Abstract

The invention comprises compounds of the formula: <FORM:0955354/C2/1> wherein R1 is a C1-5alkyl radical, and R2 is a hydrogen atom or a C1-5 alkyl radical, non-toxic salts thereof, pharmaceutical compositions thereof and a process for preparing an a -(C1-5 alkyl) histidine and its C1-5 alkyl esters by reacting an C3-7 a -nitroalkanoic acid ester with 4-methyl-imidazole, by contacting the ester in an inert solvent with sodium hydride, adding the chloromethylimidazole, agitating, until reaction is complete, isolating the a -nitroimidazole ester, reducing this with hydrogen under pressure, and Raney nickel, and if desired, hydrolyzing the resulting ester by heating with a mineral acid, and if desired, re-esterifying the resulting carboxylic acid by heating with a C1-5 alkanol in the presence of a mineral acid. Benzyl-a -bromoalkanoate is obtained by reacting benzyl alcohol with a -bromoalkanoyl bromide. Benzyl-a -nitroalkanoate is obtained by reacting benzyl-a -bromoalkanoate with sodium nitrite and glucinol in dimethylformamide. Said compositions having anti-histaminic activity are administered orally (e.g. as tablets) or parenterally.
