1429525 Determination of cholesterol BOEHRINGER MANNHEIM GmbH 15 May 1973 [17 May 1972] 23082/73 Heading G1B Cholesterol in an aqueous medium is quantitatively determined by incubating the medium with cholesterol oxidase and thereafter determining either the amount of oxygen used up or the amount of hydrogen peroxide or of cholestenone formed, on the basis of the reaction. cholesterol oxidase cholesterol + O 2 # cholestonone + H 2 O 2 (A) The amount of oxygen used up may be measured polarometrically, (B) the amount of hydrogen peroxide formed may be determined enzymatically with the use of catalase or peroxidase, (C) the amount of cholestenone is measured directly by direct absorption spectrophotometry or with the use of a Keto group-reacting compound. Bound or total cholesterol is determined by testing a sample in which the free cholesterol concentration has already been tested or a separate, untested sample, respectively, after such samples have been saponified using alcoholic potassium hydroxide solution or cholesterol esterase. Reagent Kito containing specified proportions of cholesterol oxidase and the following constituents are disclosed: (A) surface-active agent (hydroxpolyethoxydodecane) in phosphate buffer, (B) catalase, acetylacetone, methanol surface-active agent and buffer OR peroxidase, a chromogen (2, 2 -aminobenzthiazoline-sulphonic acid), surface-active agent and buffer, (C) Keto group-reacting compound (2, 4-dinitro-phenylhydrozine and surface-active agent. A reagent for saponifying esterified cholesterol (as mentioned above) may also be included.
