Abstract

A diagnostic composition for the detection of glucose comprising glucose oxidase, uroferriporphyrin of formula <FORM:0891275/III/1> and a colour forming substance oxidizable by H2O2 (formed by the oxidation of glucose). The colour forming substances specified are 2,7-diaminofluorene, O-tolidine and leucoindophenols. Complex-forming compounds containing amine donors groups, e.g. 2-amino benzothiazole nicotinic acid or pyridine, may also be incorporated in the composition. Additives as protective agents and wetting agents (e.g. Algin) and an inert dye to impart a uniform background colour may also be added, as well as a buffer system to maintain a pH range between 3 and 7 and preferably at 5,4. The test device may be a tablet or a powder but it is preferred to employ the reagent composition on bibulous carriers such as strips of filter paper. In Example 1, the impregnating solution is prepared from aqueous algin, citrate buffer, gelatin, a dye, 2:7-diamino fluorene dihydrochloride, glucose oxidase and the uroferriporphyrin. Half of the filter strip is impregnated with the diagnostic composition, the other half having a water impervious barrier of ethyl cellulose. Examples 2, 3 and 4 differ only in the choice of colour forming substance and/or the addition of 2-aminobenzothiazole. Specifications 575,612, 784,548, 886,778 and U.S.A.
