デカトリエン酸;18:3n−6;(GLA))の製造
方法及び使用方法の提供。 【解決手段】Colpidium(ナガマメガタミズケ
ムシ)からのγ−リノレン酸(GLA)の製造方法及び
その食品、化粧品及び医薬品における使用に関する。C
olpidiumは、最適培地においてGLAが高い絶
対収量になるまで発酵により培養することができる。

に関する。

ル酸(18:2 n−6)からγ−リノレン酸を合成す
るための必須酵素である。他のすべてのω−6 PUF
A及び数種のエイコサノイドホルモン(eicosan
oid hormones)がGLAから誘導される。
例えば、加齢、栄養不良又はアルコール中毒症によって
デルタ−6−デサチュラーゼの活性が過度に低下する
と、GLA及びその二次産物の供給不足が起こり、その
結果多くの障害が引き起こされ得る。

は、高級不飽和脂肪酸の獲得源としてあまり記載されて
いないし、GLAの工業上の獲得源としてほとんど全く
開発されていない。
下にある: (1.)それぞれ特徴的な脂肪酸スペクトルを有し、油
の顕著な主要成分となるものもあるという多様性; (2.)バイオリアクター発酵での培養可能性; (3.)発酵の間正確に制御され得る、つまり環境上の
諸影響から独立した培養; (4.)発酵後の規定された処理プロセス; (5.)世代時間が植物及び真菌より著しく短ければ、
より高い空間−時間収量(space−time yi
elds)が得られる。
植物又は動物起源の多細胞組織由来の細胞培養の発酵に
ついては先行技術に記載されているが、例えば発酵にお
けるその濃縮、処理及び精製は原生動物へ転用すること
はできない。

は原生動物内の他の関連種へ転用することはできない。
さらに、欠点は、多くの不飽和脂肪酸を有する広い脂肪

的に複雑であること(図1参照)、及びGLAの獲得を

用され得るならば、病原性でない、特にヒトの病原体で
はない生物源がそのために必要とされる。

高いGLA含量を有する(図3参照)。GLA分画は、
GLAに性質が似ている(ALAのような)他の脂肪酸
形態の混入がなく、その結果、GLAの精製は直に促進
され、GLAは純粋な形態でこのバイオマスから安く入
手される。この不飽和脂肪酸の工業生産がクロマトグラ
フィー処理の助けを借りて実施されているとすれば、こ
のことは特に注目される。

よれば、Colpidiumがヒトの病原体ではないた
めに好ましい。故に本発明はGLAを獲得及び単離する
ための生物源としてのColpidium属の使用に関
する。

バイオマスからの新規なGLA生産及び単離又はGLA
製造は、直接的に実施され得るか又はバイオマス、特に
脂質分画から得られる油を介して得られ、これによりG
LAを純粋な形態(以下の本発明によるGLA)で獲得
することが可能になる。GLAは、直接的に、又は酸触
媒作用若しくはエステル分解を伴う加水分解の手段によ
って(特に、糖脂質又はリン脂質のような)脂質から得
られる。

ではColpidium属の原生動物そのものの前駆体
を意味するものとして、及び本発明により発酵されて培
養される(実施例参照)、処置されたColpidiu

LAも、食品、医薬品又は化粧品の原材料及び添加物又
は補助剤として純粋な形態で使用することができる。

助剤とともに含む医薬品に関する。

糖尿病、癌及び関節炎を治療するための医薬品の生産方
法に関する。さらに本発明は、本発明によるGLAを、
好ましくは幼児栄養のためにごく純粋な形態で含む、動
物及びヒトの栄養における食品添加物に関する。

メ(chick−pea)を含む試験管において、25
℃、暗所で培養し、この培養液の200μlを3ヶ月ご
とに新しい管へ移す。 前培養: 100mlエーレンマイヤー・フラスコにおいてMM1
0mlを25℃、暗所で振盪(60rpm、Infor

ロマトグラフィーによって分析される。脂肪酸の同定及
び定量は、既知の脂肪酸メチルエステルに関して得られ
ている保持時間の比較、特にHP・ケム(Chem)ス
テーション(ヒューレット−パッカード社、ウィルミン
トン、アメリカ)によるコンピュータ支援の比較によっ
て実施される。
Aの獲得に比較して、繊毛虫類を使用するGLA生産
は、短い世代時間及びより高いGLAの絶対含量のため
に有意に高い空間−時間収量を基本的に有する。
ら得られるGLA300mg/培養液1リットルという
収量を、微生物からGLAを生産する確立した方法と比
較すると、GLA生産生物としてのこの高い潜在可能性
が確かめられる。微小藻類Spirulina pla
tensisの振盪培養からは、培養液1リットルにつ
きGLA38mgを得るのがやっとであるとかつて報告

