バクテリア、寄生虫、原生動物、菌・カビ及びウイルスに対して有効なスフィ
ンゴ脂質
本発明はバクテリア、真菌類及びウイルスに対して活性である物質としてそれ
自体公知である物質の使用に関する。特定の態様において、本発明はかかる物質
を含む化粧用及び皮膚科用調製物に関する。
健康な定温動物、殊に健康なヒトの皮膚には多数の無毒な微生物が生息してい
る。このいわゆる皮膚の微生物叢(microflora)は無害であるばかりでなく、日
和見(opportunistic)または病原性微生物に対しての防御に対して重要な保護
を構成する。
バクテリアは原始核単細胞生物に属する。これらのものは概略その形態(球形
、円筒形、曲がった円筒形)及びその細胞壁の構造(グラム−陽性、グラム−陰
性)により特徴づけることができる。更に詳細な分類には生物の生理学の考慮が
加えられる。かくて、好気性、嫌気性及び随時嫌気性のバクテリアが存在する。
ある個体は病原性微生物としてのその特性において医学的に重要なものであり、
そして他のものはまた完全に無害である。
バクテリアに対して活性である物質はかなり長い期間知られている。ペニシリ
ンの最初の発見は1929年に既になされているが、例えば、抗微生物作用を有
する物質の全てには適用できない「抗生物質」なる用語は1941年に遡ること
ができる。定温生物、即ち例えば病気の患者はしばしばいずれかの方法での使用
期間中にその代謝機能を損なうので、現代の意味における抗生物質は、あらゆる
医学に対して、そして確かにあらゆる化粧品用途に対して適当ではない。
従って、本発明の1つの目的はこの方向における従来技術を改良すること、即
ち、殊に物質の使用に伴って使用者の健康を許されない程度に損なわせることな
しにグラム−陽性及び/またはグラム−陰性バクテリアに対して活性である物質
を提供することである。
グラム−陰性微生物は、例えば大腸菌、プソイドモマス(Pseudomonas)種及
び腸内菌(Enterobacteriaceae)例えばシトロバクター菌(Citrobacter)であ
る。
またグラム−陽性微生物は、化粧品及び皮膚科学において役割を果たしている
。きたない(impure)皮膚の場合、例えばバクテリア性の二次感染は他の影響に
加えて病因学的に重要である。不潔な皮膚に関連して最も重要なものはプロピオ
ンバクテリウム・アクネス(Propionibacterium acnes)である。
きたない皮膚(にきび)及び/または面ほう(comedones)は温和な場合でも
冒されるものの安寧を損なう。殊に全ての青年がある程度はにきびに冒されるた
め、多くの人に対してこの状態を治療する必要がある。
従って、本発明の特定の目的は、きたない皮膚またはプロピオンバクテリウム
・アクネス(Propionibacterium acnes)に対して活性である物質または物質の
組合せを見いだすことであった。
他の具体例において、本発明は化粧防臭剤に関する。かかる調製物は、それ自
体は無臭である汗が殊にグラム−陽性の微生物により分解される場合に生じる体
臭を除去するために作用する。通常の化粧防臭剤は異なった活性原理に基づく。
液体防臭剤例えばエアロゾル・スプレー、ロール・オン(roll-ons)など、並
びに固体調製物例えば防臭剤スティック、粉末、粉末スプレー、
密着クレンジング(intimatec leansing)組成物などは共に公知であり、そして
通常のものである。
いわゆる抗発汗剤において、汗の生成は収斂剤、主にアルミニウム塩、例えば
ヒドロキシ塩化アルミニウム(アルミニウムクロロハイドレート)により抑制し
得る。しかしながら、皮膚タンパクを変成させることとは別に、これに用いる物
質はその投与量に依存して腋の下の部分の熱収支に劇的な効果を有するので、最
善には例外的な場合にだけ用いるべきである。
皮膚上のバクテリア叢は、化粧防臭剤中に抗微生物物質を用いることにより減
少させ得る。理想的な場合において、臭気を生成させる微生物のみを効果的に減
少させるべきである。しかしながら実際は、皮膚の全体の微生物叢が損なわれ得
ることが見いだされた。
汗の流れ自体はこれにより影響されず、そして理想的な場合において汗の微生
物分解のみが一時的に停止される。
同一の組成物中の収斂剤と抗微生物作用を有する物質との組合せも一般的であ
る。しかしながら、2つのクラスの活性成分の欠点はこの方法によっては完全に
は除去することはできない。
最後に、体臭を香りによりマスクし得るがこの方法は消費者の美的要求を少な
くとも満たすものである。何故なら、体臭及び香水の香りの混合物はやや不快な
臭いであるからである。
それにもかかわらず、殆どの化粧防臭剤及びまた殆どの化粧品は全体としてこ
れらのものが脱臭活性成分からなる場合でも香りがつけられている。また香料を
入れることは化粧製品に対する消費者により受け入れられるか、または製品にあ
る種の特徴を与える役目を果たす。
しかしながら、活性成分、殊に化粧防臭剤からなる化粧品組成物の香りづけは
活性成分及び香料成分が場合によっては相互に反応し、そして相互に不活性化さ
せ得るために少なからず問題がある。
防臭剤は次の条件を満たすべきである:
1)これらのものは信頼できる防臭の効果を有すべきである。
