【発明の属する技術】
本発明は、酸化されたアポリポタンパク質AI及び/又は同抗原を含む高密度リポタンパク質(酸化HDL)の免疫学的測定方法及びそれに使用するキットに関するものである。

【従来の技術】
これまでの疫学的研究により、高密度リポタンパク質(High Density Lipoprotein;HDL)は動脈硬化に対し予防的な効果を持つことが明らかにされている。この予防的効果はHDLの末梢からのコレステロールの搬出機能に依存すると考えられている。すなわち、アポリポタンパク質AIが肝臓、小腸などから遊離型コレステロールを受け取って生じた原始型HDLは、血流に乗って末梢へ循環する間にLCAT酵素によってコレステロールを更に取り込んで成熟型HDLとなり、この成熟型HDLが末梢にて蓄積したコレステロールを肝臓へと運搬する。

さらに、HDLの高い酸化感受性が抗動脈硬化作用に寄与していることも明らかにされつつある。すなわち、HDLは他のリポタンパク質と比較し、抗酸化物質であるユビキノール10含量が少なく、生体内においてはLDLよりも優先的に酸化される。このため、HDLは動脈硬化の原因物質である酸化LDLの生成を抑制し、さらに、酸化LDLの生体内でのクリアランスにも関与していると考えられている。

このようなHDLは、他のリポタンパク質と比較し、50%という高いタンパク質含量を持つリポタンパク質である。HDLの蛋白成分はアポリポタンパク質AI、同AII、同Cおよび同Eによって構成され、それらはHDLの構造維持や機能発現に寄与している。それらの中でもアポリポタンパク質AIは、蛋白成分の約70%を占め、HDLの主要な機能であるコレステロールの運搬を担っている。

生体内でHDLが酸化される場合、脂質の酸化と並行してアポリポタンパク質AIも酸化され、アポリポタンパク質AIが酸化されることによりHDLの凝集や分解を引き起こすことが知られている。また、酸化による構造的あるいは化学的な変性によって、HDLのコレステロールの輸送能力が減少することも知られている。

【発明が解決しようとする課題】
このように、アポリポタンパク質AIは、HDLの抗動脈硬化作用に大きく影響し、酸化に対する感受性も非常に高いことから、酸化されたアポリポタンパク質AI及び/又は同抗原を含むHDL(酸化HDL)は、脂質酸化や動脈硬化の進展と強い関連性が指摘されている生体内における酸化ストレスを鋭敏に検出するためのマーカーとなり、該マーカーの測定は動脈硬化のメカニズムの解析または臨床診断に大いに役立つものと考えられる。

従来、酸化された脂質に対する抗体とアポリポタンパク質AIに対する抗体を用いて酸化されたHDLを測定する方法(特開平6−253888号、特開平9−33525号、特許3251848号など参照)は報告されているものの、酸化されたアポリポタンパク質AI及び/又は同抗原を含むHDL(酸化HDL)を特異的に測定する方法は報告されていない。

また、上記従来法において、アポリポタンパク質AIは主にHDLに含まれるタンパク成分であり、アポリポタンパク質AIに対する抗体はHDLに対する特異性が期待されるものの、各リポタンパク質の脂質成分は同じであり、酸化された脂質に対する抗体は特定のリポタンパク質に対しての特異性に懸念が残る。

さらに、脂質を含まないアポリポタンパク質AIを主成分とする原始型HDLは、分子サイズが小さいため、間質内に分散し、動脈内膜にて酸化されることが示唆されており(Biochim. Biophys. Acta, 1483, 217−35.(2000))、酸化されたアポリポタンパク質AIを特異的に測定することは、HDLに特異的であり、かつ脂質を含まない原始型HDLと脂質を含む成熟型HDLの両方を測定しうると予想される。

【課題を解決するための手段】
本発明者らは、簡便で感度の高い酸化アポリポタンパク質AIの測定法を開発すべく鋭意研究を重ねた結果、酸化されたアポリポタンパク質AIの酸化部位に特異的に反応するに対する抗体とアポリポタンパク質AIに対する抗体との2種類の抗体を使用し、サンドイッチ法により試料中に存在する酸化されたアポリポタンパク質AI及び/又は酸化されたアポリポタンパク質AIを含む高密度リポタンパク質(酸化HDL)を特異的かつ高感度で測定できることを見いだした。

