前記変異が、トリプトファン(W)の、セリン(S)、イソロイシン(I)またはフェニルアラニン(F)による置換からなる

【請求項2】 338番のトリプトファン(W)のセリン(S)による置換、427番のトリプトファン(W)のセリン(S)での置換、及び96番のトリプトファン(W)のフェニルアラニン(F)での置換の少なくとも一つを更に有する請求項1に記載の単離精製された変異体。
【請求項3】 ヒト免疫不全ウイルス 1型(HIV−1)のgp120エンベロープ糖タンパク質の単離精製された変異体であって、
(1)427番のトリプトファン(W)のセリン(S)による置換からなるアミノ酸の変異、または
(2)338番のトリプトファン(W)のセリン(S)による置換からなるアミノ酸の変異、
を有するヒト免疫不全ウイルス 1型(HIV−1)のgp120エンベロープ糖タンパク質の単離精製された変異体。
【請求項4】 請求項1〜3のいずれかに記載の単離精製された変異体の、HIVワクチンの製造における抗原としての使用方法。
【請求項5】 ヒト免疫不全ウイルス(HIV)のgp120エンベロープ糖タンパク質(HIV gp120)の感染性を抑制するための方法であって、
前記HIV gp120を請求項1〜3のいずれかに記載の変異体を用いる工程を有する

【請求項2】 338番のトリプトファン(W)のセリン(S)による置換、427番のトリプトファン(W)のセリン(S)での置換、及び96番のトリプトファン(W)のフェニルアラニン(F)での置換の少なくとも一つを更に有する請求項1に記載の単離精製された変異体。
【請求項3】 ヒト免疫不全ウイルス 1型(HIV−1)のgp120エンベロープ糖タンパク質の単離精製された変異体であって、
(1)427番のトリプトファン(W)のセリン(S)による置換からなるアミノ酸の変異、または
(2)338番のトリプトファン(W)のセリン(S)による置換からなるアミノ酸の変異、
を有するヒト免疫不全ウイルス 1型(HIV−1)のgp120エンベロープ糖タンパク質の単離精製された変異体。
【請求項4】 請求項1〜3のいずれかに記載の単離精製された変異体の、HIVワクチンの製造における抗原としての使用方法。
【請求項5】 ヒト免疫不全ウイルス(HIV)のgp120エンベロープ糖タンパク質(HIV gp120)の感染性を抑制するための方法であって、
前記HIV gp120を請求項1〜3のいずれかに記載の変異体を用いる工程を有する
HIV gp120の感染性を抑制するための方法。
また、位置112における変異に加えて、このような変異体は、位置383のFのアラニンへの変異、場合によっては、位置427のトリプトファンのグリシンでの置換、及び/または、位置479のトリプトファンの、セリンまたはイソロイシンのような非芳香族アミノ酸での置換を含むことができる。

結論
gp120におけるW338S変異は、融合と感染を妨げるトランスコンフォメーションを引き起こす。というのも、エンベロープがトランスフェクションされた細胞の表面に現われる場合には、この変異は、融合を妨げ、このエンベロープが補足されたウィルス上に位置する場合には感染を妨げる。さらに、このトランスコンフォメーションは、主に、変異したgp120が、このgp120がCD4に複合化されることなく、AcM CG10によって認識されることから、野生gp120上で隠された部位を明らかにすることができる。この蛋白質がCD4およびCXCR4に結合される場合に、この同一AcM CG10がそのエピトープにもはやアクセスせず、抗CD4i抗体が、CD4と衝突しないという事実は、CXCR4への定着内にもたらされるのは、gp120上でAcM CG10によって認識されたエピトープであることを意味している。このような変異体は、HIV感染に対する保護目的での免疫化を行うための候補を構成する。

結論
gp120におけるW338S変異は、融合と感染を妨げるトランスコンフォメーションを引き起こす。というのも、エンベロープがトランスフェクションされた細胞の表面に現われる場合には、この変異は、融合を妨げ、このエンベロープが補足されたウィルス上に位置する場合には感染を妨げる。さらに、このトランスコンフォメーションは、主に、変異したgp120が、このgp120がCD4に複合化されることなく、AcM CG10によって認識されることから、野生gp120上で隠された部位を明らかにすることができる。この蛋白質がCD4およびCXCR4に結合される場合に、この同一AcM CG10がそのエピトープにもはやアクセスせず、抗CD4i抗体が、CD4と衝突しないという事実は、CXCR4への定着内にもたらされるのは、gp120上でAcM CG10によって認識されたエピトープであることを意味している。このような変異体は、HIV感染に対する保護目的での免疫化を行うための候補を構成する。
