Abstract
Description
(a)一般式(I)で定義される1以上の骨格を準備する段階
(b)任意段階として、上記骨格の環構造を維持しながら、上記骨格の窒素を誘導体化してアニオン交換基を導入及び/又はアミンを保護する段階、及び
(c)残基Rに結合したアミンを含む試薬の添加によるアミノ分解によって誘導体の環構造を開環させて、開環した骨格のカルボニル炭素に上記アミンを付加させる段階
を含んでなる方法である。
「分離媒体」という用語は、本明細書では、例えばクロマトグラフィーカラムの充填材として有用な材料、具体的には、ベースマトリックスに1以上のリガンドが結合したものからなる材料に用いる。ベースマトリックスは担体として作用し、リガンドはクロマトグラフィーで目標物質と相互作用する官能基を与える。
図1は、例示的な骨格としてホモシステインチオラクトンを使用して、本発明でマルチモードアニオン交換リガンドのライブラリを生成するための、基本的方法の概略図(スキーム2)である。使用した例示的な骨格は、ホモシステインチオラクトンであり、3種の異なる誘導体化した骨格が示されている。必要なら、保護段階を、誘導体化の前に含めることができる。R基に結合したアミンからなる化合物を使用して、環構造を開く段階(c)で、図1では3種の異なる例示的なリガンド構造が得られ、その第一の構造は、弱アニオン交換リガンドであり、第二の構造は、強アニオン交換リガンド、即ちその電荷がすべてのpH値で維持されるリガンドであり、第三の構造は、弱アニオン交換リガンドである。
アニオン交換体リガンドの製造は、例えば、ホモシステインチオラクトンの使用を介して実現することができ、これはいくつかの戦略的合成経路を介して考え出すことができるものである。リガンドのアニオン基は、直接開環誘導体化段階(合成経路1)又は保護、脱保護方法(合成経路2)を介して、ホモシステインチオラクトンのアミン官能基から得ることができる。アニオン基は、開環誘導体段階で、N−保護チオラクトン又は保護されていないチオラクトンに導入することもでき(合成経路3)、或いは誘導体化ホモシステインチオラクトンにも導入することができる(合成経路4)。アニオン基は、アミド基の還元によって生成することもできる(合成経路5)。N−保護/脱保護戦略を使用する場合、カップリングは、脱保護の後又は前に行うことができる。すべてのカップリングは、求核置換又はラジカル付加を介して行った。
合成経路1
実施例1a:
室温のアルゴン中、ベンジルアミン(3.7ml、33.9mmol)を、塩酸D,L−ホモシステインチオラクトン(1g、6.51mmol)のTHF(40ml)撹拌溶液に添加した。4時間後、溶液を蒸発させ、真空乾燥した。残留物をHPLC(水/アセトニトリル)で精製して、白色生成物(0.77g、52%)を得た。
室温のアルゴン中、ブチルアミン(1.6ml、16.2mmol)を、塩酸D,Lホモシステインチオラクトン(1g、6.51mmol)のTHF(40ml)撹拌溶液に添加した。2時間後、溶液を蒸発させ、真空乾燥した。残留物をHPLC(水/アセトニトリル)で精製して、白色残留物を得た。
室温のアルゴン中、2−(トリフルオロメチル)ベンジルアミン(4.5ml、32.1mmol)を、塩酸D,Lホモシステインチオラクトン(0.99g、6.47mmol)のTHF(40ml)撹拌溶液に添加した。3時間後、溶液を蒸発させ、真空乾燥した。残留物をHPLC(水/アセトニトリル)で精製して、白色固体(0.48g、25%)を得た。
実施例2a:
ホモシステインチオラクトンのアミン基の保護
塩酸D,L−ホモシステインチオラクトン(7.04g、45.82mmol)、N−ジイソプロピルエチルアミン(DIPEA、8.40ml、48.22mmol)及びジクロロメタン(DCM、200ml)を、窒素雰囲気中で混合した。反応物を、氷浴中で数分間撹拌し、次いでジカルボン酸ジ−t−ブチル(10.53g、48.25mmol)を添加した。0℃で20分間撹拌した後、反応混合物を室温まで温めた。20時間後、溶媒を真空除去し、フラッシュクロマトグラフィー(溶出剤ジクロロメタン)にかけた結果、白色固体(5.88g、59%)を得た。
上述の生成物(933mg、4.29mmol)を、THF 50mlに溶解し、アルゴンで脱ガスを行った。次いでチオフェン−2−メチルアミン(2.59g、22.88mmol)を添加し、反応混合物を4日間撹拌した。溶媒を蒸発させ、残留物を酢酸エチル(45ml)で抽出し、10%クエン酸水溶液(2×15ml)、水(10ml)で洗浄した。Na2SO4で乾燥した後、有機相を蒸発させ、得られた残留物を次の段階で直接使用した。
