Abstract
Description
標的指向ビヒクルとしてのTNT抗体
a.キメラTNT−3
TNTモノクローナル抗体(MAb)を使用して、腫瘍の壊死領域中に局在する変性中の細胞を介して固形腫瘍を標的とした(1)。この手法は、抗原による免疫調整及びその脱落の問題を回避しながら、多様な起源の癌を標的とするために使用できる。3種のキメラTNT MAbが開発された。これらのchTNT−1、chTNT−2、及びchTNT−3は、それぞれ、ヒストンDNA複合体、ヘテロクロマチンDNA、及び一本鎖DNAからなる核内抗原を標的とする(4、5、Khawliら、Cancer Biotherapy & Radiopharmaceuticals(2002)17:359−370)。腫瘍保有マウスにおける各TNT MAbの薬物動態及び生体分布特性が提示されている。3種すべてのキメラTNT MAbは以前に記載されたように産生された(同上)。
キメラTNT−1をヒトIgG1及びκ定常領域に対してマウス可変領域を遺伝子操作することによって、親のマウス抗体から初めて開発した。キメラ抗体の挙動はマウス抗体のそれと同様であったが、それが共有する35%の相同性が、ヒト抗マウス抗体(HAMA)応答の潜在能力を可能にする。この問題を回避するために、そのマウス対応物に対する同様の結合特性を示した完全なヒトTNT−1 Mab、NHS76を開発した。NHS76は、Cambridge Antibody Technologies(Cambridge、England)との共同作業で開発し、Raji Burkittリンパ腫細胞核抽出物を使用するラージscFvライブラリーをスクリーニングすることによって誘導した(Sharifiら、Hybridoma and Hybridomics、(2001)20:305−312)。これは、NS0マウスミエローマ細胞系において、Lonza Biologicals,Inc.(Slough、England)から入手したグルタミンシンテターゼ発現系を使用して発現した。
サイトカイン融合タンパク質
chTNT−3及びヒトIL−2(参考文献番号12)、マウスIFNγ(参考文献番号9)、ヒトIFNγ及びTNFα(参考文献番号10)、並びにcHTNT−3のC末端部分に遺伝子工学的に連結されたμGM−CSFからなるいくつかの融合タンパク質の産生が記載されている。これらの構築、試験、及びこの構築物を用いる免疫療法実験における使用は、以下に手短に記載される。
chTNT−3(IgG1γ)を、以前に記載されたように構築及び発現させた(参考文献番号5)。NHS76(IgG1κ)の重鎖及び軽鎖の遺伝子を有するプラスミドを、以前に記載されるように産生した(参考文献番号10)。候補scFv抗体を、制限酵素消化、ライゲーション、及びポリメラーゼ連鎖反応(PCR)を含む標準的な分子生物学技術を使用して、全抗体に転換した。
4種の融合タンパク質のサイトカイン部分の各々の生物学的活性を、インビトロアッセイによって決定した。IL−2生物活性は、IL−2依存性CTLL−2細胞の増殖を支持するその能力を試験することによって実証した(Gillisら、J.Immunol.(1978)120:2027−2031)。異なる濃度のchTNT−3/IL−2、chTNT−3、又は組換えヒトIL−2標準を、2×104個のIL−2飢餓CTLL−2細胞とともに20時間、加湿した37℃、5%CO2インキュベーター中でインキュベートした。細胞を、3H−チミジンで6時間パルス標識し、細胞試料を収集し計数した。次いで、chTNT−3/IL−2の活性を、外挿又はrIL−2曲線によって、IU/mgで決定した。
融合タンパク質の免疫反応性を、上記に記載されたように、固定化Rajiリンパ腫細胞への結合によって決定した。すべてのchTNT−3/サイトカイン融合タンパク質は、同様の結合定数(0.5〜1×109M−1)を有することが見出され、chTNT−3の結合定数(1.4×109M−1)と比較可能であった。これは、chTNT−3の重鎖へのサイトカインの遺伝子連結が抗原結合を有意に妨害しなかったことを示す。
