I.方法
A.免疫応答の調節方法
本発明は哺乳動物における免疫応答の調節方法を包含する。該方法は、こうした処置の必要な哺乳動物にセロトニン受容体とのセロトニンの相互作用の阻害剤を投与することを含んでなる。これは、本発明の教示を装備した当業者により認識されるであろうとおり、セロトニンおよび5−HT受容体の相互作用を阻害することが該受容体を含んでなるT細胞の活性化を阻害若しくは予防するからである。T細胞の阻害は、順に、本明細書に開示されるデータにより詳細に示されるとおり免疫応答の生成を予防する。
B.有用な化合物の同定方法
本発明は、セロトニンファミリー受容体とのセロトニンの相互作用を阻害する化合物の同定方法を包含する。当業者は、本明細書に提供される開示に基づき、該化合物と接触されないそれ以外は同一の細胞における同一のパラメータ(1種若しくは複数)と比較した場合にとりわけT細胞の活性化などを評価することにより、セロトニンおよびセロトニンファミリー受容体の相互作用のレベルを評価することを実施し得ることを認識するであろう。当業者は、こうした化合物が、セロトニン受容体とのセロトニンの相互作用により媒介されかつ/若しくはそれと関連する疾患、障害若しくは状態を阻害するために有用であり得ることを理解するであろう。当業者はさらに、本明細書に提供される開示に基づき、影響を及ぼされない他のセロトニン受容体型とのセロトニンの相互作用を残しつつセロトニンと特定のサブタイプ(1種若しくは複数)のセロトニン受容体との間の相互作用を減少させることが有用であるかもしれないことを認識するであろう。
C.T細胞上の異常なセロトニンタイプ2受容体により媒介される哺乳動物における疾患、障害若しくは状態の治療若しくは軽減方法
本発明は、セロトニンファミリー受容体の異常なすなわち異常発現(malexpression)により媒介される疾患、障害若しくは状態の軽減方法を包含する。疾患、障害若しくは状態がセロトニン受容体の過剰若しくは過小発現と関連している場合、該方法は、それ以外は同一であるがしかし正常な組織、すなわち治療若しくは軽減されている疾患、障害若しくは状態に関連するいかなる検出可能な臨床パラメータも表さない組織中での受容体発現のレベルに比較して増大された受容体発現により媒介される疾患、障害若しくは状態に苦しめられている患者に適切な受容体をコードする核酸に相補的なアンチセンス核酸を投与することを含んでなる。これは順に受容体発現の低下を媒介してそれにより受容体の異常発現により媒介される疾患、障害若しくは状態を軽減する。こうした疾患、障害若しくは状態は、限定されるものでないが重症筋無力症、特発性炎症性ミオパシー、慢性好中球減少症、慢性関節リウマチ、特発性血小板減少性紫斑病、自己免疫性溶血性症候群、抗リン脂質抗体症候群、炎症性腸疾患、クローン病、潰瘍性大腸炎、心筋炎、ギラン・バレー症候群、脈管炎、多発性硬化症、視神経脊髄炎(ドヴィック症候群)、リンパ球性下垂体炎、グレーヴズ病、アジソン病、副甲状腺機能低下症、1型糖尿病、全身性エリテマトーデス、尋常性天疱瘡、水疱性類天疱瘡、乾癬、乾癬性関節炎、子宮内膜症、自己免疫性精巣炎、自己免疫性勃起不全、サルコイドーシス、ヴェゲナー肉芽腫、自己免疫性難聴、シェーグレン病、自己免疫性ブドウ膜網膜炎、間質性膀胱炎、グッドパスチャー症候群および線維筋痛症を挙げることができる。
D.セロトニン受容体を介するシグナル伝達を阻害することに関する方法
本発明は、セロトニン受容体を介するセロトニンシグナルの伝達を阻害することによる細胞周期過程に影響を及ぼす方法を包含する。すなわち、当業者は、本明細書に提供される開示に基づき、5−HT受容体を介して伝達されるセロトニンシグナルの除去すなわち阻害が細胞周期過程に影響を及ぼすことを認識するであろう。これは、セロトニン作動性シグナルの阻害が、細胞周期を循環している細胞に対する大きな影響を有する、例えばシグナルの除去がアポトーシスを介する迅速な細胞死を媒介する(すなわち、細胞が細胞表面上のPIの曝露を示すアネキシンにより染色されるようになり、DNA断片化が検出され、そして細胞の大きさが増大しかつ変えられた形態を示す、など)ことを、本明細書に開示されるデータが十分に示すからである。以前の研究は、多様な神経学的障害の処置のための細胞周期を耐えていなかった非分裂細胞すなわち神経若しくは筋細胞においてセロトニン作動性シグナルを阻害することに焦点をあてていたため、これらは驚くべき結果である。いずれかの特定の論理により拘束されることを願わずに、セロトニンシグナル伝達の多様な阻害剤の使用に関する従来技術の研究が、限定されるものでないがニューロンを挙げることができる細胞周期を通過していなかった非分裂細胞を接触させることを含んでいたという事実、細胞周期に対するセロトニンシグナル伝達の除去および/若しくは阻害の影響は観察、認識若しくは理解されておらず、かつ、実際のところされていた可能性はない。従って、本明細書に開示されるデータは、セロトニン受容体の阻害を介して細胞周期に影響を及ぼす新規方法を初めて示す。
II.キット
本発明はセロトニンタイプ2受容体とのセロトニンの相互作用を阻害することに関する多様なキットを包含する。本明細書の別の場所に開示されるとおり、この相互作用を阻害することが順に免疫細胞の活性化を阻害してそれにより免疫応答を阻害するからである。従って、一局面において、本発明は哺乳動物における免疫応答を調節するためのキットを包含する。該キットは、有効量のセロトニンタイプ2受容体とのセロトニンの相互作用の阻害剤を含んでなる。こうした阻害剤は好ましくはセロトニンタイプ2受容体アンタゴニストを包含する。また、該キットはさらにアプリケーターおよびその使用のための説明資料を含んでなる。
[実施例]
トリプトファンは、細胞において新しいタンパク質を構築するために必要とされる10個の必須アミノ酸の一つである。トリプトファンの異化は飢餓をもたらし、従って、認められるT細胞阻害を説明することは、ありそうにないが、可能である。しかしながら、他の9個の必須アミノ酸のいずれもT細胞応答の制御に関与するとされていない。

