Abstract

6-aminopenicillanic acid is produced by reacting an enzyme preparation derived from a penicillin amidase producing microorganism with a penicillin of general formula <FORM:0892144/IV (b)/1> or an alkaline earth metal or alkali metal salt thereof, where R is an alkyl or alkenyl group or the group R1XCH2 where R1 is an alkyl, alkenyl, allyl or aryl group and X is O or S, or a fermentation liquor containing such a penicillin or penicillias or salt or salts. Typical penicillins are phenoxymethyl-, butylthiomethyl-, n-heptyl-, 2-pentenyl-, n-amyl- and n-butyl-penicillin, as w e as the natural mixture of penicillins produced by fermentation, including those produced in the formation of 6-aminopenicillanic acid according to Specification 870,396. The penicillin is used in the form of the free acid or a salt with a cation which will not interfere with the enzymatic hydrolysis, e.g. alkali metal and alkaline earth metal salts, e.g. sodium, calcium and potassium salts. The penicillin is added to the enzyme preparation obtained by culturing the producing organism in a suitable medium and which may be in the form of (a) a clarified brew (b) a solution of the dialysed enzyme or (c) a solution of the acetone precipitated enzyme. Specified penicillin amidase-producing organisms are (a) the fungi alternaria tenuis, aspergillus versicolor, Botrytis cinerea, Epicoccum nigrum, Fusarium roseum, Fusarium oxysporum, Mucor hiemalis, Mucor sylvaticus, Penicillium chrysogenum and Trichoderma viride (b) the yeasts Crytococcus albidus and Trichosporon sp. and (c) Streptomyces species.
