Abstract
Description
本発明は、ペプチドならびにセリンプロテアーゼ、システインプロテアーゼ、メタロプロテアーゼおよびエンドペプチダーゼ・ファミリーを含む酵素(これらの多くは悪性腫瘍の成長と転移の過程に必須である)の活性なインヒビターとしてのビス-ジフェニルホスフィン-フェロセン配位リガンドを含むシクロパラジウム化化合物ならびにそれらの類似体に関するものである。これらの酵素に作用しDNA分子の挿入に貢献することでこれらの化合物は免疫系を調節する。
薬学分野における無機化学の研究は、一連の病理の処置のため、その利用が伝統的医薬に優る明らかな利点を有するが故に、研究者の特別な注目を受けてきた。
− Xは以下の群:
ハロゲン(Cl、F、Br、I);
擬ハロゲン(N3、NCO、NCS、SCN);または
アセテート(H3C-COO-);
から選ばれる元素を表し;そして
− Yは周期表のVまたはVI族の元素、例えばN、P、As、Sb、Bi、O、S、Se、Teを表し;
− Cはパラジウム原子に共有結合した、sp2またはsp3混成を有する炭素原子を表し;C、YおよびDを含む環は3〜8個の原子で構成されることができ;
− 曲線で表されるCとYの間には、パラジウム原子を含む、3〜8個の原子で構成される、シクロパラジウム環と称する原子の連続があり;典型的には、この原子は炭素、窒素、酸素または硫黄から選ばれるが、他の態様を排除する訳ではない。他方では、この環を構成するこれら原子の各々は、他の原子または基と結合して鎖式または環式の環外可変構造を形成するが、これらに関して出願人は特段の制限をもたない;
− Lは、下記のスキーム2{式中、略図L-Lは、このビス-ジフェニルホスフィン-フェロセン化合物の構造内の2個のリンカーLの存在を表し、一方、R1、R2、R3、R4、R5、R5、R6、R7、R8、R9、R10、R11およびR12は個別に以下のラジカル:水素(H)、アルキル、アリール、ジエニル、アルコキシ、シロキシ、ヒドロキシ(OH)、アミノ(-NH2)、イミド、ハロゲン(F、Cl、Br、I)、イミノ、ニトロ(-NO2)を表し、これらは任意の順序で存在できる。}に詳細を示したビス-ジフェニルホスフィン-フェロセン化合物内部にある周期表のV族由来の供与原子(N、P、As、Sb、Bi)である配位リガンドを表す。]
− 炭素原子(A1)は、シクマ結合によりパラジウム(A5)と結合している。
− 窒素原子(A4、一般式ではYとして包括定義される)は、共有結合供与体結合によりパラジウム(A5)と結合している。
− 原子A2は炭素であり、A1と合してシクロパラジウム環の「外部」にあるベンゼン環の一部でもある。
− 窒素原子A4はシクロパラジウム環の「外部」にある2個のエチルラジカルと結合している。
− パラジウム、原子A5は、ビス-ジフェニルホスフィン-フェロセンに結合しており、ここでR1〜R12は水素原子である。
− スキーム6の点線部分はパラジウムと共にシクロパラジウム環を形成している原子を示す。
実例を補足して、このシクロパラジウム化化合物は1,1'-ビス-ジフェニルホスフィン-フェロセンリガンドと合して、提供できる可能性の1つとして以下に示される複合体構造を生成し得る:
カテプシンB
システイン-プロテアーゼは活性部位にチオール基(-SH)を有する酵素のファミリーを構成する。これらは、既知の経路により、特定のシステインと活性部位のヒスチジンの間の共同相互作用によりアミド結合を開裂することができる(米国特許5880149)。これらのタンパク質は、4またはそれ以上のスーパーファミリーを有する細菌、ウイルス、真核微生物、植物および動物に見出されている。本発明に係るシクロパラジウム化化合物によるカテプシンB酵素の阻害は、これらの化合物により示される抗腫瘍活性の主たる原因として最初に指摘された。しかしながら、本発明化合物の活性はこのシステイン-プロテアーゼに限定されず、医薬としてのその用途は癌に対する闘いに限定すべきではない。
