Abstract

Polypeptides having spasmolytic activity are obtained from animal organs of lymphoidal structure e.g. lymph ganglions of oxen or sheep or calf or lamb thymus, by hydrolysing the comminuted organs to a broth by warming with an aqueous solution of a protease e.g. papain, heating the broth to destroy the enzyme, clarifying the broth and fractionally precipitating the polypeptide from the clarified broth with alcohol, the active material being recovered from the product precipitated with the greatest concentration of alcohol. The hydrolysis may be carried out at 60 DEG C. for 15 minutes at a pH of 7.0. After filtration, the clarified broth may be treated with an equal volume of 90% ethanol, the precipitate discarded, and seven volumes of absolute ethanol added to precipitate the polypeptide. The crude polypeptide may be purified by solution in acetic acid and further precipitation with ether. Further purification may be by dialysis against water or chromatographic adsorption on an aluminium oxide and/or cellulose column.
