フッ素化芳香族化合物およびフッ素化窒素含有複素環式芳香族化合物の調製法
本発明は、式Iのフッ素化芳香族化合物
{式中
Wは NまたはCR3、
Xは NまたはCR4、
Yは NまたはCR5、
Zは NまたはCR6、
R1、R2、R3、R4、R5およびR6は同一かまたは異なり、水素、F、Cl、B
r、I、NO2、CF3、CN、CHO、COF、SO2F、SO2Cl、OCF3
、SCF3、SO2CF3、COOR7、CONR8R9、SO2R10、COR11もし
くはOR12であるか、または互いにオルト位に位置して基R1ないしR6である2
つの基が一緒に−OC−NR13−CO−となり、
R7、R8、R9、R10、R11、R12およびR13は同一かまたは異なり互いに独立
して、水素、直鎖状もしくは分枝鎖状のC1−C6アルキル、アリール、好ましく
はフェニル、またはアリールアルキルであることができ、また該アルキル基も該
アリールアルキル基も両方とも、適切な場合には3度までハロゲンで置換させる
ことができ、そして該基は好ましくはCF3およびCHalxH3-xであることが
でき、また
2個以上の基R1〜R13は、適切な場合には相互に結合して1個以上の環を形成
し、そして該環は3〜7環員を有することができる。
ただし下記を条件とする。
(i)W、XおよびYが同時にNであってはならず、
(ii)基R1〜R6の少なくとも1つがフッ素でなければならず、また
(iii)a)式Iの化合物が芳香族単核ハロゲン化化合物である場合、基R1〜R6
の残留基の少なくとも1つはNO2、CF3、CN、CHO、COOH、COC
l、COF、SO2F、SO2Cl、SO2CF3、CONR8R9、SO2R10、ま
たはCOR11であり、また
b)式Iの化合物が好ましくは芳香核に3個のハロゲン置換基を有する芳香
族多核ハロゲン化化合物である場合、基R1〜R6の残留基はさらにすべて水素で
あることができる}の調製法であって、
式Iに対応するが、その中で式Iではフッ素でなければならない基R1〜R6のそ
の1つがClまたはBrである化合物を、
式IIのフッ化物
MeF (II)
{式中、MeはNa、K、Cs、Rb、N(R14)4またはP(R15)4であり、
ここで4個の基R14は互いに独立して同一かまたは異なり、水素または直鎖状も
しくは分枝鎖状のC1−C8アルキルであることができ、そして該アルキル基はさ
らにたとえば芳香族基によって置換させることができ、また4個の基R15は互い
に独立して同一かまたは異なり、R14の意味を有することができ、さらにフェニ
ルであることもできる}と
反応媒質中で反応させることによる方法に関する。
芳香族環に結合された水素(aromatisch gebundenen Wasserstoff)をフッ素で
置換することは生物活性物質の合成およびこのような化合物の前駆物質の調製に
極めて重要である。さらに、フッ素が化合物の生物学的活性に強力かつしばしば
予期しない効果を及ぼすことは周知である。生物学的活性分子における水素のフ
ッ素による置換は、拡充され、または修正された生物学的効果を有する類似化合
物を生成することが多い。
直接フッ素化以外に、フッ素によってハロゲン(Cl、Br)を置換すること
(いわゆる「ハレックス(halex)法」)によるフッ素含有芳香族化合物の
調製は工業的に重要な極めて価値のある反応である。

特許および出版物のD1〜D5は概してKF、CsFまたは(Alk)4N+F-
を用いるハロゲン原子の直接置換によるフッ素含有芳香族化合物の調製法を記
載する。しかし、CsFまたは(Alk)4 N+F-は高コストのために工業的に
現在用いられているのはKFだけである。しかしながらこの化合物の使用は多く
の重大な欠点を招く。
一般に、ほとんどの非プロトン性有機溶剤中のKFの溶解度は極めて小さく約
10-3mol/l以下の範囲にある。これは溶解しているフッ化物イオンの濃度
が低いことを意味し、実際に長い反応時間、高エネルギー消費、および高設備費
をもたらす。さらに、高温を用いなければならない必要性を生じ、その結果副
生物の生成をもたらすことも多い。
Cl−F置換は、活性芳香族化合物の場合には通常約100〜200℃の範囲
の温度で、また非活性芳香族化合物の場合には約250〜400℃の範囲の温度
で行われる。とくに激烈な反応条件(D10参照)は活性がないかまたはごく僅