プロリル-オリゴペプチダーゼ
プロリル-オリゴペプチダーゼは30以上の残基を有するペプチドを加水分解できないセリンペプチダーゼの群に属する酵素の1ファミリーを構成する。この群は、基本型のプロリル-オリゴペプチダーゼの他にジペプチジル-ペプチダーゼIV、アシルアミナシル-ペプチダーゼおよびオリゴペプチダーゼBを含む。最近決定されたプロリル-オリゴペプチダーゼの結晶構造(80kDa)は、この酵素が、折り畳まれたα/βヒドロラーゼを伴うペプチダーゼ領域を有し、その触媒性三つ組が、大ペプチドのフィルターとして作用する領域である、稀な7枚のβ折り畳み構造を持つ中央トンネルで構成されている事を示している。プロリル-オリゴペプチダーゼは記憶喪失、鬱病および血圧の制御といった数多くの障害に関わっており、また小胞体およびミトコンドリアの膜を介したタンパク質輸送のための重要なシャペロンとしても作用していることから、このファミリーの酵素の阻害は新薬開発の重要な源である(Sharova, E. I.;Russian Journal of Plant Physiology, 49(2), 255, 2002)。
カテプシンDは酸性エンドペプチダーゼであって、乳癌の診断用因子として役立ち(Kraimps, J. L., Metaye, T., Millet, C., Margerit, D., Ingrand. P., Goujon, J. M., Levillain, P., Babin, P., Begon, F., Barbier, J., Surgery 1995 Dec., 118(6): 1036-40)、骨髄癌腫と甲状腺腫瘍において高濃度に発現される(Holm, R., Hoie, J., Kaalhus, O., Nesland, J. M., Virchows Arch 1995; 427(3): 289-94)。カテプシンDの活性は腫瘍細胞と正常細胞の両者に存在するが、正常細胞よりもそれぞれ癌細胞、腺腫および重篤な甲状腺疾患においてより高い(Metaye, T., Kraimps, J. L., Goujon, J. M., Fernandez, B., Quellard, N., Ingrand, P., Barbier, J., Begon, F., J. Clin. Endocrinol. Metab. 1997 Oct; 82(10): 3383-8)。皮膚の癌腫(メラノーマおよび神経)では、カテプシンBとLの活性が高い。カテプシンDはメラノーマでのみ高レベルに発現される。カテプシンHの活性は損傷の浸潤可能性に逆相関する(Frohlich, E., Schlagenhauff, B., Mohrle, M., Weber, E., Klessen, C., Rassner, G., Cancer, 2001 Mar 1; 91(5): 972-82)。カテプシンD、nm23、EGFRおよびLR(免疫組織化学的に検出)は、陰性結節を持つ乳癌の転移による拡延の距離に対する潜在的マーカーであって、最初の3者の組み合わせはより有望なアプローチである(Niu, Y., Fu, X., Lv, A., Fan, Y., Wang, Y., Int. J. Cancer, 2002 Apr 10; 98(5): 754-60)。免疫組織化学的分析は、癌細胞によるカテプシンDの発現が、胃および腸の腫瘍浸潤の深さに結び付いていることを示している(Ikeguchi, M., Fukuda, K., Oka, S., Yamaguchi, K., Hisamitsu, K., Tsujitani, S., Sakatani, T., Ueda, T., Kaibara, N., Oncology 2001; 61(1): 71-8)。免疫組織化学的分析によれば、カテプシンDの発現は咽喉癌細胞におけるタンパク質p53の発現に結び付いている。カテプシンDは正常な咽喉細胞には検出されなかった。