【発明の背景】
インテグリンαvβ3レセプターは、成長中でかつ遊走中の内皮細胞中で主に発現され、たとえば腫瘍血管形成時における血管細胞の血管形成表現型のマーカーとして同定されている。このαvβ3インテグリンはそれ自体、腫瘍細胞によっても発現される。このαvβ3レセプターは、したがって、腫瘍探索性分子の潜在的標的である。

腫瘍中に発生する別種のレセプターは、ソマトスタチンレセプターである。この存在は、種々の腫瘍中でおよび免疫細胞上で旧来の生化学結合法、オートラジオグラフィ、インサイチュハイブリッド形成およびRT−PCRによって明らかとなっている。リガンドをソマトスタチンレセプターに結合させると、リガンド/ソマトスタチンレセプター複合体のインターナライゼーションが結果として起こることが極めて多い。

ソマトスタチンレセプターに対する天然のリガンドは、ソマトスタチン、5種のソマトスタチンレセプターサブタイプ(亜型)(sst)全てに高いアフィニィティで結合する14または28個のアミノ酸の神経ペプチドである。ソマトスタチンはプラズマ中で容易に分解されるが、酵素分解に対して安定なソマトスタチンアナログが開発されている。臨床的に極めて広く使用されているアナログはオクトレオチドおよびランレオチドであり、これらの化合物は、sst2,3,および5に対して高いアフィニティで結合する。

【発明の開示】
本発明によって、腫瘍に対するある化合物の総体的アフィニティを向上させるため、前記化合物中において両レセプターに対してアフィニティを有するリガンドを組み合わせることを検討した。

この目的のため、本発明は、αvβ3レセプター、および(神経)ペプチドレセプター特にソマトスタチンレセプター、の両者に対して結合アフィニティを有する化合物に関し、この化合物は、少なくとも1回のアミノ酸配列Arg−Gly−Aspを含む第1ペプチド部とそれに対して任意にリンカーを介して結合した第2ペプチド部を含み、この第2ペプチド部は(神経)ペプチドである。

本発明は、ソマトスタチンに対してのみではなく他の(神経)ペプチドに対しても適用可能である。その結果、前記第2ペプチド部は、好適には、CCK、ガストリン、サブスタンスP、ボンベシン、VIP(血管作動性腸ペプチド)、PACAP(下垂体アデニレートシクレート活性化ペプチド)、ソマトスタチンおよびこれらのアナログから構成される群から選択される。アナログは、安定性または活性を向上させるため原ペプチドの修飾型であってもよいしまたはその生物活性をいまだ保持しているペプチドの部分であってもよい。

好適な態様において、前記第2ペプチド部は、好適にはオクトレオテート、オクトレオチド、ラントレオチド、バプレオチドまたはその誘導体から構成される群から選択されたソマトスタチンアナログである。

前記第1ペプチド部はいわゆるRGD−ペプチドであり、それは、Arg−Gly−Aspモチーフを少なくとも1回有するペプチドである。原RGD−ペプチドのアナログは、ヨード化用のチロシンのような付加的アミノ酸を含むことができる。RGD−アナログの好適な態様において、前記Tyrとリンカーの間にさらにAspが存在する。このAspは、RGD−ペプチド部を環化してそれをより安定にするように作用する。

ソマトスタチンアナログ(または他の(神経)−ペプチドおよびそれらのアナログ)に結合したRGD−ペプチドは、前記RGD−レセプター(αvβ3)または前記ソマトスタチンレセプターのひとつのいずれかにより、細胞に結合しかつ細胞に入る。異なる2種のレセプター、すなわち(神経)ペプチドレセプターおよびαvβ3インテグリンの化合物結合により、腫瘍細胞のような異なる標的細胞上ならびに腫瘍血管形成細胞上で前記化合物を見いだすことが期待できる。このことは、高い標的−バックグランド比に寄与できる。

