Claims (1)
- 【特許請求の範囲】 1.ガラクタナーゼ活性を示す酵素をコードするDNA配列を含むDNA構築物であ って、そのDNA配列が、 (a)エシェリキア・コリ(Escherichia coli)DSM 10355内に存在するプラス ミドpYES 2.0内にクローン化されたDNA配列のガラクタナーゼ・コーディング部 分; (b)配列番号:1内の1〜1026位又はより好ましくは55〜1026位に示される DNA配列又はその相補鎖; (c)上記DNA配列と少なくとも70%の相同性をもつ(a)又は(b)内に定 められたDNA配列のアナログ; (d)低ストリンジェンシーにおいて配列番号:1内の1〜1026位に示される DNA配列とハイブリダイズするDNA配列; (e)遺伝子コードの縮重のために、(b)又は(d)の配列とハイブリダイ ズしないが、上記DNA配列の中のいずれかによりコードされたポリペプチドと同 じアミノ酸配列をもつポリペプチドをコードするDNA配列;又は (f)(a),(b),(c),(d)又は(e)に特定するDNA配列の対立 形態(allelic form)又は断片であるDNA配列、 を含む、前記DNA構築物。 2.ガラクタナーゼ活性を示す酵素をコードする前記DNA配列が、微生物、好 ましくは糸状菌、酵母、又はバクテリアから得られることができる、請求項1に 記載のDNA構築物。 3.前記DNA配列が、サルノコシカケ科、例えばメリピラス(Meripilus)、ブジ ャーカンデラ(Bjerkandera)、又はスポンジペリス(Spongipellis)属の株、特に メリピラス・ギガンテウス(Meripilus giganteus)の株から得られることができ る、請求項2に記載の DNA構築物。 4.前記DNA配列が、メリピラス・ギガンテウス(Meripilus giganteus)CBS No .521.95株のDNAライブラリーから単離され、又はそれに基づき製造される、請 求項3に記載のDNA構築物。 5.前記DNA配列が、アスペルギルス(Aspergillus)種、特にアスペルギルス・ アクレアタス(A.aculeatus)又はアスペルギルス・ニガー(A.niger)の株、 フィトフトーラ(Phytophthora)種の株、特に、フィトフトーラ・インフェスタ ンス(P.infestans)、フィトフトーラ・メガスパーマ(P.megasperma)、フィ トフトーラ・カクトラム(P.cactorum)の株、又はタラロミセス(Talaromyces)種 の株、特にタラロミセス・ビソクラミドイデス(T.byssochlamydoides)、タラロ ミセス・エマーソニ(T.emersonii)の株、サーモアスカス(Thermoascus)種の 株、特にサーモアスカス・アウランティアカス(T.aurantiacus)の株、スポロ トリチャム(Sporotrichum)種の株、特にスポロトリチャム・セルフィラム(S .celluphilum)の株又はペニシリウム(Penicillium)種の株、特に、ペニシリウ ム・シトリナム(P.citrinum)、ペニシリウム・カメンバーチ(P.camembertii )又はペニシリウム・ロクエフォーチ(P.roquefortii)の株から得られること ができる、請求項2に記載のDNA構築物。 6.前記DNA配列は、エシェリキア・コリ(Escherichiacoli)DSM No.10355 から単離される、請求項1に記載のDNA構築物。 7.請求項1〜6のいずれか1項に記載のDNA構築物を含む組換え発現ベクタ ー。 8.請求項1〜6のいずれか1項に記載のDNA構築物又は請求項7に記載の組 換え発現ベクターを含む宿主細胞。 9.真核細胞、特に真菌細胞、例えば酵母細胞又は糸状菌細胞で ある、請求項8に記載の宿主細胞。 10.フザリウム又はアスペルギルス又はトリコデルマの株、特に、フザリウム ATCC 20334の同定特徴をもつフザリウム種、アスペルギルス・ニガー、アスペル ギルス・オリザエ、トリコデルマ・ハルジアナム又はトリコデルマ・レイセイで ある、請求項9に記載の宿主細胞。 11.サッカロミセスの株、特にサッカロミセス・セレビシエの株である、請求 項9に記載の宿主細胞。 12.メリピラス種、特にメリピラス・ギガンテウス(Meripilus giganteus)で ある、請求項9に記載の宿主細胞。 13.メリピラス・ギガンテウスCBS No.521.95株である、請求項12に記載の宿 主細胞。 14.ガラクタナーゼ活性を示す酵素の製法であって、その酵素の産生を許容す る条件下、請求項8〜13のいずれか1項に記載の細胞を培養し、そしてその培養 物からその酵素を回収することを含む、前記製法。 15.(i)相同不純物を含まず、かつ、(ii)酵素が、請求項14に記載の方法 により、そして請求項8〜11のいずれか1項に記載の宿主細胞を用いて製造され ること、を特徴とする、ガラクタナーゼ活性を示す単離酵素。 16.以下の: (a)Escherichia coli DSM 10355内に存在するプラスミドpYES2.0内にクロ ーン化されたDNA配列のガラクタナーゼ酵素コーディング部分によりコードされ たポリペプチド; (b)配列番号:2の19〜342位に示すアミノ酸配列を含むポリペプチド; (c)上記ポリペプチドと少なくとも70%の相同性をもつ(a) 又は(b)に定義されたポリペプチドのアナログ;そして (d)(a),(b)又は(c)の対立形態又は断片、から成る群から選ばれ たガラクタナーゼ活性を示す単離酵素。 17.請求項15又は16に記載の酵素を含む酵素製剤。 18.請求項15又は16に記載のガラクタナーゼ活性を示す酵素が濃縮されている 、酵素製剤。 19.a−アラビノシダーゼ、キシラナーゼ、β−ガラクトシダーゼ、a−グル クロニシダーゼ(a-glucuronisidase)、b−キシロシダーゼ、キシラン・アセチ ル・エステラーゼ、アラビナナーゼ、ラムノガラクツロナーゼ、ペクチン・アセ チルエステラーゼ、ポリガラクツロナーゼ、ペクチン・リアーゼ、ペクテート・ リアーゼ、グルカナーゼ、ペクチン・メチルエステラーゼ、ラッカーゼ、又はフ ィターゼ(phytase)をさらに含む、請求項18に記載の製剤。 20.飼料又は食品の製造における請求項15又は16に記載の酵素、請求項17〜19 のいずれか1項に記載の酵素製剤の使用。 21.植物細胞壁由来材料の粘度を減少させるための、請求項15又は16に記載の 酵素又は請求項17〜19のいずれか1項に記載の酵素製剤の使用。 22.ワイン又はジュースの製造における、請求項15又は16のいずれか1項に記 載の酵素又は請求項17〜19のいずれか1項に記載の酵素製剤の使用。 23.寄託された株エシェリキア・コリ(Escherichia coli)DSM No.10355の単 離された実質的に純粋な生物学的培養物。