2)皮膚の自然の生物学的プロセスは防臭剤により損なわれるべきではない。
3)防臭剤は過剰投与または特記されない他の使用の場合に無害でなければな
らない。
4)これらのものは繰り返しの使用後に皮膚上に濃縮されるべきではない。
5)これらのものは通常の化粧品調製物中に容易に配合されるべきである。
かくて本発明の他の目的は本分野の欠点を有しない化粧防臭剤を開発すること
であった。殊に、防臭剤は大部分は皮膚の微生物叢を保護すべきであるが、体臭
の原因となる多数の微生物を選択的に減少させるべきである。
更に良好な皮膚の許容性に特徴がある化粧防臭剤を開発することが本発明の目
的である。いずれの状況下でも防臭活性主成分は皮膚上に濃縮させるべきでない
。
大多数の可能な通常の化粧品補助剤及び添加剤、殊に防臭または抗発汗作用を
有する調製物において正確に重要である香料成分と調和する化粧防臭剤を開発す
ることが他の目的である。
またその作用が顕著に減少せずに比較的長期間、殊に少なくとも半日
間にわたって活性である化粧防臭剤を与えることが他の目的である。
最後に、1つまたはいくつかの特殊な代表例に限定されずにできるだけ一般的
に殆どの代表形態の化粧防臭剤中に配合し得る防臭用化粧原理を開発することが
本発明の目的である。
バクテリアに対して菌類またはマイコビオンツ(mycobionts)とも呼ばれる菌
・カビ(μυκηζ=菌・カビに対するギリシャ文字)は成熟核(eukaryotes)
に属する。成熟核はいわゆる原始核(prokaryotes)に対して細胞が核シェル及
び核膜により細胞質の残りのものから分離される細胞核を有する。細胞核は染色
体中に蓄積される遺伝情報を含む。
菌類の例には例えば酵母(Protoascomycetes)、カビ(Plectomycetes)、う
どんこ病(Pyrenomycetes)、わたげ状うどんこ病(Phycomycetes)及び毒茸(B
asidiomycetes)が含まれる。
毒茸を含む菌・カビは植物生物ではなく、細胞壁、細胞液で満たされた液胞及
び顕微鏡下で容易に見られる血漿流を有するものである。これらのものは植物合
成顔料を含まず、そしてC−他力栄養体である。これらのものは好気的条件下で
生長し、そして有機物の酸化によりエネルギーを得る。しかしながら、ある代表
例例えば酵母は条件嫌気的生物であり、そして発酵工程においてエネルギーを生
成させ得る。
皮膚糸状菌症はあるタイプの菌・カビ、殊に皮膚糸状菌が皮膚及び毛の小胞に
侵入する病気である。皮膚糸状菌症の症状は例えば通常かゆみまたはアレルギー
性湿疹を伴う小発疹、脱皮、亀裂及びただれである。
皮膚糸状菌は本質的に次の4つのグループに分けることができる:皮膚糸状菌
症(例えば表皮糸状菌症、黄癬、小胞子菌症及び白癬症)、酵母糸状菌症[例え
ばひこう疹及びピチロスポルム属、カンジダ感染、分
芽菌症、ブッセーブスケ(Busse-buschke)病、酵母菌症、ピエドラ・アルバ(P
iedra alba)、トルロプシス病及び砂毛症による他の糸状菌症]、カビ糸状菌症
(例えばアスペルギルス病、セファロスポリウム症、藻菌類症及びスコプラリオ
プシス症)並びに全身糸状菌症(例えば色素酵母菌症、コクシジオイデス症及び
ヒストプラズマ症)。
病原性及び条件病原性菌類には例えば酵母、カンジダ種[例えばガコウソウカ
ンジダ(Candida albicans)]及びピチロスポルム属(Pityrosporum)のものか
らのものが含まれる。ピチロスポルム種、殊に卵形ピチロスポルム(Pityrospor
um ovale)がなかんずくそれ自体頭部脂漏(=dandruff)、皮脂疹及びピチロス
ポルム・フォリクリチス(pityrosporum folliculitis)として明白である皮膚
病例えばでん風(pityriasisversicolor)、油性脂漏及び乾性脂漏の状態の脂漏
の原因と考えられる。乾癬の発達における卵形ピチロスポルムの参加が本分野で
の議論の目的である。
全てのヒトの皮膚の領域に皮膚糸状菌が感染する。皮膚糸状菌症は殆どもっぱ
ら皮膚、毛及び爪に感染する。また酵母糸状菌は粘膜及び内部器官に感染し、一
方全身的糸状菌症は通常全体の器官系に広がる。
湿度及び熱が衣服、宝石または靴により蓄積される体の部分は、殊にしばしば
感染される。かくて運動選手の足は、最も良く知られ、かつ最も広く広がる皮膚
糸状菌症の感染場所の1つである。更に、指の爪及び足の爪の部分の菌・カビ病
(爪糸状菌症)は殊に不快である。
菌・カビ及びバクテリアによる皮膚の重複感染はまたまれなことではない。
高い微生物数の1種またはそれ以上の生理学的病原体例えばブドウ球
菌、及びまたしばしば非生理学的病原体例えばガコウソウカンジダとの新たな感
染が第一の感染、即ち通常の皮膚の糸状菌数と共に生じ、そして悪影響が生じる
場合、感染された皮膚の「重複感染」が生じ得る。通常の皮膚(または他の体の
器官)の微生物叢がここに二次病原体により殆ど完全に過剰生長する。
好適に進行する場合、かかる重複感染は問題の微生物に依存して不快な皮膚の
兆候(かゆみ、みにくい外観)でそれ自体明らかになり得る。しかしながら、逆
に進行する場合、これらのものは広範囲にわたっての皮膚の破壊を引き起こし、
そして最悪の場合は患者の死をも引き起こし得る。