クリアランス試験及び生体分布試験を、すべての融合タンパク質の薬物動態クリアランスの半減期及び腫瘍の取り込みを決定するために実行した。薬物動態試験のために、BALB/cマウスに125I標識した融合タンパク質又は裸の抗体を注射し、注射の時点及びその後の選択された時点での全身活性を微小線量計で測定した。これらの試験結果を図2に示す。クリアランス時間の顕著な違いが、muIFNγ融合タンパク質と、IL−2、TNFα、及びmuGM−CSF融合タンパク質との間に記録される。chTNT−3は、これらの試験において異常に長い半減期を有することが見出された。
ケモカイン融合タンパク質
a.LEC/chTNT−3の構築、発現、及び精製
chTNT−3(IgG1κ)の軽鎖遺伝子(pEE6/hCMV−LC)及び重鎖遺伝子(pEE12/HC)を有するプラスミドを、以前に記載されたように産生した(5、12)。ヒトケモカインLEC遺伝子をRT−PCR(Stephensら、Eur.J.Immunol.(2001)31、1247−1254)によってHepG2肝臓癌細胞系から、TRIzol(Invitrogen、Carlsbad、CA)を使用してクローニングし、全RNAを得た。次いで、LECの成熟cDNAを、プライマー5’−TCTAGAATGAAGGTCTCCGAGGCTGCC−3’(配列番号4)及び5’−GCGGCCGCCTG−GGAGTTGAGGAGCTG−3’(配列番号5)を使用するPCRによって増幅し、抗体のリーダー配列の翻訳下で、XbaI及びNotIによって、chTNT−3の重鎖遺伝子のN末端に挿入した。次いで、得られる融合遺伝子を発現ベクターpEE12に挿入し、グルタミンシンテターゼ遺伝子増幅系によって規定されるように、エレクトロポレーションによって、NS0細胞にpEE6/TNT−3軽鎖とともに同時トランスフェクションした。細胞培養培地をトランスフェクションの3週間後に毎週交換し、この時点で最良の発現クローンは、以前に記載されたように(参考文献番号5)、抗原として粗DNAを使用する培養上清の間接的ELISAアッセイによって選択した。大量の融合タンパク質を産生するために、高発現クローンを8リットルの曝気攪拌フラスコ中で、5%熱不活化(68℃で1時間、断続的に攪拌しながら)透析ウシ胎児血清を含む選択培地中で増殖させ、インキュベーション及び融合タンパク質の分解の間にNS0細胞によるタンパク質分解性酵素の誘導を除去した。次いで、分泌された融合タンパク質を、タンデムプロテインA親和性クロマトグラフィー及びイオン交換クロマトグラフィーの手順によって、清澄化細胞培養上清から精製した。融合タンパク質の純度は、還元条件下でのSDS−PAGE電気泳動法によって、及び溶媒系として0.05Mリン酸緩衝液及び0.4M過塩素酸ナトリウム、pH6.1を使用するHPLCによって調べた。精製後、融合タンパク質を0.22μm Nalgene使い捨てフィルターユニットを通して濾過し、分注し、10ml滅菌チューブ中で、長期保存のために−20℃で保存した。
LEC融合タンパク質の生物活性は、96ウェル微小走化性チャンバー(Neuroprobe、Gaithersburg、MD)において製造業者のプロトコールにおいて記載されるように標的細胞の移動を測定することによって実証された。手短に述べると、LEC/chTNT−3、組換えヒトLEC、又は親のchTNT−3を、結合培地(1%BSA及び25nmol/L HEPESを含むRPMI 1640)中で0.39nMから50nMまで段階希釈した(Hedrickら、Blood(1998)91:4242−4247;Zach−Howardら、Blood(2000)96:840−845)。この溶液を微小走化性装置の下側のチャンバー中に配置した。次いで、105THP−1ヒト単球細胞を含む100μLの結合培地を上側のチャンバーに加えて、加湿した5%CO2、37℃インキュベーター中での1.5時間のインキュベーション後、移動した細胞のパーセンテージを計算して、移動指数((結合培地に曝露された細胞の平均数)で除算した(ケモカイン及び融合タンパク質に曝露された細胞の平均数))を決定した。すべてのアッセイは3重に実行した。