逆転写およびPCR
健康なドナーからの全血をCurrent Protocols in Immunologyに記述されているようにフィコール勾配を用いて分画した。末梢血リンパ球を集め、そして6ウェルプレート上にウェル当たり2x107個の細胞で平板培養した。細胞をプラスチックに1.5時間接着させ、そしてその時に懸濁細胞を取り除き、そして新しい6ウェルプレート上にウェル当たり107個の細胞で再び平板培養した。接着細胞および懸濁細胞の両方をConAで(5μg/mlで)刺激するかもしくは未処理のままにしておいた。ConA刺激後48時間で、細胞を採取し、そして製造業者の説明書(Qiagen,Chatsworth,CA)に従ってQiagen RNAeasyミニプレップシステムを用いて全RNAを抽出した。(Qiagen,Chatsworth,CA)RNAサンプルをゲルの臭化エチジウム(EtBr)染色により定量し、そして約1μgの各RNAサンプルをcDNA合成に用いた。cDNA合成は、オリゴT12−18プライマーもしくは受容体特異的リバースプライマーのいずれかを用いて、製造業者の説明書に従ってQiagen逆転写キットで行った。得られるcDNAは、5−HT受容体特異的プライマーを用いる35サイクルのPCR反応(SigmaからのTaq DNAポリメラーゼを用いる)において鋳型として用いた。50マイクロリットルのPCR反応には、25ピコモルの各特異的プライマーを用いた。PCR条件は:45秒間95℃、45秒間61.5℃、45秒間72℃であった。35PCRサイクルの後に72℃で10分の伸長工程を続けた。最終PCR生成物は、アガロースゲル電気泳動(TAEバッファー)(BMA,Rockland,MEからのNuSieve 3:1プレキャストアガロースゲル)を用いて分析した。
マクロファージ培地コンディショニング研究
Current Protocols in Immunology(1999,14.1.3−14.1.6節;Coligan et al.,eds.,1994−1997,Current Protocols in Immunology,vol.1−3,John Wiley & Sons Inc.)から改変したプロトコルを用いて、C57/B6Jマウスから得られる単球を単離した。6−8週齢のC57/B6Jマウス(Jackson Laboratories,Bar Harbor,ME)の後肢から大腿骨および頸骨を採取した。これらの骨の遠位端を取り除き、骨髄栓(marrow plug)を露出した。22ゲージ針(Becton−Dickinson,Lincoln Park,NJ)を用いて、2% FBSを補足したRPMI培地で骨髄腔をフラッシした。次に、間質細胞を取り除くために細胞懸濁液をナイロンメッシュに通した。Current Protocols in Immunology(3.1.5節)に記述されているようにACK(塩化アンモニウムカリウム溶解バッファー;0.15M NH4Cl,10mM KHCO3,0.1mM EDTA)バッファーを用いて赤血球を溶解した。
マウスT細胞の精製

