Abstract
【解決手段】 ナイーブ表現型を有する末梢CD25+CD4+T細胞を培養し、制御性T細胞を得ることを含む、制御性T細胞の製造方法が提供される。該CD25+CD4+T細胞は抗原刺激下で培養され得る。当該方法によれば、インビトロで哺乳動物の制御性T細胞を製造することが可能であり、抗原特異的な制御性T細胞を製造することも可能である。当該方法により製造された制御性T細胞は免疫調節薬として臓器移植における拒絶反応、アレルギー疾患、自己免疫疾患、移植片対宿主病(GVHD)、不妊症等の予防・治療に有用である。

Description
近年、CD25+CD4+制御性T細胞(制御性T細胞をTreg(s)という場合がある)が臓器移植後の免疫寛容に重要な働きをしているとの知見が、動物実験のレベルで、急速に集積されつつある。本発明者らは、生体肝移植後に免疫寛容が自然に成立した患者において同細胞が増加していることを確認した(非特許文献1)。また、マウスの系では、本発明者らにより、制御性T細胞をいったん分離して試験管内でドナー抗原特異的に培養し、再度体内に戻すいわゆる細胞養子免疫の手法を用いることで皮膚移植片が拒絶されないことが示されている(非特許文献2)。これらのことから、ヒトの移植医療においても、免疫寛容を積極的に誘導する有効な方法として、制御性T細胞の試験管内での増殖、患者への移入、すなわち細胞養子免疫療法が考えられる。試験管内で制御性T細胞を増殖させた場合に、その制御能が維持されるということは細胞養子免疫療法を行ううえで、必須であるが(Nat Rev Immunol., Mar;3(3), 199-210, 2003)、ヒトでは制御性T細胞を制御能を失うことなく試験管内で高倍率に増殖させたという報告がない。
ヒトCD25+CD4+制御性T細胞は、インビボで、内在性の自己抗原に対する免疫寛容において、そしてより最近では臓器移植後の移植免疫寛容において、必須な役割を果たしていることが報告されている(Annu Rev Immunol., 22, 531-562, 2004、Nat Rev Immunol., 2, 389-400, 2002、Nat Rev Immunol., 3, 199-210, 2003)。インビボにおいて、ヒトTregsは、細胞表面CD4+及びCD25high+、Foxp3発現、細胞内CTLA-4、細胞表面GITR、TNFRII、及び自己及びアロ抗原に対する不応答性/免疫抑制能等の特性に加えて、メモリー表現型−CD45RO+によって特徴付けられる(J Immunol., 167, 1245-1253, 2001、J Exp Med., 193, 1285-1294, 2001、J Exp Med., 193, 1295-1302, 2001、Blood, 98, 2736-2744, 2001、Eur J Immunol., 32, 1621-1630, 2002、Int Immunol., 16, 1643-1656, 2004、Exp Hematol, 32, 622-629, 2004)。実験的には、胸腺細胞からのCD25+CD4+細胞を除去したものを免疫不全宿主に対して養子移入すると、自己免疫疾患を発症し(J Immunol., 162, 5317-5326, 1999)、また、あるげっ歯類モデルにおいては、移植前の胸腺摘出により、Tregs介在性の移植免疫寛容が成立しなくなる(Transplantation, 76, 588-596, 2003)。これらの実験的証拠に一致して、高いレベルのFoxp3を発現しているCD25+CD4+T細胞がヒト胸腺中に存在している(Blood, 102, 4107-4114, 2003、Eur J Immunol, 35, 383-390, 2005)。これらの観察に基づけば、Tregsは胸腺内に起源し、末梢に向かって移出し、末梢免疫寛容の成立に寄与しているようである。