Claims (1)
- 【特許請求の範囲】 1.哺乳類神経上皮幹細胞の単離された純粋な集団であって、該細胞が接着支 持細胞非依存性培養培地中で自己再生することができ、かつCNSニューロン細 胞又はCNSグリア細胞、及び神経冠幹細胞に分化することができる、前記集団 。 2.神経上皮幹細胞がヒト並びに非ヒト霊長類、ウマ、イヌ、ネコ、ウシ、ブ タ、ヒツジ及びウサギからなる群から選択された1種の哺乳類から単離される、 請求項1に記載の集団。 3.神経上皮幹細胞がネスチンを発現するが、N-CAM-、GFAP-、O4- 、NF-、ChAT-、β-IIIチューブリン-、A2B5-、及びGalC-である、請求項1 に記載の集団。 4.CNSニューロン細胞がβ-IIIチューブリン+及び及びNF+である、請求 項1に記載の集団。 5.CNSニューロン細胞がChAT+である、請求項4に記載の集団。 6.CNSグリア細胞がGFAP+である、請求項1に記載の集団。 7.CNSグリア細胞がA2B5+である、請求項6に記載の集団。 8.CNSグリア細胞がA2B5+である、請求項1に記載の集団。 9.CNSグリア細胞がO4+である、請求項8に記載の集団。 10.CNSグリア細胞がGalC+である、請求項8に記載の集団。 11.神経上皮幹細胞がグリア限定前駆細胞に更に分化することができる、請 求項1に記載の集団。 12.グリア限定前駆細胞が接着支持細胞非依存性培養培地中で自己再生する ことができ、かつCNSグリア細胞には分化できるがCNSニューロン細胞には 分化できない、請求項11に記載の集団。 13.グリア限定前駆細胞がネスチンを発現しそしてA2B5+であり、かつG FAP-、O4-及びGalC-である、請求項12に記載の集団。 14.CNSグリア細胞がA2B5+及びGFAP+である、請求項12に記載の 集団。 15.CNSグリア細胞がGFAP+であり、かつA2B5-である、請求項1 2に記載の集団。 16.CNSグリア細胞がGalC+であり、かつA2B5-である、請求項12に記 載の集団。 17.哺乳類CNSグリア限定前駆体細胞の単離された純粋な集団であって、 該グリア限定前駆細胞が接着支持細胞非依存性培養培地中で自己再生することが でき、かつCNSグリア細胞には分化できるがCNSニューロン細胞には分化で きない、前記集団。 18.グリア限定前駆細胞がネスチンを発現し、A2B5+であり、かつ、GF AP-、O4-及びGalC-である、請求項17に記載の集団。 19.CNSグリア細胞がA2B5+及びGFAP+である、請求項17に記載の 集団。 20.CNSグリア細胞がGFAP+であり、かつA2B5-である、請求項17 に記載の集団。 21.CNSグリア細胞がGalC+であり、かつA2B5-である、請求項17に記 載の集団。 22.哺乳類CNS神経上皮幹細胞の純粋な集団を単離する方法であって、該 細胞が支持細胞非依存性接着培養培地中で自己再生することができ、該CNSニ ューロン細胞又はCNSグリア細胞に分化することができる、該方法において、 (a)神経管の閉鎖後であるが神経管内で細胞が分化する前の胚発生段階で哺 乳類の胚から神経管を取り出す工程; (b)哺乳類の胚から取り出した神経管を含んでいる細胞を分離する工程; (c)基層上であって、かつ有効量の線維芽細胞増殖因子及びニワトリ胚抽出 物を含む、神経上皮幹細胞の接着増殖を支持すべく調製された培地中に分離した 細胞を播種し、支持細胞非依存性培養する工程;及び (d)神経上皮幹細胞の増殖を誘導する温度及び雰囲気中で上記の播種細胞を インキュベートする工程 を含む、前記方法。 23.哺乳類の胚が霊長類、ウマ、イヌ、ネコ、ウシ、ブタ、ヒツジ及びウサ ギからなる群から選択される、請求項22に記載の方法。 24.基層がフィブロネクチンを含む、請求項22に記載の方法。 25.