上記得られた残留物を、ジクロロメタン(DCM)45mlに溶かした溶液に、トリフルオロ酢酸(5ml)を添加し、アルゴンで脱ガスを行った。
実施例3a:
室温のアルゴン中、N,N−ジメチルエチレンジアミン(1.77ml、16.26mmol)を、塩酸D,L−ホモシステインチオラクトン(1g、6.51mmol)のTHF(40ml)撹拌溶液に添加した。4時間後、溶液を蒸発させ、真空乾燥した。残留物を、カップリングに直接使用した。
実施例4a:
塩化ベンゾイル(0.87ml、7.5mmol)を5mlのDCMに溶かした溶液を、D,L−ホモシステインチオラクトン(1.15g、7.5mmol)及びDIPEA(2.6ml、15mmol)をジクロロメタン(DCM、15ml)に溶かした溶液に、0℃で1滴ずつ添加した。混合物を、室温で一晩撹拌した。溶媒を真空蒸発させ、反応残留物を酢酸エチル(30ml)で抽出した。有機相を、クエン酸10%水溶液(w/w、20ml)、K2CO3 10%水溶液(20ml)、水(20ml)で洗浄し、硫酸ナトリウムで乾燥した。濾過した後、溶媒を除去し、白色固体(1.37g、83%)を得た。N,N−ジメチルエチレンジアミン(0.68ml、6.25mmol)を、室温のアルゴン中、上記得られた白色固体(0.55g、2.49mmol)の乾燥THF(20ml)撹拌溶液に添加した。6時間後、溶液を蒸発させ、真空乾燥した結果、白色粉末が得られ、これをカップリングに直接使用した。
ゲル1aの製造:
アリル基の濃度が167μmol/mlであるSepharose(商標)6FF 2.0gを、水(5×10ml)及びジオキサン(5×10ml)で洗浄し、0.5mlのジオキサンが入っているフラスコに添加した。リガンド1a(292mg、1.3mmol)及びAIBN(2,2’−アゾイソブチロニトリル)(204mg、1.2mmol)を、1mlのジオキサンに溶解した。溶液に、窒素を10分間フラッシュし、ゲルに添加し、反応フラスコを70℃で一晩振盪させた。反応後、ゲルを濾過し、ジオキサン(5×20ml)、エタノール(5×20vol.)及び水(3×20ml)で洗浄した。ゲルに対するリガンドの投入量を、以下の手順を使用して、アミン官能基を滴定することによって測定した。ゲル1mlを0.5M HCl溶液で洗浄し、その後、1mM HCl溶液で洗浄した。ゲルをフラスコに移し、水19ml及び濃HNO3 2滴を添加した。滴定を、硝酸銀0.1M溶液を使用して行った。結果:85μmol/mlゲル。
リガンド1b(アリル基当たり、リガンド約3当量)を、水(3.5ml)に溶解し、50%(w/w)NaOH水溶液の添加によってpHを8.5に調節した。溶液を、臭素で活性化したアリルSepharose(商標)Fast Flowゲルに添加し(2g、230μmol/mlゲル)、その混合物を、振盪板上に60℃で放置した。18時間後、ゲルを濾過し、水(2×30ml)、エタノール(2×30ml)、0.2M酢酸(2×30ml)及び水(2×30ml)で洗浄した。アミン基の滴定によって測定したゲルのイオン容量は、190μmol/mlゲルであることがわかった。
アリル基の濃度が167μmol/mlであるSepharose(商標)6FF 2.0gを、水(5×10ml)及びジオキサン(5×10ml)で洗浄し、0.5mlのジオキサンが入っているフラスコに添加した。リガンド1c(240mg、0.83mmol)及びAIBN(190mg、1.14mmol)を、1mlのジオキサンに溶解した。溶液に、窒素を10分間フラッシュし、ゲルに添加し、反応フラスコを、70℃で振盪板上に放置した。19時間後、ゲルを濾過し、ジオキサン(5×20ml)、エタノール(5×20vol.)及び水(3×20ml)で洗浄した。ゲルに対するリガンドの投入量は、アミン官能基の滴定によって測定した結果、106μmol/mlゲルであった。
アリル基の濃度が230μmol/mlであるSepharose(商標)6FF 2.0gを、水(5×10ml)及びジオキサン(5×10ml)で洗浄し、0.5mlのジオキサンが入っているフラスコに添加した。リガンド2a(290mg、1.26mmol)及びAIBN(190mg、1.14mmol)を、1mlのジオキサンに溶解した。溶液に、窒素を10分間フラッシュし、ゲルに添加し、反応フラスコを、70℃で振盪板上に放置した。19時間後、ゲルを濾過し、ジオキサン(5×20ml)、エタノール(5×20vol.)及び水(3×20ml)で洗浄した。ゲルに対するリガンドの投入量は、アミン官能基の滴定によって測定した結果、142μmol/mlゲルであった。