本発明の化合物のプロトタイプは、RGD−オクトレオチドである。オクトレオチドは、ソマトスタチンレセプター(sst)2、3、および5型サブタイプに結合する安定ペプチド(プラズマ分解に対して耐性)である。他の化合物については、実施例に述べたとおりである。

sst−レセプターまたはαvβ3−レセプターを有する組織によるオートラジオグラフィ実験において、原ペプチドは両者とも1個の新規化合物に組み合わされるが、それらは両者共にそれら自体のレセプターに対して結合アフィニティを保持していることが見出された。それぞれのレセプターに対する前記新規化合物の結合は、過剰の異なる競合的アナログによってブロックできた。

(実施例1)
RGD−(神経)ペプチド結合体およびそれらの対応するDTPA−またはDOTA−誘導体合成の一般的方法
下記は、本発明の化合物調製の一般的方法である。

固相ペプチド合成(SPPS)は、Fmoc法を用いて、PEバイオシステムズ“パイオニア”(PE Biosystems “Pioneer”)シンセサイザーを用いて実施する。前記化合物の全アミノ酸から構成される線状ペプチドを、出発レジンとしてのFmoc−AA1(OtBu)−PEG−PS(PEバイオシステムズ、0.18mmol/gを載せる)により、0.1mmolスケールで調製する。ここで、開始レジンとしてAA1は、C末アミノ酸である。Fmoc保護アミノ酸(0.4mmol)をN−[(ジメチルアミノ)−1H−1,2,3−トリアゾロ[4,5−b]ピリジン−1−イルメチレン]−N−メチルメタンアミニウムヘキサフルオロホスフェートN−オキサイド(HATU)によって活性化する。全アミノ酸およびペプチド合成試薬は、市販のものを購入した。

前記ペプチドのオンボードアミド結晶化は、“アリルデブロック(Allyl Deblock)”プロトコール(Pd(PPh3)4、N−メチルモルホリン、酢酸、クロロホルム)を用いて行い、その後、7−アザベンゾトリアゾール−1−イルオキシトリス(ピロリジノ)−ホスホニウムヘキサフルオロホスフェート(PyAOP)活性化を行う。保護環化ペプチドを含むレジンを、その後、機器から取り出す。

前記レジンを、タリウムトリフルオロアセテート92mg含有ジメチルホルムアミド8ml中に懸濁させる。前記混合物を2−3時間振とうさせ、ろ過し、DMF10mL、DMG−水(1:1)10mL、DMFおよびTHF10mLによって連続的に洗浄し、前記レジンに付着した保護ペプチド(III)を得る。その後、このレジンを2個の部分に分ける。

前記ペプチドをレジンから切断し、85%TFA/5%水/5%フェノール/5%チアニソールを用いて10−12時間脱保護する。粗ペプチドを、t−ブチルメチルエーテルによる沈殿により単離し、その後遠心し、0.1%TFAを含むアセトニトリル/水勾配(溶媒A:0.1%TFA/H2O、溶媒B:0.1%TFA/10%H2O/CH3CN;勾配:95%A/5%Bに2.0分間保持した後、溶媒A(100%)から50%A/50%Bまで20分かける)を用いて逆相HPCLによって精製する。

リシンのMtt保護基は、5%TFA/5%トリイソプロピルシラン(TIPS)/90%ジクロロメタン(2×30分)処理により除去する。このレジンをジクロロメタンおよびテトラハイドロフランによって洗浄し、DIEA(35μl、0.2mmol)含有DMF(2.5mL)中に懸濁させる。別の容器では、HBTU/HOBt(0.2mmol、0.2mmol/mL溶液を1.0mL)を含有するDMF中にトリ−t−ブチルDTPA無水物またはDOTA(112mg,0.2mmol)を溶解させ、5mL溶液を得る。1時間攪拌後、活性化DTPA誘導体を先に懸濁させたレジンに添加する。

前記レジンを、タリウムトリフルオロアセテート92mg含有ジメチルホルムアミド8ml中に懸濁させる。前記混合物を2−3時間振とうさせ、ろ過し、DMF10mL、DMG−水(1:1)10mL、DMFおよびTHF10mLによって連続的に洗浄し、前記レジンに付着した保護ペプチド(III)を得る。その後、このレジンを2個の部分に分ける。

