序論 発明の分野
発明の分野は、細胞機能を調節するポリペプチド分子、このポリペプチドをコ
ードする核酸配列、ならびにこの核酸配列およびこのポリペプチドを使用する方
法である。背景
腫瘍壊死因子α(TNF-α)は、活性化マクロファージによって主として産生さ
れるサイトカインである。TNF-αは、T細胞およびB細胞増殖を刺激し、そして
内皮細胞における接着分子の発現を誘導する。このサイトカインも、感染に対す
る宿主防御に重要な役割を果たす。
TNF-α活性は、2つの異なるレセプター、TNFR-p55およびTNFR-p75によって媒
介される。これらの2つのレセプターはまた、主として活性化リンパ球により分
泌される可溶性リンホトキシンα(LT-α)によって引き起こされる活性を媒介

TNFは、特にγインターフェロン(IFN-γ)と組み合わせて、サイトカインの
何らかの他の組み合わせによって適合されない悪性細胞株を選択的に殺傷または
阻害する能力を有する。しかし、TNFの毒性副作用が、ヒトにおける有効な用量
レベルを得ることを抑制したので、TNF-α抗腫瘍治療でのガン患者の臨床試験は
、期待はずれであった。これらの毒性副作用は、TNFR-p75レセプターへのTNF結
合のせいであったが、悪性細胞における細胞傷害性活性は、TNFR-p55レセプター

発明の要旨
本発明は、腫瘍壊死因子(TNF)と相同である分子である。本発明は、この分
子、ヒトNTN-2ポリペプチド、および関連核酸に関する方法および組成物を提供

本発明のヒトNTN-2ポリペプチドに好ましい使用には、添加されたポリペプチ

発明の詳細な説明
本発明は、天然ヒトNTN-2ポリペプチド、ならびにヒトNTN-2アミノ酸配列、ま
たはアッセイ認識可能なヒトNTN-2特異的活性を有するその機能的ヒトNTN-2ポリ
ペプチドドメインを含む組換えポリペプチドを含むヒトNTN-2ポリペプチドを提
供する。従って、ポリペプチドは、開示された天然ヒトNTN-2ポリペプチドの欠
失変異体であり得、そして例えば、非ヒトNTN-2ポリペプチドとの融合産物とし

本発明のポリペプチドは、免疫原として、スクリーニングアッセイにおける標
的として、細胞増殖、分化、および/または機能などを調節するための生物活性
試薬としての使用を含む、種々の使用を見いだす。例えば、本発明は、添加され
たポリペプチドが培地の成分および/または細胞の生理機能の変化に影響を及ぼ
す細胞外表面と特異的に相互作用する条件下で、外因性ヒトNTN-2ポリペプチド
と細胞または細胞のまわりの培地とを接触させることによって、細胞外表面を含

明細書に開示されるヌクレオチド配列およびそのフラグメントを含む核酸は、天
然の染色体上で連結されるもの以外の配列に直に隣接した、または天然の染色体

開示されたヒトNTN-2ポリペプチドのアミノ酸配列は、選択された発現系(Hol
lerら(1993)Gene 136:323-328;Martinら(1995)Gene 154:150-166)につい
て最適化されたヒトNTN-2ポリペプチドをコードする核酸を逆翻訳するために使
用され、または天然のヒトNTN-2をコードする核酸配列の単離における使用のた
めの縮重オリゴヌクレオチドプライマーおよびプローブを生成するために使用さ
れる(「GCG」ソフトウエア,Genetics Computer Group,Inc.,Madison,WI)
。ヒトNTN-2をコードする核酸は、発現ベクターの一部であり得、そして例えば
、発現およびスクリーニングのため、トランスジェニック動物のため、ヒトNTN-

SSPE(0.18M NaCl、0.01M NaPO4、pH 7.7、0.001M EDTA)緩衝液中30%ホルムア

ヒトNTN-2ハイブリダイゼーションプローブは、臨床および実験室試料におい
て野生型および変異体対立遺伝子を同定することにおける使用を見いだす。変異
体対立遺伝子は、高処理能力比臨床診断のための対立遺伝子特異的オリゴヌクレ
オチド(ASO)プローブを生成するために使用される。ヒトNTN-2核酸はまた、活

するmRNAに結合し得るアンチセンス転写物を得るように配向したプロモーター配

明細書に記載のヒトNTN-2ポリペプチドに対する抗体を提供する。このヒトNTN-2
ポリペプチドに対するモノクローナル抗体の調製については、培養物中の連続細
