Abstract
1389100 3-Alkenyl derivatives of Rifamycin SV GRUPPO LEPETIT SpA 28 Dec 1972 [19 Jan 1972] 59833/72 Heading C2C Novel 3-alkenyl derivatives of rifamycin SV of the general Formula (I) and the corresponding 16, 17, 18, 19, 28, 29- hexahydro derivatives, wherein R is hydrogen, alkyl or aryl-C 1-8 alkyl; R 1 is hydrogen or acetyl; R 2 is hydrogen, C 1-8 alkyl or tert.-butoxycarbonyl; and X is oxygen, imino, N-substituted imino, oximino, O-substituted oximino, hydrazono, N-mono-substituted hydrazono or N-disubstituted hydrazono in which the two substituents are either separate or form a heterocyclic ring together with the common nitrogen atom; are prepared by reacting 3- formylrifamycin SV or its 25-desacetyl or 16, 17, 18, 19, 28, 29-hexahydro derivative with a compound of formula Z=CR 2 -CO-R wherein Z is H 2 or (C 6 H 5 ) 3 P, and R and R 2 are as defined above to give a compound (I) wherein X is oxygen, and when a compound (I) is desired in which X is imino, N-substituted imino, oximino, O-substituted oximino, hydrazono or N-substituted hydrazono, reacting the compound (I) in which X is oxygen with a primary amine, a secondary amine, hydroxylamine, an O-substituted hydroxylamine hydrazine or an N- mono or di-substituted hydrazine respectively. The compounds (I) have antibiotic, anti-viral and anti-tumour activity, and may be admixed with a pharmaceutical carrier to provide pharmaceutical compositions having such activity.