前記ペプチドをレジンから切断し、85%TFA/5%水/5%フェノール/5%チアニソールを用いて10−12時間脱保護する。粗ペプチドを、t−ブチルメチルエーテルによる沈殿により単離し、その後遠心し、0.1%TFAを含むアセトニトリル/水勾配(溶媒A:0.1%TFA/H2O、溶媒B:0.1%TFA/10%H2O/CH3CN;勾配:95%A/5%Bに2.0分間保持した後、溶媒A(100%)から50%A/50%Bまで20分かける)を用いて逆相HPCLによって精製する。

リシンのMtt保護基は、5%TFA/5%トリイソプロピルシラン(TIPS)/90%ジクロロメタン(2×30分)処理により除去する。このレジンをジクロロメタンおよびテトラハイドロフランによって洗浄し、DIEA(35μl、0.2mmol)含有DMF(2.5mL)中に懸濁させる。別の容器では、HBTU/HOBt(0.2mmol、0.2mmol/mL溶液を1.0mL)を含有するDMF中にトリ−t−ブチルDTPA無水物またはDOTA(112mg,0.2mmol)を溶解させ、5mL溶液を得る。1時間攪拌後、活性化DTPA誘導体を先に懸濁させたレジンに添加する。

反応を一晩連続させ、レジンをDMFおよびTHFで洗浄する。

前記ペプチドをレジンから切断し、10−12時間、85%TFA/5%水/5%フェノール/5%チアニソールを用いて脱保護した。粗ペプチドをt−ブチルメチルエーテルによる沈殿により単離し、その後遠心し、0.1%TFAを含むアセトニトリル/水勾配(溶媒A:0.1%TFA/H2O、溶媒B:0.1%TFA/10%H2O/CH3CN;勾配:95%A/5%Bに2.0分間保持した後、溶媒AからBまで20分かける)を用いて逆相HPCLによって精製する。

Claims (8)
- αvβ3レセプター、および(神経)ペプチドレセプター特にソマトスタチンレセプター、の両者に対して結合アフィニティを有する化合物であって、当該化合物は、少なくとも1回のアミノ酸配列Arg−Gly−Aspを含む第1ペプチド部と、それに対して任意にリンカーを介して結合した第2ペプチド部とを含み、当該第2ペプチド部は(神経)ペプチドであることを特徴とする化合物。
- 前記第2ペプチド部は、好適には、CCK、ガストリン、サブスタンスP、ボンベシン、VIP(血管作動性腸ペプチド)、PACAP(下垂体アデニレートシクラーゼ活性化ペプチド)、ソマトスタチンおよびこれらのアナログから構成される群から選択される請求項1記載の化合物。
- 前記第2ペプチド部がソマトスタチンアナログである請求項2記載の化合物。
- 前記ソマトスタチンアナログが、オクトレオテート、オクトレオチド、ラントレオチド、バプレオチドまたはその誘導体から構成される群から選択される請求項3記載の化合物。
- 前記リンカーが少なくとも1個のリシンを含む請求項1乃至4のいずれかに記載の化合物。
- 前記化合物が放射性標識されている請求項1乃至5のいずれかに記載の化合物。
- 前記放射性標識が、213Bi,186Re,188Re,77As,90Y,66Ga,67Cu,169Er,114mIn,117mSn,121Sn,127Te,142Pr、143Pr,198Au,199Au,149Tb,161Tb,109Pd,165Dy,149Pm,151Pm,153Sm,157Gd,159Gd,166Ho,172Tm,169Yb,175Yb、177Lu,105Rh,103mRh,195mPt,111Ag,124I,131Iおよび211At,99mTc,203Pb,67Ga,68Ga,72As,111In,113mIn,97Ru,62Cu,64Cu,52Fe,52mMn、51Cr,123I,131I、75Br,76Br,77Brおよび82Brから構成される群から選択された放射性同位元素による請求項6記載の化合物。
- 生物活性分子としての用途のためである請求項1乃至7のいずれかに記載の化合物。
