Claims (11)
1. Postupak za proizvodnju celulaza i hemicelulaza upotrebom soja filamentoznih gljivica u miješanom i aeriranom bioreaktoru, koji sadrži barem dvije faze:
• fazu a) rasta navedenog soja u prisutnosti barem jednog karbonatnog supstrata za rast u zatvorenom reaktoru, navedena faza rasta se provodi s koncentracijom karbonatnog supstrata za rast u rasponu 10 do 90 g/L;
• fazu b) za kontinuiranu proizvodnju celulaza, u kojoj se barem jedan karbonatni inducirajući supstrat dovodi uz brzinu dovođenja koja je konstantna tijekom perioda od barem više od 200 h, navedeni karbonatni inducirajući supstrat je barem jedna vodena otopina hemiceluloznog hidrolizata dobivena iz prethodne obrade s kiselinom lignoceluloznog supstrata, navedena vodena otopina hemiceluloznog hidrolizata nije podvrgnuta prethodnoj sterilizaciji i nije podvrgnuta ispravljanju pH, navedeni pH vodene otopine je u rasponu 0.5 do 3, masa reakcijskog volumena se održava konstantnom povlačenjem djela navedenog reakcijskog volumena, navedena faza b) se provodi pri brzini razrjeđivanja u rasponu 0.001 do 0.008 h-1,
u kojem je karbonatni inducirajući supstrat korišten u fazi b) vodena otopina hemiceluloznog hidrolizata dobivena iz prethodne obrade s kiselinom lignoceluloznog supstrata, kao smjesa s barem jednim drugim karbonatnim supstratom odabranim od inducirajućih ili ne inducirajućih šećera, u kojem je brzina dovođenja navedenog karbonatnog inducirajućeg supstrata u rasponu 35 do 140 mg karbonatnog inducirajućeg supstrata po gramu mase suhe tvari soja na sat.
2. Postupak prema zahtjevu 1, naznačen time da navedeni soj je soj Trichoderma reesei modificiran pomoću mutacije, selekcije ili genetske rekombinacije.
3. Postupak prema zahtjevu 1 ili zahtjevu 2, naznačen time da je karbonatna podloga za rast koja se koristi u navedenoj fazi rasta odabrana od ostataka glukoze, laktoze, ksiloze, dobivenih nakon etanolne fermentacije monomernih šećera enzimskih hidrolizata lignoceluloznog supstrata i ekstrakata hemicelulozne frakcije u obliku monomera dobivenih od prethodno obrađenog lignoceluloznog supstrata, koristi samostalno ili kao smjesa.
4. Postupak prema jednom od zahtjeva 1 do 3, naznačen time da kisela predobrada je kisela hidroliza, kuhanje s kiselinom ili eksplozija pare s prethodnom impregnacijom navedenog lignoceluloznog supstrata s vodenom otopinom sumporne kiseline.
5. Postupak prema jednom od zahtjeva 1 do 4, naznačen time da je trajanje faze b) za neprekinutu proizvodnju celulaza veće od barem 300 h.
6. Postupak prema zahtjevu 5, naznačen time da je trajanje faze b) za neprekinutu proizvodnju celulaza veće od barem 400 h.
7. Postupak prema jednom od zahtjeva 1 do 6, naznačen time da se brzina protoka dovoda postupno povećava u fazi b).
8. Postupak prema jednom od zahtjeva 1 do 7, naznačen time da se navedena faza b) provodi pri brzini razrjeđivanja u rasponu 0.002 do 0.008 h-1.
9. Postupak prema jednom od zahtjeva 1 do 8, naznačen time da se proizvoljna faza a') rasta i proizvodnje u reaktoru s kontinuiranom dobavom barem jednog karbonatnog inducirajućeg supstrata tijekom koje nema kontinuiranog povlačenja iz fermentora, provodi između faze a) i faze b).
10. Postupak prema zahtjevu 9, naznačen time da je navedeni karbonatni inducirajući supstrat koji je korišten u fazi a') identičan karbonatnom inducirajućem supstratu korištenom u fazi proizvodnje b).
11. Postupak prema zahtjevu 9 ili zahtjevu 10, naznačen time da se faza a') provodi kroz razdoblje u rasponu od 50 do 150 h.