かしか活性のない芳香族化合物の場合に必要とされ、多くの副生物を生成する。
この反応は溶剤中または無溶媒で行うことができる。しかし、反応媒質として非
プロトン性高極性溶剤を用いるときに最良の結果が得られる。
アセトニトリル(CH3CN)、ジメチルホルムアミド(DMFA)、ジメチ
ルアセトアミド(DMAA)、N−メチルピロリドン(NMP)、ジメチルスル
ホキシド(DMSO)、テトラメチレンスルホン(スルホランまたはTMS)、
ヘキサメチルリン酸トリアミド(HMPA)、ピリジルニトリル(PyrCN)
またはベンゾニトリル(PhCN)が溶剤として用いられている。
CH3CNおよびDMFAは低沸点(それぞれ80℃および150℃)のため
に高活性ハロゲン原子を置換させるのに適するにすぎない。高収量のフッ素含有
生成物は、無機塩を溶解させる顕著な能力を有することが公知であるDMSOを
溶剤として用いるときに達成される場合が多い。しかし、DMSOの低熱安定性
は(CH3)2Sおよび(CH3)SO2への分解をもたらす。これら分解生成物は
さらに不快臭を生じ、この不快臭は強さが分解と一致し、もしもできれば避ける
べきものである。
概して、同じ基質を用いる場合に、KFによるフッ素化の速度は、つぎのよう
に溶剤に依存して増大する:アセトニトリル(CH3CN)→N−メチルピロリ
ドン(NMP)→テトラメチレンスルホン(スルホラン)→ジメチルスルホキシ
ド(DMSO)(D6参照)。
(HMPA)はハレックス反応の溶剤として顕出した性質を示すけれども、毒
性があるために工業プロセスには適切ではない。
反応混合物中のKFの溶解度を増し、対応してフッ素化の活性を向上させるた
めに多数の触媒が用いられている。こうしてD7はクラウンエーテルまたはポリ
エチレングリコール(PEG)の添加を推奨するが、D8およびD9はそれぞれ
アンモニウム塩およびホスホニウム塩の添加を提唱する。しかし、触媒の添加は
通常コスト高を意味する。また追加の排水の汚染をも生じ、また添加した触媒が
満足のいくように触媒作用を呈しないことも多い。
したがって、本明細書に示され、論じられている技術水準の現状を考慮すると
、本発明の目的は、はじめに述べたような方法であって、特定の目標化合物を良
好な収量かつ高選択率で調製できる方法を明示することにある。この新規方法は
、大規模の工業用途に適し、環境汚染をできるだけ少なくし、同時に比較的簡単
な手段を用いて経済的に実施できるように意図される。またこの方法は、とくに
最新技術で利用可能なプロセスに現在まで付随している前記欠点が最大限に除去
されるように意図される。
この目的および詳細に述べなかった他の諸目的は、はじめに述べたような、請
求項1の権利範囲特徴部分の特徴を有する方法によって達成される。
本発明の方法の適切な修正は請求項1に従属するサブクレームで保護される。
本発明に属する使用は請求項11および12の主題である。
ハロゲン置換によるフッ素化を、式IIIの1−アルキル−2,3−ジヒドロ−
1H−ホスホル−1−オキシド、式IVの1−アルキル−2,5−ジヒドロ−1H
−ホスホル−1−オキシドおよび/または式Vの1−アルキル−ホスホラン−1
−オキシド {式中、
R16は炭素原子が1〜8個の直鎖状または分枝鎖状であることができるアルキル
基、フェニル、ベンジル、シクロアルキル、OR25またはNR26R27(式中R25
〜R27は互いに独立して同一かまたは異なり、炭素原子が1〜4個の直鎖状また
は分枝鎖状であることができるアルキル基)であり、また
R17、R18、R19、R20、R21、R22、R23およびR24は互いに独立して同一か
または異なり、水素、フェニル、または直鎖状もしくは分枝鎖状のC1-8アルキ
ル基である}を含有する反応媒質中で行うという事実によって、
得られた処理生成物の選択性、収量および品質に関して、容易に予測できなかっ
た方法で、公知の方法を改良する方法を提供することにおいて特に有利に成果が
達成される。
驚くべきことに、文献によりそれ自体公知である式III、IVおよび/またはV
の化合物を溶剤として使用すると、ハレックス反応が実質的に低い温度、したが
って従来可能であったよりもはるかに温和な条件で実施できる結果として、該反
応を強力に促進させ得ることが見出された。このことは副生物の生成が抑制され
