一定期間試験環境に馴化させた5週令のWistar ratを無作為に2群にわけ、1群にはコントロール食を、2群にはセサミン0.2% 含有食を2週間ペアフィードさせた。その後、既報に従い、単離胃還流動物を作成した。具体的には、ネンブタール麻酔下で胃・十二指腸とそれに付随する血管を摘出した後、ポリエチレンチューブを腹腔動脈と門脈に挿入し固定した。その他の動脈・静脈・膵臓(管)は結紮し取り除いた。摘出した胃はKrebs Ringer-biocarbonate buffer(37℃、pH 7.4)中に保ち、胃内腔は1%HEPESにて一定の速度で還流洗浄した。腹腔動脈側から還流液を流して門脈側で回収し、回収液中の各種消化ホルモンの濃度をRadioimmunoassay(RIA)法により測定した。 Five-week-old Wistar rats acclimated to the test environment for a certain period were randomly divided into two groups, one group being fed with a control diet and the other group being fed with a diet containing 0.2% sesamin for two weeks. A reflux solution was flowed from the celiac artery side and collected on the portal vein side, and concentrations of various digestive hormones in the collected solution were measured by a radioimmunoassay (RIA) method.
その結果、セサミン摂取群でコントロール群と比較して、ガストリン(図1)、ヒスタミン(図2)の分泌が抑制されることが明らかとなった。このことから、セサミンは胃酸の分泌を抑制する作用を有し、消化性潰瘍に対して抑制効果を示すものと考えられる。
[実施例2]製剤例

セサミン 0.01g
酢酸トコフェロール 0.25g
無水ケイ酸 20.5g
トウモロコシデンプン 179.0g
以上の粉体を均一に混合した後に10%ハイドロキシプロピルセルロース・エタノール溶液100mlを加え、常法通り練和し、押し出し、乾燥して顆粒剤を得た。
(製剤例2)カプセル剤
ゼラチン 60.0%
グリセリン 30.0%
パラオキシ安息香酸メチル 0.15%
パラオキシ安息香酸プロピル 0.51%
水 適量
上記成分からなるソフトカプセル剤皮の中に、以下に示す組成物を常法により充填し、1粒200mgのソフトカプセルを得た。
セサミン 10.0mg
グリセリン脂肪酸エステル 15.0mg
ミツロウ 15.0mg
小麦胚芽油 250mg
(製剤例3)錠剤
セサミン 0.01g
デンプン 282g
ショ糖脂肪酸エステル 9.0g
酸化ケイ素 9.0g
これらを混合し、単発式打錠機にて打錠して経9mm、重量300mgの錠剤を製造した。
(製剤例4)ドリンク剤
呈味: DL−酒石酸ナトリウム 0.1g
コハク酸 0.009g
甘味: 液糖 800g
酸味: クエン酸 12g
ビタミン:ビタミンC 10g
セサミン 1g
ビタミンE 30g
シクロデキストリン 5g
香料 15ml
塩化カリウム 1g
硫酸マグネシウム 0.5g
上記成分を配合し、水を加えて10リットルとした。このドリンク剤は、1回あたり約100mlを飲用する。