ヒト胸腺中のCD25+CD4+T細胞は、Foxp3を発現し、不応答性/免疫抑制能の特性を示すにも関わらず、CD45ROの代わりに、ナイーブ細胞の表現型であるCD45RAを呈する(非特許文献3−7)。これは、末梢中のTregsと胸腺内のCD25+CD4+T細胞は表現型として異なっていることを意味する。従って、胸腺由来のナイーブ表現型を呈するTregは、メモリー表現型を呈するTregsに発生するはずである。しかしながら、末梢におけるTregsの分化カスケードは、動物モデルにおいてもヒトにおいても理解されないままである。従って、胸腺由来の「ナイーブタイプ:未成熟Tregs」が、どのように、そしてどこで、「メモリータイプ:成熟Tregs」の特性を獲得するのかということについての疑問が生じる。
末梢血中のCD45RA+CD25+CD4+T細胞は、これまでTregsからは異なる存在と考えられていた。なぜなら、この集団は不応答性の特性も抑制性の特性も示さず、むしろ、アロ抗原及び作動性抗CD3抗体の刺激により増殖を示すからである(非特許文献8)。その代わりに、末梢では、不応答性/免疫抑制能の特性を示すCD45RA-CD25+CD4+T細胞がTregsを代表している。しかしながら、以下に言及する点は興味深い;
(1)末梢血中のCD45RA+CD25+CD4+T細胞は、胸腺、または臍帯血中のCD25+CD4+T細胞の有するナイーブ細胞の表現型であるCD45RA+を共有する(非特許文献7、9)。
(2)幾つかの研究が、末梢血から単離された時点でのみ、CD45RA+CD25+CD4+T細胞の機能的な面をみているが(非特許文献8、10、11)、インビトロ培養後のその機能の変化や、遺伝子的特徴は、いまだ詳細には研究されていない。

即ち、CD45RA+CD25+CD4+T細胞は、多くのナイーブ表現型を呈し、その頻度は年齢依存的な様式で徐々に低下した。末梢のTregsとは異なる表現型を呈するにも関わらず、この集団は非常に高いFoxp3レベルを発現し、Tregsと同様に、細胞内にCLTA-4と細胞表面にTNFRIIを発現していた。末梢血から分離された時点では、Tregsとは異なり、この集団は不応答性の特質も免疫抑制能の特質も示さず、しかしむしろ、作動性抗CD3抗体の存在下で、アロ抗原に対して増殖を示した。そして、重要なことには、該集団は、アロ抗原と培養・増殖された後に、Tregsの有する不応答性/抗原特異的免疫抑制能の特性を獲得した。
これらの観察結果に基づき、本発明者らは本発明を完成するに至った。即ち、本発明は以下に関する。
(1)ナイーブ表現型を有する末梢CD25+CD4+T細胞を培養し、制御性T細胞を得ることを含む、制御性T細胞の製造方法。
(2)該ナイーブ表現型がCD45RA+、CD45ROlow+、CD45RBhigh+、CD62Lhigh+、及びCD38+からなる群から選択される少なくともいずれかである、上記(1)記載の方法。
(3)該CD25+CD4+T細胞はFoxp3+、細胞内CTLA-4+、及びTNFRII+からなる群から選択される少なくともいずれかの表現型を有する、上記(1)記載の方法。
(4)該CD25+CD4+T細胞は霊長類由来である、上記(1)記載の方法。
(5)該CD25+CD4+T細胞は抗原存在下で培養される、上記(1)記載の方法。
(6)該抗原は免疫学的非自己細胞である、上記(5)記載の方法。
(7)該免疫学的非自己細胞は同種異系細胞である、上記(6)記載の方法。
(8)得られ得る制御性T細胞が該抗原特異的である、上記(5)記載の方法。
(9)該CD25+CD4+T細胞はT細胞増殖因子の存在下で培養される、上記(1)記載の方法。
