Claims (1)
- 【特許請求の範囲】 1. 約1cm2のフィルター面につき、約1〜10mlのウイルス含有溶液を 濾過することを含んでなる、異なる大きさのウイルスを分離する方法。 2. ウイルス含有溶液が1以上の濾過膜を通して濾過する、請求項1記載の方 法。 3. 濾過膜が、ポリフッ化ビニリデン、ポリテトラフルオロメチレン、ポリプ ロピレン、変性もしくは非変性ポリエーテルスルホン、セルロースアセテー ト、セルロースニトレート、ポリアミドまたは再生セルロース、例えば銅ア ンモニア再生セルロース、好ましくはポリフッ化ビニリデン、で構成される 、請求項2記載の方法。 4. 濾過膜が、最大約120nmまで、特に最大約60nmまで、特別には最 大約50nmまで、好ましくは約25〜60nmの細孔径を有する、請求項 2または3のいずれかに記載の方法。 5. ウイルスが、約250nmまで、好ましくは約120nmまで、特に約7 0nmまで、特別には約60nmまでの大きさのものである、請求項1〜4 のいずれか1項に記載の方法。 6. 分離されるウイルスの大きさの差が、少なくとも約5nm、特別には少な くとも約30nm、最も好ましくは約40nmである、請求項1〜5のいず れか1項に記載の方法。 7. ウイルスが、特に正二十面体対称を有する実質的球形である、請求項1〜 6のいずれか1項に記載の方法。 8. ウイルスが、約60nmまでの直径を有する第1群のウイルスから、約6 0nm〜約120nmまでの直径を有する第2群のウイルスから、および/ または約120nm〜約250nmまでの直径を有する第3群のウイルスか ら選択される、請求項1〜7のいずれか1項に記載の方法。 9. 第1群のウイルスが、フラビビリダエ(Flaviviridae)、パピローマウイル ス、パポーバウイルス、ヘパドナウイルス、ピコルナウイルス、またはパル ボウイルスに、特にAAVに、由来する、請求項8記載の方法。 10. 第2群のウイルスが、インフルエンザウイルス、レトロウイルス、アデ ノウイルス、レオウイルス、またはトガウイルスに、特にアデノウイルスに 、由来する、請求項8または9記載の方法。 11. 第3群のウイルスが、パラミクソウイルス、またはヘルペスウイルスに 、特にヘルペスウイルスに、由来する、請求項8〜10のいずれか1項に記 載の方法。 12. ウイルス含有溶液を予備精製する、請求項1〜11のいずれか1項に記 載の方法。 13. 予備精製を1以上の密度勾配の手段により、および/または1以上の予 備濾過の手段により達成する、請求項1〜12のいずれか1項に記載の方法 。 14. 予備濾過に1以上の膜フィルターを用いる、請求項13記載の方法。 15. 請求項8または9記載の第1群のウイルスを予備精製するための膜フィ ルターが少なくとも約60nmの細孔径を有し、一方、請求項8〜10のい ずれか1項に記載の第1および/または第2群のウイルスを予備精製するた めの膜フィルターが少なくとも約120nmの細孔径を有し、かつ、請求項 8〜11のいずれか1項に記載の第1群、第2群および/または第3群のウ イルスを予備精製するための膜フィルターが少なくとも約250nmの細孔 径を有する、請求項14記載の方法。 16. ウイルス含有溶液のpHが、約6〜10、好ましくは約7.0〜8.5 、特に約8.0である、請求項1〜15のいずれか1項に記載の方法。 17. ウイルス含有溶液がさらにタンパク質を含有する、請求項1〜16のい ずれか1項に記載の方法。 18. 溶液が、ウシ胎児血清または血清アルブミンの形態のタンパク質を含有 する、請求項17記載の方法。 19. タンパク質の割合が、約30%(v/v)以下、好ましくは約15%(v/v)、 特に約10%(v/v)である、請求項17または18記載の方法。 20. 約1cm2のフィルター面につき、約2〜5mlの溶液、特に約1cm2 のフィルター面につき、約2〜2.5ml以下の溶液を濾過する、請求項1 〜19のいずれか1項に記載の方法。