上記のタイプの重複感染は例えば完全に発病したAIDSでしばしば生じる二
次的病気である。低い微生物密度ではいずれの割合でもそれ自体無害であるが、
またある状況下では決定的に病原性である微生物がこのように健康な皮膚叢を過
剰生長させる。それにもかかわらず、AIDSの場合にはまた他の体の器官が重
複感染により影響される。
かかる重複感染はまた多数の皮膚病例えばアトピー性湿疹、神経皮膚炎、挫瘡
(acne)、脂漏皮膚炎または乾癬で観察される。多くの医学及び治療法例えばガ
ン病の放射線または化学療法、薬剤により誘導され、かつ副作用として生じる免
疫抑制、或いは他に全身的抗生物質治療、並びに外的化学的または物理的影響(
例えば環境汚染、スモッグ)はまた外部及び内部器官、殊に皮膚及び粘膜の重複
感染の頻度を促進する。
個々の場合において抗生物質を用いて重複感染を防除することは容易に可能で
あるが、かかる物質は不快な副作用の欠点を有する。例えば、患者はしばしばペ
ニシリンに対してアレルギー性であり、このためにか
かる場合には対応する治療は問題外である。
更に上記の抗生物質はこれらのものが二次的病原体から皮膚叢を除くばかりで
なく、それ自体生理学的である皮膚叢を重大に損なう欠点を有し、そしてこのよ
うに自然の治癒工程を再び遅延させる。
本発明の目的は、重複感染を回復させることができ、生理学的皮膚叢に重大な
損失を与えない使用により従来の欠点を除去し、そして物質及びかかる物質から
なる調製物を提供することである。
原生動物は無性的に(二または四分裂及び発芽による)、そしてまた有性的に
(生殖、ガモント及び自家生殖)増殖する明らかに分離した細胞核を有する寄生
的に生きる単細胞生物である。環境からの食物の摂取は浸透及び細胞吸収または
食作用による。増殖性の、通常移動性の状態(いわゆる栄養型)に加え、悪い状
況下では多くの原生動物はまた永久形状として胞子を形成する。
原生動物はその運動法及び器官により4つの異なるグループに分けられる:
(a)鞭毛虫類(鞭毛を有する鞭毛虫)
(b)肉質虫類/根足虫類(血漿隆起によるアメーバー状移動パターン)
(c)胞子虫類(サーペンチンまたは滑走移動パターン)
(d)繊毛虫類/有毛虫類(繊毛または鞭毛)
亜熱帯及び熱帯地域において、寄生的に生きる原生動物はしばしば剌し、そし
て吸う昆虫ばかりでなく、ほこり及びよごれ感染並びに食物連鎖により移動する
。
ある医学的及び皮膚学的に関連する原生動物には次のものがある:トリコモナ
ス症[膣トリコモナス(Trichomonas vaginalis)による]、
ランブル鞭毛虫症[ランブル鞭毛虫(Lamblia intestinalis)による]、内臓及
び静脈及び粘膜レーシュマニア症[例えばドノヴァン・レーシュマニア(leishm
ania donovanii)、熱帯レーシュマニア(L.tropica)、ブラジルレーシュマニ
ア(L.brasiliensis)、メキシコレーシュマニア(L.mexicana)、L.ディフー
サ(diffusa)またはL.ピファノイ(pifanoi)による]、トリパノソーマ病(
種々のトリパノソーマ種による)、アメーバー性赤痢及びアメーバー病[例えば
種々のアメーバー種、ヨードアメーバー・ビュッチリー(Iodamoeba butschlii
)またはネクルリア・フォウレリ(Naegleria fowleri)による]、球菌症[イ
ソスプオラ・ベリ(Isospora belli)による]及びバランチジアル赤痢[大腸バ
ランチジウム(Balantidium coli)による]。
原生動物により生じる医学的及び皮膚学的現象はヒトの健康状態をある場合に
はかなり損なう。従って感染したヒトにおいてこの状態を治療する必要性が大い
にある。かくて本発明のある目的は、原生動物に対して有効な活性成分を見いだ
すことである。
寄生虫は他の生物上(外部寄生虫)または中(内部寄生虫)にてそのものによ
って食餌し、かつ殊に病気の兆候がある(=病原性寄生虫)か、またはない(非
病原性寄生虫)単細胞または多細胞植物または動物である。生活の様式は多分期
間的な、一時的または固定的な寄生虫としてのみ非予防的または純粋に寄生的の
いずれかである。寄生虫の発育は1種またはそれ以上の異なる宿主生物に関連し
、そしてヒトは中間宿主または最終宿主であり得る。
医学的及び皮膚学的に重要である寄生虫には例えば蟻虫があり、このものはま
た吸虫類(Trematodae)、条虫類(Cestodae)及び線虫類(Ne
matodae)に分類される。ヒトの健康状態を損なう蟻虫には例えば住血吸虫症[
住血吸虫(Schistosoma)種による]、腸及び他の内部器官の条虫類感染[例え
ばチーニア(Taenia)種及び包虫(Echinococcus)種]、回虫病[回虫(Ascari
s lumbricoides)]、繞虫症[繞虫(Enterobium vermicularis)]、肺吸虫症
[肺吸虫(Paragonium)種による]、フィラリア症[バンクロフト糸状虫(Wuch
eria bancrofti)による]及び他の線虫感染[例えば鞭虫(Trichuris trichura
)または旋毛虫(Trichinella spiralis)]がある。