(10)該制御性T細胞は、CD45ROmid+ 、CD45RBhigh+、CD62Llow+、及びGITR+からなる群から選択される少なくともいずれかの表現型を有する、上記(1)記載の方法。
(11)上記(1)〜(10)のいずれか1つに記載の方法により製造され得る制御性T細胞。
(12)CD45ROmid+ 、CD45RBhigh+、CD62Llow+、及びGITR+からなる群から選択される少なくともいずれかの表現型を有するCD25+CD4+制御性T細胞。
(13)上記(11)又は(12)に記載の制御性T細胞を有効成分として含有してなる、免疫調節剤。
(14)免疫寛容誘導用である、上記(13)記載の剤。
(15)以下の工程を含む、免疫調節剤の製造方法:
(1)ナイーブ表現型を有する末梢CD25+CD4+T細胞を培養し、制御性T細胞を得る工程;
(2)該制御性T細胞を医薬として許容される担体と混合し、免疫調節剤を製造する工程。
(16)ナイーブ表現型を有する末梢CD25+CD4+T細胞を提供するための試薬を含む、制御性T細胞製造用キット。
例えば、ヒトのナイーブ表現型を有する末梢CD25+CD4+T細胞を単離・精製する場合、先ず、末梢組織(例えば末梢血液等)より、単核球分画が調製される。単核球分画の調製は、例えば、Ficoll-Hypaque (Amersham Biosciences-Uppsala)等を用いた密度勾配遠心や、アフェレーシス等により行われる。次に、蛍光色素や磁気ビーズ等により標識された、目的とする細胞の細胞表面に特異的に発現している抗原(CD25、CD4、上述のナイーブ表現型のマーカー抗原(CD45RA等)等)に対する特異的抗体により単核球分画を染色し、セルソーターや磁性カラム等を用いて目的とする分画を単離・精製する。高い精製度を達成するため、好ましくは、セルソーターが用いられる。
細胞培養は、リンパ球培養技術において通常用いられている培養条件を用いることができる。例えば、培養温度は通常約30〜40℃の範囲であり、好ましくは約37℃が例示される。CO2濃度は通常約1〜10%の範囲であり、好ましくは約5%が例示される。湿度は通常約70〜100%の範囲であり、好ましくは約95〜100%が例示される。
抗原とは、培養細胞上の抗原受容体(例えばT細胞受容体)に認識され、該受容体を介して細胞を刺激し得る物質を包括的に意味する。抗原としては、例えばペプチド、タンパク質、脂質、糖脂質等の抗原分子のみならず、免疫学的非自己細胞、抗原受容体の構成分子(CD3、TCRβ、TCRα等)や補刺激分子(CD28等)を認識する作動性抗体(例えば抗ヒトCD3抗体であるOKT-3等)やスーパー抗原などの抗原ミミックをも含む。ここで、免疫学的非自己細胞とは、同種同系(syngenic)細胞以外の細胞をいい、同種異系(allogenic)細胞及び異種(xenogenic)細胞が含まれる。免疫学的非自己細胞は、好ましくは、同種異系(allogenic)細胞である。
例えば、ある特定の免疫学的非自己細胞に特異的な制御性T細胞を製造しようとする場合は、ナイーブ表現型を有する末梢CD25+CD4+T細胞が、免疫学的非自己細胞の存在下で培養される。該免疫学的非自己細胞は、自体公知の方法、例えば放射線(ガンマ線等)照射や抗癌剤(マイトマイシンC等)処理等で不活化されていることが好ましい。免疫学的非自己細胞の種類は特に限定されず、所望の組織由来の細胞(例えば末梢血単核球細胞(PBMC)等)を使用することが可能である。例えば、同種異系のドナーからの移植に先立ち、当該ドナー由来細胞に特異的なレシピエント制御性T細胞の製造が意図されている場合には、レシピエント由来のナイーブ表現型を有する末梢CD25+CD4+T細胞が、ドナー由来の細胞の存在下で培養される。この場合、該ドナー由来の細胞は、移植されうる臓器と同一臓器由来の細胞であっても、異なる組織由来の細胞であってもよい。