るかまたは完全に避けられる結果をも生じる。
とくに、本発明の方法は下記の利点を示す:
式III〜Vの化合物は無毒かまたは極めて低毒性の高沸点で熱に安定な液体で
ある。
同時にたとえば、式IIIないしVの反応媒質中のKFの溶解度は、とくに高温
においてTMS、NMPまたはDMAA中の溶解度よりも実質的に勝り、かつD
MSO中の溶解度に匹敵し、その結果ハロゲン置換による芳香族化合物のフッ素
化はあまり手のこんだ方法を用いずに完了に至り、反応時間およびエネルギー消
費
を低減させる。
これに加えて、式III〜Vの化合物の熱応力に耐える能力は、たとえば最新技
術において公知であり、しばしば用いられるDMSOの能力よりも著しく高い。
したがって、R17〜R24が水素でR16がメチルである式Vの化合物を用いるとき
には、250℃で7日後に最高1.4%という炭素質分解生成物が見出される。
とくにこれらの理由から、ハレックス反応の反応媒質中の、または反応媒質と
しての式III〜Vの化合物の使用は、概して環境にやさしい化学的方法である。
最後に、式III〜Vの化合物は広い沸点範囲に及び、その結果期待する目標生
成物の沸点によって適当な化合物を具体的に選ぶことにより、ほとんどの場合フ
ッ素化後反応混合物を分溜することによって反応混合物を仕上げることができる
。
これらの手段によって、式III〜Vの化合物を分離することも、またリサイク
ルすることも可能である。
本発明の範囲内において、式IIIないしVの化合物は、ハレックス反応の反応
媒質中に存在するとき、すなわち、他の点では普通のフッ素化の反応媒質にハロ
ゲン置換によって付加物を形成するときに、すでにこれらの利点を生じることが
できる。本発明の利点はこのようにしてすでに達成させることができる。これに
関連して、たとえ僅かな添加でもハレックス反応に明白な影響を与えることがで
きるので式III〜Vの化合物の比率は変更可能である。本発明によって反応媒質
中に存在する式II〜Vの化合物の比率は、式Iに相当するが、その中で式Iでは
フッ素である基R1〜R6のその1つがClまたはBrである化合物を基準にして
、好ましくは1.0〜100mol%、とくには5.0〜80mol%、特に好
ましくは10〜60mol%である。
反応媒質の総重量を基準にして50重量%(重量/重量)を上回る比率がとく
に適切である。
しかし、本方法の特別の変形では、ハレックス反応の反応媒質として式IIIの
1−アルキル−2,3−ジヒドロ−1H−ホスホル−1−オキシド、式IVの1−
アルキル−2,5−ジヒドロ−1H−ホスホル−1−オキシドおよび/または式
Vの1−アルキル−ホスホラン−1−オキシドを用いることが好ましい。このこ
とは、反応媒質の大部分が前記化合物によって構成されることを意味する。つぎ
に
本発明による利点をとくに述べる。これに関連して副次的な量の他の溶剤又は希
釈剤を排除しない限りは、少量(10重量%(重量/重量)未満)に限って添加
することができる。
本発明によって希釈剤または溶剤として用いるのに適する式IIIの1−アルキ
ル−2,3−ジヒドロ−1H−ホスホル−1−オキシド、式IVの1−アルキル
−2,5−ジヒドロ−1H−ホスホル−1−オキシドおよび/または式Vの1−
アルキル−ホスホラン−1−オキシドの大部分は商品として得ることができ、し
たがって入手可能である。市販されていない式III〜Vの化合物は当業者が熟知
する方法を用いて容易に合成することができる。合成の例は、とりわけDE−A
28 26 621またはDE−A35 14 451に示されている。
たとえば、1−メチル−2,5−ジヒドロ−1H−ホスホル−1−オキシド(
化合物IVa)を調製するのにとりわけ好ましい径路はブタジエンから出発して下
記反応機構1に従って進行する。 式III〜Vの化合物を調製する他の典型的な径路はエチレンオキシドから出発
する(反応機構2参照)。
さきに述べたように、希釈剤もしくは溶剤として、または希釈剤もしくは溶剤
の添加剤として前記反応媒質を使用することができる。この新規化合物は純粋な
形で、または2種以上の化合物の混合物として用いることができる。
式III〜Vに包含される種々の化合物は種々の沸点を有する。芳香族出発化合
物および生成物の沸点に依存して、また求める生成物の仕上げに依存して、他の
反応媒質への添加剤としてであろうと、純粋な形もしくは混合物のような反応媒