更にヒト及び動物に寄生して生活し、そして宿主生物に医学的及び皮膚学的変
化を生じさせる多数の昆虫種及びダニが存在する。ヒトの健康状態を損なうこと
に関連する寄生虫病には、例えばアクロダーマチチス(accrodermatitis)[穀
物ダニ例えばシラミダニ(Pediculoides ventricosus)による]、カイセン[人
体カイセンチュウ(Sarcoptes scabii)による]、ハエ及び/またはハエ幼虫に
よる感染[例えばツエーツエーバエ(Glossina)、サシバエ(Stomoxys)、アブ
(Tabanus)、メクラアブ(Chrysops)、ヒツジウジバエ(Lucilia)、アオバエ
(Chrysomya)、コクリオミア(Cochliamya)、ニクバエ(Wohlfartia)、食人
蝿(Cordylobia)またはデルマトビア(Dermatobia)種による]、カ及び/また
はカの幼虫による感染[ヤブカ(Aedes)、マダラカ(Culex)、アノフェレス(
Anopheles)、スナカ(Phlebotomus)、ヌマカ(Culicoides)、スミリウム(Su
milium)またはヘマゴーゲス(Haemagoges)種による]、チックによる感染[例
えばペルシャダニ(Argaspersicus)及び他のナガヒメダニ種、オルニトドルス
・エラチクス(Ornithodoruserraticus)及び他のヒメダニ種並びにオロビウス
(Orobius)、リフォ
セファルス(Rhiphocephalus)、カクマダニ(Dermacentor)、チマダニ(Haema
physalis)、マダニ(Amblyomma)及びマダニ(Ixodes)による]、ポロセファ
ロシス(Porocephalosis)[舌虫(Porocephalus)種による]、ノミによる感染
[例えばヒトノミ(Pulexirritans)、イヌノミ(Cenocephalides canis)、エ
ジプトノミ(Xenopsylla cheopsis)、ネズミノミ(Nosophyllus fasciatus)ま
たはスナノミ(Sarcopsylla penetrans)による]、シラミによる感染[例えば
ケジラミ(Phthirius pubis)、ヒトジラミ(Pediculosus humanus)またはアタ
マジラミ(Pediculosus capitis)による]、半翅虫による感染[例えばトコジ
ラミ(Cimex lectularius)、タイワントコジラミ(Cimex hemipterus)、パン
ストロンギルス・メジスツス(Panstrongylus megistus)、ロドニウス・プロリ
クス(Rhodnius prolixus)、トリアトーマ・ジミダタ(Triatoma dimidata)、
トリアトーマ・インフェスタンス(Triatoma infestans)、トリアトーマ・ソル
ジダ(Triatoma sordida)またはブラジルサシガメ(Triatoma brasiliensis)
による]並びにダニによる感染[例えばニキビダニ(Demodex folliculorum)及
び他の毛嚢虫(Demodex)種並びにデルマミスクス(Dermamyskus)、ニクダニ(
Glyciphagusdomesticus)、シラミダニ(Pyemotes)種、ヒゼンダニ(Sarcoptes
)種またはケダニ(Trombicula)種による]がある。
ここにヒトに寄生する寄生虫が、また宿主生物例えばヒトの健康及び安寧を継
続的に損ない得るバクテリア、菌類、原生動物及びウイルスのキャリアとなり得
ることは加えて重要である。従って寄生虫病に対して有効であり、そして医学的
または皮膚学的兆候を改善し得る活性成分を見いだすことが必要である。従って
更にこの必要性を満たすことが本発
明の目的である。
原始核及び成熟核細胞に対し、ウイルス(ウイルス=毒に対するラテン語)は
生合成に宿主細胞を必要とする生物学的構造体である。細胞外ウイルス(”vi
rions”とも呼ばれる)は追加の脂質含有ケーシング(エンベロープ)に囲
まれ得る単鎖または二重鎖核酸配列(DNAまたはRNA)及びタンパクシェル
[キャプシド(capsid)と呼ばれる]からなる。また核酸及びキャプシドの全シ
ステムは核キャプシドと呼ばれる。ウイルスは通常臨床的基準により分類される
が、現在は通常その構造、その形態及び殊に核酸配列により分類される。
医学的に重要なウイルスの属には例えばインフルエンザウイルス[オロトミキ
ソウイルス(Orthomyxoviridae)属]、狂犬病ウイルス(例えば狂犬病、ラブド
ウイルス属)、エンテロウイルス(例えばA型肝炎、ピコルナウイルス属)及び
ヘパドナウイルス[例えばB型肝炎、ヘパドナウイルス(Hepadnaviridae)属]
がある。
殺ウイルス剤(virucides)、即ちウイルスを殺す物質はウイルスがそれ自身
代謝を有していないために、真の意味では存在しない。このためにまた、ウイル
スを生物として分類すべきか否かについての議論もある。未感染の細胞を損傷さ
せない薬理学的介在はいずれの割合でも困難である。ウイルスに対する攻撃に可
能な作用機構は例えば複製に重要な宿主細胞中に存在する酵素をブロックするこ
とにより主にその複製を妨害することである。更に、宿主細胞中へのウイルス核