カテプシンDの高発現は、腫瘍の浸潤性成長に有意に結び付いている(Ikeguchi, M., Sakatani, T., Ueta, T., Fukuda, K., Oka, S., Hisamitsu, K., Yamaguchi, K., Tsujitani, S., Kaibara, N., J. Clin. Pathol.. 2002 Feb; 55(2): 121-6)。ラットによる検定は、カテプシンDが、乳癌中のヒトカテプシンDの発現ベクターの移動を介する腫瘍転移プロセスと直接結び付いている事を示している。当該シクロパラジウム化化合物が包含される該酵素のインヒビターは、腫瘍の転移の低下または阻害に直接影響を及ぼす(Marcel Garcia, Nadine Platet, Emmanuelle Liaudet, Valerie Laurent, Danielle Derocq, Jean-Paul Brouillet, Henri Rochefort, Stem Cells 1996; 14: 642-650)。
アンギオテンシン変換酵素(ACE)
アンギオテンシン変換酵素は、その部位の1つに亜鉛を含むメタロ-プロテアーゼの群の一部であって、アンギオテンシンIからアンギオテンシンIIへの変換に関係している。この酵素は、領域Cおよび領域Nと称する二つの領域を持つ、即ち二つの活性中心を持っている。アンギオテンシンIIは造血系(HPC)由来の前駆細胞の増殖を誘導し、一方アンギオテンシンIはこの増殖を阻害する。他方では、ECA酵素の基質でもあるペプチド アセチル-N-Ser-Asp-Lys-Pro(AcSDKP)は造血系由来の親細胞の増殖の調節物質である。
− Chisi et al(Inhibitory Action of the Peptide AcSDKP …., Stem Cells 1997; 15: 455-460)は、造血細胞の増殖をインビトロで調節するための、AcSDKPと適当なACEインヒビターの組み合わせの使用を記載している。有効なACEインヒビターであるカプトプリルの存在下におけるペプチドAcDSKPの作用により、該酵素の活性部位Nに対する効果と共に、幼若細胞周期の顕著な阻害が認められる。
本発明化合物の作用経路は主として可逆的であり、よって低い細胞毒性と望ましくない副作用の低下がもたらされる。
azida(N3)基を含む本発明に係るパラジウム複合体を、該複合体のイオンまたは分子特性およびそれらの単核または二核性に対する正確な構造特性決定のために、この基が赤外領域に「分光プローブ」を構成するよう合成した。この基は、2040-2060cm-1(非対称性伸縮)および1400-1500cm-1(対称性伸縮)の領域にIVの基準振動モードを持ち、前記属性を促進する。この複合体は抗腫瘍および酵素インヒビターとしての生物学的効果をも現すが、これらは細胞呼吸周期に影響を及ぼす望ましくない細胞毒性と副作用を示すため、医薬として使用するための最も適切な化合物とはならない。azida基を含む合成された全複合体について塩化物(Cl-)イオンを伴う類似体を取得したが、前記の二次的効果を示さず、同じように活性な薬物ともならなかった。
実施例1に記載の出発複合体から、本発明の特定態様に従い、1,1'-ビス-ジフェニルホスフィン-フェロセン(dppf)との反応により、数多くのシクロパラジウム化複合体を合成した。得られる生成物は、使用した化学量論および溶媒に依存する。1または2個の金属中心を持ち、パラジウムイオン(II)に関して単歯または二歯のビホスフィン配位リガンドを含む分子およびイオン複合体を単離した。
