Abstract
Pharmacologically active 1,2-benzothiazino(3,4-d)pyrazole- S,S-dioxides of formula (I) are new. In (I) R is opt. branched 2-4C alkyl substd. by -N(R2)2 or is 1-ethyl-pyrrolidin-2-ylmethyl; R1 is H, halogen or 1-4C alkyl; each R2 is H, 1-4C alkyl or 4-7C cycloalkyl, or -N(R2)2 is a satd. 5-7 membered heterocyclic ring contg. 1-3 N, O and/or S heteroatoms. Cpds. (I) have antiinflammatory, antipyretic and analgesic properties with low toxicity and gastric ulcerogenicity c.f. piroxicam and naproxen, and are potentially useful as antirheumatic drugs. Test cpds. gave no gastric lesions at 100mg/kg p.o., and at this dose their antipyretic and analgesic props. are comparable to piroxicam and naproxen at 10 and 30 mg/kg p.o., (resp.), which doses are ulcerogenic. At these doses antiinflammatory activity is generally somewhat less for (I) than piroxicam but potency is 0.8-2.4 times that of naproxen.
