Abstract

Compositions for detecting glucose comprise (a) glucose oxidase, (b) a metalloporphyrin, (c) a heterocyclic compound containing a ring nitrogen atom singly and doubly bonded to two carbon atoms and (d) a colour-forming substance oxidisable by hydrogen peroxide in the presence of (b) and (c). Specified metalloporphyrins (b) are hemin and metalloprophyrins in which the metal atom is Fe, Co, Ni, Mn, Mg, Cu, Zn or a complex such as VO. Specified heterocyclic compounds (c) are 2-aminobenzthiazole, pyridine, bipyridyl, bipyridyl-pyridine and nicotinic acid. Specified colour-forming substances (d) are 2:7-diaminofluorene, o-tolidine and leucoindophenols. Protective agents, wetting agents, a background dye and buffering mixtures (preferred pH is 3-7) may also be present in the composition, and the composition is prepared by separately preparing solutions of (a) and (d) and of (b) and (c) and thereafter impregnating a bibulous material with the first mentioned solution, drying, impregnating with the second solution and using wet or subsequently drying. In one example, only one end of a strip was impregnated with the diagnostic composition, the remainder having a water-impervious barrier of ethyl cellulose. In examples, the following combinations were employed: (1) first solution: aqueous algin, citrate buffer, gelatine, red substituted xanthone dye, 2:7-diaminofluorene dihydrochloride (in aqueous ethanol) and glucose oxidase (catalase present being destroyed by heating); second solution: hemin and 2-aminobenzthiazole in aqueous ethanol containing polyvinyl alcohol (wetting and stabilizing agent), (2) first solution: citrate buffer, gelatine, o-tolidine dihydrochloride (in ethanol) and glucose oxidase; second solution: hemin dissolved in pyridine and diluted with ethanol, (3) first solution as (2); second solution: hemin, nicotinic acid or 2-aminobenzthiazole in aqueous ethanol containing polyvinyl alcohol, (4) first solution as (1); second solution as (3), using nicotinic acid.
