Description

ホルモン

N末端欠失
構築
Δ1−9−hPRL、Δ1−10−hPRL、Δ1−11−hPRL、Δ1−12−hPRL、Δ1−13−hPRL、およびΔ1−14−hPRLアナログをコードする発現プラスミドの構築を、ポリメラーゼ連鎖反応(PCR)を使用して行なった。プラスミドpT7L−hPRL(PARISら、Biotechnol. Appl. Biochem.、12、436〜449、1990)を、テンプレートとして使用した。5’プライマーの配列は、N末端の9コドンを欠失したhPRL cDNA(Δ1−9−hPRL)からN末端の14コドンを欠失したhPRL cDNA(Δ1−14−hPRL)の5’配列に対応する。ATGコドン(メチオニン開始配列)を含む独特なNdeI制限部位(CATATG)を、5’プライマーに挿入した。Cys11をコードするTGCコドンを、セリンをコードするTCCに変異させた。5’プライマーの配列は以下のとおりである(5’→3’):

組換えWT hPRLおよびhPRLアナログを、以前に記載された(PARISら、Biotechnol. Appl. Biochem.、12、436〜449、1990;GOFFINら、Mol. Endocrinol.、6、1381〜1392、1992)ようにして、E.coli BL21(DE)の1リットル培養物において過剰発現させそして精製した。簡潔に述べると、細菌培養物のOD600が約0.9に達したときに、2mMイソプロピルチオールガラクトシド(IPTG)を4時間使用して過剰発現を誘発した(4時間後OD600が約2.5)。細胞粉砕機(cell disintegrator:Basic Z、Cell D、Roquemaure、France)を使用して、細胞溶解を行なった。タンパク質を不溶性封入体として過剰発現させ、不溶性封入体を8M尿素に溶解し(55℃にて5分間、次いで室温にて2時間)、そして50mMのNH4HCO3(pH8)に対する連続透析(72時間、4℃)によってタンパク質をリフォールディングさせた。
pT7L−G129R−hPRLプラスミド(G129R変異を含む)(GOFFINら、J. Biol. Chem.、269、32598〜32606、1994)由来のEcoRI−BglIIフラグメントを、pT7L−Δ1−9−hPRLおよびpT7L−Δ1−14−hPRL発現ベクター中の対応するEcoRI−BglIIフラグメントと置換することによって、アナログΔ1−9−G129R−hPRLおよびΔ1−14−G129R−hPRLをコードする発現プラスミドを構築した。インサートの存在について、得られたクローンを分析し、次いで、配列決定して、期待された変異を照合した。BL21(DE3)細菌を使用するアナログ発現およびタンパク質精製を上記のように行なった。
結合研究
ヒトPRLRに対する種々のhPRLアナログのアフィニティーを、このレセプターとの結合について[125I]―hPRLと競合する能力によって推定した。以前に記載された手順(KINETら、J. Biol. Chem.、274、26033〜26043、1999)に従って、(ヒトPRLRを発現する)HL5細胞の細胞ホモジネートを使用して結合アフィニティーを決定した。
hPRLアナログの相対的結合アフィニティーを、競合曲線から算出される(S字の直線部分に回帰)WT hPRLのIC50に対するそのIC50の比として算出した。図2Aに示された結果は、二連で(in duplicate)行なった少なくとも3つの独立した実験の代表である。
Gly129→Arg変異を含む3つのアナログを用いて得られた代表的な競合曲線を、図2Bに示す。WT hPRL(−●−);単一変異体G129R−hPRL(−◆−);二重変異体Δ1−9−G129R−hPRL(−■−);二重変異体Δ1−14−G129R−hPRL(−▲−)。
実験プロトコル
Nb2細胞増殖アッセイ
乳腺刺激ホルモンについての参照バイオアッセイは、ラットNb2リンパ腫細胞のラクトゲン誘導増殖である。ラットNb2リンパ腫細胞を、P.W.GOUT(Vancouver, Canada)から入手し、そして以前(BERNICHTEINら、Endocrinology、142、3950〜3963、2001)に記載されたようにして培養した。10%HSと、10%熱不活化FCSと、2mMグルタミンと、50U/mlペニシリンと、50μg/mlストレプトマイシンと、100mMのβ―メルカプトエタノールとを補充したRPMI 1640中でNb2細胞を慣用手法に従って維持した。最初(TANAKAら、J. Clin. Endocrinol. Metab.、51、1058〜1063、1980)に記載されたように、しかし僅かに改変して(BERNICHTEINら、Endocrinology、142、3950〜3963、2001)、増殖アッセイを行なった。簡潔に述べると、ホルモンを含む200μlの最終容量で、開始日に2×104細胞/ウェルを使用して、96ウェルプレートにおいてアッセイを行なった。10μlのWST−1テトラゾリウム塩(ROCHE, Meylan, France)を添加してホルモン刺激した3日後に、細胞増殖を評価した。この生存試薬(survival reagent)は、生存細胞のミトコンドリアによって代謝される。このことから、血球計によりカウントされる細胞数に比例して(BERNICHTEINら、Endocrinology、142、3950〜3963、2001)、450nmにて測定されるOD(OD450)が増加することが導かれる。三連または四連で(in triplicate or quadruplicate)少なくとも3回、この実験を行なった。