酸の放出を防止することができる。本開示に関連し、「抗ウイルス」または「ウ
イルスに対して活性」、「殺ウイルス」などなる用語は個々の場合における物質
の実際の作用機構には無関係に、予防的または治療的かのいずれかで有
害なウイルス感染の結果から単細胞または多細胞生物を保護する物質の特性を意
味するものと理解される。
しかしながら、従来はウイルスに対して活性であり、そして更に宿主生物に対
して損傷を全く与えないか、または認め得る程度に与えない物質が欠けていた。
かくて本発明の目的はこの乏しい事態を改善し、即ち予防的または治療的かの
いずれかで有害なウイルス感染の結果から単細胞または多細胞生物を効果的に保
護する物質を見いだすことである。
驚くべきことに、抗バクテリア性、抗真菌性、抗ウイルス性活性成分並びに原
生動物及び寄生虫に対して有効な活性成分としてのスフィンゴ脂質の使用が従来
の欠点を克服し、そしてこの中にこの問題の解決が存在することが見いだされた
。
スフィンゴ脂質並びに製薬物及び化粧品におけるその使用はそれ自体公知であ
る。スフィンゴ脂質は次の構造式を有するスフィンゴシンまたはフィトスフィン
ゴシンの構造をベースとする:
本発明によるスフィンゴ脂質は有利には一般的塩基構造
の特徴があり、ここにR1及びR3は相互に独立して炭素原子1〜28個を有する
飽和もしくは不飽和の、分枝鎖状もしくは非分枝鎖状のアルキル基であり、R2
は水素原子、炭素原子1〜28個を有する飽和もしくは不飽和の、分枝鎖状もし
くは非分枝鎖状のアルキル基、糖基、非エステル化リン酸塩基または有機基でエ
ステル化されたリン酸塩基、非エステル化硫酸塩基または有機基でエステル化さ
れた硫酸塩基よりなる群から選ばれ、そしてYは水素原子、ヒドロキシル基また
は他のヘテロ官能基である。
本発明により有効であるスフィンゴ脂質にはこれらの天然のセラミド、セレブ
ロシド、ガングリオシド、スフィンゴリン脂質、殊にスフィンゴミエリン、スフ
ィンゴスルファチド及びグリコスフィンゴシド並びに化学合成により得られる同
族体が含まれ、その例は下記のものである:
セラミド: R1及びR3はアルキル基であり、そしてR2=Hである。
スフィンゴリン脂質:
R1及びR3はアルキル基であり、そしてR4はリン酸塩または有機リン酸基で
ある。
スフィンゴミエリンは有機ホスホリル化されたスフィンゴ脂質
である。
R2を糖基の基から選ぶ場合、モノグリコシルセラミド及びジ−、トリ−また
は一般にオリゴグリコシルセラミド間で通常区別がある。モノグリコシルセラミ
ドを通常セレブロサイドと呼ばれる: オリゴグリコシルセラミドは通常ガングリオシドと呼ばれる。
好適なスフィンゴ脂質はセラミドI、II、III及びIV、グリコシルセラ
ミド、ラクトシルセラミド及びガングリオシドGM1、2及び3である。
驚くべきことに、本発明により施用する場合、スフィンゴ脂質はグラム−陽性
及びグラム−陰性バクテリア、マイコビオンツ(mycobionts)、寄生虫、原生動
物及びウイルスの生長を防止することが見いだされた。そのグラム−陽性バクテ
リアに対する作用が殊に有利である。
加えて、本発明により施用する場合、スフィンゴ脂質は殊にピチロスポルム(
Pityrosporum)種の酵母、即ち卵形ピチロスポル(Pityrosp
orium ovale)の生長を防止することができる。
更に本発明により施用する場合、スフィンゴ脂質は脂漏の症候、殊にふけの発
達を防止し、そして既に存在する脂漏の症候、殊にふけを除去することが見いだ
された。
更に、本発明により施用する場合、スフィンゴ脂質は化粧防臭剤中の消臭活性
成分として、そしてきたない皮膚、挫瘡の温和な形態及びプロピオンバクテリウ
ム・アクネ(Propionibacterium acne)に対しての使用に殊に適する。
最後に、本発明により施用する場合、スフィンゴ脂質はこれらのものをこれら
の調製物に加える場合にグラム−陽性及びグラム−陰性バクテリア、マイコビオ
ンツ(mycobionts)、原生動物、寄生虫及びウイルスによる攻撃による有機物、
殊に化粧品及び皮膚学的調製物の腐敗を防止し得ることが見いだされた。
かくてまた本発明は適当ならば適当な化粧用または皮膚学的担体中のスフィン
ゴ脂質を菌類に汚染された部分と接触させることを特徴とする菌類の防除方法、
及びスフィンゴ脂質を活性量でこれらの有機生成物に加えることを特徴とする菌
類による攻撃からの有機生成物の保護方法に関する。
従って従来の分野は本発明による使用を全く与えるものでなかった。
更に、本発明により施用する場合、スフィンゴ脂質はふけの発達の原因となる
微生物卵形ピチロスポルム(Pityrosporium ovale)、及び関連する微生物に対
して殊に良好な作用を有することは驚くべきことである。従ってふけに対して用
いられる調製物例えば抗ふけシャンプーは本発明の好適な具体例である。
本発明により、スフィンゴ脂質は好ましくは化粧用または皮膚学的組成物にお
いて組成物の全重量をベースとして0.005〜50.0重量%、殊に0.01
〜20.0重量%の含有量で用いる。組成物は各々の場合に組成物の全重量をベ
ースとして有利には0.02〜10.0重量%、殊に好ましくは0.02〜5.