マウスマイトジェン刺激
Current Protocols in Immunology(1999,3.1.5および3.12.2−3.12.4節)に記述されているように6−8週齢のBALB/c/BYJマウスからの一次脾臓T細胞を得た。全ての薬剤は、細胞懸濁液の添加の前に、様々な濃度で96ウェルU底プレート上で事前に平板培養した。全ての実験条件は、少なくとも三重反復でアッセイした。精製された一次細胞をウェル当たり100,000個の細胞で薬剤を含んでなるウェルにおいて平板培養した。他に示さない限り、ConAは1μg/mlの最終濃度になるように細胞懸濁液に加えた。総容量は、ウェル当たり200μlであった。コントロールサンプルには、ConA刺激を与えなかった。細胞を60時間増やした。1μCiのトリチウム化チミジンを各ウェルに加え、そして12時間後にプレートを半自動式PHD(Brandel,Gaithersburg,MD)ハーベスターを用いてConAの添加後72時間で採取した。
マウス混合リンパ球反応(MLR)
MLRは、本質的にCurrent Protocols in Immunology(1999,3.12.6−3.12.7節)に記述されているように行った。すなわち、C57/B6Jマウスから得られる一次脾臓細胞を刺激物質として用いた。それらは、Current Protocols in Immunology(1999,3.1.5節)に記述されているように精製し、そしてHahnemann Hospital(Philadelphia,PA)血液銀行施設で放射線照射した(35Gy)。BALB/c/BYJから得られる一次脾臓細胞をナイロンウールカラムを用いて接着細胞から除去し、そしてレスポンダー(responder)として用いた。
ヒトT細胞の精製
全ての必要な説明同意書(informed concern from)に記入した後に様々な健康なドナーから血液を得、標準方法に従ってフィコール−ヒパーク(Ficoll−Hypaque)(Sigma)勾配を用いて末梢血単核細胞(PBM)を単離した。細胞をフィコール−血清界面から集め、そして残りのフィコールを除くために十分に洗浄した。細胞を洗浄し、そして次にスピン培地(2%ウシ胎仔血清、1% pen−strepおよび1% L−Gluを補足したRPMI 1640倍地)に再懸濁した。接着細胞を除くために細胞をフラスコにおいて37℃、5% CO2で約4時間インキュベーションした。懸濁細胞を集め、そして感作培地(10%ヒト血清(Sigma)、1% pen−strepおよび1% L−Gluおよびベータ−MKEを補足したRPMI 1640倍地)に再懸濁した。
薬剤処理

サイトスピン

組織化学
標準的な染色プロトコルに従って、サイトスピンしたスライドをヘマトキシリンおよびエオシン、続いて核染色液、ビス−ベンズアミドで染色し、すすぎ、そして標本にした。同じスライド視野の整合する像を作製するためにスライドを明視野および蛍光灯の両方の下で視覚化した(例えば、図28および29を参照)。
ヒトマイトジェン刺激

ヒト混合リンパ球反応のプロトコル
二人の健康な関係のないドナーからの血液を上記のように採取した。他の所ですでに記述したように末梢血単核細胞を単離した。すでに記述したように両方のドナーで接着細胞を懸濁細胞から分離した。両方のドナー(ドナーAおよびB)からの懸濁細胞をレスポンダーとして用いた。ドナーの各々からの残りの接着細胞を異なるドナーに対する刺激物質として用いた(接着Bに対して懸濁Aおよび懸濁Bに対して接着A)。刺激物質細胞を35Gyで放射線照射した。薬剤は、三重反復で96ウェルU底プレート上に事前に平板培養した。次に、10%ヒト血清を補足したRPMI中200,000個の「刺激物質」細胞を各ウェルにおいて平板培養した。次に、同じ培地中200,000個の「レスポンダー」細胞を200μl/ウェルの最終容量になるように刺激物質上に平板培養した。バックグラウンドコントロールには刺激物質細胞もレスポンダー細胞も与えなかった。プレートを37℃および5% CO2でインキュベーションした。1μCiのトリチウム化チミジンを4.5日後に各ウェルに加え、そして12時間後にプレートを採取した。
マウス同種移植モデル
使用するインビボ検証スクリーニングは、Zhan et al.,2000に記述されているような当該技術分野で認められているマウス同種異系拒絶モデルであった。このアッセイでは、5x106個のP815細胞([H−2d]DBA/2肥満細胞腫)を実験の0日目にC57BL/6J(H−2b)マウスの腹腔に注射する完全なMHCミスマッチ系を用いた。これらのマウスは、典型的には、P815細胞に応答して強い細胞傷害性Tリンパ球(CTL)反応を引き起こす。最初のP815細胞接種の後、マウスにCTL応答を発生させた(これは通常約10〜14日かかる)。研究の6および8日目に、試験薬剤のボーラス静脈内(iv)注射を投与した。次に、図に示すように実験の14日目にマウスを殺し;そして同種異系CTL応答を以前に記述されているようにアッセイした(Tretiakova et al.,2000,Nature Biotechnology 18:984−988)。処理したおよび未処理の動物からの一次脾臓細胞を直接CTL読み取りに用いた。新しく得た一次脾臓細胞を[3H]標識した標的細胞(P815細胞)と100:1、50:1、25:1および12.5:1の比率で37℃で3.5時間インキュベーションし、そしてPHDハーベスターを用いて採取した。%致死=(S−E)/S(ここで、Sはエフェクター細胞のない場合に標的細胞により保持されるDNAの量であり、そしてEはエフェクター細胞の存在下で保持されるDNAの量である(分当たりのカウント;cpm単位で表される))の式を用いて特異的致死のパーセンテージを決定した。