即ち、後述の実施例に示される様に、本発明の方法により獲得され得る細胞は、細胞接触が許容される条件で反応性T細胞と共に培養され、T細胞受容体を介する刺激を受けたときに、当該反応性T細胞の活性化を阻害し得る能力(免疫抑制能)を有する。さらに、獲得される細胞は、不応答性の特性をも有し得る。
重要なことに、本発明の方法において、ナイーブ表現型を有する末梢CD25+CD4+T細胞が、抗原存在下で培養された場合、獲得され得る制御性T細胞は、培養時に用いられた抗原に特異的であり得る。
また、本発明の方法により製造され得る制御性T細胞は、CD45RA-CD25high+CD4+ T細胞と比較するとCD62Lの発現が弱く(CD62L弱陽性(CD62Llow+):発現レベルはCD45RA-CD25high+CD4+T細胞の通常約1/3〜1/30倍程度)、またGITR陽性(GITR+)の表現型を有し得る。
また、本発明の方法により製造され得る制御性T細胞は、CD45RA-CD25high+CD4+ T細胞と比較するとCD45RBの発現が強く(CD45RB強陽性(CD45RBhigh+))、発現レベルはCD45RA-CD25high+CD4+T細胞の通常約1.5〜8倍(例えば2.5〜4.5倍)程度で有り得る。
従って、本発明はCD45ROmid+、CD45RBhigh+、CD62Llow+、及びGITR+からなる群から選択される少なくともいずれかの表現型を有するCD25+CD4+制御性T細胞を提供するものである。
本発明の制御性T細胞の投与量は、投与対象、対象臓器、症状、投与方法などにより差異はあるが、通常、成人の患者(体重60Kgとして)においては、例えば非経口投与の場合、一日につき約6x109個程度を上限とする有効量を投与するのが好都合である。他の動物の場合も、60kg当たりに換算した量を投与することができる。
(細胞調製)
0〜69歳の範囲の異なる年齢の健常ボランティアから静脈血が獲得された。20歳より若い対象者は、軽度の先天性の心疾患のため、京都大学病院小児科にてフォローされ、該対象者のために親の許可を得た。末梢血単核球細胞(PBMC)がFicoll-Hypaque (Amersham Biosciences-Uppsala)密度勾配遠心により単離された。細胞は、アロフィコシアニン(APC)抱合抗CD4モノクローナル抗体(mAb)、フィコエリスリン(PE)抱合抗CD25 mAb、及びフルオレッセインイソチオシアナート(FITC)抱合抗CD45RA mAbにより、30分間、4℃にて暗中で染色された後に、以下の6つの異なる細胞分画が、BD FACSAriaセルソーター(Becton Dickinson)を用いて単離された(図1参照)。

上述の(a)〜(e)に示された細胞分画は、図1C中に示された定義に従って単離された。
全てのmAbsはBecton Dickinsonから購入された。それぞれの分画の純度は>98%であった。機能アッセイをするための細胞分離のために、CD4+細胞が、プレートに結合した細胞を除去した後に、抗CD4 mAbマイクロビーズ及び磁性細胞ソーティング(MACS)システム磁性カラム(Miltenyi Biotec GmbH)を用いて、積極(positively)単離された。CD4+ 細胞の純度は>95%であった。
対象者由来のPBMC、或いは増殖されたT細胞株が、30分間、4℃、暗中にて、多様な抗体とインキュベートされ、洗浄され、BD FACSAriaセルソーター及びFACSDiVA ソフトウェア(Becton Dickinson)を用いて解析された。使用された抗体は、perCP抱合抗CD4 mAb、ビオチン抱合抗CD25 mAb、FITC抱合抗CD45RA mAb、PE抱合抗CD45RB mAb、抗CD45RO mAb、抗CD62L mAb、抗CD38 mAb、抗GITR mAb、及び抗TNFRII mAbを含む。CTLA-4の細胞内染色を行うために、細胞は、細胞表面染色、固定、及びCytofix/Cytoperm溶液(Becton Dickinson)による浸透化処理の後に、PE抱合抗CTLA-4により細胞内染色された。
