Abstract

Polyvinyl alcohol with a degree of polymerization of 600 to 800 is partially esterified with phthalic anhydride to produce a product with a phthalyl content of 60-70% by weight and an acetyl content of 1,6 to 6,0% by weight which, being substantially insoluble in the stomach juices but soluble in the intestinal fluids is used as an enteric coating for solid medicinal oral dosage units. Esterification is performed by refluxing in an inert solvent, e.g. pyridine. picoline or methyl ethyl ketone. The reaction mixture is forced into aqueous mineral acid to precipitate the product which is usually applied as a coating from a solution e.g. 95% ethanol.ALSO:A solid medicinal oral dosage unit consists of a core containing at least one medicinal agent coated with an enteric film of polyvinyl alcohol having a degree of polymerization of 600 to 800 partially esterified with phthalic anhydide. The esteric coating has a phthalyl content of 60-70% by weight, an acetyl content of 1,6 to 6,0% by weight. It is applied from a solution, e.g. in 95% ethanol, the solution being applied in several increments to build up a layer of desired thickness. Solvent is removed by a stream of warm air and the units are dusted, e.g. with talc between layers while the coating is sticky to prevent their sticking together. In example I, tablets containing brewers yeast concentrate, thiamine hydrochloride, riboflavin, niaciamide, hyridoxine hydrochloride, vitam B12, vitamin C and vitamin D are coated with a film of polyvinyl alcohol acetate phtlalate (phthalyl content 65%, acetyl content 4,8%) applied as a 30% w/v solution in 90% denatured alcohol. Example II is similar using tablets which contain ferrous sulphate, edible bone flour, vitamin A acetate, vitam D, vitamin B1, riboflanin, niacinamide, vitamin C and roduim iodide. In both examples a standard sugar coating was applied over the entire layer.
