本出願は、2003年1月31日に提出された米国仮特許出願60/443,979号に基づく優先権を主張するものである。前記の出願の内容全体を、引用をもってここに援用することとする。
免疫応答は、身体全体の組織に常在する樹状細胞(DC)及びマクロファージ(MG)を含む専門的な抗原提示細胞(APC)のレベルにより、惹起される。DCは、Tリンパ球と相互作用し、従って強力な免疫応答を誘導する、高レベルの細胞表面分子及び相補性受容体を発現する。更にDCは、免疫応答を惹起すると共に、細胞性免疫及び体液性免疫の両方の増幅で頂点に至るサイトカイン、ケモカイン及びプロテアーゼを分泌する。
T細胞に提示し、そしてMHCクラスII分子は抗原を特異的CD4+ T細胞に提示する。
vivoでの寿命は短い。
al. (62)は、ある抗原に特異的な抗体が、抗原に対する体液性免疫応答を高めることができたことをマウスで示したが、これはおそらく、APC上に発現したIgGに対するFc受容体(FcγR)に免疫複合体を送達することによる。Wernersson及び共同研究者
(63)は、免疫応答の亢進における個々のFcγRの役割を免疫複合体をin vivoで用いて研究した。彼らの研究では、FcγRIは、免疫応答の亢進を媒介するのに充分であることが実証された。しかしながら、このような免疫複合体は、抗原提示に関与していない数多くの細胞上のFc受容体にも結合するため、APCを特異的に標的とするものではなく、従って、抗原送達の効率を低下させてしまう。
(66, 67)。加えて、DC上のFcRに結合した免疫複合体はMHCクラスIの関係でプロセッシング及び提示されることが示された (64, 65)。更に、数多くのこのようなFcR標的決定法には制約がある。なぜなら、FcRは血小板及び好中球など、数多くの非APC上で発現するからである。理想的には、APCを特異的に狙い、また効果的なMHCクラスII限定TH応答だけでなく、効果的なMHCクラスI限定CTL応答を誘導することのできるワクチンがあれば、いくつかの疾患を治療する上で優れた効験を上げるであろう。
本発明は、数多くの疾患の効果的な治療に必要な抗原特異的T細胞媒介性免疫応答を生じさせるための抗体ベースのワクチン及び方法を提供するものである。具体的には、強力な抗原特異的細胞傷害性Tリンパ球(CTL)応答が、抗原提示細胞(APC)上に発現した特定の受容体に結合する抗体を用いて、一種以上のタンパク質抗原をAPCの標的に決定することにより、誘導される。好適な受容体には、樹状細胞(DC)及びマクロファージの両方の上に発現するC-レクチン、特にヒトマンノース受容体、がある。本発明により実証されるように、抗体−抗原複合体を用いてマンノース受容体を標的に決定すると、MHCクラスI及びクラスII経路の両方を通じて、抗原のプロセッシングが起きる。このように、抗原特異的CTL(例えばCD8+T細胞)や、ヘルパT細胞(例えばCD4+T細胞)を含む他の重要な免疫エフェクタT細胞が誘導される。

本発明は、特定の細胞受容体を狙う抗体を用いて抗原を抗原提示細胞(APC)の標的に設定することにより、重要なT細胞媒介性免疫応答を生じさせることができるという発見に基づく。具体的には、癌及び感染性疾患など、数多くの疾患の効果的な治療のためには、ワクチンは、MHCクラスIの関係で抗原を認識するCD8+T細胞により主に媒介される、強力な抗原特異的細胞傷害性Tリンパ球(CTL)応答を惹起せねばならない。最適な免疫処理のためには、これには、好ましくは、MHCクラスII経路の関係で抗原を認識する、例えばCD4+T細胞などの抗原特異的T細胞の誘導を含め、他の重要なエフェクタT細胞機能が伴っているとよい。このように、効果的なワクチンは、抗原特異的CTLを、好ましくは他のT細胞媒介性免疫応答と組み合わせて、複数のMHC経路を通じて誘導せねばならない。