クローンHL5は、ヒトPRLRおよび(STAT5のDNA結合エレメントである(KINETら、J. Biol. Chem.、274、26033〜26043、1999)乳腺刺激ホルモン応答エレメント(LHRE)の6反復配列の制御下に、ルシフェラーゼ遺伝子をコードする配列を含む)PRL応答性レポーター遺伝子をコードするプラスミドで安定にトランスフェクトされた293 HEK線維芽細胞である。
Ba/F3細胞は、増殖についてインターロイキン−3(IL−3)要求性のマウスpro−Bリンパ系細胞である。hPRLRをコードするプラスミドを使用するトランスフェクションおよびG−418処理と増殖培地中のIL−3のhPRLへの置換とを含む二重選択の後、Ba/F3−hPRLR細胞を得た。hPRLRをコードするプラスミドを使用して、細胞をトランスフェクトし(エレクトロポレーション)、そしてレセプターを安定に発現する集団を、G−418処理後に選択した。10%熱不活化FCSと、2mMグルタミンと、50U/mlペニシリンと、50μg/mlストレプトマイシンと、500〜1000μg/mlのG−418と、IL−3に代えて10ng/mlのWT−hPRLとを補充したRPMI 1640培地中でBa/F3−hPRLR細胞を維持した。リガンドとしてWT hPRLを使用して、バイオアッセイの最適条件(細胞数、飢餓時間および培地など)を決定した。条件は以下のとおりであった。増殖アッセイの前では、細胞を、1%FCS RPMI培地(添加物を含む)において6時間飢餓にし、次いで96ウェルプレートにおいて、5×104細胞/ウェルの密度にて、100μlの最終容量で、同じ培地(ホルモンを除く)に播種した。アゴニスト作用実験では、100μlの[2×]ホルモンを添加し、アンタゴニスト作用実験では、アンタゴニスト特性について試験すべき50μlの[4×]ホルモンおよび50μlの[4×]WT hPRL(10ng/mlの最終濃度)を添加した。10μlのWST−1を使用したホルモン刺激の3日後に、細胞増殖をモニターした。3連または4連で少なくとも3回実験を行なった。
N末端欠失アナログ
アゴニスト作用
Nb2細胞増殖アッセイ
以前の報告によれば、単量体hPRLは、古典的なNb2細胞増殖アッセイにおいて、1〜2ng/mlにて最大効果を有する細胞増殖を誘導する。
Nb2細胞を用いるアッセイとは対照的に、Ba/F3細胞を用いるhPRLR媒介増殖アッセイは、これらアナログの異なる細胞分裂活性を示した。WT hPRLは、用量依存的様式でこの細胞集団の増殖を誘導し、ほぼ10ng/mlで最大効果を有した。このことは、慣用培養培地中のIL−3を10ng/mlのhPRLに置換することによる細胞選択と相関している。
3回の実験の代表として、二連で実施された1つの典型的な実験データを図5に示す。図5は、漸増濃度(μl/ml)のhPRL(−●−)、Δ1−9−hPRL(−□−)、Δ1−14−hPRL(−△−)、Δ1−10−hPRL(−−*−−)、Δ1−11−hPRL(−−○−−)、Δ1−12−hPRL(−−□−−)、Δ1−13−hPRL(−−◇−−)の存在下でのhPRL転写活性(WT hPRLの最大効果を100%として、何%の活性であるか)を示す。
本来のアゴニスト活性に一致して、いずれのN末端欠失変異体も、本研究に使用した3つのバイオアッセイのいずれにおいても、アンタゴニスト活性を示さなかった(データは示さず)。
すべての実験に、単一変異体(G129R−hPRL)ならびに二重変異体Δ1−9−G129R−hPRLおよびΔ1−14−G129R−hPRLが含まれる。
8週齢の野生型雌性balb−c/Jを、10μgのhPRLまたは異なる比のhPRL/アンタゴニスト(G129R−hPRLまたはΔ1−9−G129R−hPRL)で処置した。ホルモンの腹腔内(IP)注射の60分後、慣用プロトコルに従って、マウスを屠殺し、肝臓を迅速に採取し、切り分けそしてホモジナイズし、次いで細胞溶解物を調製した。70μgの溶解物を、10%SDS−PAGEにロードし、続いてニトロセルロース膜に液成分を移した。膜を、5%脱脂乳で1時間室温にてブロックし、そして十二分に洗浄した後、活性形態のErk1およびErk1 Mapキナーゼ(スレオニン202およびチロシン204でリン酸化されている)に対して特異的な一次モノクローナル抗体と共に一晩インキュベートした。十二分に洗浄後、膜を、西洋わさびペルオキシダーゼ(HRP)結合抗マウス二次抗体と共に1時間インキュベートした。イムノブロットを、増強化学発光(ECL)に続いてオートラジオグラフィーによって顕在化させた。