リットルにつきGLA500mgの最大収量をもたら
し、Tetrahymena thermophila
(テトラヒメナ サーモフィラ)(Kiy,T.,19
93;ミュンスター大学の学位論文)の脱脂乳培地での
大量培養(mass culturing)では培養液
1リットルにつきGLA1.1gを達成した。しかしな
がら、後者の微生物由来の脂質組成はColpidiu
m由来の脂質組成より複雑なので、その結果、テトラヒ
メナからGLAを製造して獲得することは複雑なのであ
る。図面の説明 図1は、Tetrahymena thermophi
la(テトラヒメナサーモフィラ)の脂肪酸スペクト
ル、つまり「分析方法」の節に記載した方法によって得
られたテトラヒメナ・サーモフィラの脂肪酸スペクトル
のガスクロマトグラフを示す図である。GLAは18.
2分後に溶出する。これは全脂肪酸含量の16.3%
(w/w)の比率を有する。

4779〜87による様々な繊毛虫種の系統発生図を示
す図である。
31℃での発酵を示す図である。それぞれの接種量は5
x104細胞/ml。最短世代時間は、最大細胞密度
1.4x106ml-1、25℃(20.8時間)で得ら
れる。一方、22℃では、45.4時間の世代時間で
1.5x106ml-1の細胞密度が得られる。
ロマトグラムを示す図である(図3に対応する)。GL
A含量は全脂質の19%であるが、これは乾燥バイオマ
ス(DBM)1gあたりGLA5mg、又は培地1リッ
トルあたりGLA50mgである。
発酵経過でGLA産生が進展する様子を示す図である。
GLA絶対量が最大になるのは、細胞密度1.4x10
6ml-1の前期安定増殖期である。最適増殖条件(25
℃)下で発酵すると、C.campylum培養物の相
対脂肪酸組成は、高比率の飽和脂肪酸から高いGLA含
量へ移行する。遅延期にはGLA15%が存在する。最
大相対GLA量として、22℃又は25℃での発酵でも
約160時間後の対数期に19%のGLAが得られる。
しかし、この場合は、25℃より増殖が緩やかなために
約50時間遅れている。さらに増殖すると、相対GLA
含量は再び低下する。
は、培養温度25℃、発酵185時間後の安定期であ
り、DBM1gにつきGLA13mg、つまり培養液1
リットルにつきGLA290mgである。このことは、
温度22℃、発酵220時間後ではDBM1gにつき最
大でもGLA7mg、培養液1リットルにつきGLA1
70mgであることと対照的である。
量は対数増殖期に達成されるが、最高絶対GLA含量が
達成されるのは、増殖期から安定期へ移行してからであ
る。このことは、安定期で最大細胞密度に達成すること
に加えて、全脂肪含量が増加することによって説明され
る(Erwin,J.,Bloch,K.,1963;

- 【請求項1】 γ−リノレン酸の獲得及び単離のための
生物源としてのColpidium属の使用。 - 【請求項2】 Colpidium属を培養及び発酵す
る、請求項1に記載のγ−リノレン酸の製造方法。 - 【請求項3】 発酵及び培養を19〜28℃で実施す
る、請求項2に記載の製造方法。 - 【請求項4】 バイオマス及び/又は油及び/又は脂質
からγ−リノレン酸を単離する、請求項1〜3のいずれ
か1項に記載のγ−リノレン酸の製造方法。 - 【請求項5】 Colpidium由来のγ−リノレン
酸を含むバイオマス及び/又は油。 - 【請求項6】 好ましくは脱脂粉乳2%、ビタミン及び
炭素源及び更なる補助剤及び添加物を含む、請求項2及
び3に記載の培養及び発酵用の複合無菌培地。 - 【請求項7】 請求項1〜5のいずれか1項に記載のγ
−リノレン酸、及び適切ならば、製剤学的に許容される
添加物及び/又は補助剤を含む医薬品。 - 【請求項8】 請求項1〜5のいずれか1項に記載のγ
−リノレン酸を製剤学的に許容される添加物及び/又は
補助剤とともに製剤化することを含む、ヒト及び/又は
動物の心臓血管系障害、糖尿病、癌及び関節炎を治療す
るための医薬品の生産方法。 - 【請求項9】 請求項1〜5のいずれか1項記載のγ−
リノレン酸の、動物又はヒトの栄養における食品添加物
としての使用。 - 【請求項10】 請求項1〜5のいずれか1項記載のγ
−リノレン酸の、化粧品基剤及び化粧品添加物としての
使用。