U.S.A. 86:10029-10033を参照されたい)。このようなフレームワーク配列は、生殖細胞系抗体遺伝子配列を含む公共のDNAデータベースから得ることができる。これらの生殖細胞系配列は、B細胞成熟中のV(D)J接合により形成される、完全に集合した可変遺伝子を含まないこととなるため、成熟抗体遺伝子配列とは異なるであろう。生殖細胞系遺伝子配列はまた、高親和二次レパートリ抗体の配列からも、可変領域全体にわたって均一に、個々で異なるであろう。例えば、体細胞変異は、フレームワーク領域のアミノ末端部分ではあまり頻繁には起きない。例えば、体細胞変異は、フレームワーク領域1のアミノ末端部分と、フレームワーク領域4のカルボキシ末端部分では、あまり頻繁には起きない。さらに、多くの体細胞変異は、本抗体の結合特性を大きくは変化させない。この理由で、もとの抗体のものと同じ結合特性を有するインタクト組換え抗体を作り直すためにも、特定の抗体の全DNA配列を得る必要はない(全ての目的のために引用をもってここに援用されたWO
99/45962を参照されたい)。CDR領域にわたる部分的重鎖及び軽鎖配列があれば、この目的には充分であることが多い。この部分的配列を用いて、どの生殖細胞系可変及びジョイニング遺伝子セグメントが、組換えられた抗体可変遺伝子に寄与したかを決定する。次に、この生殖細胞系配列を用いて、可変領域の消失部分を充填する。重鎖及び軽鎖リーダ配列は、タンパク質の成熟中に切断されるため、最終的な抗体の特性には寄与しない。これが理由で、発現コンストラクトのための対応する生殖細胞系リーダ配列を用いる必要がある。消失配列を加えるために、クローニングされたcDNA配列を合成オリゴヌクレオチドにライゲーション又はPCR増幅法により組み合わせることができる。代替的には、可変領域全体を、一組の短い、重複のあるオリゴヌクレオチドとして合成し、PCR増幅で組み合わせて、完全に合成の可変領域クローンを作製することもできる。このプロセスは、消失又は含有又は特定の制限部位や、あるいは、特定のコドンの最適化など、いくつかの長所を有する。

Nizard, P. et al. (1998) FEBS Lett. 433:83-88)。目的の抗原を発現すると共に、表面に抗体が繋がれているような細胞は、腫瘍細胞又は微生物病原体など、この細胞に対するCTL応答などの特異的免疫応答を誘導するために用いられよう。
別の局面では、本発明は、本発明のワクチン複合体の1つ又は組合せを、薬学的に許容可能な担体と一緒に調合された形で含有する医薬組成物などの治療用組成物を提供するものである。本発明のワクチン複合体は、対象のT細胞との相互作用に向けて、対象の血流に送達されるように投与される。このようなT細胞の標的決定は、本複合体を直接用いるか、あるいは、ワクチン複合体で予め標的決定された細胞を用いることにより、in vivo 又はex vivo のいずれでも達成することができる。

本発明のワクチン複合体は、多種の疾患及び状態を治療及び/又は防止(例えば免疫化)するために用いることができる。
vivo)することにより、本複合体を対象に間接的に投与することもできる。本複合体をAPCに接触させ、送達することで、これらが投与前にAPCによりプロセッシング及び提示されるようにすることは、抗原又は細胞の「装備」とも呼ばれる。抗原をAPCに装備する技術は当業で公知であり、その中には、例えば、Gunzer
and Grabbe, Crit Rev Immunol 21 (1-3):133-45 (2001)及び Steinman, Exp Hematol
24(8): 859-62 (1996)がある。
治療しようとする疾患又は状態、投与される特定の複合体、対象の大きさ、又は当該疾患もしくは状態の重篤度などの因子に応じて様々であろう。当業者であれば、不要な実験を要せず、特定の多重特異的分子の有効量を経験的に決定することができる。
方法及び材料
全血及びロイコパックからのDCの作製: ヘパリン処理済み全血又はアフェレーシス製剤のフィコール・ハイパックによる密度勾配遠心分離により、ヒト末梢血単核細胞(PBMC)を得た。次に、プラスチック製培養皿への付着又は水簸法により単球を単離し、サイトカイン(10ng/ml GM-CSF及び2ng/ml