全てのmAb、ストレプトアビジン−APCは、Beckton Dickinson、Immunotech、又はGenzyme/Techneより購入された。
上述の様にセルソーターで分離されたCD45RA+CD25+CD4+又はCD45RA+CD25-CD4+ T細胞(1 x 10^5 細胞/well)が、96ウェルプレートに、1ウェルあたり130μlの増殖培地(10% FBS又はヒトAB型血清、HEPES、ピルビン酸ナトリウム、L−グルタミン、2-ME、及び抗生物質・抗真菌薬(antibiotics-antimycotics)が補充されたRPMI-1640)中で、200 U/mlヒトIL-2の存在下、2 x 10^5 細胞/wellの放射線照射(40Gy)された同種異系のPBMCで刺激された。3〜4日毎に、培地の半分がIL-2を含有する新鮮な増殖培地と交換された。必要であれば、細胞が分けられ、繰り返し同種異系のPBMCで、7〜10日毎に再刺激された。細胞数が十分増加したら、細胞が回収され、48ウェルプレート中で放射線照射された同種異系のPBMC(1 x 10^6 細胞/well)と共に培養された(0.5 x 10^5 細胞/well)。増殖されたT細胞株は3回洗浄された後で機能アッセイのために使用された。
表示された数の、単離されたばかりのCD45RA-CD25high+CD4+T細胞、CD45RA+CD25+CD4+T細胞、又は増殖されたT細胞株が、5 x 10^4 個の単離されたCD4+ T細胞及び1 x 10^5 個の放射線照射(40Gy)された同種異系のPBMCと、0.5μg/mlの抗ヒトCD3抗体(OKT-3)と共に、あるいはそれなしで、丸底96ウェルマイクロプレート中、10%FBSを含む増殖培地中で、それぞれ、刺激された。中和アッセイのために、IL-10R(5μg/ml)、TGF-β(0.5μg/ml)、CTLA-4(5μg/ml)、又はPD-1(5μg/ml)に対する中和抗体が添加された。トランスウェル試験のために、等しい数の、単離されたばかりのCD4+T細胞と1 x 10^6 個のT細胞株が共培養されるか、CD4+T細胞とT細胞株が同種異系PBMC(3 x 10^6個)と共にチャンバーの底と上に分離培養された。培養物は5日又は7日間(それぞれ、OKT-3と共に、あるいはそれなしで)インキュベートされ、最後の16時間の間に[3H]タイミジンでラベルされた。培養物が回収され、[3H]タイミジン取込が1450 Microbeta TriLux(Wallac)により測定された。
全RNAが、CD4+T細胞分画から、Isogen試薬(Nippon Gene)を用いて、製造者提供の指示書に従い抽出され、そして、10 mM DTT、0.5 mM dNTPs、1 x M-MLV RT緩衝液、40 ngランダムプライマー p(dN)6、6U リボヌクレアーゼインヒビター、及び40U M-MLV逆転写酵素 (Invitrogen)を含む、20μl 反応溶液中で、37℃で1時間インキュベートし、引き続き70℃で10分間加熱することによりcDNAが合成された。FOXP3 mRNAの発現レベルは、ABI/PRISM7700シークエンス検出システム(Applied Biosystems)及びQuantiTect Probe PCR kit(Quiagen)を用いて、リアルタイムPCRにより定量された。FOXP3特異的プライマー及び内部蛍光Taqmanプローブが、既述のようにデザインされた(非特許文献8参照)。20xプライマー及びHPRTのためのプローブ混合液は、Applied Biosystemsより購入された。FOXP3 mRNAの発現レベルは、ハウスキーピング遺伝子であるHPRT mRNAのそれにより標準化された。データはトリプリケイトウェルからの平均値で表された。標準偏差は<5%であった。
