Abstract

Prolonged acting granules of a medicament consist of granules of the medicament coated with a composition comprising a non-toxic, impermeable cellulose ether or cellulose ester which is very sparingly soluble in the digestive fluids of the alimentary tract and a substance which is readily soluble in the digestive fluids of the alimentary tract and is present as 5 to 30% by weight of the coating. The granules of the medicament are prepared in a conventional manner. The medicament may be mixed with a diluent such as sucrose or lactose before granulating. The granules are coated by rotating them in a coating pan and spraying on to them a solution in a volatile solvent, e.g. acetone or alcohol of the film-forming substance and the readily soluble substance. The granules are used as such or put up into capsules or caskets or compressed into tablets. They may be mixed with uncoated granules or an inert substance such as lactose, sucrose, glucose or dicalcium phosphate and then compressed into tablets. In examples: (a) granules of acetyl-salicylic acid are coated with ethyl cellulose and polyethylene glycol 1000 and a mix of coated and uncoated granules tabletted; (b) granules of ephedrine hydrochloride, sodium chloride and sucrose are coated with cellulose acetate, polyethylene glycol 1000 and polyoxy-ethylene sorbitan monolaurate, and a mixture of coated and uncoated granules are tabletted; (c) granules of acetylsalicylic acid are coated with ethyl cellulose and dioctyl sodium sulpho-succinate and granules of p-ethoxyacetanilide and methyltheobromine are likewise coated, the granules mixed and tabletted; (d) acetyl-salicylic acid, p-ethoxyacetanilide and methyl-theobromine are coated as in (c), mixed with uncoated granules and tabletted.