IL-4)の培養基への添加により、未熟DCに分化させた。DCを5日目から7日目までの間、採集し、フローサイトメトリで分析した。この方法で調製されたDCは、CD14−、HLA-DR+、CD11c+マンノース受容体+であり、高レベルの MHC クラスI及びII、CD80 及びCD86を発現した。
P.L. and Stevens V. (1999) Oncology Reports 6:7-17にレビュー)。第一に、妊娠の他には、この抗原の発現は、生殖細胞腫瘍や、数多くの腺癌に主に限られている(表3)。また、hCGは、システイン・ループ成長因子スーパーファミリの一メンバであり、成長因子として、血管新生及び/又は転移促進作用物質として、あるいは、免疫機能の抑制物質として、癌の樹立又は進行において役割を果たしていると思われる。従って、機能的hCGの発現を制限する免疫治療法は、更なる治療上の利益をもたらすであろう。
と一緒に、そして全クラスIIHLA分子 (20μg/ml)を遮断するためにL243と一緒に、30分間、室温でプレインキュベートした。結合しなかったmAbは遠心分離で取り除かれた。
fで60分間、37℃で標識した。標識された標的をRPMI媒質中で3回、洗浄し、ペレットを再懸濁させて、3×104細胞/mlの細胞濃度にした。抗原特異的CTLを96 ウェルV底プレートに滴定して、100:1 (エフェクタT細胞、E:対標的、T)乃至12.5:
1以下にした。一定数の標識済み標的を添加(100μl/ウェル又は3,000標的細胞/ウェル)し、プレートを低速(180×g)で遠心分離し、37℃でインキュベートした。4時間後、100乃至120μlの上清を採集し、放出される放射活性をγ-カウンタ計数(パーキン・エルマ社、ウォラック・インスツルメンツ)で判定した。CTL活性を計算し、以下の等式:

を用いて特異的溶解(致死)率で表した。但し式中、実験的(cpm)とは、CTL(E)及び標的(T)を含有するウェルからの放射活性(放出されるクロム)を言い;自発的(cpm)とは、0.1mlの媒質のみ(即ちCTLを添加せず)に入れた標的の入ったウェルからの放射活性を言い、最大放出とは、0.1mlの界面活性剤溶液(Igepal
CA 630;syn. NP-40;RPMI媒質に溶かした5%溶液)の存在下の標的の入ったウェルからの放射活性を言う。よく制御された実験条件下では、自発的な放出値は、最大放出の10%以下であるはずである。MHC遮断分析のために、標識された標的を、全クラスIの遮断のためにHLA特異的mAbであるW6/32と、そして全クラスIIHLA分子の遮断のためにL243と(20μg/ml)、30分間、室温でプレインキュベートした。結合しなかった
mAb を遠心分離で取り除き、mAbで被覆された標的をCTLに加えた。アイソタイプの一致するmAbをコントロールとして用いた。
ワクチン複合体のデザイン: このコンストラクトは、樹状細胞上のヒトマクロファージマンノース受容体に結合する完全ヒト抗体であるB11にβhCG抗原を連結することにより、作製された。連結は、図3に示すように、遺伝子融合により、抗体の重鎖に抗原を共有結合させることにより、行われた。
C末端ペプチドに特異的なmAb(シグマ社)を用いて別々にプローブした。その結果から、形質転換したCHO細胞がB11-βhCGワクチン複合体を特異的に発現したことが、融合産物の適切な大きさ及び組成を証左として、裏付けられた。
ワクチン複合体のデザイン: 樹状細胞上のヒトマクロファージマンノース受容体に結合すると共に、完全ヒトB11抗体のVL及びVHフラグメントを含有する一本鎖抗体であるB11一本鎖融合体(ScFv)に、βhCG抗原を連結することにより、二番目のコンストラクトを作製した。連結は、図1に示すように、遺伝子融合により、B11
ScFvのカルボキシ末端に該抗原を共有結合させることによりなされた(ここではB11sfv-βhCGコンストラクトと呼ぶ)。