(異なる世代におけるCD4でゲートされたCD45RA+CD25+CD4+T細胞の割合)
図1AにCD45RA+CD25+CD4+T細胞分画の代表的なFACSプロファイルを示す。20歳未満の子供では、CD45RA+CD25+CD4+T細胞分画は、大きく、密集したクラスターを示したが(図1A;左上及び右上)、20歳を超えた成人においては、子供と比較して、同一の細胞のより小さく、密度の低いクラスターを示した(図1A;左下及び右下)。
図1Bに示されるように、CD4+T細胞中のCD45RA+CD25+細胞の割合は、5歳未満の幼児において最も高く、年齢依存的な様式で徐々に低下し、60歳を超えた成人において最も低かった。
Foxp3の発現レベルが以下の6つの異なる細胞分画について測定された。
(a) CD45RA+CD25+CD4+T細胞
(b) CD45RA-CD25high+CD4+T細胞
(c) CD45RA-CD25low+CD4+T細胞
(d) CD45RA-CD25-CD4+T細胞
(e) CD45RA+CD25-CD4+T細胞、及び
(f) 全CD4+T細胞
これらの細胞は、セルソーターを用いて、図1Cに示されたゲートに従ってPBMCから単離された。CD45RA-CD25high+CD4+(b)及びCD45RA+CD25+CD4+(a)T細胞分画が、CD45RA-CD25low+CD4+(c)、CD45RA-CD25-CD4+(d)、CD45RA+CD25-CD4+(e)、及び全CD4+(f)T細胞分画と比較してより高いFoxp3レベルを発現していた(図1D)。CD45RA+CD25+CD4+細胞分画は、Tregsからは異なるものと考えられてきたにもかかわらず、CD45RA-CD25high+CD4+細胞分画のそれよりさえも高いFoxp3レベルを発現していた。異なった個体からの2回以上の試験が行われ、同様の結果が得られた。
CD45RA+CD25+CD4+(a)、CD45RA-CD25high+CD4+(b)、及びCD45RA+CD25-CD4+(e)T細胞(図1C参照)が、ナイーブ/メモリー細胞表現型、並びに細胞内CTLR-4及びTNFRII(図2)について調べられた。CD45RA-CD25high+CD4+(b)T細胞はCD45RO+、CD45RBlow+、CD62Llow+、及びCD38-を示した。これは、CD45RA-CD25high+CD4+T細胞は、表現型としてはメモリー型であることを示している。しかしながら、全体としては反対の傾向がCD45RA+CD25+CD4+(a)T細胞において観察された。例えば、CD45RA+CD25+CD4+T細胞はCD45ROlow+、CD45RBhigh+、CD62Lhigh+、及びCD38+であった。これは、CD45RA+CD25+CD4+T細胞は、表現型としてはナイーブであることを示す。CD45RA+CD25+CD4+T細胞におけるこれらの4つのナイーブ/メモリー細胞表現型は、典型的なナイーブ細胞であるCD45RA+CD25-CD4+T細胞(e)のそれと同等であった。CD45RA+CD25+CD4+T細胞は、表現型としてはナイーブであったが、この細胞はTregs(これはCD45RA-CD25high+CD4+細胞に相当する)に特異的な細胞内CTLR-4及びTNFRIIを示した。これらの2つの細胞分画とは異なり、典型的なナイーブ細胞であるCD45RA+CD25-CD4+細胞は、細胞内CTLA-4及びTNFRIIを示さなかった。異なった個体からの2回以上の試験が行われ、同様の結果が得られた。