125I−標識された融合タンパク質を、以前に記載されたように(5、7)、改変クロラミン−T法を使用して調製した。手短に述べると、1mCi(37MBq)の放射性ヨード及び20μLのクロラミン−Tの水溶液(2mg/mL)を、100μL PBS中の100μg LEC/chTNT−3を含む5mL試験管に加えた。2分後、20μLのメタ重亜硫酸ナトリウムで溶液をクエンチした。各反応混合物を、Sephadex G−25カラムを使用して精製し、代表的には90〜95%収率を回収した。放射性標識された抗体を注射のためにPBSで希釈し、4℃で保存し、そして放射性標識後2時間以内に投与した。放射性ヨード標識された抗体を、シリカゲル含浸させたグラスファイバーからなる分析用インスタント薄層クロマトグラフィー(ITLC)システム(Gelman Sciences、Ann Arbor、MI)を使用して分析した。細片(2×20cm)を、使用前の110℃、15分間の加熱によって活性化し、1μLの試料をスポットし、風乾し、そしてメタノール/H2O(80:20)を用いて約10cm溶離させ、再度風乾し、半分に切断し、そして結合したタンパク質及び遊離の放射性ヨードを決定するために計数した。ITLC分析は、0のRf値(MAb結合)及び99%より高い放射性化学物質の純度を明らかにした。インビトロ血清安定性もまた、以前に記載されたように評価した(5)。手短に述べると、放射性標識されたMAbをマウス血清中で37℃、48時間インキュベートする。トリクロロ酢酸沈殿及び遠心分離後、MAb結合放射能をγカウンターで測定した。約95%の活性がトリクロロ酢酸沈殿であり、この時間の間では放射性ヨードの放出は実質的に検出されなかった。
精製LEC/chTNT−3のアビディティー定数を決定するために、固定化細胞ラジオイムノアッセイを、以前に記載されたように実行した(12)。手短に述べると、Rajiリンパ腫細胞をPBSで1回洗浄し、EMグレード2%パラホルムアルデヒド(Polysciences、Washington、PA)で室温で10分間固定し、そしてPBSで再度洗浄し、その後0.2%アジ化ナトリウムを含むPBS中で4℃にて保存した。10から110ngの125I−標識LEC/chTNT−3を、106固定化Raji細胞と室温で1時間インキュベートした。この細胞を、1%BSAを含むPBSで3回洗浄して、いかなる未結合の抗体をも除去し、γカウンターで計数した。結合した融合タンパク質の量を、残存する結合した細胞の放射能(cpm)及び融合タンパク質の比活性から決定した。データのScatchardプロット分析を使用して傾きを得た。この傾きから、平衡又はアビディティー定数Kが、式K=−(傾き/n)によって計算された。ここで、nは抗体の価数(IgGについては2)である。LEC/chTNT−3は、chTNT−3(1.4×109M−1)と同様の結合定数(1.0×109M−1)を有することが見出された(5)。これは、chTNT−3の重鎖の可変領域へのLECの遺伝子連結が抗原結合を有意に妨害しなかったことを示す。
6週齢雌性BALB/cマウスを使用して、125I−LEC/chTNT−3の薬物動態クリアランスを決定した。遊離の放射性ヨードの甲状腺の取り込みをブロックするために事前に2〜3日間、飲用水中にヨードカリウムを与えたマウスの群(n=5)に、0.1mL接種物を使用して、125I−標識融合タンパク質(30〜40μCi/マウス)の静脈内注射を投与した。注射の時点及び注射後の選択された時点での全身活性を、CRC−7微小線量計で測定した。データを分析し、PSTRIP薬物動態プログラム(MicroMath,Inc.、Salt Lake City、UT)によって半減期を決定した。結果を平均±標準偏差として表現し、有意なレベル(P値)を、Wilcoxon順位和検定を使用して決定した。これらの試験から、125I−LEC/chTNT−3が3時間±20分(p≦0.01)のT1/2を有することが見出されたことが決定された。
免疫療法試験
a.LEC/chTNT−3治療
6週齢雌性BALB/cマウスの群(n=7)に、University Animal Care Committee−認可プロトコールの下で、約107のMAD109細胞、Colon26細胞、又はRENCA細胞を含む0.2mL接種物を左側腹に皮下注射した。腫瘍を5〜7日間、それらが直径約0.5cmに達するまで増殖させた。次いで、腫瘍保有マウスの群を、LEC/chTNT−3(20μg)、PBS、又はchTNT−3(20μg)を伴う、0.1mL接種物で静脈内治療した。すべての群を1日5回治療し、1日おきに三次元のキャリパー測定によって腫瘍増殖をモニターした。腫瘍体積は式:長さ×幅×高さによって計算された。結果を平均±標準偏差として表現し有意なレベル(P値)を、Wilcoxon順位和検定を使用して決定した。