抗体で標的決定されたワクチンの、APC表面上のそのコグネート受容体による認識は、この送達プラットフォームの最初のステップである。βhCG-B11及びB11sfv-βhCGコンストラクトが、MRを発現している培養ヒトDCに特異的に結合することを実証するために、フローサイトメトリ研究が用いられてきた(図4)。
B11 HuMAb染色樹状細胞を用いて行われた免疫組織化学検査では、意外な交差反応性は見られなかった(結果は図示せず)。これらの研究をβhCG-B11で繰り返したが、結果は同一だった。
βhCG-B11コンストラクトがDCによるプロセッシングを受けて、DC上のMHCクラスI及びクラスII分子を介して(交差提示)βhCG抗原をT細胞に提示する能力を評価した。具体的には、ワクチンに曝露してあるDCと一緒に正常T細胞のプールを培養することにより、βhCG-B11コンストラクトを用いて、抗原特異的T細胞を惹起した。次に、その結果得られる「感作」T細胞を分析して、それらの活性(増殖及び致死)及び特異性を調べた。T細胞の特異性は、βhCG抗原を有する標的細胞に応答したT細胞活性を、抗原陰性コントロールに比較することにより、実証することができる。存在する場合の細胞傷害性T細胞(CTL)は、βhCG関連抗原を提示する標的のみを致死させ、抗原を欠くか、あるいは、無関係の抗原を提示しているコントロール標的は生かすはずである。CTL媒介性抗原認識は、常に、当該ペプチドを担持するMHC分子の関係でのみ、起きるため、MHC:ペプチド-CTL相互作用がMHC特異的mAbで遮断されれば、クラスI又はクラスIIの提示が裏付けられる。
U/ml のIL-2を2日目(48時間目)に添加。再刺激を前の通りに7日目、14日目、及び21日目に行ったが、例外として、βhCG-B11濃度は半分(それぞれ10、5及び2.5μg/ml)にした。装備していない、又は、βhCG-B11、B11sfv-βhCG、もしくはB11で装備された51Cr標識済みDCに対する反応性(バルクで、又は精製済みT細胞下位集団で)についてT細胞を検査した。MHC特異性が、HLA特異的mAbの存在下で確認された。
抗体(B11)のみで処理された標的細胞は、細胞傷害活性に対して感受性でなかったことから、ワクチンの抗原部分のみがCTL活性を惹起することができることが実証された。これらの結果は、βhCG-B11コンストラクトが効率的なCTL活性を誘導すること、そして具体的には、このCTL活性は、標的決定している抗体(B11)ではなく、βhCG抗原に向かうことを示している。
(コントロール)と一緒に、又は、βhCG-B11コンストラクト を装備したDCと一緒に、培養した。増殖を測定するために、DNA合成を培養から5日後に3H-チミジンを用いて分析した。データは、コントロールに対する、増殖の倍増(刺激指数)で表された。CTL活性で見られるように、T細胞をDCのみ(即ち抗原なし)で刺激した場合には、何の応答もみとめられなかった。複合体を形成させていない抗体(抗MR
B11 mAb)のみで標的決定されたDCは、βhCG-B11コンストラクトで惹起されるT細胞の増殖を誘導しなかった。T細胞の増殖能は、抗MHCクラスIやクラスII特異的mAbの両方の存在下で著しく遮断されたことから、CD4+及びCD8+T細胞の両方が応答性であったことが実証された。これらのデータは、MHC区画とのMRの同時局在と一致して、DCによるβhCG-B11コンストラクトの取り込みにより、ワクチンがMHCクラスI及びクラスIIプロセッシング経路に到達可能になることを示している。
未熟DCは、飲作用又は受容体媒介性飲食作用の機序により、可溶性抗原を取り込むことができる(55)。抗原内部移行の機序は、その細胞内での運命を決定すると共に、それに対する免疫応答の質に影響すると考えられる(54、55、56)。MRを通じた内部移行は、急速な、クラスリン媒介性内部移行事象だと解説されている
(57, 58)。MR自体はその細胞質側の尾内に2つの推定クラスリン標的決定配列を有し、マンノシル化金粒子の内部移行により、クラスリンで被覆された孔が電子顕微鏡により位置特定されている(58,