0重量%、極めて殊に有利には0.5〜3.0重量%のスフィンゴ脂質からなる
。
また純粋なスフィンゴ脂質の代わりにまたスフィンゴ脂質を含むことを特徴と
する物質を用いることが有利である。
本発明によれば、スフィンゴ脂質は難無く通常の化粧用または皮膚学的調製物
中に、有利にはポンプ・スプレー、エアロゾル・スプレー、クリーム、軟膏、チ
ンキ剤、ローション、ネイルケア製品(例えばネイルワニス、ネイルワニス除去
剤、ネイルバルサム)など中に配合し得る。
本発明によれば、スフィンゴ脂質は極めて殊に有利には抗ふけシャンプーの形
態である。
またスフィンゴ脂質を他の活性物質例えば他の抗微生物剤、抗糸状菌剤または
抗ウイルス物質と一緒にすることが可能であり、そしてある場合に有利である。
本発明による組成物は緩衝液を加えることが有利である。3.5〜7.5の範
囲のpH値が有利である。4.0〜6.5の範囲内のpH値を選択することが殊
に好ましい。
本発明による化粧用及び/または皮膚学的調製物は通常の組成物を有すること
ができ、そして皮膚学的処理の意味か、またはケア化粧品の意味における処理に
おいて皮膚及び/または毛髪の処理に用いることができる。しかしながらまたこ
れらのものは装飾用化粧品においてメーキャッ
プ製品に使用し得る。
使用のために、本発明による化粧用及び皮膚学的調製物は化粧品及び皮膚学的
製品に通常の方法にて適量で皮膚及び/または毛髪に施用する。
サンスクリーン(sunscreen)の状態であるこれらの化粧用及び皮膚学的調製
物が有利である。これらのもは有利には加えて少なくとも1つのUVAフィルタ
ー及び/または少なくとも1つのUVBフィルター及び/または少なくとも1つ
の無機顔料からなる。
UV線に対する皮膚の保護用の本発明による化粧品調製物は種々の形態例えば
このタイプの調製物に通常用いられる形態であり得る。例えば、これらのもは溶
液、油中水(W/O)タイプもしくは水中油(O/W)タイプの乳液、または例
えば水中油中水(W/O/W)タイプの多成分乳液、ゲル、ヒドロ分散体、固体
スティック或いはまたエアロゾルであり得る。
本発明による化粧品調製物はかかる調製物中で通常用いられる化粧品補助剤例
えば保存剤、殺バクテリア剤、酸化防止剤、香料、発泡防止剤、着色剤、着色効
果を有する顔料、糊料、界面活性剤、乳化剤、皮膚軟化剤、湿潤剤及び/または
軟釈剤、脂肪、油、ワックス或いは他の化粧品調製物の通常の成分例えばアルコ
ール、ポリオール、重合体、発泡安定剤、電解質、有機溶媒またはシリコーン誘
導体からなり得る。
化粧用または皮膚学的調製物が溶液またはローションである場合、使用し得る
溶媒は次のものである:
−水または水溶液;
−油例えばカプリン酸またはカプリル酸のトリグリセリド、好ましくはヒマシ
油;
−脂肪、ワックス並びにたの天然及び合成脂肪物質、好ましくは脂肪酸と低炭
素数のアルコール例えばイソプロパノール、プロピレングリコールもしくはグリ
セリンとのエステル、または脂肪アルコールと低炭素数のアルカン酸もしくは脂
肪酸とのエステル;
−低炭素数のアルコール、ジオールまたはポリオール及びそのエーテル、好ま
しくはエタノール、イソプロパノール、プロピレングリコール、グリセリン、エ
チレングリコール、エチレングリコールモノエチルもしくはモノブチルエーテル
、プロピレングリコールモノメチルエーテル、モノエチルエーテルもしくはモノ
ブチルエーテル、ジエチレングリコールモノメチルエーテルもしくはモノエチル
エーテル及び同族体生成物。
殊に、上記の溶媒の混合物を用いる。アルコール性溶媒の場合、更に水が成分
であり得る。
本発明によれば、好適な使用し得る酸化防止剤は化粧用及び/または皮膚学的
施用に適するか、または通常である全ての酸化防止剤である。
酸化防止剤は有利にはアミノ酸(例えばグリシン、ヒスチジン、チロシン、ト
リプトファン)及びその誘導体、イミダゾール(例えばウロカニン酸)及びその
誘導体、ペプチド例えばD,L−カルノシン、D−カルノシン、L−カルノシン
及びその誘導体(例えばアンセリン)、カロテノイド、カロテン(例えばα−カ
ロテン、β−カロテン、リコペン)及びその誘導体、リポイック酸及びその誘導
体(例えばジヒドロリポイック酸)、アウロチオグルコース、プロピルチオウラ
シル及び他のチオール(例えばチオレドキシン、グルタチオン、システイン、シ
スチン、シスタミン並びにそのグリコシル、N−アセチル、メチル、エチル、プ
ロピル、アミル、ブチル並びにラウリル、パルミトイル、おれいる、γ−
リノレイル、コレステリル及びグリセリルエステル)及びその塩、チオジプロピ
オン酸ジラウロリル、チオジプロピオン酸ジステリル、チオジプロピオン酸及び
その誘導体(エステル、エーテル、ペプチド、脂質、ヌクレオチド、ヌクレオシ
ド及び塩)及び極めて低い許容量の(例えばpmol〜μmol/kg)のスル
ホキシム化合物(例えばブチオニン−スルホキシム、ホモシスチン−スルホキシ
ム、ブチオニンスルホン、ペンタ−、ヘキサ−及びヘプタチオニン−スルホキシ
ム)、並びに更に(金属)キレート剤(例えばα−ヒドロキシ−脂肪酸、パルミ
チン酸、フィチン酸、ラクトフェリン)、α−ヒドロキシ酸(例えばクエン酸、
乳酸、マレイン酸)、ギ酸、バイル酸、バイル抽出物。ビリルビン、ビリバジン
、EDTA、EGTA及びその誘導体、不飽和脂肪酸及びその誘導体(例えばγ
