Abstract
Description

3 滅菌処理(例えば、2000ppm、またはそれ以上の過酢酸処方法を用いて、55〜57℃の温度で10〜30分間)の工程。
(増殖培地)
適切な増殖培地が、プリオンモデルの培養のために開発された。増殖培地は、寒天またはブロスベース(broth base)、例えばOxoidから入手されるMycoplasmaTM寒天またはブロスベースを含む。ヘモグロビンの供給源、例えば洗浄および溶解したヒトまたは他の動物由来の赤血球もまた存在する。上で議論したように、赤血球は、使用前にプリオンが増殖培地中に存在しないことを確認するため、好ましくは試験される。培地はまた、プロテアーゼの供給源も好ましくは含む。プロテアーゼの供給源は、単離されたプロテアーゼ、他の酵素(例えば、リパーゼ)も含み得る酵素抽出物、または粉砕動物組織(例えば、膵臓組織をホモジェナイズすることで得られるパンクレアチン)であり得る。Tween 80のような分散剤が、好ましくは存在する。酢酸タリウムは、培地汚染を少なくするための任意の成分である。これらの成分は、水(好ましくは蒸留したあるいは他の精製水)とブレンドされる。

1)最小阻害濃度(Minimum inhibitory concentrations)(MIC):この方法は、試験タンパク質の培養を阻害するための、広範な種々の活性物質の迅速な評価を可能にする。マイクロウェルプレートの簡易設置において、活性の段階希釈が行われ、そして増殖培地中のプリオンモデルの標準低濃度(例えば、<105実体形成単位(efu’s))が、各々の希釈液に加えられる。37℃で48〜72時間インキュベートされた後、増殖があることが、ウェルの底の黒い沈殿物により示され、増殖を阻害するための活性物質の最も低い濃度が、MICとして記録される。この方法は、抗プリオン活性に対する可能な薬物標的または殺生剤の同定に使用され得る。
2)タイムキル実験(Time kill experiments):この方法は、長時間にわたる活性効率または処方効率の決定を可能にする。試験材料(例えば、液体処方物、生成物、または活性物質)は、様々な濃度および様々な環境条件下(例えば、pH、温度、固体の存在等)で調製される。次いで、既知の濃度でのプリオンモデルの培養は、試験液体および評価のための時間にわたって除去されたアリコートに直接加えられる。このアリコートは、試験材料の活性をさらに阻害するため、好ましくは中和される。評価は、アリコートの段階希釈および選択された試験条件下での長時間にわたる試験タンパク質の減少を測定するための選択培地上へのプレーティングを含み得る。

