一部分は、前記標的識別特性に応じて前記信号光を出力するように作動する請求項19ないし23のいずれか1のアッセイ装置。
- 前記カップは、さらに少なくとも一つの境界と、前記各円周方向の導波管の少なくとも1つの隣接する対を包含し、前記境界が、前記円周方向の導波管の前記隣接する対の間に位置する請求項19ないし24のいずれか1のアッセイ装置。
- 前記境界の少なくとも1つは、無効参考ゾーンを包含する請求項25のアッセイ装置。
- 前記カップは、さらに前記側壁に接続されたベースを包含し、このベースは、少なくとも2つの容器を包含し、これら容器の各々は、少なくとも前記円周方向の試験セグメントの各々のための貯蔵液を保持できるように作動する請求項19ないし26のいずれか1のアッセイ装置。
- 前記カップは、さらに少なくとも2つの境界を包含し、これら境界の少なくとも2つは、前記境界の隣接する対の少なくとも1つの対を形成し、また、前記各容器と、これに対応する前記各円周方向の試験セグメントとは、前記境界の前記隣接する対の各々の間に位置する請求項27のアッセイ装置。
- このアッセイ装置は、さらに、前記カップを回転させるように作動する回転装置を包含し、前記境界の各々の前記隣接する対は、前記カップが前記回転装置によって回転する際に、前記各容器のからの前記貯蔵液を、前記円周方向の各試験セグメントの前記対応する1つに流れるように作動する請求項28のアッセイ装置。
- 前記導波管の前記内表面は、少なくとも2つの軸方向の試験セグメントを包含し、これら軸方向の試験セグメントに対応する前記導波管の前記外表面の夫々の部分は、前記検体に応じて前記信号光をそれぞれ出力するように作動する請求項1ないし29のいずれか1のアッセイ装置。
- 前記導波管は、少なくとも2つの円周方向の導波管を包含し、各円周方向の導波管は、円周方向の弧の幅、外表面および少なくとも2つの試験セグメントを含む内表面を包含し、前記軸方向の試験セグメントに対応する前記円周方向の導波管の前記外表面の夫々の部分は、前記検体に応じて前記信号光をそれぞれ出力するように作動する請求項1ないし18のいずれか1のアッセイ装置。
- 前記カップの前記内部ボリュームは、前記各軸方向の試験セグメントの少なくとも一部分にある検体検出コーティングを包含する請求項30もしくは31のアッセイ装置。
- 前記カップはさらに、少なくとも2つのセンサー導波管を包含し、各センサー導波管は、前記各軸方向の試験セグメントの上の前記検体検出コーティングの少なくとも一部分を包含し、前記各センサー導波管は、前記導波管検査光の入力の少なくとも一部分を受けるように作動し、また、前記各センサー導波管は、前記各軸方向の試験セグメントの上の前記検体検出コーティングの少なくとも一部分が、前記導波管検査光の入力の少なくとも一部分を使用することで検査を受けさせるよう作動する請求項32のアッセイ装置。
- 前記各センサー導波管はさらに、前記円周方向の導波管の各々の少なくとも一部分を包含する請求項33のアッセイ装置。
- 前記試液内には少なくとも2種類の前記検体があり、前記信号光の前記各出力は、前記検体の種類に応じて放たれる請求項30ないし34のいずれか1のアッセイ装置。
- 前記検体は、少なくとも2つの標的識別特性を包含し、前記信号光の各出力は、前記標的識別特性のうちの1つに応じて放たれる請求項30ないし35のいずれか1のアッセイ装置。
- 前記カップは、さらに、少なくとも1つの境界と、少なくとも1つの前記軸方向の試験セグメントの隣接ペアを包含し、前記境界は、前記軸方向の試験セグメントの前記隣接する対のそれぞれの間に位置する請求項30ないし36のいずれか1のアッセイ装置。
- 前記少なくとも1つの境界は、無効参考ゾーンを包含する請求項37のアッセイ装置。
- このアッセイ装置はさらに、前記カップのためのカバーと、このカバーと前記カップの間の液体密封シールとを包含する請求項1ないし38のいずれか1のアッセイ装置。
- 前記カバーは、カバーインレットポート、カバーアウトレットポートおよび環状フランジを包含し、この環状フランジは、フランジインレットポートおよびフランジアウトレットポートを包含し、
組み合わさった場合、前記カバーと前記カップとは、前記環状フランジと前記カップの前記側壁との間に周辺の流れ室を規定し、さらに、組み合わさった場合、前記カバーと前記カップとは、液体が連続的に前記カバーの中に、前記カバーインレットポートを通り、前記フランジインレットポートを通り、前記周辺の流れ室を通り、前記フランジアウトレットポートを通り、前記カバーアウトレットを出て流れるように作動する請求項39のアッセイ装置。 - このアッセイ装置はさらに、前記カップの少なくとも一部分に熱を伝えるるよう作動するヒーターと、このヒーターと前記カップとが互いに取り付けられるよう作動する取り付け装置とを包含する請求項1ないし40のいずれか1のアッセイ装置。
