Abstract

A derivative of vitamin B12, said derivative being antagonistic to vitamin B12 and being a compound in which one particular amide group of vitamin B12 is replaced by a carboxylic acid group, is obtained in a process which comprises fermenting the vitamin B12 antagonist-producing microorganism Propionibacterium shermanii PBS 77 to produce a fermentation broth containing said derivative of vitamin B12, and then separating the said derivative of vitamin B12 from the vitamin B12 and other ingredients of the fermentation broth. The separation includes one of the following steps: (a) solvent extraction using a water insoluble organic solvent at pH7 to remove the vitamin B12 in the solvent layer followed by solvent extraction using a water insoluble organic solvent of the aqueous layer at pH 2-4.5 to obtain said derivative in the solvent layer; (b) adsorbing the derivative of vitamin B12 on to an alkaline anion exchange resin; (c) electrodialysis. A pharmaceutical composition containing said derivative of vitamin B12 together with a pharmaceutical carrier or excipient, is suitable for intramuscular injection against polycythaemia.