試験タンパク質を、上記の処方を有する、補充Mycoplasmaブロスベースをプレーとした培地皿上に接種し、そして37℃で培養し、接種したスポットは、48時間後に、好気性、微好気性または嫌気性条件下で分離した褐色コロニーを生じる。4℃では増殖は観察されない。
試験タンパク質を分析して、アミノ酸(以下に記述される)、および少なくとも2つのペプチドを含むことを決定した。
プリオンモデルの培養物を、ブロス中で増殖し、(生理食塩水中で5回ボルテックスすることで)勢いよく洗浄し、そして乾かした。サンプルを、総アミノ酸分析に供した。サンプルを、6N HCl中で26時間110℃にて加水分解し、0.01N HCl中に溶解し、そしてクロマトグラフィーにより分析した。
プリオンモデルタンパク質を、可溶化し、そしてタンパク質ゲルを、上清上で動かした。SDS−PAGEを、タンパク質を分離するために使用した。拡散したタンパク質のバンドの存在が、色素領域上(<10kDa)で観察された。このバンドを、ウエスタンブロッティングによってメンブラン上に移し、そしてCCF(N末端配列分析の分子生物学コア)にかけた。シグナルは弱かったが、以下のとおりの二つのペプチド配列を示した:
タイムキル試験の有効性を示すため、および試験タンパク質の反応と公知のプリオンの反応との間の相関を確立するため、試験タンパク質の対数減少を、プリオンに対し有効であることが知られている活性物質を用いて測定した。対数減少は、存在する生物の元の数の対数(この場合、試験タンパク質の数、あるいは、サンプル中の試験タンパク質の濃度)と残存している生物の数の対数との間で異なる。良好な相関関係が、そのような活性物質について見出された。実施例は以下に与えられる:
a)過酢酸研究
以前に有効なプリオシダル薬剤として提唱された、特定の過酢酸処方物(STERIS Corp.、Mentor、Ohioから入手されるSTERIS 20TM)は、プリオンモデルに対して有効であることが見出された。STERIS 20TMは、緩衝液、界面活性剤、キレート剤、および防食剤を含む過酢酸ベースの滅菌剤である。これらの試験の結果は、処方物の温度が重要な因子であることを示している。温度および活性物質濃度は、タンパク質およびプリオン両方の不活化において驚くべきかつ有意な効果を有することが見出された。
タンパク質分解研究を、プリオンモデルのサンプルを滅菌剤(例えば過酢酸)およびコントロール溶液(例えば、水またはトリス緩衝生理食塩水、TBS)にさらすことにより行った。さらした後、過酢酸を、チオ硫酸ナトリウム(STS)を用いて直ちに中和し、そしてサンプルは、ゲル電気泳動により分離する。ドデシル硫酸ナトリウムポリアクリルアミドゲル電気泳動(SDS−PAGE)は、タンパク質を培地と分離する場合に有効である。さらに、HLC分析は、モデルの主成分はコラーゲンの単純アミノ酸構造に類似した短いペプチド構造であることを示している。分離したタンパク質を、ウエスタンブロット法によりニトロセルロースに移した。プリオンモデルのような特定のタンパク質の存在を、タンパク質に特異的なモノクローナル抗体を用いた免疫ブロッティングにより検出し、そして免疫染色されたタンパク質の強度の割合により定量した。過酢酸は、試験された特定の処方物(STERIS 20TM)中で使用する場合、全タンパク質の量を減少させ、より小さなペプチドを生成した。このことは、プリオンモデルの目的に対して不活化であると見なされる。
異なる温度および濃度下における、過酢酸処方物の効果をプリオンモデルを使用し、懸濁液を試験することにより研究した。過酢酸処方物を、1000、1500、および2000mg/L過酢酸で調製し、そして約50℃に維持した。試験タンパク質懸濁液のアリコートを、直接各処方物に添加し、そしてサンプルを除去および段階希釈により定量し、改変マイコプラズマ寒天にプレーティングした。
LpHTMは、STERIS Corporation、Mentor、OHにより販売されるフェノール処方物である。この組成物は、以前にインビボ研究においてスクレーピーに対して有効であると記載された(Ernst&Race、Comparative Analysis of Scrapie Agent Inactivation Methods、J.Virological Methods、41、pp.193〜202(1993))。著者は、濃度および露出時間に依存したスクレーピー感染性の減少を記述した。例えば、0.9%の濃度で、感染性が除去され、対数減少により測定した時、0.5時間後には5ログ、16時間では7ログを越えた。同様に9%では、7ログを越える減少が、0.5時間で観察された。
スクリーニングツールとしてタンパク質分解を使用する試験は、約2000mg/Lまたはそれ以上の過酢酸濃度が、試験タンパク質に対する効果に好ましいことを示している(図3)。これらの試験を、STERIS 20TMを用いて行った。
プリオンモデルのMIC(増殖阻害)試験における様々な活性物質(多くがプリオンに対する可能な効果のこれまでの報告を有する)の効果を研究した。

(実施例6:除去試験)
様々な洗浄剤を評価した。器具を、ウシ血清アルブミン(BSA−タンパク質)で汚染した。タンパク質の除去をより困難にするため、器具を、タンパク質を変成させるために110℃で1時間加熱した。次いで、器具を、自動洗浄機内で、1オンス/ガルの洗浄剤および高い洗浄温度(150℃)を用いて洗浄した。洗浄サイクル後、残存固体の視覚的実験を行った。評価した洗浄剤は、有効性が減少する順に以下に列挙した。全ての洗浄生成物は、STERIS Corp.、Mentor、OHより入手された。

上記の有効性順序はまた、一般的に生成物のアルカリ度に伴う(水は除外して)。最も有効な洗浄生成物、CIP 100TMもまた最も高いアルカリ度を有する。最も効果のない、CIP 220TMは、酸生である。
プリオンモデルは、増殖培地中のヘモグロビンまたは赤血球(RBC)成分を吸着/使用するために見出された。全赤血球に関するプリオンモデルの効果の研究を行った。RBCを、生理食塩水中で3回洗浄し、そして40×(約100/視野)のペトロフハウザー(Petroff Hauser)下で細胞を数えるための濃度で再懸濁した。総赤血球およびまだインタクトな(形態学において変化していない)細胞の割合を、プリオンモデル(IFDO)とともに、または水(RBC)もしくはパンクレアチン(Panc−これは、RBCを溶解する予備実験において示されている)とともにインキュベーションした後に長時間にわたってモニタリングした。
予備的なタイムキル実験を、プリオンモデルを用いて行った。結果は、ログ数字(log10)として表され、ログ数字が低くなるにつれて、残存するプリオンが少なり、従って、有効な処理となる。