- このアッセイ装置はさらに、前記カップの少なくとも一部分から熱を取り除くよう作動するクーラーと、このクーラーと前記カップとが互いに取り付けられるよう作動する取り付け装置とを包含する請求項1ないし41のいずれか1のアッセイ装置。
- 試液中の検体を検出する方法であって、
(a)光学アッセイカップが側壁と内部ボリュームを含み、この内部ボリュームが前記側壁によって少なくとも部分的に規定され、前記側壁が内表面と外表面を持つ導波管を包含する光学アッセイカップを含む光学アッセイ装置を準備する工程と、
(b)前記カップの内部ボリュームに前記試液を与える工程と、
(c)前記導波管に導波管検査光の入力を与える工程と、
(d)前記内部ボリュームの少なくとも一部分を、前記導波管検査光の入力の少なくとも一部分を使用して検査させる工程と、
(e)前記導波管の前記外表面から前記検体に応じて出る信号光の出力を検出する工程とを具備する、方法。 - この方法はさらに、前記工程(b)を実行した後であって、前記工程(c)、(d)、および(e)を実行する前に、前記カップを回転させる工程を具備する請求項43の方法。
- この方法はさらに、前記工程(b)を実行した後であって、前記工程(c)、(d)、および(e)のうちの少なくとも1つを実行す間に、前記カップを回転させる工程を具備する請求項43の方法。
- 前記工程(c)はさらに、前記導波管検査光を、暗視野検査光に選定する工程を包含し、前記工程(d)はさらに、前記検査を暗視野検査に選定する請求項43ないし45のいずれか1の方法。
- 前記工程(a)はさらに、前記側壁が反射体を含むように選択する工程を包含し、前記工程(c)はさらに、少なくともいくらかの前記暗視野検査光を与えるように前記反射体を使用する工程を包含する請求項46の方法。
- 前記工程(c)はさらに、前記導波管検査光を、エバネセント検査光に選択する工程を包含し、また、前記工程(d)はさらに、前記検査をエバネセント検査に選択することを包含する請求項43ないし45のいずれか1の方法。
- 前記工程(a)はさらに、前記側壁が反射体を含むように選択する工程を包含し、前記工程(c)はさらに、前記エバネセント検査光の少なくともいくらかを前記反射体が与えるように使用する工程を包含する請求項48の方法。
- この方法はさらに、
(f)適当な時に、前記導波管の前記内表面の少なくとも一部分に検体検出コーティングを与える工程とを具備する請求項43ないし49のいずれか1の方法。 - 前記工程(f)はさらに、前記検体検出コーティングが液体検出レイヤーを含むように選択する工程を包含する請求項50方法。
- 前記工程(f)はさらに、前記液体検出レイヤーが前記試液の少なくとも一部分を含むように選択する工程を包含する請求項51の方法。
- 前記工程(f)はさらに、前記検体検出コーティングが非液体検出レイヤーを含むように選択する工程を包含する請求項50の方法。
- 前記工程(f)はさらに、前記検体検出コーティングが液体検出レイヤーと非液体検出レイヤーとを含むように選択する工程を包含する請求項50の方法。
- 光学アッセイカップと、ルミネセンス検出中に前記カップを回転させるように作動する回転装置とを包含し、前記カップは、側壁と内部ボリュームを包含し、前記側壁は、外表面を包含し、前記内部ボリュームは、少なくとも部分的に前記側壁により規定され、前記側壁は、試液と検体のためのルミネセンス指標剤を受け取るよう作動し、
前記外表面と前記側壁の少なくとも一部分とは、前記ルミネセンス検出の間、前記検体に応じてルミネセンス信号光の出力を放つよう作動する、ルミネセンス検出によって試液中の検体を検出するための光学アッセイ装置。 - 試液内の検体を検出するための合成センサー導波管であって、
表面を有する導波管と、この導波管の前記表面の少なくとも一部分の上にある検体検出コーティングとを具備し、
前記検体検出コーティングの少なくとも一部分は、この合成センサー導波管の使用中、前記試液と接触するよう作動し、
この合成センサー導波管は、導波管検査光の入力を受け取り、前記検体に応じた信号光の出力を放つように作動し、前記導波管検査光の入力の少なくとも一部分は、暗視野検査光の入力を包含し、前記検査は、暗視野検査を包含する、合成センサー導波管。
