Abstract

A vaccine comprises the Komarov KH strain of Newcastle disease virus cultivated on mammalian kidney tissue culture of one mammalian species, e.g. calf or more than one mammelian species, e.g. calf and pig. In general, more than one serial passage is used, e.g. 21 passages through calf kidney culture or 10 serial passages on calf kidney tissue culture followed by 11 serial passages on pig kidney tissue culture. Pieces of kidney are treated with a buffered saline solution of trypsin. The freed cells are suspended in Hank's solution, centrifuged and incubated in a nutrient medium comprising lactalbumin hydrolyate, yeast extract, calf serum, sodium penicillin and streptomycin sulphate in Hank's solution, at 37 DEG C. for 3 days, fresh nutrient medium added and incubation continued until a complete mono-layer developes. This is inoculated with KH strain of Newcastle disease virus in thick embryo allantoic fluid and diluted 1/100 with above nutrient fluid and incubated for three days at 37 DEG C. The culture is then frozen, thawed, shaken and centrifuged. The supernatant is inoculated into another kidney kidney tissue culture and the process repeated for the desired number of serial passages. The resultant culture fluid may be formulated into a vaccine.