[Pd(C2,N-dmpa)(L)]X(L=二座リガンド)型のキレート化ビホスフィンリガンドを示すイオン化合物を、出発シクロパラジウム化複合体とビホスフィンリンカー(L-L)間の反応から合成した。ビホスフィンリンカーと各パラジウム複合体間のモルreason 2:1を使用した。即ち、二量体シクロパラジウム化化合物0.2mmolをエルレンマイヤー中のアセトン50mlに部分的に溶解した。次いで(L-L) 0.4mmolを得られた懸濁液に添加した。混合物を室温で1時間絶えず振盪した。最終混合物の溶媒をほぼ乾燥圧力下に蒸発させ、得られた固体をヘキサンを添加して沈殿させた。この固体を引き続きEt2Oで洗浄し、減圧乾燥した。
カテプシンBおよびパパインはCalbiochem Co.社により供給され、活性酵素の濃度はE-64として知られるシステインプロテアーゼインヒビターを用いるtitulationによって決定した。カテプシンBとパパインは、10μM MMTSを含有する50mM酢酸ナトリウム緩衝液(pH5.0)内に4℃で梱包されていた。蛍光生成基質アミドメチルクマリルZ-Phe-Arg-MCA、トリプシンおよび非可逆的パパインインヒビターE-64はSigmaにより供給された。
カテプシンBとパパイン・スーパーファミリーに属するその他のシステインプロテアーゼは高度に保存された折り畳み構造を含んでいる(Turk, V.; Bode, W., Lysosomal cysteine proteinases and their inhibitors cystatins. Innovations in Proteases and their inhibitors. Aviles, F. X.(Ed.), Walter de Gruyter & Co., Berlin, Germany, 1993)。これらの酵素がペプチド基質に結合し、チオラート-イミダゾリウムのイオン対を利用してそのタンパク質分解活性を働かせる。システイン残基とヒスチジンの間のこの結び付きが、システインの活性部位に高い求核性を付与する(Michaud, S.; Gour, B. J., Cathepsin B inhibitors as potential anti-metastatic agents. Exp. Opin. Patents., 1998, 8, 645-672)。基質のアミド結合の破壊は、中間体アシル酵素の形成を含む。ミカエリスの非共有複合体の形成の後、活性部位のチオラートがペプチド結合を攻撃してオキシアニオンを形成し、これがグルタミン残基によるいわゆる「オキシアニオンチャネル」中で安定化される。四面体中間体の崩壊は、結果としてアシル酵素を生成し、生成物を放出する。その後このアシル酵素の加水分解が触媒性イオン対を再生し、新たな生成物、即ちカルボン酸を放出する。実施例5は、本発明化合物による酵素阻害検定を示している。

円二色性技術による研究(図2)は、時間および薬物濃度に依存する、DNA分子中のシクロパラジウム化化合物の惹起する構造変化を示している。これは、それらがアポトーシスと細胞周期調節プロセスに関与している事を意味している。
本発明化合物を、被験動物としてのラットと、充実性、浸潤性および転移性腫瘍のモデルとしてのWalker-256乳癌、「ウォーカー腫瘍」を用いるインビボモデルで試験したところ、この化合物は大きな活性を示した。実施例6ならびに図3、4および5に実験および得られた結果を示す。
エールリッヒ腹水腫瘍(EAT)
本発明化合物で処置した動物の防御可能性を観察する目的で、処置したまたはしなかったエールリッヒ腹水腫瘍を持つ動物を考慮して生存延長曲線を描いた。

リガンド1,1'-ビス-ジフェニルホスフィン-フェロセンを含有するシクロパラジウム化化合物で連続4日間処置した動物の骨髄の顆粒球およびマクロファージの造血前駆体(CFU-GM)の数を、1:2の構造(スキームC(一般的な構造)による)で評価した時(この方法は実施例8に示した)、1mg/kg未満の用量で骨髄毒性の欠如が確認できた。何故なら対照群に比して処置動物では顆粒球/マクロファージの造血前駆体(CFU-GM)の数に有意差が無く、よってこの用量未満では骨髄毒性が無い事が示された。同様の結果が同用量のインビトロ試験(正常動物細胞を当該化合物と共にインキュベート)で確認された。他方では、5mg/lより高い用量では用量依存的なCFU-GM数の減少が惹起され、骨髄に対するこの化合物の効果が用量依存的であることを示した。