細胞系

HMBA 細胞における最終分化の誘導
DNA断片化
実験処置の後に細胞(8x105個)をよく冷えたPBSで洗浄し、そして遠心分離(500xg、5分)によりペレットにした。細胞ペレットを溶解バッファー(1% IGEPAL−CA630,20mM EDTA,50mM Tris−HCL,pH7.5)において氷上で5分間インキュベーションした。溶解物を約1,600xgで5分間遠心分離した。50μLの溶解バッファーを上清に加えた。抽出物を1% SDSにし、そして5μg/μLのRNアーゼAで2時間処理し(57℃)、続いて2.5μg/μLのプロテイナーゼKで2時間処理した(37℃)。RNAおよびタンパク質の消化後、半分容量の10M酢酸アンモニウムを加え、そして2.5容量のエタノールを用いてDNAを沈殿させ、70%エタノールにおいて洗浄し、風乾させ、そしてTEバッファーに溶解した。DNAフラグメントは、Siegel et al.(1998,Proc.Natl.Acad.Sci.USA 95:162−166)から適用したプロトコルを用いて1.5%アガロースゲルにおける電気泳動により分離した。
フローサイトメトリー
初期のアポトーシスを起こした細胞は、各実験条件にあらかじめさらした細胞を共染色することにより検出した。簡潔に言えば、細胞を本明細書の他の所に記載するような様々な濃度の各実験もしくはコントロールアポトーシス誘導因子の存在下で時間経過の間インキュベーションした。インキュベーションの後に、細胞をよく冷えたPBSで洗浄し、アネキシンV−Alexa flour 488およびヨウ化プロピジウム(Molecular Probes,Eugene,OR)で染色し、そして二色フローサイトメトリーにより分析した。アネキシンV+且つPI―の細胞を初期のアポトーシスを起こした細胞とみなした。骨髄腫細胞の既知の最終分化因子、HMBA(ヘキサメチレン−ビス−アセトアミド)、およびトポイソメラーゼインヒビター、カムトテシンをそれぞれ5mMおよび2μMでアポトーシスの陽性コントロールとして用いた。
本明細書の他の所に開示するデータは、セロトニン受容体を介したシグナルの伝達を阻害することが、細胞における検出可能な変化を誘導するか、媒介することができ、もしくはそれと関連することを初めて示す。さらに特に、ある種のセロトニンアンタゴニストと接触させた細胞は、変化した細胞形態および/もしくは光学顕微鏡検査を包含する当該技術分野において既知である様々な方法により検出可能な他の改変された物理的特性を示した。

- 前記阻害剤が血液脳関門を実質的に横断しない、請求項29に記載の方法。

- 前記自己免疫疾患が、重症筋無力症、特発性炎症性ミオパシー、慢性好中球減少症、慢性関節リウマチ、特発性血小板減少性紫斑病、自己免疫性溶血性症候群、抗リン脂質抗体症候群、炎症性腸疾患、クローン病、潰瘍性大腸炎、心筋炎、ギラン・バレー症候群、脈管炎、多発性硬化症、視神経脊髄炎(ドヴィック症候群)、リンパ球性下垂体炎、グレーヴズ病、アジソン病、副甲状腺機能低下症、1型糖尿病、全身性エリテマトーデス、尋常性天疱瘡、水疱性類天疱瘡、乾癬、乾癬性関節炎、子宮内膜症、自己免疫性精巣炎、自己免疫性勃起不全、サルコイドーシス、ヴェゲナー肉芽腫、自己免疫性難聴、シェーグレン病、自己免疫性ブドウ膜網膜炎、間質性膀胱炎、グッドパスチャー症候群および線維筋痛症よりなる群から選択される、請求項29に記載の方法。
