Claims (18)
1. Ekspresijski vektor za ekspresiju tirozinaze (Tyr1) koji treba biti izložen na površini bakterijske stanice, pri čemu navedeni ekspresijski vektor sadrži ekspresijsku kasetu koja kodira tirozinazu (Tyr1) kao putnika u sustavu beta-autotransportera tipa V, pri čemu navedena tirozinaza (Tyr1) sadrži aminokiselinski slijed SEQ ID NO: 1 kada se eksprimira.
2. Ekspresijski vektor prema zahtjevu 1, pri čemu je navedena tirozinaza (Tyr1) kodirana slijedom nukleinske kiseline koji sadrži SEQ ID NO: 2 ili slijedom koji ima barem 80 %, kao što je 85, 90 ili 95 % jednakosti sa SEQ ID NO: 2.
3. Ekspresijski vektor prema bilo kojem prethodnom zahtjevu, pri čemu navedena ekspresijska kaseta kodira protein koji je tirozinaza (Tyr1) kao putnik u sustavu beta-autotransportera AIDA (engl. “Adhesin Involved in Diffuse Adherence”, adhezin uključen u difuzno pridržavanje) tipa V, kao što je sustav beta-autotransportera AIDA-I tipa V (SEQ ID NO: 3) ili kodira protein koji ima barem 95 % jednakosti sa SEQ ID NO: 3.
4. Ekspresijski vektor prema zahtjevu 3, pri čemu je protein kodiran slijedom nukleinske kiseline koji sadrži SEQ ID NO: 5 ili slijedom koji ima barem 80 % jednakosti, kao što je 85 %, 90 % ili 95 % jednakosti sa SEQ ID NO: 5.
5. Ekspresijski vektor prema bilo kojem od zahtjeva 1 – 4, pri čemu je navedeni ekspresijski vektor pAIDA1-Tyr1 (SEQ ID NO: 4) ili ekspresijski vektor koji ima barem 80 % jednakosti slijeda s istim, kao što je 85 %, 90 % ili 95 % jednakosti sa slijedom SEQ ID NO: 4.
6. Stanica domaćina koja sadrži ekspresijski vektor prema bilo kojem od zahtjeva 1 – 5, poput stanice E.coli, kao što je soj E.coli K12, najbolje E.coli 0:17K12∆OmpT.
7. Postupak za proizvodnju jedne ili više bakterijskih stanica, barem djelomično presvučenih sastavom koji adsorbira kemijske tvari, a da navedeni postupak sadrži sljedeće korake:
a) odabir i uzgoj jedne ili više bakterijskih stanica koje sadrže ekspresijski vektor prema bilo kojem od zahtjeva 1 – 5 ili stanicu domaćina prema zahtjevu 6.
b) induciranje ekspresije tirozinaze (Tyr1) iz navedenog ekspresijskog vektora kako bi se omogućilo izlaganje navedene tirozinaze (Tyr1) na površini navedene jedne ili više bakterijskih stanica,
c) dodavanje odgovarajuće količine tirozina ili njegovog funkcionalnog ekvivalenta navedenoj jednoj ili više bakterijskih stanica kako bi se omogućilo stvaranje navedenog sastava koji adsorbira kemijske tvari barem djelomično na površini navedene jedne ili više bakterijskih stanica, i nakon toga
d) pribavljanje stanica iz koraka c).
8. Postupak prema zahtjevu 7, pri čemu se uzgoj stanica izvodi pri pH od oko 7 – 8, kao što je pri približno pH 7,5, a po izboru pri čemu se induciranje u koraku b) izvodi dodavanjem IPTG-a ili laktoze u kulturu bakterijskih stanica, na primjer u količini od oko 200 µM IPTG-a po indukciji.
9. Postupak prema bilo kojem od zahtjeva 7 – 8, pri čemu se ekspresija tirozinaze (Tyr1) može nastaviti tijekom barem dvije generacije stanica nakon induciranja u koraku b), kao na primjer 2, 3, 4, 5, ili 6 generacija.
10. Postupak prema bilo kojem od zahtjeva 7 – 9, pri čemu je tirozin u obliku praškaste smjese ili je sadržan u vodenoj bazičnoj otopini koja ima pH oko 9 – 12, na primjer otprilike pH 10.
11. Postupak prema bilo kojem od zahtjeva 7 – 10, pri čemu sastav koji adsorbira kemijske tvari sadrži melanin i/ili njegov funkcionalni ekvivalent.
12. Postupak prema bilo kojem od zahtjeva 7 – 11, pri čemu kemijska tvar sadrži farmaceutsku tvar ili jedan ili više proizvoda razgradnje ili jedan ili više spojeva teških metala.
13. Jedna ili više bakterijskih stanica, barem djelomično presvučenih sastavom koji adsorbira kemijske tvari, a koji se može dobiti postupkom prema bilo kojem od zahtjeva 7 – 12.
14. Upotreba jedne ili više bakterijskih stanica, barem djelomično presvučenih sastavom koji adsorbira kemijske tvari prema zahtjevu 13, za uklanjanje jedne ili više kemijskih tvari iz vode, kao što je otpadna voda.
15. Upotreba prema zahtjevu 14, pri čemu su navedena jedna ili više bakterijskih stanica suspendirane u otopini ili prisutne u biofilmu, po izboru gdje se navedeni biofilm nalazi na čvrstom nosaču, kao što je stacionarni čvrsti nosač ili pokretni čvrsti nosač.
16. Uređaj, kao što je MBR (engl. “Membrane Bioreactor”, membranski bioreaktor), koji sadrži jednu ili više bakterijskih stanica prema zahtjevu 13, suspendirane u otopini.
17. Uređaj, kao što je MBBR (engl. “Moving bed biofilm reactor”, reaktor s biofilmom s pomičnim slojem), koji sadrži jednu ili više bakterijskih stanica prema zahtjevu 13, koje se nalaze u biofilmu na čvrstom nosaču.
18. Postupak za uklanjanje jedne ili više kemijskih tvari iz vode, poput otpadnih voda, koji sadrži korake:
a) Osiguravanje jedne ili više bakterijskih stanica prema zahtjevu 13, ili biofilm koji sadrži navedenu jednu ili više bakterijskih stanica,
b) Omogućavanje prolaska vode kroz jednu ili više bakterijskih stanica ili navedeni biofilm koji sadrži navedenu jednu ili više bakterijskih stanica, adsorbirajući pritom sve kemijske tvari prisutne u toj vodi,
c) Uklanjanje svih adsorbiranih kemijskih tvari iz jedne ili više bakterijskih stanica ili biofilma koji sadrži navedenu jednu ili više bakterijskih stanica, ispiranjem vodenim pripravkom kiselog pH, da bi se nakon toga omogućila ponovna upotreba jedne ili više bakterijskih stanica ili biofilma koji sadrži navedenu jednu ili više bakterijskih stanica i nakon toga
d) Po želji ponavljanje koraka b) i c).