−リノレン酸、リノール酸、オレイン酸)、ホール酸及びその誘導体、ユビキノ
ン及びユビキノール及びその誘導体、ビタミンC及び誘導体(例えばパルミチン
酸アスコルビル、Mgリン酸アスコルビル、酢酸アスコルビル)、トコフェロー
ル及び誘導体(例えばビタミンE酢酸塩)、ビタミンA及び誘導体(ビタミンA
パルミチン酸塩)並びにベンゾイン樹脂の安息香酸コニフェリル、ルチン酸及び
その誘導体、フェルラ酸及びその誘導体、ブチル化されたヒドロキシトルエン、
ブチル化されたヒドロキシアニソール、ノルジヒドログアイアック樹脂酸、ノル
ジヒドログアイアレチン酸、トリヒドロキシブチルフェノン、尿酸及びその誘導
体、マンノース及びその誘導体、亜鉛及びその誘導体(例えばZnO、ZnSo4
)、セレン及びその誘導体(例えばセレニウムメチオニン)、スチルベン及び
その誘導体(スチルベンオキシド、トランススチルベンオキシド)並びに本発明
により適するこれら上記の
活性成分の誘導体(塩、エステル、エーテル、糖、ヌクレオチド、ヌクレオシド
、ペプチド及び脂質)よりなる群から選ばれる。
調製物中の酸化防止剤(1つまたはそれ以上の化合物)の量は調製物の全重量
をベースとして好ましくは0.001〜30重量%、殊に好ましくは0.05〜
20重量%、殊に1〜10重量%である。
ビタミンE及び/またはその誘導体が酸化防止剤である場合、調製物の全重量
をベースとして0.001〜10重量%の範囲のその特殊な濃度を選ぶことが有
利である。
ビタミンAもしくはビタミンA誘導体またはカロテンもしくはその誘導体が酸
化防止剤である場合、調製物の全重量をベースとして0.001〜10重量%の
範囲のその特殊な濃度を選ぶことが有利である。
本発明によるエマルジョンは例えば特定の脂肪、油、ワックス及び他の脂肪物
質、並びに水及び例えばかかるタイプの調製物に通常用いられる乳化剤からなる
ことが有利である。
本発明によるゲルは通常油−アルコール性ゲルの場合は好ましくは二酸化ケイ
素またはケイ酸アルミニウムであり、そして水−アルコール性またはアルコール
性ゲルの場合には好ましくはポリアクリレートである糊料の存在下における低炭
素数のアルコール例えばエタノール、イソプロパノール、1,2−プロパンジオ
ール、グリセリン及び水並びに上記の油からなる。
本発明による固体スティックは例えば天然または合成ワックス、脂肪アルコー
ルまたは脂肪酸エステルからなる。好ましくはリップ・ケア・スチック及び防臭
剤スティック(「Deo-Sticks」)である。
エアロゾル容器から噴霧し得る本発明による化粧用または皮膚学的調
製物に適する抛射剤は通常公知の、容易に揮発する、液化された抛射剤例えばそ
れ自体または相互の混合物として用いられる炭化水素(プロパン、ブタン、イソ
ブタン)である。また圧縮空気が有利である。
本分野に精通せる者にはそれ自体無毒であり、かつ原理的に本発明に適するが
、環境または他の付随する状況に対する有害な効果のために避けるべき抛射ガス
、殊にフルオロカーボン及びクロロフルオロカーボン(CFCs)があることは
明らかに公知である。
本発明による調製物は好ましくはまたUVB領域においてUV照射を吸収する
物質からなることができ、ここに皮膚を紫外線照射の全領域から保護する化粧用
調製物を提供するためにフィルター物質の全量は調製物の全量をベースとして例
えば0.1〜30重量%、好ましくは0.5〜10重量%、殊に1〜6重量%で
ある。またこれらのものはサンスクリーン(sunscreen)としても使用し得る。
UVBフィルターは油溶性または水溶性であり得る。挙げ得る油溶性物質の例
には次のものがある:
−3−ベンジリデンカンファー及びその誘導体例えば3−(4−メチルベンジ
リデン)カンファー;
−4−アミノ安息香酸誘導体、好ましくは4−(ジメチルアミノ)安息香酸2
−エチルヘキシル、4−(ジメチルアミノ)安息香酸アミル;
−桂皮酸のエステル、好ましくは4−メトキシ桂皮酸2−エチルヘキシル、4
−メトキシ桂皮酸イソペンチル;
−サリチル酸のエステル、好ましくはサリチル酸2−エチルヘキシル、サリチ
ル酸4−イソプロピルベンジル、サリチル酸ホモメチル;
−ベンゾフェノンの誘導体、好ましくは2−ヒドロキシ−4−メトキ
シベンゾフェノン、2−ヒドロキシ−4−メトキシ−4’−メチルベンゾフェノ
ン、2,2’−ジヒドロ−4−メトキシベンゾフェノン;
−ベンザルマロン酸のエステル、好ましくは4−メトキシベンザルマロン酸ジ
(2−エチルヘキシル);
−2,4,6−トリアニリノ−(p−カルボ−2’−エチル−1’−ヘキシル
オキシ)−1,3,5−トリアジン。
水溶性物質には有利には次のものがある:
−2−フェニルベンズイミダゾール−5−スルホン酸及びその塩例えばナトリ
ウム、カリウムまたはトリエタノールアンモニウム塩;
−ベンゾフェノンのスルホン酸誘導体、好ましくは2−ヒドロキシ−4−メト
キシベンゾフェノン−5−スルホン酸及びその塩;
−3−ベンジリデンカンファーのスルホン酸誘導体例えば4−(2−オキソ−
3−ボルニリデンメチル)ベンゼンスルホン酸。2−メチル−5−(2−オキソ
−3−ボルニリデンメチル)ベンゼンスルホン酸及びその塩。
勿論本発明により使用し得る与えられるUVBのリストは限定されるものでは
ない。
また本発明による調製物中の化粧用及び/または皮膚学的調製物中に通常存在
