Claims (1)
- 【特許請求の範囲】 1.母方の血球細胞と胚又は胎児赤血球細胞又はその両方を含んだ試料中にお ける胚又は胎児赤血球細胞を同定する方法であって、 この方法は、 どちらの細胞又は細胞群が成人肝臓成分を含んでいるか又は発現しているかを 決定することを含む方法。 2.母方の血球細胞と胚又は胎児赤血球細胞又はその両方を含んだ試料中にお ける胚又は胎児赤血球細胞を分離する方法であって、 この方法は、 成人肝臓成分を含んでいるか又は発現している前記細胞を分離することを含む 方法。 3.前記試料は、妊娠メス由来の血液試料である請求項1または2記載の方法 。 4.前記妊娠メスはヒト女性であり前記試料は妊娠3ヵ月内に採取されたもの である請求項3記載の方法。 5.前記胚又は胎児赤血球細胞は有核巨大芽細胞系である請求項1または2記 載の方法。 6.前記成分はたんぱく質である請求項1〜5のいずれか一つに記載の方法。 7.前記母方血液の母方細胞には実質的に前記成分が無い請求項1〜6のいず れか一つに記載の方法。 8.前記たんぱく質は、ミクロソームのグルコース−6−ホスファターゼ酵素 、前記リン酸又はグルコース又はグルコース−6−ホスフェイトランスポーター 、ウリジンジホスフェイトグルクロノシル転移酵素、シトクロームP450イソ 酵素、ニコチンアミドアデニンジヌクレオチドホスフェイトシトクロームP45 0還元酵素、グルコーストランスポーター2、P−グライコプロテイン、MDR P、MRP、γ−グルタミルトランスペプチダーゼ、リポプロテイン受容体、ア ルカリホスファターゼ、胆汁塩運搬体、胆汁酸運搬体、ホルモン受容体、多有機 イオン運搬体、ビリルビン運搬体、又はビリルビン複合運搬体を含むグルコース −6−ホスフェイト系の他のたんぱく質成分のうちいずれか一つである請求項1 または2のいずれかに記載の方法。 9.前記成人肝臓成分が細胞表面に露出された成分である先行する請求項のい ずれか一つに記載の方法。 10.前記細胞表面に露出された成分は成人肝臓細胞膜たんぱく質である請求 項9記載の方法。 11.成人肝臓細胞膜たんぱく質はグルコーストランスポーター2、P−グラ イコプロテイン、MDRP、MRP、γ−グルタミルトランスペプチダーゼ、リ ポプロテイン受容体、アルカリホスファターゼ、胆汁塩運搬体、胆汁酸運搬体、 ホルモン受容体、多有機イオン運搬体、ビリルビン運搬体、又はビリルビン複合 運搬体のいずれかである請求項10記載の方法。 12.前記試料は、前記成人肝臓成分に結合する結合部分に接触し、前記胚又 は胎児細胞は、前記結合部分に結合される効果によって前記試料中から同定され 又は分離される先行する請求項のいずれか一つに記載の方法。 13.前記試料は酵素の基質に接触されていて、前記酵素は成人肝臓成分であ り、前記胚又は胎児細胞は前記基質上の前記酵素の働きによって形成される生成 物によって試料から同定され、または分離される請求項1〜11のいずれか一つ に記載の方法。 14.前記結合部分は抗体、またはその断片もしくは派生物である請求項12 記載の方法。 15.前記結合部分は固形支持体に固定されている請求項12または14に記 載の試料から胚又は胎児赤血球細胞を分離する方法。 16.前記結合部分は検出可能に標識されている又は検出可能である請求項1 2または14に記載の方法。 17.前記標識は前記細胞の分離を促進する請求項16に記載の試料から胚又 は胎児赤血球細胞を分離する方法。 18.前記生成物は蛍光又は着色されている請求項13に記載の方法。 19.胎児の異常性を決定する方法であって、 この方法は、 前記請求項のいずれか一つに記載の方法に基づいて胚又は胎児細胞を同定又は 分離すること、 そして前記胎児の異常性について胚又は胎児細胞を分析すること を含む方法。 20.遺伝物質の分析によって前記胎児細胞の異常性を決定する請求項19記 載の方法。 21.染色体の異常が検出される請求項20に記載の方法。 22.前記DNA中の突然変異が検出される請求項20に記載の方法。 23.(a)細胞が成人肝臓成分を含んでいる又は発現しているかどうかを決 定するための手段と、 (b)細胞の異常性を分析する手段と を含む胎児異常性検定のための設備一式。 24.請求項19〜22のいずれか一つによる方法において請求項12に定義 される結合部分の使用。 25.細胞が胚または胎児赤血球細胞を同定または分離するための成人肝細胞 成分を含んでいるか、または発現しているかどうかを決定するための手段の使用 。 26.ここに記載される母方血液中の胚又は胎児細胞を分析することによって 胎児異常性を検出するあらゆる新規な方法。 27.ここに記載される胚もしくは胎児細胞を分離または同定するあらゆる新 規な方法。