質としてであろうと、特別に作った化合物または式III〜Vの幾つかの化合物の
最適混合物を、それぞれ個々の場合に選ぶことができる。
本発明によって用いるのが好ましい化合物には、とりわけ下記のものがある:
1−メチル−2,3−ジヒドロ−1H−ホスホル−1−オキシド;
1−エチル−2,3−ジヒドロ−1H−ホスホル−1−オキシド;
1−n−プロピル−2,3−ジヒドロ−1H−ホスホル−1−オキシド;

1−n−ブチル−2,3−ジヒドロ−1H−ホスホル−1−オキシド;
1−イソブチル−2,3−ジヒドロ−1H−ホスホル−1−オキシド;
1−sec−ブチル−2,3−ジヒドロ−1H−ホスホル−1−オキシド;

式III〜Vの化合物中本発明の範囲内でとくに好ましいものは、R16がメチル
でR17〜R24が水素である式III、IVおよび/またはVの1種以上の化合物の中
でハロゲン置換によってフッ素化が好適に行われる1−メチル−2,3−ジヒド
ロ−1H−ホスホル−1−オキシド(化合物IIIaに等しい)、1−メチル−2
,5−ジヒドロ−1H−ホスホル−1−オキシド(化合物IVaに等しい)および
/または1−メチルホスホラン−1−オキシド(化合物Vaに等しい)である。
この新規方法は、とくに好適な態様において、ハロゲン置換によるフッ素化を
式Vの1−アルキル−ホスホラン−1−オキシドで行うという実施にさらに修正
し、そして1−メチル−ホスホラン−1−オキシド(化合物Vaに等しい)が極
めてとくに好適である。
この新規方法は式Iに対応する多数の化合物に適用することができ、そしてこ
の点に関し大きな分子中の芳香族サブユニット(untcreinheit)として式Iの構
造を含む基質(Substrate)も包含される。
本発明の範囲内で、式VIの化合物
{式中
R1’〜R5’は同一かまたは異なり、水素、F、Cl、Br、I、NO2、CF3
、CN、CHO、COF、SO2F、SO2Cl、OCF3、SCF3、SO2CF3
、COOR7、CONR8R9、SO2R10、COR11もしくはOR12であるか、ま
たは互いにオルト位に位置して基R1’〜R5’である2つの基が一緒に−OC−
NR13−CO−となり、そしてR7〜R13は式Iと同じであり、ただし、式VIの
化合物が芳香族単核ハロゲン化化合物である場合、基R1’〜R5’の少なくとも
1つの基はNO2、CF3、CN、CHO、COOH、COCl、COF、SO2
F、SO2Cl、SO2CF3、CONR8R9、SO2R10またはCOR11であり、
また式VIの化合物が好ましくは芳香核に3個のハロゲン置換基を有する芳香族多
核ハロゲン化化合物である場合、R1’〜R5’の残留
基はさらにすべて水素であることができる}を、方法を好適に修正して、式VII
の化合物
(式中、HalはClまたはBrであり,R1’〜R5’は式VIに示された意味を
有する)から調製する。
本発明によれば、式VIIの化合物
(式中、
R1’〜R4’は同一かまたは異なり、水素、F、Cl、Br、I、NO2、CF3
、CN、CHO、COF、SO2F、SO2Cl、OCF3、SCF3、SO2CF3
、COOR7、CONR8R9、SO2R10、COR11もしくはOR12であるか、ま
たは互いにオルト位に位置して基R1’〜R4’である2つの基が一緒に−OC−
NR13−CO−となり、そしてR7〜R13は式Iと同じであり、ただし、式VIII
の化合物が複素環式芳香族単核ハロゲン化化合物である場合、基R1’〜R4’の
少なくとも1つの基はNO2、CF3、CN、CHO、COOH、COCl、CO
F、SO2F、SO2Cl、SO2CF3、CONR8R9、SO2R10、COR11で
あり、また式VIIIの化合物が好ましくは芳香核に3個のハロゲン置換基を有する
複素環式芳香族多核ハロゲン化化合物である場合、基R1’〜R4’の残留基はさ
らにすべて水素であることができる)も、
下式IXの対応する塩素化または臭素化化合物
(式中、R1’〜R4’は式VIIIにおけると同じ意味を有し、またHalはClま
たはBrである)から極めてうまく調製される。
最後に、とくに好ましい態様において、この新規方法は、式X、XIおよびX
IIまたはXIIIの化合物(式中、
R1’〜R3’は同一かまたは異なり、水素、F、Cl、Br、I、NO2、C