するUVAフィルターを用いることが有利であり得る。かかる物質は好ましくは
ジベンゾイルメタンの誘導体、殊に1−(4’−t−ブチルフェニル)−3−(
4’−メトキシフェニル)プロパン−1.3−ジオン及び1−フェニル−3−(
4’−イソプロピルフェニル)プロパン−1,3−ジオンである。またこれらの
組合せを含む調製物も本発明の目的物質である。UVBフィルター物質に与えら
れた同量のUVA
フィルター物質を使用し得る。
また本発明による化粧用及び/または皮膚学的調製物はUV照射に対しての皮
膚の保護に対して化粧品産業で通常用いられる無機顔料を含有し得る。これらの
ものはチタン、亜鉛、鉄、ジルコニウム、ケイ素、マンガン、アルミニウム、セ
リウムの酸化物及びその混合物、並びに酸化物が活性剤である改質物である。二
酸化チタンをベースとする顔料が殊に好ましい。上記の組合せに与えられる量を
使用し得る。
ヘア・ケアに対する化粧用調製物は例えばシャンプー組成物、シャンプー前も
しくは後にか、パーマ処理の前もしくは後にか、または毛髪の着色もしくは脱色
の前もしくは後に毛髪をすすぐ時に用いる調製物、毛髪をブロー乾燥するか、ま
たはセットするための調製物、着色または脱色用調製物、スタイリング及びトリ
ートメントローション、ヘアラッカーまたはパーマ用組成物である。
化粧用調製物は活性成分並びにヘアケア及びヘアトリートメントに対するこの
タイプの調製物に通常使用される補助剤からなる。
用いる補助剤には保存剤、界面活性剤、発泡防止剤、乳化剤、糊料、脂肪。油
、ワックス、有機溶媒、殺バクテリア剤、香料、その役目が毛髪または着色剤自
体を着色する着色剤または顔料、電解質及び毛髪がべとつくのを防止する調製物
がある。
シャンプー組成物またはウォッシュ、シャワーもしくはバス用調製物である化
粧用調製物は好ましくは少なくとも1つの陰イオン性、非イオン性または両性界
面活性剤またはその混合物、少なくとも1つの水性媒質中のスフィンゴ脂質及び
この目的に通常用いられる補助剤からなる。
本発明により有利に使用し得る界面活性剤の例には通常のセッケン例
えばナトリウムの脂肪酸塩、アルキル硫酸塩、アルキルエーテル硫酸塩、アルカ
ン−及びアルキルベンゼンスルホン酸塩、スルホ酢酸塩、スルホベタイン、サル
コシネート、アミドスルホベタイン、スルホ琥珀酸塩、スルホ琥珀酸モノエステ
ル、アルキルエーテルベタイン及びアミドベタイン、脂肪酸アルカノールアミド
及びポリグリコールエーテル誘導体がある。
界面活性剤はシャンプー組成物またはウォッシュ、シャワーもしくはバス用調
製物中で1乃至50重量%間の濃度で存在し得る。
化粧用または皮膚学的調製物を例えば着色前または後にか、シャンプー前また
は後にか、2回のシャンプー工程間にか、或いはパーマ処理の前または後にすす
ぎ、そして用いられるローションの状態で用いる場合、このものは例えば必要に
応じてその濃度が0.1乃至10重量%間、好ましくは0.2乃至5重量%間に
あり得る界面活性剤、好ましくは非イオン性または陽イオン性界面活性剤からな
る水性または水性−アルコール性溶液からなる。またこの化粧用または皮膚学的
調製物はこの目的に用いられる通常の補助剤からなるエアロゾルであり得る。
すすぎだされないローションの状態の化粧用調製物、殊に毛髪をセットするた
めのローション、毛髪をブロー乾燥する場合に用いるローション、スタイリング
及びトリートメントローションは一般に水性、アルコール性または水性−アルコ
ール性溶液であり、そして少なくとも1つの陽イオン性、陰イオン性、非イオン
性または両性重合体またはその混合物、及び1つまたはそれ以上のスフィンゴ脂
質からなる。用いるスフィンゴ脂質の量は例えば0.1乃至10重量%間、好ま
しくは0.1乃至3重量%間である。
少なくとも1つのスフィンゴ脂質からなる毛髪のトリートメント及びケア用の
化粧用及び皮膚学的調製物は非イオン性または陰イオン性のタイプの乳液の状態
であり得る。水からなるばかりでなく、非イオン性乳液は例えばポリエトキシル
化またはポリプロピル化され得る油または脂肪アルコール、或いは2つの有機成
分の混合物であり得る油または脂肪アルコールからなる。これらの乳液は必要に
応じて陽イオン性界面活性剤からなる。陰イオン性乳液は好ましくはセッケンタ
イプのものであり、そして陰イオン性またはまたは陽イオン性の特性を有する少
なくとも1つの本発明によるエトキシル化またはプロポキシル化された有機化合
物からなる。
毛髪のトリートメント及びケア用の化粧用及び皮膚学的調製物は少なくとも1
つのスフィンゴ脂質及びこの目的に通常用いられる溶媒に加えてまた有機糊料例
えばアラビアゴム、キサンタンゴム、アルギン酸ナトリウム、セルロース誘導体
、好ましくはメチルセルロース、ヒドロキシメチルセルロース、ヒドロキシエチ
ルセルロース、ヒドロキシプロピルセルロースもしくはヒドロキシプロピルメチ
ルセルロース、または無機糊料例えばケイ酸アルミニウム例えばベントナイト、
或いはポリエチレングリコール及びポリエチレングリコールステアレートまたは
ジステアレートの混合物からなるゲルの状態であり得る。糊料はゲル中において
例えば0.1乃至30重量%間、好ましくは0.5乃至15重量%間の量で存在
する。
毛髪用の生成物中のスフィンゴ脂質の量は調製物の全重量をベースとして好ま
しくは0.01乃至10重量%間、殊に0.5乃至5重量%間である。