長期持続液体培養系における1:2構造(スキームC(一般的な構造)による)のリガンド1,1'-ビス-ジフェニルホスフィン-フェロセンを含有するシクロパラジウム化化合物の効果を、骨髄ストロマの形成に及ぼす該化合物の効果を評価できる実施例9に示した方法に従って評価した時、これらの細胞を該化合物に長時間暴露すると、この液体培養の上清由来の非付着細胞の数(図7)、およびこの培養の上清から得た細胞コロニー数(図8)の両者が減少する事が確認できた。他方では、対照に比してシクロパラジウム化生成物の存在下では、骨髄ストロマ由来の付着細胞数に顕著な増加があった(図9)。この応答標準は、骨髄の多能性細胞(幹細胞)が細胞周期に入るのを阻害するよう働く、アンギオテンシン変換酵素(ECA)、例えばカプトプリルを阻害する薬物で観察されている。故に、細胞周期の一時的中断は、化学療法の骨髄毒性効果に対する骨髄細胞の優れた防御に結びつき得る。
この試験に使用する本発明に係るシクロパラジウム化化合物の濃度を固定用量試験の実施により決定する。この試験では、被験物質を、統治機関による分類に従って先に定められた用量から選択した特定の用量でマウスに投与するが、最大被験濃度は2000mg/kgを超えてはならない。LD50試験ではなく固定用量試験を使用する利点は、固定用量試験では、評価された標準は必ずしも動物の死を必要とせず、したがって各実験に用いられる動物の苦痛および数を最小にできる事である(Barros & Davion, 1996)。
ミトコンドリアオキシ還元系による細胞増殖と細胞毒性の両者が評価できる、MTT-テトラゾリウム(ジフェニルテトラゾリウム3-(4,5-ジメチアゾール-2-イル-2,5-ブロミド))還元の方法による細胞毒性の評価を使用した。この方法により、テトラゾリウムが生存細胞によって還元され、Formazan塩を生成し、これを可溶化して(Mosmman, 1983)ELISAを介して560nmの吸光度を読み取る。
マウスメラノーマB16F10-Nex2細胞に対するシクロパラジウム化化合物の細胞毒性効果を証明するため、インビトロ生物検定を実施し、この細胞を薬物と共に24時間インキュベートした。雌性マウスC57Bl/6におけるインビボ検定もまた実施し、105の腫瘍細胞を皮下移植した。腫瘍の移植後、10μM濃度のシクロパラジウム化化合物を週に3回腹腔内に適用することにより、処置を開始した。
この結果を実施例12および図13、14、15および16に示す。
甲状腺腫瘍
WRO、NPAおよびARO系由来の甲状腺腫瘍細胞培養で検定を実施した。[Pd(C2,N-(S(-)dmpa)(L)X]型の分子化合物は、WRO系で0.48μg、NPA系で0.59μg、そしてARO系で0.60μgの阻害用量(IC50)を示した。化合物[Pd2(C2,N-(S(-)dmpa)2(μ-L)X2]は類似の阻害用量(IC50)を示した。dmpa由来のエナンチオマーR(+)により製造した類似のシクロパラジウム化化合物については以下の阻害用量(IC50)が得られた:WRO系で5.56μg、NPA系で6.04μg、そしてARO系で7.57μg。さらに本発明者等は、[Pd2(C2,N-(R(+)dmpa)2(μ-L)X2]型の化合物について、これらの阻害用量が著しく変化しないことを観察した。記載した結果から、R(+)エナンチオマーに関しては、阻害用量IC50はS(-)エナンチオマーで観察された用量に比して約10倍高いことが明らかとなった。この事実は、該化合物の惹起する細胞毒性プロセス由来のエナンチオ選択的依存性を示しており、dmpa由来のS(-)異性体を構造内に有する該化合物の高い有効性を示すものである。
これらの結果はさらに、免疫調節物質および免疫抑制剤としての本発明化合物の活性を示すものである。