Claims (20)
- プリオンまたはプリオン関連疾患に対する活性の潜在的な処理を評価する方法であって、該方法は、以下:
該処理にプリオンモデルを供する工程であって、該プリオンモデルは、プリオンを攻撃するように設計された処理に対して、プリオンの応答と類似した応答を示すことが示されているモデルである、工程:および
プリオンまたはプリオン関連疾患の処理効果の指標として、該プリオンモデルにおける該処理効果を評価する、工程、
により特徴付けられる、方法。 - 前記プリオンモデルが、回腸流動体依存性生物(IFDO)を含むことによってさらに特徴付けられる、請求項1に記載の方法。
- 前記処理が、汚染除去プロセスを含むことによってさらに特徴付けられる、請求項1および請求項2のいずれか一項に記載の方法。
- 前記処理が、50〜58℃の温度での、過酢酸溶液によるプリオンモデルの処理を含むことでさらに特徴付けられる、請求項1〜3のいずれか一項に記載の方法。
- 請求項3および4のいずれか1項に記載の方法であって、以下:
血管中に含まれるプリオンモデルであって、該血管は、経路を含み、該経路によって汚染除去剤が該プリオンモデルと接触し、該経路は、該汚染除去剤のアクセスに対するチャレンジを提供し、該チャレンジは、該汚染除去剤によって汚染除去されるデバイスによって与えられるのと少なくとも同じくらいのチャレンジである、プリオンモデル
によってさらに特徴付けられる、方法。 - 前記処理が、微生物を粉砕する際に有効であることが知られているプロセスに、プリオンモデルを供することを包含することによりさらに特徴付けられる、請求項1〜5のいずれか一項に記載の方法。
- 前記プロセスが、過酸化水素および過酸からなる群のうち、少なくとも一つと前記プリオンモデルとを接触させることを包含することにより、さらに特徴付けられる、請求項5および6のいずれか一項に記載の方法。
- 前記処理が、プリオンモデルで汚染された医療用デバイスまたは薬学的デバイスを、ナイシン、マンガン、および硝酸銀のうち少なくとも一つを含む組成物で処理することを包含することによりさらに特徴付けられる、請求項1〜7のいずれか一項に記載の方法。
- 前記プリオンモデルが基質に塗布されており、そして前記処理が以下:
該基質を、タンパク質除去する洗浄組成物で洗浄する、工程:および、
該洗浄された基質を酸化剤と接触させる、工程、
を包含することによりさらに特徴付けられる、請求項1〜8のいずれか一項に記載の方法。 - 前記評価する工程が、以下:
増殖培地中で残存のプリオンモデルを培養する工程;および
プリオンモデルの増殖を検出する、工程、
を包含することによりさらに特徴付けられる、請求項1〜9のいずれか一項に記載の方法。 - 前記検出する工程が、培養されたプリオンモデルを、タンパク質を検出する手段に供することを含むことにより、さらに特徴付けられる、請求項10に記載の方法。
- プリオンによって汚染され得るアイテムを処理する方法であって、該方法は、該アイテム上で生存可能なプリオンのレベルを減らすために、ナイシン、マンガン、および硝酸銀のうち少なくとも一つを含む組成物で該アイテムを処理する工程により特徴付けられる、方法。
- 前記アイテムが、動物の消費または人間の消費用の食品を含むことにより、さらに特徴付けられる、請求項12に記載の方法。
- 前記アイテムが、医療用デバイスまたは歯科用デバイスを含むことにより、さらに特徴付けられる、請求項12に記載の方法。
- 前記組成物が、IFDOを攻撃する際有効であることが見出されている組成物により、さらに特徴付けられる、請求項12〜14のいずれか一項に記載の方法。
- プリオン関連疾患に対する活性用に提唱された薬物またはプリオシダル活性用に提唱された処理方法または化学物質をスクリーニングする方法であって、該方法は、以下:
プリオンモデルを、提唱された薬物、化学物質またはプロセスにさらす、工程;および
インビトロで任意の残存プリオンモデルを培養する工程であって、該プリオンモデルは、薬物、化学物質、またはプロセスに対してプリオンと類似した応答を示すことが示されている、工程
によって特徴付けられる、方法。 - 前記培養する工程が、以下:
ヘモグロビンを含む培地中で前記残存プリオンモデルを増殖させること、および、
残存するプリオンモデルの量の指標として該培地の色の変化を観察すること、
を包含することによりさらに特徴付けられる、請求項16に記載の方法。 - 前記培養されたプリオンモデルを、タンパク質を検出する手段に供する工程により、さらに特徴付けられる、請求項16および請求17のいずれか一項に記載の方法。
- 前記手順がタンパク質フラグメントを検出することにより、さらに特徴付けられる、請求項18に記載の方法。
- プリオンに汚染された対象物またはプリオン関連疾患を有する対象物を処理する方法であって、該方法は、以下:
IFDOに汚染されたサンプルを、提唱された処理剤で処理する工程であって、該IFDOは、プリオンに対すると類似の様式で、他の処理剤に対して応答することが示されている、工程:および
該処理剤がIFDOを攻撃する際に効果がある場合、該対象物を、有効量の処理剤で処理する、工程