この測定系を用いて酸化アポリポタンパク質AI及び/又は酸化されたアポリポタンパク質AIを含む高密度リポタンパク質(酸化HDL)と臨床症状との関連に関して検討した結果、糖尿病合併症(腎症、神経障害など)において酸化アポリポタンパク質AIが有意に増加し、酸化アポリポタンパク質AIが生体内の酸化ストレスの度合いを測定するためのマーカーのみならず、糖尿病合併症の診断又は予知のマーカーとしても利用可能であることを確認し、本発明を完成させた。

日本において、中高年の4人に1人が糖尿病の発症の可能性を潜在的に有していると考えられており、糖尿病は、続発する合併症により、患者のQOL並びに医療経済にも多大な影響を及ぼす疾患である。糖尿病の発症や合併症の併発のメカニズムについては不明な点も多く、その発症に酸化ストレスが関与しているも指摘されているが(Diabetes Metab Res Rev.17:189−212.(2001)、Diabetes Res.Clin. Pract. 45:147−51(1999)、 Am. J. Med. 30:17S−26S (1999))、酸化ストレスを指標にした糖尿病合併症の予防、予知に関する研究に関する報告は、現時点で見あたらない。このような状況下、酸化アポリポタンパク質AIが糖尿病合併症の診断又は予知のマーカーとしても利用可能であることは、まったく予想外のことである。

したがって、本発明は、抗体として酸化されたアポリポタンパク質AIの酸化部位に特異的に反応する抗体と、アポリポタンパク質AIに対する抗体との2種類の抗体を使用し、サンドイッチ法により試料中に存在する酸化されたアポリポタンパク質AIを特異的に測定する方法及びキットに関するものである。

【発明の実施の形態】
本発明で使用する試料は、酸化されたアポリポタンパク質AIを含有するものであれば特に限定されず、具体的には血液自体または血清、血漿などの血液画分を試料として使用することができる。このような試料中には、酸化されたアポリポタンパク質AI及び/又は酸化されたアポリポタンパク質AIを含むHDL(酸化HDL)の形態で存在する可能性があり、本発明の方法によれば、いずれの形態であっても測定することが可能である。

本発明で使用する抗体は、測定対象の抗原によって異なり、たとえば、酸化されたアポリポタンパク質AI及び/又は酸化されたアポリポタンパク質AIを含むHDL(酸化HDL)の場合には、酸化されたアポリポタンパク質AIの酸化部位に特異的に反応する抗体とアポリポタンパク質AI抗体との2種類の抗体を使用する。また、測定対象の抗原が酸化されたアポリポタンパク質AIを含むHDL(酸化HDL)単独の場合には、酸化されたアポリポタンパク質AIの酸化部位に特異的に反応する抗体と高密度リポタンパク質(HDL)の構成成分(脂質またはタンパク質)に対する抗体との2種類の抗体を使用する。このような抗体は、抗体自体あるいはそれらの活性フラグメント〔F(ab’)2、Fab’など〕であってもかまわない。

ここで、酸化されたアポリポタンパク質AIの酸化部位に特異的に反応する抗体とは、酸化されたアポリポタンパク質AIを構成するアミノ酸の酸化部位や酸化によるアポリポタンパク質AIの構造的変化を認識し、これと結合する抗体を意味する。特に、本発明においては、酸化されたアポリポタンパク質AIの酸化部位に特異的に反応する抗体としてモノクローナル抗体が好ましく、このような特異性を有するモノクローナル抗体を固相に結合させた固相化抗体の形態で使用することにより、より高感度に酸化されたアポリポタンパク質AIを測定することが可能である。

このような酸化されたアポリポタンパク質AIの酸化部位に特異的に反応する抗体は、常法により作製することができる。たとえば、酸素ラジカル発生剤などを用いて調製した酸化アポリポタンパク質AIを抗原とし、常法によりモノクローナル抗体を調製し、得られた抗体の中から酸化していないアポリポタンパク質AIとは結合せず、酸化型のアポリポタンパク質AIとに結合する抗体を選出すればよい。

また、アポリポタンパク質AIに対する抗体とは、アポリポタンパク質AIが酸化変性を受けた場合でもその結合能が変化しない抗体であり、このような抗体はHDL中のアポリポタンパク質AIと遊離したアポリポタンパク質AIの両方を認識する。さらに、高密度リポタンパク質(HDL)の構成成分(脂質またはタンパク質)に対する抗体とは、HDLが酸化変性を受けた場合でもその結合能が変化しない抗体であり、このような抗体はHDL中のタンパク質及び/又は脂質を認識する。
このような抗体としては、モノクローナル抗体、ポリクローナル抗体のいずれであってもかまわない。