リガンド3a(アリル基当たり、リガンド約3当量)を水(3.5ml)に溶解し、50%(w/w)NaOH水溶液の添加によってpHを8.5に調節した。溶液を、臭素で活性化したアリルSepharose(商標)Fast Flowゲルに添加し(2g、230μmol/mlゲル)、その混合物を、振盪板上に60℃で放置した。18時間後、ゲルを濾過し、水(2×30ml)、エタノール(2×30ml)、0.2M酢酸(2×30ml)及び水(2×30ml)で洗浄した。アミン基の滴定によって測定したゲルのイオン容量は、156μmol/mlゲルであることがわかった。
リガンド4a(0.32g、1.02mmol)を、DMSO/水:80/20(2.5ml)に溶解し、50%(w/w)NaOH水溶液の添加によってpHを9に調節した。溶液を、臭素で活性化したアリルSepharose(商標)Fast Flowゲルに添加し(2g、230μmol/mlゲル)、その混合物を、振盪板上に60℃で放置した。18時間後、ゲルを濾過し、水(2×30ml)、エタノール(2×30ml)、0.2M酢酸(2×30ml)及び水(2×30ml)で洗浄した。アミン基の滴定によって測定したゲルのイオン容量は、133μmol/mlゲルであることがわかった。
・イオン交換体に関する実験参照手順
材料
緩衝液
緩衝液1:20mMピペラジン+0.25NaCl pH6.0
タンパク質溶液:
BSA:緩衝液1中、4mg/mL
すべての緩衝液及びタンパク質溶液は、使用前に、0.45μm Millipore Millex HAフィルターに通して濾過した。
すべての実験は、Unicorn 3.1ソフトウェアを備えたAKTA(商標)Explorer 100クロマトグラフィーシステム(Amersham Biosciences AB)を使用して、室温で行った。サンプルを、150mLスーパーループを介してカラムに注いだ。全体を通し、1mL/分(約300cm/時)の流量を使用した。10mmフローセルを使用して、280nmで吸光度を測定して、溶出物を連続的にモニタした。
プロトタイプのアニオン交換体のそれぞれを、HR5/5カラム(充填床容積=1mL)に充填し、20カラム体積の20mMリン酸緩衝液(pH6.8)で平衡にした。タンパク質混合物(6mg/mlコンアルブミン、4mg/mlラクトアルブミン及び6mg/ml大豆トリプシンインヒビター)50μlをカラムに注ぎ、100%の前述の緩衝液及び2M NaClに至る線形勾配(20カラム体積)で溶出した。流量は、0.3ml/分(100cm/時)に調節した。
プロトタイプのアニオン交換体のそれぞれを、HR5/5カラム(充填床容積=1mL)に充填し、適切なpH及び塩濃度の緩衝液で平衡にした。システムの空隙容量は、非結合条件下、適切なタンパク質溶液をカラムに加えることによって決定した。溶出物のA280が、加えられたタンパク質のA280の10%に到達するまでの時間を、このシステムの空隙容量とした(分で表す)。
最大吸光度の10%のレベルでのブレイクスルー(Qb10%)を、以下の関係式を使用して計算した。

Claims (13)
- 1以上のマルチモードアニオン交換リガンドの製造方法であって、
(a)一般式(I)で定義される1以上の骨格を準備する段階
(b)任意段階として、上記骨格の環構造を維持しながら、上記骨格の窒素を誘導体化してアニオン交換基を導入及び/又はアミンを保護する段階、及び
(c)残基Rに結合したアミンを含む試薬の添加によるアミノ分解によって誘導体の環構造を開環させて、開環した骨格のカルボニル炭素に上記アミンを付加させる段階
を含んでなる方法。 - 式(I)のA、B及びXが炭素原子であり、mが1である、請求項1記載の方法。

- 得られた開環生成物をそのチオール基を介して、反応性基を含むベースマトリックスに固定化する段階(d)をさらに含む、請求項1乃至請求項6のいずれか1項記載の方法。

- マルチモードアニオン交換リガンド又はベースマトリックスに複数のマルチモードアニオン交換リガンドが結合した分離媒体であって、当該リガンド又は媒体が請求項1乃至請求項9のいずれか1項記載の方法で製造したものである、リガンド又は媒体。
- 請求項11記載の分離媒体を充填したアニオンクロマトグラフィー用クロマトグラフィーカラム。
- 液体から目標物質を分離する方法であって、請求項11記載の分離媒体を準備し、媒体を液体に接触させてその表面に目標物質を吸着させることを含む方法。