59)。クラスリン依存的飲食作用は、素早い高張ショック又はK+枯渇により、特異的に破壊することができる (61)。マンノシル化抗原、又は、マンノース受容体に結合させたB11が、クラスリンで被覆された孔を通じて内部移行したのかどうかを判定するために、未熟DCをAIM5培地に入れた氷上で、400mMのショ糖と一緒に又は無しで、30分間、B11
mAb又はマンノシル化BSAのいずれかの存在下でインキュベートした。次に細胞を37℃まで温め、20分間、内部移行させた。洗浄及び固定後、細胞を共焦点顕微鏡法(データは図示せず)で分析した。B11をMRに結合させた場合、その取り込みは高張ショックで阻害されたことから、その内部移行機序はクラスリンで被覆された孔を通じてであることが示された。対照的に、マンノシル化BSAの取り込みは高張ショックでは阻害されなかったことから、その内部移行機序は、クラスリンで被覆された孔の形成に依存していなかったことが示された。B11の濃度の20倍高いそれでも、マンノシル化BSA
FITCによる表面染色は比較的に弱かった。その後の研究では、内部移行したマンノシル化 BSA FITC は非特異的な液相トレーサと同じ位置に特定されたが、他方、内部移行したB11を含有するベシクルには、この非特異的トレーサは含まれていなかった(データは図示せず)。B11-FITCとは対照的に、
マンノシル化BSA-FITC及び液相トレーサの両方の取り込みとも、PI3K阻害剤ワートマニンによる予備処理で概ね遮断された(データは図示せず)。これらの結果は、マンノシル化BSAの大半は未熟樹状細胞に非特異的マクロピノサイトーシス機序を通じて取り込まれたことを示しており、マンノシル化抗原に対する免疫応答の質は、MRに特異的に標的決定された抗原とは大きく異なることを示唆している。
単球由来DCを、37℃のB11sfv-βhCG 又はβhCG-B11のいずれかのPBS-BSA緩衝液溶液に45分間、曝露し、CD40Lの存在下で一晩、成熟させた。次に、採集されたDCを洗浄し、マウス抗βhCG
、続いてヤギ抗ヒトIgG (Fc)-PE 複合体で染色した。染色された細胞をフローサイトメータ(BD-LSR)で分析した。ほぼ10,000の事象を各試料について採集した。バックグラウンドの自発蛍光及びアイソタイプ適合抗体の染色をコントロールとして役立てた。平均蛍光強度(MFI)(データは図示せず)に基づくと、DC上に発現したMRへのB11sfv-βhCG
結合はβhCG-B11のそれと同様である。
DCにより提示されたβhCG-B11に対して生じたCTLを、βhCG-B11及びB11sfv-βhCGに曝露した自己由来DC標的に対して検査し、他方、未処理のDC又はB11に曝露したDCをコントロールとして役立てた。抗原曝露後、上清中の放射活性の放出を測定する4時間検定で、標的を51クロムで標識し、CTLと混合した。この実験において、βhCG-B11特異的T細胞は、MHCクラスI分子上に抗原を提示する4つの標的のうち2つを認識する。DCが抗原を欠いた場合、標的の致死は何らみとめられなかった(図11)。このように、DCによるβhCG-B11の取り込みの結果、CTLにより認識されるβhCG由来T細胞エピトープが生ずると思われる。

Claims (49)
- ヒト抗原提示細胞(APC)に結合するモノクローナル抗体を、βヒト絨毛性性腺刺激ホルモン(βhCG)に連結させて含む分子複合体。
- 前記抗体がヒト樹状細胞上に発現したC型レクチンに結合する、請求項1に記載の分子複合体。
- 前記抗体がヒトマンノース受容体に結合する、請求項1又は2に記載の分子複合体。
- 前記抗体がヒト、ヒト化及びキメラ抗体から成る群より選択される、上記請求項のいずれかに記載の分子複合体。
- 前記抗体が、全抗体、Fabフラグメント及び一本鎖抗体から成る群より選択される、上記請求項のいずれかに記載の分子複合体。
- 前記複合体が組換え融合タンパク質である、請求項1に記載の分子複合体。