上記アポリポタンパク質AIに対する抗体およびHDLの構成成分に対する抗体は、対応する抗原(アポリポタンパク質AI、HDLまたはその構成成分)を用いて常法により抗体(ポリクローナル抗体、モノクローナル抗体)を調製することができる。さらに、このような抗体は既に市販されており、この市販品を利用することも可能である。

本発明は、上述のような抗体を使用し、サンドイッチ法により試料中の酸化アポリポタンパク質AI及び/又は同抗原を含むHDL(酸化HDL)を測定しようとするものである。

サンドイッチ法自体は、従来行われている方法、手順など何等変わるものではない。しかし、上述したように、酸化されたアポリポタンパク質AIの酸化部位に特異的に反応するモノクローナル抗体を固相に結合させた固相化抗体を固相試薬として使用することにより、より高感度に酸化されたアポリポタンパク質AI及び/又は同抗原を含むHDL(酸化HDL)を測定することが可能である。

固相試薬を調製する際に使用する担体としては、通常使用されているもの、たとえば、ポリ塩化ビニル、ポリスチレン、スチレン−ジビニルベンゼン共重合体、スチレン−無水マレイン酸共重合体、ナイロン、ポリビニルアルコール、ポリアクリルアミド、ポリアクリロニトリル、ポリプロピレン、ポリメチレンメタクリレートなどの合成有機高分子化合物、デキストラン誘導体(セファデックスなど)、アガロースゲル(セファロース、バイオゲルなど)、セルロース(ペーパーディスク、濾紙など)などの多糖類、ガラス、シリカゲル、シリコーンなどの無機高分子化合物を例示することができ、これらはアミノ基、カルボキシル基、カルボニル基、水酸基、スルヒドリル基などの官能基が導入されたものであってもよい。

担体の形状は、平板状(マイクロタイタープレート、ディスクなど)、粒子状(ビーズなど)、管状(試験管など)、繊維状、膜状、微粒子状(ラテックス粒子など)、カプセル状、小胞体状などいずれの形状であってもよく、測定法に応じて好適な形状の担体を適宜選択すればよい。

担体と抗体の結合は、物理的吸着法、イオン結合法、共有結合法、包括法など公知の方法〔たとえば、「固定化酵素」(千畑一郎編、昭和50年3月20日、(株)講談社発行)参照〕を採用することができ、とりわけ、物理的吸着法は簡便である点で好ましい。また、上記結合は直接行ってもよく、両物質の間に他の物質を介して行ってもよい。

このようにして得られた固相試薬は、非特異的結合を抑制するため、ゼラチン、BSAなどの通常のブロッキング剤を用いてブロッキング処理を施してもよい。

また、検出に使用する標識剤としては、放射性同位体(たとえば32P、3H、14C、125Iなど)、酵素(たとえばβ−ガラクトシダーゼ、ペルオキシダーゼ、アルカリホスファターゼ、グルコース−6−リン酸デヒドロゲナーゼ、カタラーゼ、グルコースオキシダーゼ、乳酸オキシダーゼ、アルコールオキシダーゼ、モノアミンオキシダーゼなど)、補酵素・補欠分子族(たとえば、FAD、FMN、ATP、ビオチン、ヘムなど)、フルオレセイン誘導体(たとえば、フルオレセインイソチオシアネート、フルオレセインチオフルバミルなど)、ローダミン誘導体(たとえば、テトラメチルローダミンBイソチオシアネートなど)、ウンベリフェロンおよび1−アニリノ−8−ナフタレンスルホン酸などの蛍光色素、ルミノ−ル誘導体〔たとえば、ルミノール、イソルミノール、N−(6−アミノヘキシル)−N−エチルイソルミノールなど〕などを例示することができる。

抗体と標識剤との結合は、成書〔たとえば、「続生化学実験講座5免疫生化学研究法」(株)東京化学同人、(1986年発行)第102〜112頁〕に記載されているような公知の方法から適宜選択して実施すればよい。

このような固相試薬と抗体試薬を用いての測定手順は、通常のサンドイッチ法の手順をそのまま採用すればよい。すなわち、固相試薬と被検サンプルを反応させ、必要によりBF分離後、さらに抗体試薬を反応させる(ツーステップ法)か、固相試薬、被検サンプル及び抗体試薬を同時に反応させ(ワンステップ法)、以後のそれ自体公知の方法によりサンプル中の酸化されたアポリポタンパク質AIを検出または定量することができる。