温度が約37℃であり、かつ、雰囲気が約5%のCO2と約95%の空気を 含む、請求項22に記載の方法。 26.培地がNEP培地を含む、請求項22に記載の方法。 27.哺乳類CNSグリア限定前駆細胞の純粋な集団を単離する方法であって 、該細胞が接着支持細胞非依存性培養培地中で自己再生することができ、かつ、 CNSグリア細胞には分化できるがCNSニューロン細胞には分化できない該方 法において、 (a)哺乳類CNS神経上皮幹細胞の集団を単離する工程; (b)神経上皮幹細胞が分化を開始するのに十分な期間、有効量のニワトリ胚 抽出物を欠いている培地中で該細胞をインキュベートする工程; (c)インキュベートした細胞をグリア限定前駆細胞に特徴的な抗体を使用す る特異的な抗体捕獲に付して、捕獲された細胞亜集団を生じさせる工程;そして (d)捕獲細胞亜集団を、有効量の線維芽細胞増殖因子及び血小板由来増殖因 子を含み、該集団の接着増殖を支持すべく調製された培地中でインキュベートす る工程 を含む、前記方法。 28.CNS神経上皮幹細胞集団の単離が: (1)神経管の閉鎖後であるが神経管内で細胞が分化する前の胚発生段階で哺 乳類の胚から神経管を取り出す工程; (2)哺乳類の胚から取り出した神経管を含んでいる細胞を分離する工程; (3)基層上に、かつ有効量の線維芽細胞増殖因子及びニワトリ胚抽出物を含 み、神経上皮幹細胞の接着増殖を支持すべく調製された培地中に分離した細胞を 播種し、支持細胞非依存性培養する工程;そして (4)神経上皮幹細胞の増殖を誘導する温度及び雰囲気中で播種細胞をインキ ュベートする工程 を含む、請求項27に記載の方法。 29.哺乳類の胚が霊長類、ウマ、イヌ、ネコ、ウシ、ブタ、ヒツジ及びウサ ギからなる群から選択される、請求項28に記載の方法。 30.基層がフィブロネクチンを含む、請求項28に記載の方法。 31.温度が約37℃であり、かつ、雰囲気が約5%のCO2と約95%の空気を 含む、請求項28に記載の方法。 32.哺乳類運動ニューロンの集団を産生させる方法であって、 (a)哺乳類CNS神経上皮幹細胞の集団を単離する工程;及び (b)細胞が分化を開始するのに十分な期間、細胞の増殖及びニューロンの分 化を促進する培地中で神経上皮幹細胞をインキュベートする工程、 を含む、前記方法。 33.培地がラミニン被覆プレートと、有効量のニワトリ胚抽出物を欠いてい るNEP培地を含む、請求項32に記載の方法。 34.神経上皮幹細胞をインビトロで分化するように誘導し、それによって哺 乳類神経冠幹細胞を産生させる工程を含む、哺乳類神経冠幹細胞の産生方法。 35.誘導が神経上皮幹細胞をラミニン被覆基質上に再播種する工程を含む、 請求項34に記載の方法。 36.誘導がマイトジェンを除去する工程を更に含む、請求項35に記載の方 法。 37.マイトジェンがFGFである、請求項36に記載の方法。 38.誘導がニワトリ胚抽出物を除去する工程を更に含む、請求項35に記載 の方法。 39.誘導がマイトジェンを除去する工程を含む、請求項34に記載の方法。 40.マイトジェンがFGFである、請求項39に記載の方法。 41.誘導がニワトリ胚抽出物を除去する工程を含む、請求項34に記載の方 法。 42.誘導が細胞に背方化剤を添加する工程を含む、請求項34に記載の方法 。 43.背方化剤が骨形態形成タンパク質からなる群から選択される1種のメン バーである、請求項42に記載の方法。 44.背方化剤がBMP-2である、請求項43に記載の方法。 45.背方化剤がBMP-4である、請求項43に記載の方法。 46.背方化剤がBMP-7である、請求項43に記載の方法。 47.末梢神経系の細胞を産生させる方法であって、 (a)神経上皮幹細胞をインビトロで分化するように誘導し、それによって神 経冠幹細胞を産生させる工程、及び (b)神経冠幹細胞をインビトロで分化するように誘導し、それによって末梢 神経系細胞を産生させる工程、 を含む、前記方法。