chTNT−3/サイトカイン融合タンパク質を、COLON26結腸腺癌、MAD109肺腺癌、及び以下の図3において示されるように、RENCA腎臓細胞癌を含むBALB/cマウスの異なる固形腫瘍モデルにおいて試験した。これらの試験のために、6週齢雌性BALB/cマウスに、左側腹に5×106腫瘍細胞を皮下注射した。腫瘍を5〜7日間、それらが直径約0.5cm(0.2cm3)に達するまで増殖させ、マウスを群に分け(n=5)、4日間連続して毎日皮下注射した。次いで、腫瘍の増殖を、キャリパー測定によって1日おきに17日目まで追跡し、そこで実験を終了した。0.1ml接種物中の融合タンパク質の用量は、chTNT−3/muGM−CSFがより毒性であり5μg/用量の使用を必要とする以外は、20μgであった。融合タンパク質の組み合わせた使用される場合、これらは0.1mlで接種物を維持するためにあらかじめ混合された。対照群は生理食塩水のみ又は20μgのchTNT−3を受容し、これは、3つの腫瘍モデルの増殖曲線に効果がなかった。4つの個々の実験からの対照群の試験は、増殖曲線が非常に再現性が高いことを示した。図3に示される3つの腫瘍モデルにおけるこれらの実験の結果は、これらのモデルのうちの2つにおいて、chTNT−3/IL−2、chTNT−3/TNFα、及びchTNT−3/IFNγの組合せを受容するマウスが、大部分の腫瘍退行を示し、これは17日目で対照群の約80%であると見積もられることを実証した。個々の融合タンパク質はこれらの腫瘍モデルにおいて様々な有効性の程度を有したが、この組合せ、又はchTNT−3/TNFαの代わりにmuTNT−3/muGM−CSFが置換されたものが最も有効であった。キメラ抗体はこれらの融合タンパク質のいくつかの構築において使用されたので、治療は1回の4日間の期間に限定された。この限定のために、腫瘍は、処置の中断の直後に対照群のペースと同じペースで増殖し始めた。それゆえに、これらの処置治療効果は一過性であり、より持続する効果及び腫瘍の完全な退行のためには第2又は第3の処置過程の追加が必要と思われる。
6週齢雌性BALB/cマウスに、約5×106MAD109マウス肺腺癌細胞を皮下接種した。5日後、腫瘍が直径約0.5cmに達し、マウスに、20μgのLEC/chTNT−3単独、又は20μgのTNT−3/IFNγ、chTNT−3/TNFα、chTNT−3/GM−CSF若しくはchTNT−3/IL−2を伴う、0.1mlの接種物を注射した。すべての群を1日5回治療し、1日おきに三次元のキャリパー測定によって腫瘍増殖をモニターした。腫瘍体積は式:長さ×幅×高さによって計算された。結果を平均±標準偏差として表現した。図4において示されるように、LEC/chTNT−3及び4種のchTNT−3サイトカイン融合タンパク質の各々を用いる併用療法は最小限の改善のみを生じたが、chTNT−3/IL−2を含む組合せは17日までに増殖曲線の平板化を示した。
a.LEC/chTNT−3の免疫組織化学及び組織試験
BALB/cマウスの群に、上記のように左側腹に107腫瘍細胞を注射した。腫瘍移植の7日後、マウスを、LEC/chTNT−3(20μg)、PBS、又はTNT−3(20μg)を用いて、1日5回静脈内治療した。次いで、各群からのマウスを、腫瘍移植の10日後、12日後、14日後、及び16日後に屠殺し、腫瘍を切除し、そしてパラフィン包埋のために10%中性緩衝化ホルマリン(VWR Scientific、West Chester、PA)中に固定するか、又は凍結切片のためにO.C.T.化合物(Lab−Tek Products、Naperville、III)中で液体窒素中で急速冷凍するかのいずれかであった。MAD109腫瘍保有マウスからのパラフィン包埋切片を、形態学的試験のために、ヘマトキシリン及びエオシン(H&E)で染色した。免疫組織化学試験のために、Colon26腫瘍保有マウスからの腫瘍の凍結切片を、リンパ、PMN、及び樹状細胞亜集団を染色するための、ビオチン化抗CD4+、抗CD8+、抗CD11b+、抗汎内皮、抗CD11c+、抗CD19+、抗CD3e+、及び抗45R+(BD PharMingen、San Diego、CA)抗血清で染色した。次いで切片をHRP−結合体化ストレプトアビジンとともにインキュベートし、比色定量用作用剤で発色させ、その後hr & Eで染色した。顕微鏡による知見を、Optronixデジタルカメラを使用して記録した。