本発明の方法及びキットは、ヒトを含む動物の生体内における酸化ストレスの度合い及び該酸化ストレス関連疾患の検出に利用可能である。より具体的には、後述実施例に示すように、例えば、糖尿病合併症(腎症、神経障害など)などの陽性群においては、酸化アポリポタンパク質AIの有意な増加が認められることから、血中の酸化アポリポタンパク質AI及び/又は酸化アポリポタンパク質AIを含むHDL(酸化HDL)を測定し、その測定値を健常人の数値あるいは当該疾患の基準値と比較することで、動脈硬化、糖尿病合併症(腎症、神経障害など)などの発症や進展を検出することが可能である。

【発明の効果】
本発明により簡便で実用的な酸化アポリポタンパク質AI及び/又は酸化アポリポタンパク質AIを含むHDL(酸化HDL)の測定法及びキットを初めて提供することが可能となった。また、本発明の方法及びキットは、サンプル中の酸化アポリポタンパク質AI及び/又は酸化アポリポタンパク質AIを含むHDL(酸化HDL)を定量するために十分な感度を有し、生体内における酸化ストレスを検出することが可能となり、動脈硬化、糖尿病合併症(腎症、神経障害など)などの酸化ストレス関連疾患のメカニズムの解析または臨床診断に利用可能である。

【実施例】
以下、本発明を実施例をあげて具体的に説明するが、本発明はこれらによって何等限定されるものではない。
実施例1
<方法>
(1)酸化アポリポタンパク質の調製
ヒトアポリポタンパク質AI,AII,B,CII,E(Chemicon,U.S.)500mgを、酸素ラジカル発生剤である2,2’−azobis(2−amidinopropane)dihydrochloride(4mM)(AAPH;和光純薬)に添加し、37℃、3時間反応させて酸化アポリポタンパク質を得た。

(2)抗酸化アポリポタンパク質AI抗体の作製
常法に従い、酸化アポリポタンパク質AIを免疫したBALB/Cマウスの脾細胞とミエローマ細胞SP2/O−Ag14を細胞融合して得られたハイブリドーマから酸化されたアポリポタンパク質AIに強く結合する抗体を産生するクローンを複数選出し、モノクロナール抗体を得た。このような抗体のうち、今回はモノクロナール抗体3C11(FERM P−18759:平成14年3月12日 独立行政法人産業技術総合研究所 特許生物寄託センターに寄託)を用いた。

(3)酸化リポタンパク質の調製
リポタンパク質はヒトEDTA血漿より密度勾配比重遠心法により精製した。得られたリポタンパク質はゲル濾過カラムクロマトグラフィー(Biochem.J.,139, 89−95.(1974))でさらに分画し、目的のリポタンパク質以外のリポタンパク質の混入がみられないフラクションのみを実験に用いた。得られたリポタンパク質の蛋白量をLowryらの方法により定量し、1mg/mlの濃度に調整後、これらに1μMとなるよう硫酸銅を添加し、37℃で3時間インキュベートした。

(4)抗体の特異性試験
酸化アポリポタンパク質AIに対する抗体3C11並びにアポリポタンパク質AIに対する抗体(ヤマサ社製)の抗原特異性を固相化した各抗原に対する免疫活性により検討を行った。すなわち、各抗原(1μg/ml:リン酸緩衝生理食塩水(PBS)、pH7.4)を96穴マイクロプレート(Nunc Immunoplate I)に100μl毎分注し、4℃で1晩反応させた。抗体液を吸引除去後、プレートをPBSで洗浄し、300μlの0.5%スキムミルクを含むPBSにより各ウェルをブロッキングした(一昼夜、4℃)。ブロッキング液を吸引除去後、1%ウシ血清アルブミンを含むPBSにて希釈した40ng/mlの抗酸化アポリポタンパク質AI抗体3C11と1ng/mlの抗アポリポタンパク質AI抗体(ヤマサ社製)を1時間反応させ、0.05% Tween20を含むPBSで各ウェルを洗浄後、抗マウスIgG抗体西洋ワサビペルオキシダーゼ標識体を100μl加え室温で1時間反応させた。
反応後、0.05% Tween20を含むPBSで洗浄し、発色液(SAT−BLUE、同仁化学)を100μl毎分注し発色させた。1N硫酸にて反応を停止後、450nmによって吸光度を測定した。