- 前記抗体が、FR1、CDR1、FR2、CDR2、FR3、CDR3及びFR4配列を含むヒト重鎖可変領域と、FR1、CDR1、FR2、CDR2、FR3、CDR3及びFR4配列を含むヒト軽鎖可変領域とを含み、但しこの場合:
(a)前記ヒト重鎖可変領域CDR3配列が、配列番号 15及びその保存的改変を含み;そして
(b)前記ヒト軽鎖可変領域CDR3配列が、配列番号18及びその保存的改変を含む、
上記請求項のいずれかに記載の分子複合体。 - 前記ヒト重鎖可変領域CDR2配列が配列番号14及びその保存的改変を含み;そして前記ヒト軽鎖可変領域CDR2配列が配列番号17及びその保存的改変を含む、請求項6に記載の分子複合体。
- 前記ヒト重鎖可変領域CDR1配列が配列番号13及びその保存的改変を含み;そして前記ヒト軽鎖可変領域CDR1配列が配列番号16及びその保存的改変を含む、請求項7又は8に記載の分子複合体。
- 前記抗体が:
(a)ヒトVH5-51生殖細胞系配列(配列番号30)を由来とする重鎖可変領域;及び
(b)ヒトVk-L15 (配列番号32)生殖細胞系配列を由来とする軽鎖可変領域、を含む、
請求項1に記載の分子複合体。 - 前記抗体が、それぞれ配列番号4及び配列番号8に示されたアミノ酸配列、又は、前記抗体がヒト樹状細胞への結合能を保持しているように配列番号4又は配列番号8に充分相同なアミノ酸配列、を含むヒト重鎖及びヒト軽鎖可変領域を含む、上記請求項のいずれかに記載の分子複合体。
- いずれか又は両方の鎖をβhCGに連結させたヒト抗体重鎖及びヒト抗体軽鎖、を含む分子複合体。
- 前記重鎖がβhCGに連結してあると共に、配列番号2に示されたアミノ酸配列を含む、請求項12に記載の分子複合体。
- 前記軽鎖が配列番号6に示されたアミノ酸配列を含む、請求項12又は13に記載の分子複合体。
- ヒト抗原提示細胞(APC)に結合するモノクローナル抗体をβヒト絨毛性性腺刺激ホルモン(βhCG)に連結させて含む分子複合体であって、但し前記抗体が:
(a)ヒトVH5-51生殖細胞系配列 (配列番号30)を由来とする重鎖可変領域;及び
(b)ヒトVk-L15 (配列番号32)生殖細胞系配列を由来とする軽鎖可変領域、
を含む、分子複合体。 - ヒト抗原提示細胞(APC)に結合するヒト一本鎖抗体を、βヒト絨毛性性腺刺激ホルモン(βhCG)に連結させて含む分子複合体であって、配列番号12に示されたアミノ酸配列を含む、分子複合体。
- T細胞媒介性免疫応答が抗原に対して生ずるように、APCにより内部移行してプロセッシングされる、上記請求項のいずれかに記載の分子複合体。
- 前記T細胞応答が細胞傷害性T細胞により媒介される、請求項17に記載の分子複合体。
- 前記T細胞応答がCD4+及びCD8+T細胞の両方により媒介される、請求項17又は18に記載の分子複合体。
- 前記T細胞応答が、MHCクラスI及びMHCクラスII経路の両方を通じて誘導される、請求項17乃至19のいずれかに記載の分子複合体。
- 上記請求項のいずれかに記載の分子複合体と、薬学的に許容可能な担体とを、選択的にはアジュバントと組み合わせて含む組成物。
- βhCGに対するT細胞媒介性免疫応答を誘導又は亢進する方法であって、抗原に対するT細胞媒介性応答が誘導又は亢進される態様で前記抗原がプロセッシングされ、T細胞に提示されるように、上記請求項のいずれかに記載の分子複合体をAPCに接触させるステップを含む、方法。
- 前記T細胞応答がCD4+及びCD8+T細胞の両方により媒介される、請求項22に記載の方法。
- 前記T細胞応答が細胞傷害性T細胞及び/又はヘルパT細胞により媒介される、上記請求項のいずれかに記載の方法。
- 前記T細胞応答が、MHCクラスI及びMHCクラスII経路の両方を通じたT細胞への抗原の交差提示により誘導される、上記請求項のいずれかに記載の方法。
- 前記βhCG抗原が腫瘍細胞により発現される、上記請求項のいずれかに記載の方法。
- 前記腫瘍細胞が、結腸、肺、膵臓、乳房、卵巣、及び生殖細胞を由来とする腫瘍細胞から成る群より選択される、請求項26に記載の方法。
