Abstract

A composition suitable for use as a substitute for blood plasma extract in producing antiserum titration curves and standard antigen inhibition curves for the determination of carcinoembryonic antigen in human plasma is disclosed. [US3852415A]

Claims (16)

1. An aqueous buffer solution suitable for use as a plasma substitute during the production of antiserum titration ' .curves-and-s-tanda-rd-an-tigen—i-nhibi-tion-curves~for-th~e~~d¾"ter-mination of carcinoembryonic antigen in human plasma having a pH of about 6.5 comprising per liter, a. salt of ethylenediamine tetraacetic acid with an alkali metal in an amount equivalent to about 1.3 grams per liter of disodium ethylenediamine tetraacetic acid dihydrate, a preservative to inhibit the growth of microorganisms, and an alkali metal hydroxide.
2. An aqueous buffer solution as claimed in claim 1 wherein the alkali metal of the salt is the same as the alkali metal of the hydroxide.
3. An aqueous buffer solution as claimed in claim 1 or claim 2 wherein the alkali metal is sodium.
4. An aqueous buffer solution as. claimed in any one of the preceding claims wherein the salt is disodium ethylenediamine tetraacetic acid dihydrate.
5. An aqueous- buffer- solution- as~ claimed i an one of the preceding claims wherein the salt is present in an amount equivalent to 1.25 to 1.32 gm/liter of disodium ethylenediamine tetraacetic acid dihydrate. 43378/2
6. An aqueous buffer solution as claimed in anyone of the preceding claims having a pH of from 6.45 to 6.55.
7. An aqueous buffer solution as claimed in any one of the preceding claims also containing a proteinaceous material which is inert to CEA antibody-antigen reactions.
8. An aqueous buffer solution as claimed in claim 7, containing, as the proteinaceous material, bovine serum albumin.
9. An aqueous buffer solution as claimed in claim 8 containing 70 ϋΐβΓβ/ϋΐβΓ of bovine serum albumin.
10. An aqueous buffer solution as claimed in any one of the preceding claims wherein the preservative is sodium azide.
11. .11. An aqueous buffer solution as claimed in claim 10 containing 0.17 gm/liter of sodium azide. 43378/2
12. A concentrate suitable for the preparation, by the addition of deionized or distilled water, of an aqueous buffer solution as claimed in any one of the preceding claims.
13. A concentrate as claimed in claim 12 having an pH of about 6.25 comprising, a salt of an organic acid with an alkali metal in an amount equivalent to about 13 grams per liter of disodium ethylenediamine tetra- acetic acid dihydrate, a preservative to inhibit, the growth of microorganisms* and an alkali metal hydroxide.
14. An aqueous buffer solution in concentrated or diluted form as claimed in any one of the preceding claims substantially as hereinbefore described with particular reference to Examples 1 and 2.
15. A process for the preparation of a carcinoembryonic antigen antiserum titration curve or a carcinoembryonic antigen standard antigen inhibition curve, wherein in place of blood plasma extract a buffer solution as claimed in any one of claims 1 to 11 is employed.
16. A process for the preparation of a carcinoembryonic antigen antiserum titration curve or an carcinoembryonic antigen standard antigen inhibition curve substantially as hereinbefore described with particular reference to the foregoing Examples 5/ 6, 8 and 9· AN/PAJHNj