(5)ELISA法
モノクロナール抗酸化リアポリポタンパク質AI抗体3C11溶液を96穴マイクロプレート(Nunc Immunoplate I)に100μl毎分注し、4℃で1晩反応させた。抗体液を吸引除去後、PBSで洗浄し、300μlの0.5%スキムミルクを含むPBSにてブロッキング(4℃、一晩)を行った。反応後ブロッキング液を吸引除去し、プレートを乾燥した。この96穴マイクロプレートの各ウエルに検体あるいは標準濃度溶液を100μl毎加え、37℃、3.5時間反応した。0.05% Tween20を含むPBSで洗浄した後、標識二次抗体として抗アポリポタンパク質AI抗体の西洋ワサビペルオキシダーゼ標識体を100μl加え37℃、1.5時間反応した。
反応後、0.05% Tween20を含むPBSで洗浄し、発色液(SAT−BLUE、同仁化学)を100μl毎分注し発色した。1N硫酸にて発色を停止後、450nmによって吸光度を測定した。なお、血清検体は1%ウシ血清アルブミン並びに0.15M塩化ナトリウムを含む50mMトリス塩酸(pH7.4)によって20倍に希釈したものを使用した。また、標準液としては40mMAAPHにより酸化したアポリポタンパク質AIを用いた。

<結果>
(1)抗原特異性試験
固相に結合させた抗原を用いて抗酸化アポリポタンパク質AI抗体3C11の反応性特異性を試験したところ、本抗体が酸化されたアポリポタンパク質AIと酸化されたHDLに特異性を示すことが明らかとなった(図1,2)。このことから、酸化されたアポリポタンパク質AIに対する抗体はHDL中の酸化されたアポリポタンパク質AIにも反応する抗体であることが明らかとなった。より具体的には、酸化HDLを固相したときの吸光度は1.309であり、未酸化のHDLのそれより6倍高く、また、酸化アポリポタンパク質AIを固相したときの吸光度は1.009であり、未酸化のアポリポタンパク質AIのそれより4倍高かった。なお、他のアポリポタンパク質並びにHDL以外のリポタンパク質においては、その吸光度は最大で0.11であり、交差反応性は認められなかった。一方、抗アポリポタンパク質AI抗体は、HDL及びアポリポタンパク質AIの酸化によってその吸光度に有意な変化はなく、この抗体が酸化に関与する部分以外を認識していることが明らかとなった。

(2)標準曲線
酸化アポリポタンパク質AIの標準液を使用して作成した上記ELISA法の標準曲線を図3に示す。標準曲線は2ng/mlから40ng/mlまで直線性を示し、なおかつゼロ点に収束する良好な反応性が認められた。

(3)本ELISA法の特異性の解析
本ELISA法にてアポリポタンパク質AI並びにその酸化物を測定した。測定された結果より、これら1mgあたりの酸化アポリポタンパク質AI濃度(ng/mg(タンパク質))を算出し、その反応性を比較した(図4)。その結果、4mMAAPHにて酸化したアポリポタンパク質AIは未酸化のそれと比較し、12倍の特異性を有し、この測定系が酸化されたアポリポタンパク質AIを特異的に認識していることが明らかとなった。また、40mMAAPHにて酸化したアポリポタンパク質AIは未酸化のそれと比較し、730倍であり、より特異性が向上した。

Blood was collected from a type II diabetic patient, and the obtained serum was used as a sample. The diagnosis of type II diabetes was in accordance with WHO criteria (Diabet Med. 15: 539-53. (1998)).
The oxidized apolipoprotein AI in the serum sample was measured by the above-mentioned ELISA method using a 3C11 monoclonal antibody. Statistical analysis used a one-tailed test of the student-t test, and P <0.05 was set as the significance level.

The clinical characteristics of the patients from whom the samples were taken are summarized in Table 2. Using such a sample, the concentration of oxidized apolipoprotein AI in the serum was measured. As shown in FIG. 6, a significant increase in the concentration of oxidized apolipoprotein AI was observed in the diabetic nephropathy and neuropathy disease positive groups. Was observed.
This suggests that oxidative stress may be increased in patients with these diseases, suggesting that oxidized apolipoprotein AI can be used as an in vivo oxidative stress marker and a marker for diabetic complications. .