- 前記分子複合体を樹状細胞にin vivoで接触させる、上記請求項のいずれかに記載の方法。
- 前記分子複合体を樹状細胞にex vivoで接触させる、上記請求項のいずれかに記載の方法。
- 樹状細胞の増殖を刺激するサイトカイン、選択的にGM-CSF又はFLT3-Lに、前記樹状細胞を接触させるステップを更に含む、上記請求項のいずれかに記載の方法。
- 免疫刺激性作用薬、選択的にはCTLA-4に対する抗体に、前記樹状細胞を接触させるステップを更に含む、上記請求項のいずれかに記載の方法。
- 上記請求項のいずれかに記載の分子複合体を、選択的にはアジュバント、樹状細胞の増殖を刺激するサイトカイン、及び/又は免疫刺激性作用薬と組み合わせて、投与するステップを含む、対象を免疫する方法。
- 抗原と、抗原提示細胞(APC)に結合するモノクローナル抗体との複合体を形成するステップと、
前記抗原に対する細胞傷害性T細胞応答を誘導又は亢進する態様で、前記抗原が内部移行し、プロセッシングされ、そしてT細胞に提示されるように、前記複合体をin vivo 又は ex vivo でAPCに接触させるステップと
を含む、抗原に対する細胞傷害性T細胞応答を誘導又は亢進する方法。 - 更に前記抗原に対するヘルパT細胞応答を誘導又は亢進する、請求項33に記載の方法。
- 前記T細胞応答がCD4+及びCD8+T細胞の両方により媒介される、請求項33又は34に記載の方法。
- 前記T細胞応答が、MHCクラスI及びMHCクラスII経路の両方を通じて誘導される、請求項33乃至35のいずれかに記載の方法。
- 前記抗体が、ヒト樹状細胞上に発現したC型レクチンに結合する、請求項33乃至36のいずれかに記載の方法。
- 前記抗体がヒトマンノース受容体に結合する、請求項33乃至37のいずれかに記載の方法。
- 前記抗体が、ヒト、ヒト化及びキメラ抗体から成る群より選択される、請求項33乃至38のいずれかに記載の方法。
- 前記抗体が全抗体、Fabフラグメント及び一本鎖抗体から成る群より選択される、請求項33乃至39のいずれかに記載の方法。
- 前記抗体が、FR1、CDR1、FR2、CDR2、FR3、CDR3及びFR4配列を含むヒト重鎖可変領域と、FR1、CDR1、FR2、CDR2、FR3、CDR3及びFR4配列を含むヒト軽鎖可変領域とを含み、但し:
(a)前記ヒト重鎖可変領域 CDR3配列が、配列番号 15及びその保存的改変を含み;そして
(b)前記ヒト軽鎖可変領域CDR3配列が配列番号 18及びその保存的改変を含む、
請求項33乃至40のいずれかに記載の方法。 - 前記ヒト重鎖可変領域CDR2配列が配列番号14及びその保存的改変を含み、そして前記ヒト軽鎖可変領域CDR2配列が配列番号17及びその保存的改変を含む、請求項41に記載の方法。
- 前記ヒト重鎖可変領域CDR1配列が配列番号13及びその保存的改変を含み、そして前記ヒト軽鎖可変領域CDR1配列が配列番号16及びその保存的改変を含む、請求項41又は42に記載の方法。
- 前記抗体が、それぞれ配列番号4及び配列番号8に示されたアミノ酸配列、又は、前記抗体が樹状細胞への結合能を保持しているように配列番号4又は配列番号8に充分相同なアミノ酸配列、を含むヒト重鎖及びヒト軽鎖可変領域を含む、請求項41乃至43のいずれかに記載の方法。
- 前記抗原が腫瘍細胞又は病原性生物により発現される、請求項33乃至44のいずれかに記載の方法。
- 前記抗原が、βhCG、Gp100、前立腺関連抗原及びPmel-17から成る群より選択される、請求項33乃至45のいずれかに記載の方法。
- 樹状細胞を、アジュバント、樹状細胞の増殖を刺激するサイトカイン、及び/又は免疫刺激性作用薬に接触させるステップを更に含む、請求項33乃至46のいずれかに記載の方法。
- 前記複合体が対象に in vivo 投与される、請求項33乃至47のいずれかに記載の方法。
- 前記対象が前記抗原に対して免疫される、請求項48に記載の方法
