Abstract
The invention comprises steroids of the formula: <FORM:0953543/C2/1> (wherein X1 is chlorine or fluorine, X2 is chlorine or bromine, and R is hydrogen or alkyl) and their esters; and their preparation by reduction of the corresponding 3-ketones with a complex light metal hydride followed, if desired, by esterification or hydrolysis of the product. Lists of suitable hydrides for the reduction and acids for the esterification are provided. The acids include mono- and di-basic carboxylic acids which may contain halogens or nitrogen, sulphonic acids, phosphoric acids and sulphuric acid. Examples describe the reduction of 9a :11b -dihalo-testosterones and -17a -alkyl-testosterones and 17-esters thereof according to the invention, esterification of the products to form 3-mono-esters and 3:17-di-esters of saturated and unsaturated, mono- and di-basic aliphatic and cycloaliphatic acids, and of tetrahydrophthalic, benzoic and sulphuric acids (the last named as semisulphates prepared by the action of liquid sulphur trioxide in pyridine), and hydrolysis of D 4-9a , 11b -dichloro-3b , 17b -dihydroxyandrostene-17-acetate to the free 17-ol. Methyl esters and alkali metal and amine salts of the semi-esters with dibasic acids are also described. The androgens of the invention may be made up into pharmaceutical compositions with suitable carriers. They may take the form of oily or aqueous solutions (e.g. in ampoules), suspensions, emulsions, implants, tablets, dragees, ointments or creams, and may contain preserving, stabilizing, wetting or emulsifying agents, salts for regulating the osmotic pressure or buffers.
