Abstract

1338124 Phenothiazines MAY & BAKER Ltd 23 Aug 1972 [25 Aug 1971] 39854/71 Heading C2C Compounds of the general formula (R<SP>6</SP>=Me, Et) and their acid addition salts are prepared by (a) reacting a compound of the formula (A = CO or protected CO) with (X<SP>1</SP>=reactive ester residue, carbamate, mono- or dialkylcarbamate or alkyl carbonate), (b) decarboxylating a compound of the formula or (c) reacting a compound of the formula (X<SP>2</SP>=reactive ester residue) with dimethylamine, followed in each case by removal of the protecting group if present and (optionally) salt formation. The preparation of 8-chloro-2-propionylphenothiazine, 2 - acetyl - 8 - chlorophenothiazine anil, 2 - acetyl - 8 - chlorophenothiazin - 10 - ylcarbonyl chloride, 2 - acetyl - 8 - chloro - 10 - (3 - dimethylamino - 2 - methylpropyloxycarbonyl)phenothiazine and its hydrochloride and 2-acetyl-8-chloro-10-(3- chloro - 2 - methylpropyl) phenothiazine is described. The above compounds are useful in the relief of upper motor neurone (pyramidal) spasticity, and may be administered in the form of pharmaceutical preparations containing them in association with a carrier.