PBMCから単離されたときの、CD45RA+CD25+CD4+T細胞分画の機能的特性が詳細に調べられた(図3)。分離されたCD45RA+CD25+CD4+T細胞は、同種異系PBMC及び作動性抗CD3抗体の存在中では、不応答性ではなく、むしろ増殖性であった(図3A、レーン3、黒色カラム)。一方、CD45RA-CD25high+CD4+T細胞は、その分画が単離された時点では、明らかに不応答性であった(図3A、レーン3、白色カラム)。
更に、CD45RA+CD25+CD4+T細胞は、アロ抗原の存在中で1:1の比率で共培養されたときの同系(syngenic)CD4+T細胞の増殖をほとんど抑制しなかった(図3A、レーン4、黒色カラム)が、一方、CD45RA-CD25high+CD4+T細胞は、全CD4+T細胞単独でのMLR(図3A、レーン2)と比較して、著しく全CD4+T細胞の増殖を抑制し、免疫抑制能を示した(図3A、レーン4、白色カラム)。
同様の試験が作動性抗CD3抗体の存在しない条件で行われた(図3B)。作動性抗CD3抗体の不在中では、CD45RA+CD25+CD4+T細胞及びCD45RA-CD25high+CD4+T細胞の両方とも、アロPBMCに対して不応答性であった(図3B、第3レーン)。抗CD3抗体が存在しているときと同様に、CD45RA-CD25high+CD4+T細胞は、CD4+T細胞の増殖を抑制したが(図3B、第4〜7レーン、白色カラム)、CD4+T細胞単独のMLR(図3B、第2レーン)と比較すると、CD45RA+CD25+CD4+T細胞はわずかに抑制した(図3B、第4〜7レーン、黒色カラム)。
CD45RA+CD25+CD4+T細胞は、IL-2の存在中、10% FBSを含有する増殖培地中で、同種異系のPBMCの繰り返しの刺激により、40日間で、100倍以上の数に増殖した。10% ヒト血清を含有する増殖培地を用いたこの分画の培養は、細胞の数を80日間で約100倍に増加させた(図4A)。
CD45RA+CD25+CD4+T細胞分画から獲得された、増殖された細胞株(CD45RA+CD25+CD4+T細胞株という)は、CD45RA+CD25+CD4+T細胞自体とは異なり、不応答性及び抑制性の特性を示した。増殖培養で使用された同種異系PBMCにより刺激されたときの同系全CD4+T細胞の増殖を、増殖された細胞株は用量依存的に抑制した(図4B)。
一方、CD45RA+CD25+CD4+T細胞株を樹立する際に使用された同種異系PBMCとは異なる同種異系PBMC(第三者のPBMC)により刺激されたときの同系全CD4+T細胞の増殖は、該CD45RA+CD25+CD4+T細胞株によってほとんど抑制されなかった(図4C)。このことは、CD45RA+CD25+CD4+T細胞株の免疫抑制能は抗原特異的であることを示す。
また、同系全CD4+T細胞が、CD45RA+CD25+CD4+T細胞株と、5:1の割合で、同種異系PBMC(増殖培養に用いられたもの)の存在中で共培養され、IL-10R、TGF-β、CTLA-4、又はPD-1に対する抗体が共培養物に添加された。しかし、いずれの抗体もCD45RA+CD25+CD4+T細胞株の免疫抑制能を、解除させなかった。(図4D)。
CD45RA+CD25+CD4+T細胞株及び同系全CD4+T細胞の同種異系PBMCとの共培養の間、CD45RA+CD25+CD4+T細胞株の同系全CD4+T細胞への接触をトランスウェルチャンバー系を用いて回避した。同種異系PBMCをCD45RA+CD25+CD4+T細胞株とともに配置したか否かに関わらず、CD45RA+CD25+CD4+T細胞株の免疫抑制能は、細胞−細胞接触の回避によりなくなった(図4E)。この結果は、CD45RA+CD25+CD4+T細胞株の抑制性特性は細胞−細胞接触を要することを示唆する。