これらの腫瘍モデルの抑制のために部分的な原因であるT細胞の亜集団を評価するために、枯渇化試験を、上記の治療試験とともに実行した。マウスの群に、上記のようにColon26結腸癌を移植し、腫瘍が直径0.5cmに達すると、これらは5日毎に、CD4+(0.5mg/用量、クローンGK1.5)、CD8+(0.5mg/用量、クローンH35)、又はNK細胞(0.35mg/用量、抗−アシアロ−GM)に特異的な細胞傷害性抗血清を受容し、FACS分析によって示されるように、これらの各々のT細胞亜集団の各々を末梢循環中で<2%まで減少した。特定の亜集団がLEC/chTNT−3免疫療法のための道具であるならば、LEC/chTNT−3によって誘導される観察される腫瘍抑制が妨害され、腫瘍は対照治療マウスと同様の増殖曲線を有する。図5に示されるように、これらの抗血清各々単独による枯渇化は、chTNT−3治療対照の腫瘍の増殖から、その腫瘍の増殖を変化しなかった。対照的に、LEC/chTNT−3治療マウスは、50%より高い阻害を示した。予測されるように、LEC/chTNT−3治療と組み合わせたCD8+及びNK枯渇化治療は、LEC/chTNT−3の抗腫瘍活性を無効にし、このことは、これらのリンパ球亜集団がLEC機能において非常に重要であることを示す。対照的に、LEC/chTNT−3治療と組み合わせたCD4+枯渇化を受けるマウスは完全な治癒に至り、予想外であり且つ潜在的に非常に意味のある知見である。

Claims (23)
- 個体において腫瘍のサイズを減少させるか、又は癌細胞の増殖を阻害する方法であって、肝発現ケモカイン(LEC)に連結された癌標的指向分子を含む有効量の癌療法剤を前記個体に投与する工程を含み、前記癌療法剤が前記個体の癌細胞又は腫瘍に局在する方法。
- 癌に罹患している個体において転移性癌の発生を減少させるか、又は阻害する方法であって、肝発現ケモカイン(LEC)に連結された癌標的指向分子を含む有効量の癌療法剤を前記個体に投与する工程を含み、前記癌療法剤が前記個体の癌細胞又は腫瘍に局在する方法。
- 個体における免疫調節性T細胞の活性を減少させる工程をさらに含む、前記請求項のいずれかに記載の方法。
- 前記免疫調節性T細胞の活性を減少させる工程が、個体から免疫調節性T細胞をエキソビボで除去することを含む、請求項3に記載の方法。
- 前記免疫調節性T細胞の活性を減少させる工程が、個体における免疫調節性T細胞をインビボで枯渇化又は不活性化させることを含む、請求項3に記載の方法。
- 前記免疫調節性T細胞の活性を減少させる工程が、免疫調節性T細胞に結合する少なくとも1種の抗体を使用して達成される、請求項3に記載の方法。
- 前記少なくとも1種の抗体がIL−2レセプターに特異的である、請求項6に記載の方法。
- 前記IL−2レセプターに特異的な抗体がCD25に特異的な抗体である、請求項7に記載の方法。
- 前記免疫調節性T細胞の活性を減少させる工程が、前記癌療法剤を投与する前に実行される、請求項3から8までのいずれかに記載の方法。
- 癌に対する細胞傷害活性を有するT細胞を投与する工程をさらに含む、前記請求項のいずれかに記載の方法。
- 前記T細胞を投与する工程が、個体からT細胞を除去し、そのT細胞を活性化し、次いで前記個体に活性化T細胞を投与することを含む、請求項10に記載の方法。

- 前記抗体が細胞内抗原に特異的である、請求項13に記載の癌療法剤。
- 前記抗体が核内抗原に特異的である、請求項16に記載の癌療法剤。
- 前記抗体が、マウス抗体TNT−1、TNT−2、TNT−3又はNHS76のマウス型、キメラ型、ヒト化型、又はヒト型である、請求項12、13、16、又は17に記載の癌療法剤。
- 前記癌標的指向分子及びLECが共有結合で結合されている、請求項12に記載の癌療法剤。
- 前記癌標的指向分子が遺伝子融合によってLECに連結されたタンパク質である、請求項12から19までのいずれかに記載の癌療法剤。
- LECがそのC末端で前記癌標的指向分子のN末端に融合される、請求項20に記載の癌療法剤。
- 前記癌標的指向分子が抗体であり、且つLECが前記抗体の軽鎖若しくは重鎖のN末端に融合されるか、又はLECが前記抗体の軽鎖及び重鎖のN末端に融合される、請求項12から20までのいずれかに記載の癌療法剤。
- 請求項12から22までのいずれかに記載の癌療法剤及び薬学的に許容可能な担体を含む組成物。