F3、CN、CHO、COF、SO2F、SO2Cl、OCF3、SCF3、SO2C
F3、COOR7、CONR8R9、SO2R10、COR11もしくはOR12であるか
、または、可能ならば、互いにオルト位に位置して基R1’ないしR3’である2
つの基が一緒に−OC−NR13−CO−となり、そしてR7〜R13は式Iと同じ
である)を、式Xa、XIaおよびXIIaまたはXIIIaの対応する塩素化また
は臭素化化合物(式中、R1’〜R3’は式X〜XIIIに示された意味を有し、またHalはCl
またはBrである)から調製するのに用いることもできる。
とくに式VIIIのピリジン誘導体のような窒素含有複素環式芳香族化合物の場合
には、下記の式VIIIaによって暗示できるような大きな分子構造の1成分である
フッ素化芳香族化合物もうまく調製できることはいうまでもない。

新規方法の範囲内において、ハロゲン置換によるフッ素化を、ほぼ室温から、
反応媒質の沸点または式Iの化合物に相当する出発化合物の沸点(どちらの沸点
が低いかに依存して)までの範囲の温度で行うのがさらに好ましい。これに関連
して反応速度に関し甘受しなければならない損失もなしに、通常、従来公知の方
法の場合よりも温度を低く保つことができることを強調しなければならない。
適切な態様では、新規ハレックス法は約40℃〜約270℃の範囲の温度で行
われるハロゲン置換によるフッ素化によって代表される。とくに好ましい修正で
は反応温度は約60℃〜約220℃の範囲にある。
原則として本発明によって使用出来るフッ素化剤には式II(MeF)(式中、
Meはアルカリ金属、アンモニウムもしくはホスホニウム、またはアンモニウム
もしくはホスホニウムから誘導されるアルキルアンモニウム基、アルキルホスホ
ニウム基もしくはアリールホスホニウム基)のすべての化合物が包含される。フ
ッ化ナトリウム、フッ化カリウム、フッ化セシウム、フッ化テトラメチルアンモ
ニウムおよび他のフッ化テトラアルキルアンモニウム(アルキルはエチル、n−
プロピルまたはn−ブチル)のような化合物が適切である。
フッ化テトラフェニルホスホニウムもとくに適切である。しかし、フッ素を供
給し、本発明の範囲内でうまく使用出来る式II(MeF)の化合物の中では低コ
ストのためにフッ化カリウムがとくに好ましい。
フッ素化剤は所望のハロゲン置換度を得るのに十分な量で使用される。式Iの
化合物に相当する出発化合物の量を基準にして化学量論的に用いるのが好ましい
。出発化合物ににおいて代わるべきハロゲン原子のモル量を基準にして過剰量、
とくに好ましくは1.1〜2.0倍量を使用することも好ましい。
適切な場合には、触媒効果を有する化合物を加えることによって、本発明によ
って用いられる反応媒質の、それ自体とりわけ高い効率をさらに向上させること
ができる。このために、たとえばはじめに引用した参考文献から当業者に公知の
触媒はすべて通常使用することができる。使用可能な触媒には、とりわけ(R”
)4N+Hal-(式中、HalはClまたはBrであり、R”は直鎖状または分
枝鎖状のC1−C4アルキルである);(R”’)4P+Hal-(式中、Halは
ClまたはBrであり、R”’は直鎖状もしくは分枝鎖状のC1−C4アルキル、
またはアリール、好ましくはフェニルである);クラウンエーテル;ポリエチレ
ングリコールおよび/またはピリジニウム塩が含まれる。本発明の方法は、触媒
活性量の塩化テトラメチルアンモニウム、塩化テトラブチルアンモニウム、臭化
テトラフェニルホスホニウム、18−クラウン−6、PEGおよび/またはR28
R29N−p−ピリジニウム−R30(式中、R28〜R30は好ましくは炭素原子が1
〜8個の直鎖状または分枝鎖状であることができるアルキル基である)を使用す
るとき、とくに有利であることが証明される。
すでに暗示されているように、フッ素化後の反応混合物の仕上げは反応混合物
の分溜によって好適に行うことができ、溶剤を単離してリサイクルすることがで
きる。水性仕上げの場合には、混合物を過剰の水中に注入して、得られた生成物
を濾別するかまたは有機溶剤で抽出する。
これに関連して、式III〜Vの化合物は、連続抽出器でジクロロメタンまたは
トリクロロメタンを用いて、反応溶液から連続的に、簡単かつ完全に抽出するこ
とができる。
本発明による方法を用いると、望ましい目標生成物が極めて高純度で得られる