これらの同じ検定において、本発明者等は、多くの骨髄ストロマコロニーが赤血球系由来の特性を示す事を証明したが、それは赤血球系に対する刺激活性の可能性を示している。
本発明の目的のため、受容者は動物または人間、特に人間とすることができる。
本発明に係るシクロパラジウム化化合物および少なくとも一つの本発明化合物を含む組成物は、該投与に対して当分野で知られる任意の薬学上許容し得る用量を用いることにより、経口的に、注射手段により、特に腹腔内に投与することができる。或る特定の態様では、該化合物および本発明化合物を含む組成物は、注射として供給された場合に消化系による変化を受けない。
「造血系」とは、血液細胞の形成に関係する系を意味する。
「受容者血液の総体積」とは、受容者の生体に循環している血液の総体積を意味する。人間の場合、この体積は6および8リットルの間である。
本発明の具体的態様の単なる実例となる実施例を以下に述べるが、付記した請求項に内包した範囲を除けば、その範囲にいかなる限定も創成してはいない。

抗腫瘍アッセイ - ウォーカー腫瘍
抗腫瘍活性アッセイのために、その全てが1%ジメチルスルホキシド(DMSO)を含む、pH = 7.4の、水溶液、生理食塩液、およびリン酸緩衝液中に化合物を希釈した。試験動物の血液の総量(30 mlと推定される)中のそれらの濃度が、合計で生体内で15 μMの薬物濃度になるように、化合物の同等分量の適用がなされ、しれが動的ステップに非常に有益であるとみなされた。
腹膜内、皮下、そして腫瘍移植部位への直接的な適用、さらに本発明パラジウム化合物と一緒に、腫瘍細胞を用いた腫瘍移植によって、実験をおこなった。
エールリッヒ腹水腫瘍を成長させるために、保持マウスの腹膜腔由来の、106細胞を含む腫瘍細胞の懸濁液0.1 mlを、マウスに腹腔内接種する。エールリッヒの腫瘍を持つマウスの腹膜から腹水を取り出した後に、ノイバウエル・チェンバでトリパンブルー染色材を使って、細胞の数および生存率を測定した。
エールリッヒ腫瘍細胞を保持動物の腹膜腔から取り出し、そして細胞生存率を測定するためにトリパンブルー溶液中に希釈した(1:100)。濃度を1×106細胞/動物に、生理食塩液で調節した。そして、0.2 mlのこの溶液(1×106細胞を含む)を研究される動物に接種した。腫瘍接種の72時間後に、シクロパラジウム化化合物による処置を開始し、そして連続した4日間続けた。死亡率を管理するために前記動物を毎日観察した。
マウスの骨髄由来の造血前駆体(CFU-C)をクローン培養
これらのクロージェニック細胞を数えるために、培養物中にある全ての多能性細胞が増殖するように誘導され、そして培養条件が、それらの結果的な重ね合わせによる各々のペトリ板上での多すぎるコロニーを避けるように、各々のコロニーの同定を可能にするように、調節されることが重要である。同様に、成長刺激因子(GSF)が最大濃度を上回って使われることが重要である。我々は、顆粒球およびマクロファージ-のために組み換え造血性増殖因子 - GM-CSFを使う。さらに、様々なバッチと市場で入手可能な商標の活性に見られた変動性のために、ウシ胎児血清の選択は慎重にされるべきである。
長期持続骨髄細胞液体培養手順による骨髄ストロマ形成の評価
この培養は、骨髄のストロマ細胞由来の接着細胞の単層の形成に完全に依存している。接着細胞のこの層の形成は、ストロマ細胞の細胞外マトリックスの形成に関与する造血性増殖因子の合成および分泌によって生じる。よって、より未熟な細胞が接着細胞のその層の中に残っていて、そして造血を維持するという目的で、培地に放出される。このため、液体培地中の始原細胞の数は、外因性造血性増殖因子の存在により、効果的なクローン培養後に定量される(Spooncer et al, 1993)。
長期持続液体培養培地中に得られた始原細胞からの造血前駆体(CFU-C)のクローン培養
