Abstract
The invention comprises 10, 11-dihydro -11-oxo-5H-dibenzo[b, e] [1, 4]-diazepine derivatives of the general formula: <FORM:0959995/C2/1> (wherein X represents a hydrocarbon chain of 2 or 3 carbon atoms, Y represents a C2-4 dialkylamino radical, a pyrolidino or piperidino radical or a pyrrolidino or piperidino radical in which one -CH2- group is replaced by -O-, -NH-, -N(CH3)- or -N(C2H5)-, R1 represents a hydrogen atom or a methyl, ethyl or benzyl group, and R2 and R3 represent, interchangeably, hydrogen or chlorine atoms or methyl, ethyl, methoxy, or ethoxy or alkylmercapto groups) and their non-toxic therapeutically useful acid addition and C1-3 alkyl quaternary ammonium salts, and the preparation thereof by cyclizing, by allowing to stand or preferably by heating, a compound of the general formula: <FORM:0959995/C2/2> (wherein R represents a hydrogen atom or a lower alkyl group and R1 has one of the meanings of R1, preferably hydrogen) or a quaternary ammonium derivative thereof, and, if desired, when R1 is hydrogen, treating the product with a reactive methyl, ethyl or benzyl compound. and/or, if desired, converting the product into an acid addition salt, or into a quaternary ammonium salt if it is not already in this form. o-Aminodiphenylamine -o1- carboxylic acids and esters of the general formula (II) above (R1=H) are prepared by reducing the corresponding o-nitro compounds. Pharmaceutical preparations, useful as parasympatholytics, antihistaminics, spasmolytics, tranquillizers, psychic energizers and psycho stimulants, comprise the compounds (I) or their non-toxic acid addition or quaternary ammonium salts, in admixture with a pharmaceutical organic or inorganic solid or liquid carrier suitable for enteral, parenteral or topical administration, and, if desired, preserving, stabilizing, wetting or emulsifying agents, salts for varying the osmolic pressure, buffers, or additional therapeutically useful substances. They may take the form of tablets, dragees, salves, creams, solutions, suspensions or emulsions.
