Abstract
A composition for detecting glucose in urine comprises glucose oxidase, an oxidation-catalyst, and an indicator which is oxidized by hydrogen peroxide, changes colour on oxidation and has the general formula AR-X-AR wherein Ar is a phenyl group which may have one or more hydroxy and methoxy substituents and X is the radical -(CH=CH)m-CH=(N-N)n=CH-(CH=CH)p-in which m, n and p are each each to 0 or to 1, and a catalyst for causing the oxidation of the indicator by hydrogen peroxide. The glucose oxidase converts glucose to gluoconic acid in the presence of atmospheric oxygen and at the same time forms hydrogen peroxide. Suitable indicators are 4,41-dihydroxystilbene, 4,41-dihydroxybenzaldazine, vanillinazine, 3,31,5,51-tetramethoxy-4, 41-dihydroxybenzaldazine, 3,31-dimethoxy-4,41-dihydroxystilbene, and bis-gamma(4-hydroxyphenyl)acroleinazine. Splinters, sticks or strips of wood, paper or cellulose acetate may be impregnated with the composition; or the composition may be applied to glass, metal or plastic using gelatin as adhesive. Such test pieces will change colour when moistened with a glucose-containing liquid. Suitable catalysts are peroxidase, catalase, blood, hemin, uroferriporphrin chloride, and a mixture of potassium iodide and sodium molybdate. A buffer composition, e.g. citric acid/sodium citrate may be added to the test composition in order to keep the pH of the reactants at a predetermined value. Gelatine, dog plasma, bovine albumin, egg albumin and casein may be incorporated in the compositions. Specification 808,742 is referred to.