8週齢の授乳中(10〜12日目)の野生型雌性balb−c/Jを、200μgブロモクリプチンで処置(IP)して、内因性PRLの循環レベルを減少させた(ブロモクリプチンは、ドーパミンアナログの、下垂体プロラクチン分泌の天然阻害剤である)。5時間後、マウスを、10μgのhPRL単独か、または1:100比のhPRL:Δ1−9G129R−hPRLかで、30分間処置(IP)した。次いで、慣用プロトコルに従って、マウスを屠殺し、第4乳腺(左右)を取り出し、プールし、そしてホモジナイズした。総細胞溶解物(2mg)を、ポリクローナル抗STAT5抗体またはポリクローナル抗STAT3抗体をそれぞれ使用する免疫沈降に(4℃にて一晩、回転下で)使用した。次いで、免疫複合体を、回転下で1時間のインキュベーションによって、20μlのプロテインAセファローススラリを用いて捕獲した。プロテインA複合体を遠心分離により沈降させ、ペレットを洗浄し、次いで還元サンプル緩衝液中で5分間煮沸させた。免疫沈降サンプルを、実施例4に記載のように、7.5%SDS−PAGE、続いてウエスタンブロットで分析した。

アゴニスト(PRL;G129R−hPRL)
このインビボバイオアッセイは、遍在性メタロチオネインプロモーターの制御下でヒトPRLまたはG129R−hPRLを発現するトランスジェニックマウス(雄性)(TgMet−hPRLおよびTg Met−G129R)か、または前立腺特異的プロバシンプロモーターの制御下でラットPRLを発現するトランスジェニックマウス(雄性)(Tg Prob−rPRL)を使用する。

前立腺特異的プロバシンプロモーターの制御下でラットPRLを発現するトランスジェニックマウス系統の7ヶ月齢の雄に、1mgのΔ1−9G129R−hPRLを60分間注入(IP)した。次いで、マウスを屠殺し、尿生殖路を取り出し、そして前立腺を顕微鏡下で細かく切り分けて、背外葉から腹葉を分離した。慣用プロトコルに従って、組織をホモジナイズし、そして細胞溶解物を調製した。実施例4に記載されるように、70μgの溶解物を10%SDS−PAGEにロードし、続いてウエスタンブロットした。

Claims (12)
- アンタゴニストが、以下の変異:
a)N末端の14アミノ酸内の変異または変異のセットであって、Cys4とCys11との間のジスルフィドブリッジの形成を防止する、変異または変異のセット、および
b)プロラクチンの結合部位2内に立体障害を起こす変異または変異のセット
を有する哺乳動物プロラクチン変型体である、哺乳動物プロラクチンレセプターのアンタゴニスト。 - 変異a)がプロラクチンのN末端の少なくとも4残基の欠失を含む、請求項1に記載のプロラクチン変型体。
- 変異a)がプロラクチンのN末端の9残基の欠失を含む、請求項2に記載のプロラクチン変型体。
- 以下の変異:
−N末端の少なくとも9残基の欠失であって、N末端の14残基までの欠失、および
−G129R置換
を有する、請求項3に記載のプロラクチン変型体。 - ヒトプロラクチンの変型体である、請求項1から4のいずれかに記載のプロラクチン変型体。
- 請求項1から5のいずれか1つに記載のプロラクチン変型体をコードするポリヌクレオチド。
- 請求項6に記載のポリヌクレオチドを含む、発現カセット。
- 請求項6に記載のポリヌクレオチドを含む、組換えベクター。
- 請求項6に記載のポリヌクレオチドより形質転換された宿主細胞。
- 請求項6に記載のポリヌクレオチドで形質転換されたトランスジェニック非ヒト哺乳動物。
- または請求項6に記載のポリヌクレオチドを含む、治療組成物。
- PRLが媒介する効果を含む疾患を処置または防止する治療組成物を得るための、請求項1から5のいずれかに記載のプロラクチン変型体または請求項6に記載のポリヌクレオチドの使用。