典型的なナイーブ細胞であるCD45RA+CD25-CD4+T細胞分画、又はCD45RA+CD25+CD4+T細胞分画に起源する増殖された細胞株のために、表現型解析が行われた(図5)。CD45RA+CD25+CD4+T細胞株上のCD25、GITR、及びCTLA-4の強度は、CD45RA+CD25-CD4+T細胞株と比較して明らかに高かった。CD45RA+CD25+CD4+T細胞上のCD62Lの強度も、実質的に高かった。これは、CD45RA+CD25-CD4+T細胞上のCD62Lが不在であることとは対照をなしている。従って、CD45RA+CD25+CD4+T細胞株の表現型特性は、Tregsのそれと類似しているが、ナイーブT細胞のそれとは異なっていることが示唆された。
CD45RA+CD25+CD4+T細胞株のCD45ROの発現レベルは、培養前の末梢CD45RA+CD25+CD4+T細胞と比較すると、約6.7倍に上昇しているが、末梢血で自然に存在しているTregs(CD45RA-CD25high+CD4+T細胞)と比較すると発現レベルは低く、約0.2倍であった。また、CD45RA+CD25+CD4+T細胞株のCD45RBの発現レベルは、末梢に自然に存在しているTregs(CD45RA-CD25high+CD4+T細胞)に比較して高く、約3.6倍であった。(なお、発現レベルの比較は、縦軸に細胞数を、横軸に発現強度(蛍光強度)を取ったグラフ中のヒストグラムのピークにおける発現強度を比較することにより行った。)
従って、CD45RA+CD25+CD4+T細胞株は、機能的には制御性T細胞の要件を満たしているが、末梢血に自然に存在しているTregとは表現型が異なることが示唆された。
Claims (16)
- ナイーブ表現型を有する末梢CD25+CD4+T細胞を培養し、制御性T細胞を得ることを含む、制御性T細胞の製造方法。
- 該ナイーブ表現型がCD45RA+、CD45ROlow+、CD45RBhigh+、CD62Lhigh+、及びCD38+からなる群から選択される少なくともいずれかである、請求項1記載の方法。
- 該CD25+CD4+T細胞はFoxp3+、細胞内CTLA-4+、及びTNFRII+からなる群から選択される少なくともいずれかの表現型を有する、請求項1記載の方法。
- 該CD25+CD4+T細胞は霊長類由来である、請求項1記載の方法。
- 該CD25+CD4+T細胞は抗原存在下で培養される、請求項1記載の方法。
- 該抗原は免疫学的非自己細胞である、請求項5記載の方法。
- 該免疫学的非自己細胞は同種異系細胞である、請求項6記載の方法。
- 得られ得る制御性T細胞が該抗原特異的である、請求項5記載の方法。
- 該CD25+CD4+T細胞はT細胞増殖因子の存在下で培養される、請求項1記載の方法。
- 該制御性T細胞は、CD45ROmid+ 、CD45RBhigh+、CD62Llow+、及びGITR+からなる群から選択される少なくともいずれかの表現型を有する、請求項1記載の方法。
- 請求項1〜10のいずれか1項に記載の方法により製造され得る制御性T細胞。
- CD45ROmid+ 、CD45RBhigh+、CD62Llow+、及びGITR+からなる群から選択される少なくともいずれかの表現型を有するCD25+CD4+制御性T細胞。
- 請求項11又は12に記載の制御性T細胞を有効成分として含有してなる、免疫調節剤。
- 免疫寛容誘導用である、請求項13記載の剤。
- 以下の工程を含む、免疫調節剤の製造方法:
(1)ナイーブ表現型を有する末梢CD25+CD4+T細胞を培養し、制御性T細胞を得る工程;
(2)該制御性T細胞を医薬として許容される担体と混合し、免疫調節剤を製造する工程。 - ナイーブ表現型を有する末梢CD25+CD4+T細胞を提供するための試薬を含む、制御性T細胞製造用キット。
