Abstract

Description
本発明は、目的のアミノ酸配列に付着した不安定なアミノ酸配列を含有する融合タンパク質をコードする核酸(例えば、DNA免疫プラスミド)に関し、ここで目的アミノ酸配列の免疫原性は、不安定なアミノ酸配列の存在によって増加される。本発明はまた、目的の付着したアミノ酸配列分泌型融合タンパク質(例えば、ケモカインまたはサイトカインおよび目的の付着したアミノ酸配列を含有するもの)をコードする核酸に関し、ここで目的アミノ酸配列の免疫原性は、分泌配列に付着している結果として、増加する。本発明はまた、ワクチンとしてまたは遺伝子治療における使用のために、コードされたタンパク質の免疫原性を増加させる方法に関する。
ヒト免疫不全ウイルス1型(HIV−1)および関連するサル免疫不全ウイルス(SIV)に対する細胞性免疫応答は、HIV−1およびSIV感染を制御する際、ならびに疾患の進行を遅らせる際に重要な役割を果たすことが示されている。感染した個体における初期のHIV−1感染の封じ込めは、ウイルス特異的細胞傷害性Tリンパ球(CTL)応答の出現と関連する(1,2,3)。慢性的に感染した個体において、HIV−1に対する高頻度CTL応答はまた、低いウイルス付加および緩慢な疾患の進行と関連する(4,5)。HIV−1特異的CTL応答はまた、特定の高度に強く曝露された血清陰性の個体において実証されている(6,7,8)。また、CTL応答の維持に重要であり得る、HIV特異的な強い増殖応答は、長期にわたり無症状のまま経過する個体(long−term nonprogressor)において同定されている(9,10)。
非Met N末端を有するタンパク質は、細胞内において、標的タンパク質のコード配列がユビキチンのコード配列のC末端に対してインフレームで融合される融合構成物を使用して発現されている。ユビキチンは、通常、細胞内でポリタンパク質としてとして産生され、これがユビキチン加水分解酵素によって各ユビキチンサブユニットのC末端で切断され、個々のユビキチン分子を生じる。これらの同一のユビキチン加水分解酵素はまた、ユビキチン標的融合タンパク質をユビキチンのC末端で切断し、標的のN末端を曝露する。最近の研究において、ToberyおよびSilicianoは、nefのN末端のMetまたはArgのいずれか(それぞれ、UbMNefおよびUbRNef)を用いてHIV−1
nefへのユビキチン融合物を作製した(37)。UbMNefおよびUbRNefを発現するためのワクシニアウイルスを使用するインビトロ実験において、両方のベクターは、同程度の量のnefおよびN末端にArg残基およびより短い半減期(t1/2=15分と10時間)を有する形態のnefの発現を誘導した。さらに、迅速に分解されるUbRNefを発現するワクシニアウイルスを有するマウスの免疫化は、安定なUbMNefを発現するワクシニアベクターを用いる免疫化よりもさらに活発なnef特異的CTL応答を誘導した。ToberyおよびSilicianoは、迅速な細胞質における分解のための抗原のターゲッティングによるnef特異的CTL応答の増加は、HIVに対するワクチン接種のための魅力的なストラテジーを提示すると結論付けている(37)。

(ベクター)
HIV−1またはSIVの抗原を発現するDNAベクターが、本明細書中の実施例において用いられる。
1)全長gagを発現するプラスミド(p55)またはgagの一部を発現するプラスミド(p37)あるいはgagおよびプロテアーゼを発現するプラスミド(p55gagpro)。P55は、細胞より部分的に放出されるgag粒子を産生する。P37は、細胞より部分的に放出されるが、粒子を形成しない。P55gagproはまた、プロテアーゼを産生し、従って、そのgagは、プロセシングされて、p17、p24、p6およびp7を形成する。
3)プロテアソームの効率的な分解を与える配列へのgagの融合物を発現するプラスミド。

本発明者らは、gagに対するMCP−3融合が、非改変gagベクター(上記のような1型)と比較して、gagに対する免疫応答を劇的に増加することを見出した(図を参照のこと)。この特性は、これらの細胞からのより効率的なgag分泌の結果の一部であり得る。なぜなら、本発明者らは、tPAのリーダー配列を有する分泌されたgagが、分泌および免疫原性においてより効率的であることを、最近示したからである(Qiuら、J.Virol.2000)。

HIV抗原およびSIV抗原の改変された形態が、DNAワクチン接種後に異なる免疫応答を示すことを示唆する以前のデータに基づいて、本発明者らは、12匹のマカクにおけるSIV gagおよびSIV envについての3つの異なるDNAワクチンベクターの免疫原性を研究した。使用したDNAを、以下の表1に示す:

生後遺伝子治療において、新規遺伝子形成は、間接的な手段(例えば、標的細胞を体から取り出し、次いで新規遺伝子情報を保有するウイルスベクターを用いてこの標的細胞を感染させ、次いでこの標的細胞をその体に再移植すること);または、直接的な手段(例えば、DNA処方物をリポソームにカプセル化すること;ウイルスエンベロープレセプタータンパク質を含むタンパク質リポソームにDNAを捕捉すること;DNAのリン酸カルシウム共沈殿;およびDNAをポリリジン−糖タンパク質キャリア複合体に結合すること)によって、組織に導入される。さらに、リン酸カルシウム共沈殿物の形成の有りまたは無しでの直接的な肝臓内注射後での、クローン化したウイルスDNA配列のインビボ感染性もまた、記載されている。安定性を増強するエレメントを含有するmRNA配列もまた、陽イオン性脂質小胞の使用によって、Xenopus laevis胚において効率的に翻訳されることが示されている。例えば、J.A.Wolffら、Science 247:1465−1468(1990)およびその中に引用される参考文献を参照のこと。
