Description
当業者ならば、X線結晶学によって求められた一組の構造座標が標準誤差内にはないことがわかるであろう。本発明の目的では、タンパク質主鎖原子(N、α-C、CおよびO)の標準偏差(root mean square deviation)が、主鎖原子を用いて表1に挙げた構造座標に重ね合わせた場合に0.75Å未満であるヒトIII型3a-HSDまたはヒトIII型3a-HSD変異体に関するいずれの構造座標セットも同一であるものとする。
A. NADPとの複合体におけるヒトIII型3a-HSDの構造
本発明ははじめてポリエチレングリコール溶液からNADPの存在下で成長させたヒトIII型3a-HSDの結晶を提供する。この結晶は斜方六面体空間群対称を有し、0.5 x 0.5 x 0.2mmに達した。この結晶の単位格子は以下の寸法:a=b=c=108.5+/-1Å、α=β=γ=85.1°+/-1°である。結晶性ヒトIII型3a-HSDのX線結晶学によって決定されるヒトIII型3a-HSD 3α-HSDも構造座標は表1に示されている。結晶充填からはヒトIII型3a-HSDが単一のダイマーとして結晶化することが明らかである(表1のデータはダイマーに関するものである)。

本発明のヒトIII型3a-HSDと結合する化合物のデザインは一般に2つの因子を考慮する必要がある。1つには、その化合物がヒトIII型3a-HSDと物理的かつ構造的に会合しなければならない。ヒトIII型3a-HSDとその基質との会合において重要な非共有結合分子相互作用としては水素結合、ファンデルワールスおよび疎水性相互作用がある。
RT-PCRはGeneAmp PCR System 9700 (PE Appliad Biosystems)にて行う。第一鎖cDNA合成を45℃30分間の1サイクルで行った後、94℃2分間RT変性を行う。PCRは94℃15秒間の変性、55℃30秒間のプライマーアニーリング、および68℃2分間のプライマー伸長の40サイクルの後、72℃10分間の最終伸長1回で行う。
NMRスクリーニング
NMR分光光度法を用いるとIII型3a-HSDの阻害剤を発見およびデザインすることができる。これはリガンドおよび標的タンパク質間の相互作用を、たとえその相互作用が弱いものでしかなくとも、ミリモル範囲の親和性しか持たない場合でも検出できるというNMRの周知の能力によるものである(Diercks, T.; Coles, M.; Kessler, H. Applications of NMR in drug discovery. Curr. Opin. Chem. Biol. 2001, 5, 285-291; Hajduk, P. J.; Meadows, R. P.; Fesik, S. W. NMR-based screening in drug discovery. Q. Rev. Nature Reviews Drug Discovery 2002, 1, 211-219)。この目的で、化合物の混合物に3a-HSD溶液を加え、化合物のNMR特性(緩和率など)を測定し、3a-HSDの存在下と不在下で比較する。緩和率の上昇は3a-HSDとその化合物の間の結合親和性を示す。この作用は、3a-HSDの側鎖がTEMPO(2,2,6,6-テトラメチルピペラジン-N-オキシル)またはPROXYL(2,2,5,5-テトラメチルピロリジン-N-オキシル)などの常磁性中心でスピンラベルされていれば約15倍増強させることができる(Jahnke, W.; Ruedisser, S.; Zurini, M. 2-アセチルベンゾフランは3a-HSDと弱くしか結合しないが、進歩的な化合物であり、すなわち、これは化学修飾を受けやすい、非常に小さく可溶性であって「薬物様」の化合物であるのでその効力を向上させることができる。これをNMRにより達成するにはいくつかの方法がある。1つの可能性としては、部分構造または類似性検索により化合物を選択し、それらをNMRで試験することである。もう1つの可能性としては、NMRスクリーニングによって2-アセチルベンゾフランの近傍で結合する別の化合物を同定し、両化合物を化学的に結合させて高親和性リガンドを得ることである(Shuker, S. Current Topics in Medicinal Chemistry 2002, 2, 1337-1348 (in press))。しかし、3a-HSDの結晶構造から座標を得ることができれば、一番良い方法は、NMRスクリーニングから得られた結果を実施例3に記載のようなin silico GOLDドッキングと合成することである。2-アセチルベンゾフランのようなNMRスクリーニングヒットはGOLDドッキング実験のインプットとなり、in silicoでドッキングされた化合物を選択の指標となる。ドッキング自体、またその結果のスコアリングは完全に信頼できるものではないので、ドッキングの結果はNMRにより実験的にバリデートする必要がある。従って、NMRおよびドッキングは化合物の効力の劇的な向上をもたらす反復サイクルとなる。