長期持続液体培養系から非接着細胞を得た後に、これらを培地RPMI 1640 (Sigma)により洗浄し、2×105細胞/mlの最終濃度で再懸濁した。先の項目に記載のとおり、その後これらの細胞をクローン培養(CFU-C)プロトコールに供した。
急性毒性についての評価
アッセイした化合物を、10%のDMSOを含む生理食塩液中に希釈し、そして0.2 mlのその溶液を動物に腹腔内投与する。これにより、動物10匹の群(5匹の雄と5匹の雌)を様々な量の検討される化合物で処理した。対照動物には、10%のDMSOを含む生理食塩液だけを与えた。処置終了後に、前記動物は14日間の観察は続いた。
シクロパラジウム化化合物の急性毒性についての評価。動物に、以下に記載の化合物の1服用量を腹腔内投与し、そしてこの手順の後14日間観察した。
白血病株HL-60とK-562、および癌原遺伝子BCL-2の発現における生体外の細胞毒性。
2 mMのL-グルタミンと20%のウシ胎児血清を補った培地RPMI-1640 (Sigma)中で維持した細胞株HL-60とK-562からの細胞を、前もって洗浄(3回)して、そしてRPMI培地中に再懸濁した。その後、それらを96穴(オリフィス)の培養プレート(1×106細胞/オリフィス)上で、検討される化合物の9つの濃度(160, 80, 40, 20, 10, 5, 2.5, 1.25および0.65 μg/ml)と一緒にインキュベートした。それらをウェット・オーブン(空気中5%のC02)内で72時間インキュベートした。この期間の後に、MTT溶解(5 mg/ml)(Sigma)を加え、そして培養プレートをさらに4時間インキュベートした。その後、MTT安定化溶液(0.04 Nのイソプロピル酸)を加えて、そして30分後、ELISAの読み取りを3つのコピーで560 nmにておこなった(Mosmann, 1983)。検討される化合物の存在下での細胞生存百分率を以下の通り計算した:
メラノーマを用いてなされる生物学的アッセイ
生体外アッセイ
げっ歯類のメラノーマB16F10-Nex2細胞のシクロパラジウム化の細胞障害効果を確認するために生体外アッセイをおこなった。異なる濃度下で24時間前記細胞をインキュベートした。細胞生存率を、MTTを用いたアッセイによって測定した。図13に示されるように、100%の有効性は、エタンビホスフィンを含む薬物(薬物1, 2および3)の1.25 μMの濃度下で観察される。同じ有効性は、ビス-ジフェニルホスフィン-フェロセンを含む薬物で、しかしより高い濃度である10 μM(薬物6)で、そして官能化アルキンを含むシクロパラジウム化化合物(薬物10)については、約20 μMで、示される。図13は、細胞生存百分率の減少が薬物なしの細胞を用いた対照と比較して棒の上に表される、前述の細胞障害効果を示す。

- 前記疾患が、組織崩壊により引き起こされる疾患、例えば関節炎、筋ジストロフィー、腫瘍浸潤、糸球体腎炎、寄生虫による骨感染、パラサイトミー、歯周病および腫瘍転移;心房性ナトリウム利尿因子の分解の関与する心疾患;炎症性疾患、例えば気管支炎、関節リウマチ、骨粗鬆症、急性膵炎および癌の進行;障害、例えば記憶喪失、鬱の制御および血圧;糖尿病、トリパノソーマ症、シャーガス病、食餌障害、神経性過食症および食欲不振;アルコール中毒、サイトカインおよびサイトカイン・レセプター、例えばインターロイキン-6(IL-6)、腫瘍壊死因子α(TNF-α)、インターフェロン-γ(INFN-γ)、IL-1アンタゴニスト・レセプター(IL-1RA)、IL-10および顆粒球-マクロファージコロニー増殖刺激因子(GM-CSF)の産生に関係する疾患、心理的ストレス、HIVウイルスによって引き起こされる感染症(AIDS)に対する闘病;多発性硬化症および自己免疫脳脊髄炎を包含するミエリン崩壊を引き起こす中枢神経系の炎症性疾患;癌腫瘍、自己免疫疾患、腹水もしくは固形腫瘍、乳房の腫瘍、骨髄腫、アデノーマ、甲状腺腫、メラノーマ、胃腫瘍、腸腫瘍、食道腫瘍および甲状腺腫瘍を含む腫瘍、ならびに神経芽細胞腫を含む、請求項83に記載の使用。
